| 1 | Artificial Intelligence Applications in the Development of Autonomous Vehicles:A Survey显示文摘The advancement of artificial intelligence(AI)has truly stimulated the development and deployment of autonomous vehicles(AVs)in the transportation industry.Fueled by big data from various sensing devices and advanced computing resources,AI has become an essential component of AVs for perceiving the surrounding environment and making appropriate decision in motion.To achieve goal of full automation(i.e.,self-driving),it is important to know how AI works in AV systems.Existing research have made great efforts in investigating different aspects of applying AI in AV development.However,few studies have offered the research community a thorough examination of current practices in implementing AI in AVs.Thus,this paper aims to shorten the gap by providing a comprehensive survey of key studies in this research avenue.Specifically,it intends to analyze their use of AIs in supporting the primary applications in AVs:1)perception;2)localization and mapping;and 3)decision making.It investigates the current practices to understand how AI can be used and what are the challenges and issues associated with their implementation.Based on the exploration of current practices and technology advances,this paper further provides insights into potential opportunities regarding the use of AI in conjunction with other emerging technologies:1)high definition maps,big data,and high performance computing;2)augmented reality(AR)/virtual reality(VR)enhanced simulation platform;and 3)5G communication for connected AVs.This paper is expected to offer a quick reference for researchers interested in understanding the use of AI in AV research. | Yifang Ma Zhenyu Wang Hong Yang Lin Yang | 2020 | IEEE/CAA Journal of Automatica Sinica2020,7,2: | 16 |
| 2 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. | 3 | Variation in cis-regulation of a NAC transcription factor contributes to drought tolerance in wheat显示文摘Drought is a major environmental factor limiting wheat production worldwide,and developing drought-tolerant cultivars is a central challenge for wheat breeders globally.Therefore,it is important to identify genetic components determining drought tolerance in wheat.In this study,we identified a wheat NAC gene(TaNAC071-A)that is tightly associated with drought tolerance by a genome-wide association study.Knockdown of TaNAC071-A in wheat attenuated plant drought tolerance,whereas its overexpression significantly enhanced drought tolerance through improved water-use efficiency and increased expression of stress-responsive genes.This heightened water-saving mechanism mitigated the yield loss caused by water deficit.Further candidate gene association analysis showed that a 108-bp insertion in the promoter of TaNAC071-A alters its expression level and contributes to variation in drought tolerance among wheat accessions.This insertion contains two MYB cis-regulatory elements(CREs)that can be directly bound by the MYB transcription activator,TaMYBL1,thereby leading to increased TaNAC071-A expression and plant drought tolerance.Importantly,introgression of this 108-bp insertion allele,TaNAC071-AIn-693,into drought-sensitive cultivars could improve their drought tolerance,demonstrating that it is a valuable genetic resource for wheat breeding.Taken together,our findings highlight a major breakthrough in determining the genetic basis underlying phenotypic variation in wheat drought tolerance and showcase the potential of exploiting CRE-containing indels for improving important agronomical traits. | Hude Mao Shumin Li Bin Chen Chao Jian Fangming Mei Yifang Zhang Fangfang Li Nan Chen Tian Li Linying Du Li Ding Zhongxue Wang Xinxiu Cheng Xiaojing Wang Zhensheng Kang | 2022 | Molecular Plant2022,15,2: | 9 |

| 6 | DELAUNAY TRIANGULATION METHOD OF CURVED SURFACES BASED ON RIEMANNIAN METRIC显示文摘A method for quality mesh generation of parametric curved surfaces is proposed. It is shown that the main difference between the proposed method and previous ones is that our meshing process is done completely in the parametric domains with the guarantee of mesh quality. To obtain this aim, the Delaunay method is extended to anisotropic context of 2D domains, and a Riemannian metric map is introduced to remedy the mapping distortion from object space to parametric domain. Compared with previous algorithms, the approach is much simpler, more robust and speedy. | 9 | Simulated annealing spectral clustering algorithm for image segmentation显示文摘The similarity measure is crucial to the performance of spectral clustering. The Gaussian kernel function based on the Euclidean distance is usually adopted as the similarity measure. However, the Euclidean distance measure cannot fully reveal the complex distribution data, and the result of spectral clustering is very sensitive to the scaling parameter. To solve these problems,a new manifold distance measure and a novel simulated annealing spectral clustering(SASC) algorithm based on the manifold distance measure are proposed. The simulated annealing based on genetic algorithm(SAGA), characterized by its rapid convergence to the global optimum, is used to cluster the sample points in the spectral mapping space. The proposed algorithm can not only reflect local and global consistency better, but also reduce the sensitivity of spectral clustering to the kernel parameter, which improves the algorithm's clustering performance. To efficiently apply the algorithm to image segmentation, the Nystr¨om method is used to reduce the computation complexity. | 12 | Antisense Oligodeoxynucleotide Inhibits Expression of Re-com binantPorcine Follicle-Stim ulating Horm one Receptor显示文摘To assess the role of follicle stimulating hormone receptor(FSHR) gene expression in regulating expression of FSHR protein in the plasma membrane, the