| 2 | The investigation for the relationship among serum leptin, erythrocyte membrane Ca^(2+)-ATPase activity and hypertensive disorder complicating pregnancy显示文摘Objective: To study the significance of Leptin and the activity of erythrocyte membrane Ca2+-ATPase(EMCA) in the development of hypertensive disorder complicating pregnancy. Methods: Radioimmunoassay was used to test the level of serum Leptin,and the activity of EMCA was determined chemically in 38 pregnant women with hypertensive disorder complicating pregnancy and 36 normotensive pregnant women. Results: The level of serum Leptin in hypertensive disorder complicating pregnancy(gestational hypertension: 13.76 ± 3.46 ng/ml; preeclampsia:15.76 ± 5.47 ng/ml; eclampsia: 18.32 ± 6.38 ng/ml)was significantly higher than that in normotensive pregnant women(11.33 ± 2.93 ng/ml),respectively. The average EMCA activity of patients with hypertensive disorder complicating pregnancy(gestational hypertension:1.65 ± 0.24 μmol·pi/mg·h ; preeclampsia:1.37 ± 0.19 μ mol·pi/mg·h; eclampsia:1.12 ± 0.14 μ mol·pi/mg·h) was significantly lower than that of normotensive pregnant women(1.83 ± 0.38 μ mol·pi/mg·h),respectively. There was a negative correlation between the level of serum Leptin and the activity of RMCA in hypertensive disorder complicating pregnancy(r = -0.63). Conclusion: Inhibition of EMCA activity of erythrocyte in hypertensive disorder complicating pregnancy may increase cytoplasmic free calcium, which contributes to the development of hypertensive disorder complicating pregnancy. | 3 | Effect of artesunate on human endometrial carcinoma HEC-1B cells显示文摘Objective:To observe the effect of the artesunate(ART) on cellular proliferation in vitro,to search for the possible anti-tumor mechanism of ART on endometrial carcinoma at the molecular level and to provide the experimental and theoretical foundations for the clinical applications of ART.Methods:The cell proliferation was observed by microscope;MTT was used to examine the effects of ART on proliferation of HEC-1B cells,and flow cytometric analysis was used to detect cell cycle and apoptosis.The human endometrial carcinoma HEC-1B cells were conventionally cultured;ART was administered with a concentration of 40 μg/ml before the total RNA were extracted.mRNA expression of Survivin,Caspase-3,N-Cadherin,E-Cadherin,Fibronectin1 and Cox-2 were detected using RT-PCR.Results:ART reduced proliferation in human endometrial carcinoma cell line HEC-1B in a dose-and time-dependent effect.The cells of G0/G1 stage were significantly increased(P<0.05),but the cells of G2/M stages were significantly decreased(P<0.05),so it has shown that the cell cycle was probably blocked in G0/G1 stage.After intervention with ART at 20 and 80 μg/ml for 48 h,cellular apoptosis rate respectively was(36.42±0.77)% and(11.77±0.58)%,and the difference was statistically significant compared with the control([6.64±0.19]%,P<0.01).The expression of Cox-2 mRNA in the ART group was lower than those of control group,yet the expression of Caspase-3 and E-Cadherin mRNA in the ART group was higher than those of control group.Conclusion:ART can inhibit HEC-1B cell growth and proliferation in a dose-and time-dependent manner.Furthermore,ART can induce apoptosis in a dose-dependent manner.ART is able to downregulate Cox-2 mRNA expression and to upregulate E-Cadherin and Caspase-3 mRNA expression.So we can conclude that ART could induce the endometrial carcinoma HEC-1B cell apoptosis and inhibit tumor cell proliferation. | Wang Lijuan Yang Yucong Gou Wenli | 2010 | Journal of Medical Colleges of PLA(China)2010,25,3: | 2 |
| 4 | Effect of selective uterine artery embolization on symptomatic uterine fibroids显示文摘Objective: To evaluate the outcomes of selective uterine artery embolization (SUAE) in treatment of symptomatic uterine fibroids using Color Doppler ultrasonography (CDUS). Methods: As the procedure SUAE for fibroids, prospective data of the initial 65 consecutive women treated from March 2007 to September 2009 were collected. The follow-up period from 1 week to 30 months, using questionnaires, we investigated the fibroid-related physical and psychological symptoms, and the cumulative rates of symptom control, gynecologic interventions, and complications. The volumes of uterine and uterine fibroids were calculated by CDUS pre- and post-SUAE for 6 months. And we tested the levels of internal hormone including FSH, LH and E2 before and after SUAE for 3, 6 months. Results: SUAE was performed successfully in all patients. There was no peri-operative morbidity in all procedures. Compared with pre-SUAE volumes of uterine [(322±112) cm3] and uterine fibroids [(125±46) cm3], their volumes in post-SUAE were (144±72) cm3 and (51±11) cm3 (P<0.01), and reduction rate was 58% and 61%, respectively. At 3, 6 months after SUAE, levels of FSH, LH, and E2 were not decreased than that of in pre-SUAE (P>0.05). After SUAE for 24 months, the fibroids related physical and psychological symptoms such as bleeding, pain and quality of life were significantly improved (P<0.01). 