1晚期非小细胞肺癌免疫检查点抑制剂新型生物标志物:血液PD-L1动态检测显示文摘背景免疫检查点抑制剂(immune checkpoint inhibitors,ICIs)具有良好疗效已成为恶性肿瘤治疗领域备受瞩目的翘楚。然而,仅有小部分患者获得长期临床获益。如何选择生物标志物来筛查ICIs治疗优势人群仍是一大热点。方法收集51例晚期恶性肿瘤患者的肿瘤组织和配对血样本,进行PD-L1表达的相关性分析。40例晚期非小细胞肺癌(non-small cell lung cancer,NSCLC)患者接受ICIs治疗,2个月时检测血液PD-L1(bloodPD-L1,bPD-L1)表达的动态变化,包括PD-L1 mRNA、外泌体PD-L1(exosomalPD-L1,exoPD-L1)蛋白和可溶性PD-L1(solublePD-L1,sPD-L1)。应用R软件计算最佳截断值,用于预测最佳客观缓解(best objective response,b OR)、无进展生存期(progression-free survival,PFS)和总生存期(overall survival,OS)。结果51例不同恶性肿瘤患者中,组织PD-L1(tissuePD-L1,tPD-L1)表达阳性患者的PD-L1mRNA表达水平显著升高。40例接受ICIs治疗的晚期NSCLC患者中,bPD-L1 mRNA上调≥2.04的患者b OR、PFS和OS更佳;21例接受ICIs治疗的晚期NSCLC患者中,exoPD-L1上调≥1.86的患者临床获益更大。sPD-L1动态变化与ICIs疗效无显著相关性。PD-L1 mRNA和exoPD-L1联合检测更有利于预测ICIs治疗疗效。结论bPD-L1表达与tPD-L1表达存在正相关;晚期NSCLC患者ICIs治疗初期PD-L1 mRNA和(或)exoPD-L1表达升高,可作为评估临床获益的新型生物标志物。Qiao YANG Mingjing CHEN Jiaoyang GU Kai NIU Xianlan ZHAO Linpeng ZHENG Zihan XU Yongxin YU Feng LI Lingxin MENG Zhengtang CHEN Wenlei ZHUO Luping ZHANG Jianguo SUN 杨峤(译) 孙建国(译) 2021中国肺癌杂志2021,24,12:10
2Digital Evaluation of Sitting Posture Comfort in Human-vehicle System under Industry 4.0 Framework显示文摘Most of the previous studies on the vibration ride comfort of the human-vehicle system were focused only on one or two aspects of the investigation. A hybrid approach which integrates all kinds of investigation methods in real environment and virtual environment is described. The real experimental environment includes the WBV(whole body vibration) test, questionnaires for human subjective sensation and motion capture. The virtual experimental environment includes the theoretical calculation on simplified 5-DOF human body vibration model, the vibration simulation and analysis within ADAMS/VibrationTMmodule, and the digital human biomechanics and occupational health analysis in Jack software. While the real experimental environment provides realistic and accurate test results, it also serves as core and validation for the virtual experimental environment. The virtual experimental environment takes full advantages of current available vibration simulation and digital human modelling software, and makes it possible to evaluate the sitting posture comfort in a human-vehicle system with various human anthropometric parameters. 3Generation of coexisting high-energy pulses in a mode-locked all-fiber laser with a nonlinear multimodal interference technique显示文摘We demonstrate a passively mode-locked all-fiber laser incorporating a piece of graded-index multimode fiber as a mode-locking modulator based on a nonlinear multimodal interference technique, which generates two types of coexisting high-energy ultrashort pulses [i.e., the conventional soliton(CS) and the stretched pulse(SP)]. The CS with pulse energy as high as 0.38 n J is obtained at the pump level of 130 mW. When the pump increases to175 mW, the high-energy SP occurs at a suitable nonlinear phase bias and its pulse energy can reach 4 n J at a 610 mW pump. The pulse durations of the generated CS and SP are 2.3 ps and 387 fs, respectively. The theory of nonlinear fiber optics, single-shot spectral measurement by the dispersive Fourier-transform technique, and simulation methods based on the Ginzburg–Landau equation are provided to characterize the laser physics and reveal the underlying principles of the generated CS and SP. A rogue wave, observed between the CS and SP regions, mirrors the laser physics behind the dynamics of generating a high-energy SP from a CS. 4Adaptive tracking control of an MEMS gyroscope with H-infinity performance显示文摘Microelectromechanical systems (MEMSs) pose unique measurement and control problems compared with conventional ones because of their small size,low cost,and low power consumption.The vibrating gyroscope is one of those MEMS devices that have significant potential in many industry applications.When the MEMS gyroscope system is considered simultaneously with the coupling terms,the exogenous disturbances and the parameter variations,the controller design of this system becomes very challenging.This paper investigates the primary control problem of a perturbed vibrating MEMS gyroscope.A nonlinear robust adaptive control scheme is proposed for the drive axis of a vibrating MEMS gyroscope.By combining the dynamic surface control (DSC) method with the H-infinity disturbance attenuation technique,a simpler systematic design procedure is developed.The derived H-infinity controller has a simplified structure,and it can drive the drive axis to resonance,regulate the output amplitude of the drive axis to a desired value,and attenuate the generalized disturbances.The features of the derived controller are discussed and illustrated by the simulation of a closed-loop system.The analysis and simulation show that the obtained controller possesses good adaptability and robustness to system uncertainties.Li, Wenlei Liu, Peter X. 2011控制理论与应用(英文版)2011,9,2:5
