| 1 | Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently,the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however,the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown.In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo,accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment.The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death,indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together,our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis,which will hopefully provide a prospective compound for bladder cancer treatment. | Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 23 |
| 2 | Probabilistic stability analyses of undrained slopes by 3D random fields and finite element methods显示文摘A long slope consisting of spatially random soils is a common geographical feature. This paper examined the necessity of three-dimensional(3 D) analysis when dealing with slope with full randomness in soil properties. Although 3 D random finite element analysis can well reflect the spatial variability of soil properties, it is often time-consuming for probabilistic stability analysis. For this reason, we also examined the least advantageous(or most pessimistic) cross-section of the studied slope. The concept of'most pessimistic' refers to the minimal cross-sectional average of undrained shear strength. The selection of the most pessimistic section is achievable by simulating the undrained shear strength as a 3 D random field. Random finite element analysis results suggest that two-dimensional(2 D) plane strain analysis based the most pessimistic cross-section generally provides a more conservative result than the corresponding full 3 D analysis. The level of conservativeness is around 15% on average. | 3 | Effect of butanol on flotation separation of quartz from hematite with N-dodecyl ethylenediamine显示文摘In order to investigate the effect of butanol on quartz flotation when N-dodecyl ethylenediamine(ND)was used as collector, single mineral flotation and artificial mixed mineral(hematite and quartz were mixed at a mass ratio of 3:2) separation were conducted in the laboratory. Experimental results indicated that addition of butanol could improve the collecting performance of ND on quartz and enhance the floatability of quartz. Best flotation recovery of quartz was obtained when butanol was mixed with ND at a mass ratio of 1:1. Moreover, the molecular structure of alcohols had a significant effect on mineral recovery. Best separation efficiency could be obtained when tert-butanol was added as it had the largest cross-sectional area. Zeta potential measurements indicated that alcohols could strengthen electrostatic adsorption between quartz and collector. | 4 | Influence of groundwater drawdown on excavation responses e A case history in Bukit Timah granitic residual soils显示文摘Performances of a braced cut-and-cover excavation system for mass rapid transit(MRT) stations of the Downtown Line Stage 2 in Singapore are presented. The excavation was carried out in the Bukit Timah granitic(BTG) residual soils and characterized by significant groundwater drawdown, due to dewatering work in complex site conditions, insufficient effective waterproof measures and more permeable soils. A two-dimensional numerical model was developed for back analysis of retaining wall movement and ground surface settlement. Comparisons of these measured excavation responses with the calculated performances were carried out, upon which the numerical simulation procedures were calibrated. In addition, the influences of groundwater drawdown on the wall deflection and ground surface settlement were numerically investigated and summarized. The performances were also compared with some commonly used empirical charts, and the results indicated that these charts are less applicable for cases with significant groundwater drawdowns. | 7 | Extraction mechanism of rare earths from chloride acidic solution with ammonium-bifunctionalized ionic liquid extractants显示文摘A series of ammonium-bifunctionalized ionic liquid extractants(ammonium-Bif-ILEs) combined with a number of anions, including carboxylic acids and 1,3-diketonates, have been prepared and studied in this report. Their extraction behavior and properties toward rare earth ions(REs(Ⅲ)) are systematically investigated in chloride media as a function of important parameters such as aqueous phase p H, salting-out agent concentrations, and extraction temperature. The separation performance of ammonium-Bif-ILEs toward REs(Ⅲ) are systematically discussed. The results demonstrate that ammonium-Bif-ILEs have a synergistic effect between the cation and anions in separation of REs(Ⅲ). The influences of different anions on separation factor(b) values are further studied. | 8 | Genetically modified CD7-targeting allogeneic CAR-T cell therapy with enhanced efficacy for relapsed/refractory CD7-positive hematological malignancies:a phase I clinical study显示文摘Chimeric antigen receptor(CAR)-T cell therapy against T cell malignancies faces major challenges including fratricide between CAR-T cells and product contamination from the blasts.Allogeneic CAR-T cells,generated from healthy donor T cells,can provide ready-to-use,blast-free therapeutic products,but their application could be complicated by graft-versus-host disease(GvHD)and host rejection.Here we