1First record of Thalassiosira curviseriata Takano (Bacillariophyta) and its bloom in the East China Sea显示文摘A bloom caused by a diatom, Thalassiosira curviseriata Takano (Bacillariophyta), is recorded in the East China Sea for the first time in China during a red tide investigation cruise (MC2005-2) from 27 March to 12 April 2005. This bloom was developed with the competition of Chaetoceros debilis and Skeletonema spp. The highest cell density of T. curviseriata, which has reached 1.27×106 cells/dm3, was found in the surface and middle water layers of Stas ZD, ZB in the East China Sea (27.22°~29.48°N, 121.53°~122.98°E)in early spring in 2005. During the blooming period of T. curviseriata, the population with high cell density was found in the water area with temperature of 10~15 ℃ and salinity of 29.0~33.5. The percentage of the predominant species, T. curviseriata, has reached 95.8% of total diatom cells at one time in the middle water layer. The morphological characteristics of T. curviseriata, were observed with light microscope (LM) and transmission electronic microscope (TEM). The cells are 5.0~12.6 μm in diameter, connecting each other by mucilaginous thread to form spiral and curved chains. Description and LM and TEM images of T. curviseriata are presented. T. curviseriata is ecologically characterized by eurythermy and euryhalinity, and its population variation is affected mainly by silicate, the ratios of phosphorus to silicon and nitrogen to silicon.XIE Wenling LI Yang GAO Yahui 2008Acta Oceanologica Sinica2008,27,6:7
2Development and effectiveness of pseudotyped SARS-CoV-2 system as determined by neutralizing efficiency and entry inhibition test in vitro显示文摘With the development of the COVID-19 epidemic,there is an urgent need to establish a system for determining the effectiveness and neutralizing activity of vaccine candidates in biosafety level 2(BSL-2)facilities.Previously,researchers had developed a pseudotyped virus systemfor SARS-CoV andMERS-CoV,based onHIV-1 core,bearing virus spike protein.During the development of a pseudotyped SARS-CoV-2 system,a eukaryotic expression plasmid expressing SARSCoV-2 spike(S)protein was constructed and then co-transfectedwith HIV-1 based plasmid which containing the firefly luciferase reporter gene,into HEK293T cells to prepare the pseudotyped SARS-CoV-2 virus(ppSARS-2).We have successfully established the pseudotyped SARS-CoV-2 system for neutralization and entry inhibition assays.Huh7.5 cell line was found to be the most susceptible to our pseudotyped virus model.Different levels of neutralizing antibodies were detected in convalescent serum samples of COVID-19 patients using ppSARS-2.The recombinant,soluble,angiotensin-converting enzyme 2 protein was found to inhibit the entry of ppSARS-2 in Huh7.5 cells effectively.Furthermore,the neutralization results for ppSARS-2 were consistent with those of live SARS-CoV-2 and determined using the serum samples fromconvalescent patients.In conclusion,we have developed an easily accessible and reliable tool for studying the neutralizing efficiency of antibodies against SARS-CoV-2 and the entry process of the virus in a BSL-2 laboratory.Ren Yang Baoying Huang Ruhan A Wenhui Li Wenling Wang Yao Deng Wenjie Tan 2020Biosafety and Health2020,2,4:5
3Qualitative and quantitative assessment of related substances in the Compound Ketoconazole and Clobetasol Propionate Cream by HPLC-TOF-MS and HPLC显示文摘Related substances in pharmaceutical formulations are associated with their safety, efficacy and stability. However, there is no overall study already published on the assessment of related substances in the Compound Ketoconazole and Clobetasol Propionate Cream. In this work, a reliable HPLC-TOF-MS qualitative method was developed for the analysis of related substances in this preparation with a quick and easy extraction procedure. Besides the active pharmaceutical ingredients, two compounds named ketoconazole impurity B′ optical isomer and ketoconazole impurity E were identified. Furthermore, a new HPLC method for qualitative and quantitative assessment on related substances and degradation products, which were found in the stability test, was established and validated. The single standard to determine multi-components method was applied in the quantitative analysis, which was an effective way for reducing cost and improving accuracy. 4Genetic basis of high aroma and stress toleranee in the oolong tea cultivar genome显示文摘Tea plants(Camellia sinensis)are commercially cultivated in>60 countries,and their fresh leaves are processed into tea,which is the most widely consumed beverage in the world.Although several chromosome-level tea plant genomes have been published,they collapsed the two haplotypes and ignored a large number of allelic variations that may underlie important biological functions in this species.Here,we present a phased chromosome-scale assembly for an elite oolong tea cultivar,'Huangdan',that is well known for its high levels of aroma.Based on the two sets of haplotype genome data,we identi fi ed numerous genetic variations and a substantial proportion of allelic imbalance related to important traits,including aroma-and stress-related alleles.Comparative genomics revealed extensive structural variations as well as expansion of some gene families,such as terpene synthases(TPSs),that likely contribute to the high-aroma characteristics of the backbone parent,underlying the molecular basis for the biosynthesis of aroma-related chemicals in oolong tea.Our results uncovered the genetic basis of special features of this oolong tea cultivar,providing fundamental genomic resources to study evolution and domestication for the economically important tea crop.Pengjie Wang Jiaxin Yu Shan Jin Shuai Chen Chuan Yue Wenling Wang Shuilian Gao Hongli Cao Yucheng Zheng Mengya Gu Xuejin Chen Yun Sun Yuqiong Guo Jiangfan Yang Xingtan Zhang Naixing Ye 2021Horticulture Research2021,8,1:4