5Decaying evolution dynamics of double-pulse mode-locking显示文摘Taking advantage of the dispersive Fourier transformation technique, the decaying evolution processes of doublepulse mode-locking in a single-walled carbon-nanotube-based Er-doped fiber laser are observed in detail for the first time to our knowledge. The decaying dynamics of the double-pulse mode-locking state is analyzed in the spectral and temporal domains. We reveal that the two pulses in one cluster disappear either simultaneously or one by one during the decaying processes of double-pulse mode-locking states. 6Thermoplastic micro-formability of TiZrHfNiCuBe high entropy metallic glass显示文摘High entropy metallic glasses(MGs) have attracted tremendous attentions owing to high entropy that benefits the probing of new MG-forming systems. However, the micro-formability of high entropy MGs is lack of investigation in comparison with these conventional MG counterparts, which is crucial to the development of this kind of metallic alloys. In this work, the thermoplastic mciro-formability of TiZrHfNiCuBe high entropy MG was systemically investigated. Time-Temperature-Transformation(TTT)curve was first constructed based on isothermal crystallization experiments, which provides thermoplastic processing time of the supercooled high entropy MGs. By comparison with the deformation map,Newtonian flow was found beneficial to the thermoplastic formability. While the thermoplastic forming becomes arduous with reducing mould size to tens micrometer, because of the strong supercooled TiZrHfNiCuBe high entropy MG(fragility = 27). Fortunately, the micro-formability of TiZrHfNiCuBe high entropy MG could be improved by vibration loading, as demonstrated by finite-element-method simulation. 7Particle image velocimetry for an automatic cooling device using temperature-sensitive magnetic fluid显示文摘An automatic cooling device has been developed by using a temperature-sensitive magnetic fluid as the coolant. A particle image velocimetry (PIV) system was used to measure the flow velocity of the fluid inside the loop. The efficiency of the device under varying conditions such as the heat load and the power of cooling were ex- perimentally investigated. The effect of cooperation between external magnetic field and thermal field on the performance of the device was studied. As expected, a continuous flow induced by the thermal and magnetic field was observed in the loop, where heat was transferred by the circulating magnetic fluid. 8Gedunin Degrades Aggregates of Mutant Huntingtin Protein and Intranuclear Inclusions via the Proteasomal Pathway in Neurons and Fibroblasts from Patients with Huntington’s Disease显示文摘Huntington's disease(HD) is a deadly neurodegenerative disease with abnormal expansion of CAG repeats in the huntingtin gene. Mutant Huntingtin protein(m HTT) forms abnormal aggregates and intranuclear inclusions in specific neurons, resulting in cell death. Here,we tested the ability of a natural heat-shock protein 90 inhibitor, Gedunin, to degrade transfected m HTT in Neuro-2 a cells and endogenous m HTT aggregates and intranuclear inclusions in both fibroblasts from HD patients and neurons derived from induced pluripotent stem cells from patients. Our data showed that Gedunin treatment degraded transfected m HTT in Neuro-2 a cells, endogenous m HTT aggregates and intranuclear inclusions in fibroblasts from HD patients, and in neurons derived from induced pluripotent stem cells from patients in a dose-and time-dependent manner, and its activity depended on the proteasomal pathway rather than the autophagy route. 11Repercussions of COVID-19 pandemic on solid waste generation and management strategies显示文摘It has been over ten months since the beginning of the 2019 coronavirus disease(COVID-2019),and its impact on solid waste management,especially medical waste,is becoming clearer.This study systematically reviews the potential influences of the COVID-19 pandemic on medical waste,personal protection equipment waste and municipal solid waste(MSW),and discusses the corresponding measures and policies of solid waste management in typical countries.The results show that the generation of medical waste from the pandemic increased significantly,with 18%‒425%growth.It is estimated that the daily output of COVID-19 medical waste increased from 200 t/d on Feb.22 to over 29000 t/d at the end of September 2020 throughout the world.The use of personal protective equipment will continue to grow in the long-term,while the blockade and isolation measures greatly reduced the volume of commercial waste,especially for tourist cities,and part of this waste was transferred to household waste.Residents’attitudes and behavior toward food waste have changed due to the COVID-19 pandemic.In response to the pandemic,international organizations and several countries have issued new policies and guidelines and adjusted their management strategies for medical waste and MSW treatment.The pandemic has brought specific challenges to the disposal capacity of medical waste worldwide.It has also brought about the stagnation of policies related to the reduction of plastic products and waste recycling.This study will provide some useful information for managers and governmental officials on effective solid waste management during and after the COVID-19 pandemic.Yangyang Liang Qingbin Song Naiqi Wu Jinhui Li Yuan Zhong Wenlei Zeng 2021Frontiers of Environmental Science & Engineering2021,15,6:1