90.8% (59/65) women had resolution of symptoms and no significant post-procedural symptoms. Parts of patients discharge necrotic fibroids. 86% (34/40) complications were minor, requiring no therapy. 9.2% (6/65) had significant post-procedural symptoms, requiring therapy in hospital or out-patient department. Conclusion: SUAE contributes to a long-term significant improvement of all investigated uterine fibroid-related symptoms and markedly improves women's health-related quality of life. SUAE that no impair on changes of internal hormone, under suitable conditions, represents an attractive alternative to hysterectomy for the treatment of symptomatic fibroids and is a minimally invasive, new method of good clinical prognosis. | 5 | Effect of behavior training on learning and memory of young rats with fetal growth restriction显示文摘Objective: To investigate the effect of behavior training on the learning and memory of young rats with fetal growth restriction (FGR). Methods: The model of FGR was established by passive smoking method to pregnant rats. The new-born rats were divided into FGR group and normal group, and then randomly subdivided into trained and untrained group respectively. Morris water maze behavior training was performed on postnatal months 2 and 4, then learning and memory abilities of young rats were measured by dark-avoidance testing and step-down testing. Results: In the dark-avoidance and step-down testing, the young rats’ performance of FGR group was worse than that of control group, and the trained group was better than the untrained group significantly. Conclusion: FGR young rats have descended learning and memory abilities. Morris water maze test was proceeded on postnatal month(PM2/4)as a behavior training method,then the learning-memory of rats was detected through dark-avoidance and step-down tests.The expressions of NR2B and GluR1 subunits in hippocampal CA1 and CA3 areas were detected by immunohistochemical method.Results:In the dark-avoidance and step-down tests,the performance record of rats with FGR was worse than that of control rats,and the behavior-trained rats was better than the untrained rats,when the FGR model and training factors were analyzed singly.The model factor and training factor had significant interaction(P<0.05).The expressions of NR2B and GluR1 subunits in hippocampal CA1 and CA3 areas of rats with FGR reduced.In contrast,the expressions of GluR1 and NR2B subunits in CA1 area of behavior-trained rats increased,when the FGR model and training factors were analyzed singly. | 10 | Impairments of spatial learning and memory in rat offspring with fetal growth restriction显示文摘Objective:Throughout the world,fetal growth restriction(FGR) is one of the most severe complications occurring during pregnancy. It is subsequently associated with neurologic abnormalities in chldren. Our aim was to investigate the spatial learning and memory ability of rat offspring born with FGR. Methods:A rat model of FGR was constructed using the method of passive smoking. Spatial learning and memory were studied in rat offspring born with FGR by assessing the animals’ performance using the Morris water maze task. Results: At 1-and 2-months of age,both female and male offspring rats showed impairment of performance,while at 4 months of age,only female rats showed impaired performance. The FGR offspring spent a longer time swimming and used inefficient strategies(P < 0.05,respectively). However,there were no significant maze performance FGR effects in the 4 month old male rats. In all groups of FGR offspring,irrespective of age or sex,the time spent in the platform quadrant by the rat was significantly less than that in the control group(P < 0.05). Conclusion: The Morris water maze performance decreased in rat offspring born with FGR.