developed healthy donor-derived,CD7-targeting CAR-T cells(RD13-01)with genetic modifications to resist fratricide,GvHD and allogeneic rejection,as well as to potentiate antitumor function.A phase I clinical trial(NCT04538599)was conducted with twelve patients recruited(eleven with T cell leukemia/lymphoma,and one with CD7-expressing acute myeloid leukemia).All patients achieved pre-set end points and eleven proceeded to efficacy evaluation.No dose-limiting toxicity,GvHD,immune effector cell-associated neurotoxicity or severe cytokine release syndrome(grade≥3)were observed.28 days post infusion,81.8%of patients(9/11)showed objective responses and the complete response rate was 63.6%(7/11,including the patient with AML).3 of the responding patients were bridged to allogeneic hematopoietic stem cell transplantation.With a median follow-up of 10.5 months,4 patients remained in complete remission.Cytomegalovirus(CMV)and/or Epstein-Barr virus(EBV)reactivation was observed in several patients,and one died from EBV-associated diffuse large B-cell lymphoma(DLBCL).Expansion of CD7-negative normal T cells was detected post infusion.In summary,we present the first report of a Phase I clinical trial using healthy donor-derived CD7-targeting allogeneic CAR-T cells to treat CD7^(+)hematological malignancies.Our results demonstrated the encouraging safety and efficacy profiles of the RD13-01 allogeneic CAR-T cells for CD7^(+)tumors. | Yongxian Hu Yali Zhou Mingming Zhang Houli Zhao Guoqing Wei Wengang Ge Qu Cui Qitian Mu Gong Chen Lu Han Tingting Guo Jiazhen Cui Xiaoyan Jiang Xiujun Zheng Shuhui Yu Xiaolong Li Xingwang Zhang Mingxi Chen Xiuju Li Ming Gao Kang Wang Cheng Zu Hao Zhang Xiaohong He Yanbin Wang Dongrui Wang Jiangtao Ren He Huang | 2022 | Cell Research2022,32,11: | 2 |

| 14 | Structure and Electrochemical Hydrogen Storage Properties of as-Milled Mg-Ce-Ni-Al-Based Alloys显示文摘At room temperature,crystalline Mg-based alloys,including Mg2 Ni,MgNi,REMg12 and La2 Mg17,have been proved with weak electrochemical hydrogen storage performances.For improving their electrochemical property,the Mg is partially substituted by Ce in Mg-Ni-based alloys and the surface modification treatment is performed by mechanical coating Ni.Mechanical milling is utilized to synthesize the amorphous and nanocrystalline Mg1-xCexNi0.9Al0.1(x=0,0.02,0.04,0.06,0.08)+50 wt%Ni hydrogen storage alloys.The effects made by Ce substitution and mechanical milling on the electrochemical hydrogen storage property and structure have been analyzed.It shows that the as-milled alloys electrochemically absorb and desorb hydrogen well at room temperature.The as-milled alloys,without any activation,can reach their maximal discharge capacities during first cycling.The maximal value of the 30-h-milled alloy depending on Ce content is 578.4 mAh/g,while that of the x=0.08 alloy always grows when prolonging milling duration.The maximal discharge capacity augments from337.4 to 521.2 mAh/g when milling duration grows from 5 to 30 h.The cycle stability grows with increasing Ce content and milling duration.Concretely,the S100 value augments from 55 to 82%for the alloy milled for 30 h with Ce content rising from 0 to 0.08 and from 66 to 82%when milling the x=0.08 alloy mechanically from 5 to 30 h.The alloys’electrochemical dynamics parameters were measured as well which have maximum values depending on Ce content and keep growing up with milling duration extending. | Yanghuan Zhang Zhenyang Li Wei Zhang Wengang Bu Yan Qi Shihai Guo | 2020 | Acta Metallurgica Sinica(English Letters)2020,33,5: | 1 |

| 18 | Characterization of nanoparticle mixed 316L powder for additive manufacturing显示文摘Nanoparticles reinforced steels have many advantaged mechanical properties.Additive manufacturing offers a new method for fabricating nanoparticles reinforced high performance metal components.In this work,we report the application of low energy ball milling in mixing nanoparticles and micron 316 L powder.With this method,0.3 and 1.0 wt% Y2 O3 nanoparticles can be uniformly distributed on the surface of 316 L powder with the parameters of ball-to-powder ratio at 1:1,speed at 90 rpm and 7 h of mixing.The matrix 316 L powders remain spherical in shape after the mixing process.In the meantime,the effect of low energy ball milling and the addition of Y2 O3 nanoparticles on the powder characteristics(flowability,apparent density and tap density) are also studied.Results show that the process of low energy ball milling itself can slightly decrease the flowability and apparent density of the 316 L powder.The addition of 0.3 and 1.0 wt% Y2 O3 nanoparticles can also decrease the flowability,the tap density and the apparent density compared with the original 316 L powder.All of these changes result from the rough surface of the mixed powder produced by ball milling and the addition of Y2 O3 nanoparticles.The powder’s rough surface can increase the coefficient of friction of powders.The mixture of 316 L powder and Y2 O3 nanoparticles can be successfully used for selective laser melting(SLM).The relative density of SLM 316 L-Y2 O3 is measured at 99.5%.However,Y2 O3 agglomerations were observed which is due to the poor wettability between 316 L and Y2 O3. | Wengang Zhai Wei Zhou Sharon Mui Ling Nai Jun Wei | 2020 | Journal of Materials Science & Technology2020,47,12: | 0 |
