1Evodiamine induces extrinsic and intrinsic apoptosis of ovarian cancer cells via the mitogen-activated protein kinase/phosphatidylinositol-3-kinase/protein kinase B signaling pathways显示文摘OBJECTIVE: To explore the effects of evodiamine on ovarian cancer cells and the mechanisms underlying such effects.METHODS: Human ovarian cancer cells HO-8910 PM were treated with evodiamine at 0, 1.25,2.5, and 5 μM for 1-4 d. 3-(4,5-Dimethiylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay was used to detect the growth inhibition rate of evodiamine-treated HO-8910 PM cells. The cell cycle was observed via propidium iodide(PI) staining. Apoptosis induction was assessed via Annexin V-fluorescein isothiocyanate/propidium iodide(Annexin V-FITC/PI) double staining assay. To verify the mechanism of apoptosis, caspase-dependent apoptotic pathway-related protein was detected by Western blot analysis. The expression levels of mitogen-activated protein kinase(MAPK)and/or phosphatidylinositol-3-kinase(PI3K)/pro-tein kinase B(Akt) pathway-related proteins were also investigated.RESULTS: Evodiamine significantly inhibited the proliferation of HO-8910 PM cells in a dose- and time-dependent manner. Evodiamine induced G2/M arrest with an increase of cyclin B1 level, and promoted cell apoptosis with a decrease of B cell lymphoma/lewkmia-2(Bcl-2) and an increase of Bcl-2-associated X protein(Bax) level. In addition,evodiamine treatment led to the activation of caspase-8, caspase-9, and caspase-3 and the cleavage of poly(ADP-ribose)-polymerase(PARP). Evodiamine targeted the MAPK and/or PI3K/Akt pathways by reducing the expression and activity of PI3 K, Akt, and extracellular signal-regulated kinase mitogen-activated protein kinase(ERK1/2 MAPK)and the activity of p38 MAPK.CONCLUSION: Evodiamine can inhibit the growth of ovarian cancer cells by G2/M arrest and intrinsic and extrinsic apoptosis. 2The pattern of plasma BCAA concentration and liver Bckdha gene expression in GK rats during T2D progression显示文摘Background: This study was conducted to measure the concentration of branched chain amino acid(BCAA) in different species and detect the expression pattern of the liver Bckdha gene in Goto-Kakizaki(GK) rats during type 2 diabetes(T2D)progression.Methods: We measured the concentration of BCAA in GK rats, induced T2D cynomolgus monkeys and T2D humans by liquid chromatography tandem mass spectrometry, and used real-time quantitative PCR to analyze the gene expression of Bckdha and Bckdk, which encode the rate-limiting enzymes in catabolism of,respectively, branched chain amino acids and branched chain α-keto acid dehydrogenase kinase.Results: In this study, we showed that GK rat BCAA concentrations were significantly reduced at 4 and 8 weeks(P < 0.05 and P < 0.01, respectively), while the expression of Bckdha in GK rat liver was increased at 4 and 8 weeks(1.62-fold and1.93-fold, respectively). The BCAA concentrations were significantly reduced in diet-induced T2D cynomolgus monkeys(P < 0.01), but significantly increased in T2D humans(P < 0.001).Conclusions: Our results showed that BCAA concentrations changed at different times and by different amounts in different species and during different periods of T2D progress, and the significant changes of BCAA concentration in the three species indicated that BCAA might participate in the progress of T2D. The results suggested that the increased expression of Bckdha in GK rat liver might partially explain the reduced plasma BCAA concentration at 4 and 8 weeks. 3Strategies to improve the fluorescent signal of the tripartite sfGFP system显示文摘Bimolecular fluoresce nee compleme ntati on(BiFC)is a popular method used to detect proteinprotein interactions.For a BiFC assay,a fluorescent protein is usually split into two parts,and the fluoresce nee is recovered upon the in teracti on between the fused proteins of interest.As an elega nt extensi on of BiFC,a tripartite superfold green fluoresce nt protein(sfGFP)system that has the advantages of low background fluorescence and small fusion tag size has been developed.However,the tripartite system exhibits a low fluorescence signal in some cases.To address this problem,we proposed to in crease the affinity betwee n the two parts,G1-9 and G11,of the tripartite system by adding affinity pairs.Among the three affinity pairs tested,LgBiT-HiBiT improved both the signal and sign a I-to-noise(S/N)ratio to the greatest exte nt.More strikingly,the direct covalent fusion of G11 to G1-9,which converted the tripartite system into a new bipartite system,enhanced the S/N ratio from 20 to 146,which is superior to the bipartite sfGFP system split at 157/158 or 173/174.Our results implied that the 10th p-strand of sfGFP has a low affinity and a good recovery efficiency to construct a robust BiFC system,and this concept might be applied to other fluorescent proteins with similar structure to construct new BiFC systems.Jing Shen Wenlu Zhang Chunyang Gan Xiafei Wei Jie Li Yuxue Sun Yi Yuan Xuefei Cai Quanxin Long Jing Cui Haitao Guo Ailong Huang Jieli Hu 2020Acta Biochimica et Biophysica Sinica2020,52,9:0

6Experimental study of channel electron multiplier显示文摘Background Channel electron multiplier(CEM)can be used to measure extremely few charged particles,such as electrons and ions.The CEM is widely used in particle detection,so it is very important to study their performance parameters.Purposes Test and analyze the performance parameters of the CEM,such as resistance,gain,and pulse output for a single entrance photoelectron.Methods The heated tantalum filament is used as a stable and adjustable planar electron source to test the performance of the CEM in the analog mode.The performance parameters of the CEM in the pulse counting mode are tested by using the ultraviolet LED to excite the gold photocathode to generate a single photoelectron.Results and conclusions The gain of the CEM in the analog mode can reach more than 106 and the gain in the pulse counting mode can be two orders of magnitude higher.The curved helical channel has a greater advantage than the ordinary straight channel,which is conducive to weakening the ion feedback phenomenon.Binting Zhang Cuiping Li Shulin Liu Baojun Yan Baoqiang Li Wenlu Wei Huaxing Peng 2023Radiation Detection Technology and Methods2023,7,1:0