6Emerging roles of activating transcription factor (ATF) family members in tumourigenesis and immunity: Implications in cancer immunotherapy显示文摘Activating transcription factors, ATFs, are a group of bZIP transcription factors that act as homodimers or heterodimers with a range of other bZIP factors. In general, ATFs respond to extracellular signals, indicating their important roles in maintaining homeostasis. The ATF family includes ATF1, ATF2, ATF3, ATF4, ATF5, ATF6, and ATF7. Consistent with the diversity of cellular processes reported to be regulated by ATFs, the functions of ATFs are also diverse. ATFs play an important role in cell proliferation, apoptosis, differentiation and inflammation-related pathological processes. The expression and phosphorylation status of ATFs are also related to neurodegenerative diseases and polycystic kidney disease. Various miRNAs target ATFs to regulate cancer proliferation, apoptosis, autophagy, sensitivity and resistance to radiotherapy and chemotherapy. Moreover, ATFs are necessary to maintain cell redox homeostasis. Therefore, deepening our understanding of the regulation and function of ATFs will provide insights into the basic regulatory mechanisms that influence how cells integrate extracellular and intracellular signals into genomic responses through transcription factors. Under pathological conditions, especially in cancer biology and response to treatment, the characterization of ATF dysfunction is important for understanding how to therapeutically utilize ATF2 or other pathways controlled by transcription factors. 12Whole-genome Duplication Reshaped Adaptive Evolution in A Relict Plant Species,Cyclocarya paliurus显示文摘Cyclocarya paliurus is a relict plant species that survived the last glacial period and shows a population expansion recently.Its leaves have been traditionally used to treat obesity and diabetes with the well-known active ingredient cyclocaric acid B.Here,we presented three C.paliurus genomes from two diploids with different flower morphs and one haplotype-resolved tetraploid assembly.Comparative genomic analysis revealed two rounds of recent whole-genome duplication events and identified 691 genes with dosage effects that likely contribute to adaptive evolution through enhanced photosynthesis and increased accumulation of triterpenoids.Resequencing analysis of 45 C.paliurus individuals uncovered two bottlenecks,consistent with the known events of environmental changes,and many selectively swept genes involved in critical biological functions,including plant defense and secondary metabolite biosynthesis.We also proposed the biosynthesis pathway of cyclocaric acid B based on multi-omics data and identified key genes,in particular gibberellinrelated genes,associated with the heterodichogamy in C.paliurus species.Our study sheds light on evolutionary history of C.paliurus and provides genomic resources to study the medicinal herbs.Yinquan Qu Xulan Shang Ziyan Zeng Yanhao Yu Guoliang Bian Wenling Wang Li Liu Li Tian Shengcheng Zhang Qian Wang Dejin Xie Xuequn Chen Zhenyang Liao Yibin Wang Jian Qin Wanxia Yang Caowen Sun Xiangxiang Fu Xingtan Zhang Shengzuo Fang 2023Genomics, Proteomics & Bioinformatics2023,21,3:1

14Numerical simulation of gas–liquid flow in the bubble column using Wray–Agarwal turbulence model coupled with population balance model显示文摘In this paper,an improved computational fluid dynamic(CFD)model for gas-liquid flow in bubble column was developed using the one-equation Wary-Agarwal(WA)turbulence model coupled with the population balance model(PBM).Through 18 orthogonal test cases,the optimal combination of interfacial force models,including drag force,lift force,turbulent dispersion force.The modified wall lubrication force model was proposed to improve the predictive ability for hydrodynamic behavior near the wall of the bubble column.The values simulated by optimized CFD model were in agreement with experimental data,and the errors were within±20%.In addition,the axial velocity,turbulent kinetic energy,bubble size distribution,and the dynamic characteristic of bubble plume were analyzed at different superficial gas velocities.This research work could provide a theoretical basis for the extension of the CFD-PBM coupled model to other multiphase reactors..Hongwei Liang Wenling Li Zisheng Feng Jianming Chen Guangwen Chu Yang Xiang 2023Chinese Journal of Chemical Engineering2023,58,6:1