effects of a porcine FSHR cDNA antisense oligodeoxynucleotide (ODN) on FSHR mRNA levels and 125 I FSH binding were determined in Chinese hamster ovary cells expression recombinant porcine FSHR (pFSHR CHO cells). An 18 mer phosphorothioate endcapped antisense ODN that corresponded to the region surrounding the translation initiation codon of the porcine FSHR cDNA was synthesized. An 18 mer nonsense sequence of identical nucleotide composition, which had little homology to known DNA sequences, was synthesized for use as a control. pFSHR CHO cells were cultured in 24 well plates (10 5 cells/well) in the absence or presence of 1 20 μmol/L antisense or nonsense ODN for 24 h and then assayed for porcine FSHR mRNA, using quantitative reverse transcription and competitive polymerase chain reaction, and for 125 I FSH binding activity. Treatment with 10 μmol/L antisense ODN caused a paradoxical increase in porcine FSHR mRNA from 0.89±0.06 to 1.64±0.08 ng/mg total RNA ( P <0.05). Transfection with lipofectamine and 0.33 μmol/L antisense ODN caused an increase in porcine mRNA from 0.95±0.08 to 1.53±0.07 ng/mg total RAN. This was probably due to upregulation of mRNA synthesis resulting from inhibition of porcine FSHR protein translation. The nonsense ODN had no effect on porcine FSHR mRNA. Antisense, but not nonsense, ODN (10 μmol/L) inhibited membrane binding of 125 I FSH by 13.6± 0.8 % ( P <0.05) in 24 h. Treatment of cells with antisense ODN (10 μmol/L) for 48 h resulted in a 76±1.5 % ( P <0.05) inhibition of 125 I FSH binding. In contrast, transfection with lipofectamine and 0.33 μmol/L antisense ODN at 0 h caused a 76.1±1.3 % ( P <0.05) reduction in binding within 24 h. Binding had returned to 52.3±2.3 % ( P < 0.05) of normal by 48 h. These results indicate that an antisense ODN corresponding to the region of the translation start site of the porcine FSHR cDNA is an effective specific inhibitor of porcine FSHR synthesis and that inhibition of receptor synthesis causes a decrease in functional membrane bound FSHR. | ZHU Changhong 1, Mark D. | 14 | Stereotactic surgery for refractory epilepsy complicated by mental disorders A retrospective case analysis显示文摘BACKGROUND: Anti-epilepsy and anti-psychosis drugs have traditionally been used in the clinic to treat epilepsy complicated by mental disorders. However, there is still no effective therapy for refractory epilepsy patients suffering from persistent mental disorders. OBJECTIVE: To explore the therapeutic effects of stereotactic multi-element localization and multi-target radiofrequency ablation on patients with refractory primary epilepsy and mental disorders. DESIGN: A retrospective case analysis. SETTING: Department of Neurosurgery, the 454 Hospital of Chinese PLA. PARTICIPANTS: Between June and November 2004, 13 patients with refractory primary epilepsy compli-cated by persistent mental disorders were admitted to the Department of Neurosurgery, the 454 Hospital of Chinese PLA. The patient group consisted of nine males and four females, with an average age of 25 years (range 18-39 years), and a course of disease ranging 3-11 years. Diagnosis of mental disorders was in ac-cordance with Chinese Classification of Mental Disorders. Written informed consent was obtained from all patients and their families, and the treatment protocol was approved by the Ethics Committee of the Hospital. METHODS: Under venous inhalation anesthesia, the disease targets, including bilateral corpus callosum, bilateral amygdala, and bilateral medial septal area, as well as unilateral Forel-H area, were coagulated by RFG-3CF radiofrequency thermocoagulation at 75-80 ℃ for 60-70 seconds. During thermocoagulation, the targets were identified using deep-brain microelectrodes and localized according to electrophysiology and electric resistance values. MAIN OUTCOME MEASURES: One year post-surgery, epileptic seizures were assessed on the basis of the Tan classification, and psychogenic (non-epileptic) seizures were evaluated using a 5-grade system. RESULTS: All enrolled 13 epileptic patients were included in the final analysis. The results of follow-up evaluations demonstrated that epilepsy was well-controlled. In total, nine patients did not experience relapse, and the frequency of seizures was reduced by 75% in two patients, and 50%-74% in another two patients. Psychogenic seizures were ameliorated in 11 cases (class 1, recovery) and improved significantly in two cases (class 2, significant improvement). | 15 | Involvement of a cytoplasmic glyceraldehyde-3-phosphate dehydrogenase GapC-2 in low-phosphate-induced anthocyanin accumulation in Arabidopsis显示文摘Phosphorous is one of the essential mineral elements for plant growth and development.Typically, the shoots of plant seedlings usually turn a dark-brown or purple colour under low-Pi stress. Using protein 2-D gel and peptide mass fingerprinting mapping (PMF) methods, a cytoplasmic glyceralde-hyde-3-phosphate dehydrogenase GapC-2 was identified as a low-Pi responsive protein in Arabidopsisplants. Expression of AtGapC-2 protein was significantly decreased after 4 d of low-Pi stress. Two in-dependent T-DNA insertion lines of GapC-2 gene (At1g13440) showed a hypersensitive phenotype inresponse to low-Pi stress compared with wild type plants, while the transgenic complementation linesof the mutants showed a similar phenotype to the wild type. | 17 | Targeting lactate dehydrogenase A (LDHA) exerts antileukemic effects on T-cell acute lymphoblastic leukemia显示文摘Background:T-cell acute lymphoblastic leukemia(T-ALL)is an uncommon and aggressive subtype of acute lymphoblastic leukemia(ALL).In the serum of T-ALL patients,the activity of lactate dehydrogenase A(LDHA)is increased.We proposed that targeting LDHA may be a potential strategy to improve T-ALL outcomes.The current study was conducted to investigate the antileukemic effect of LDHA gene-targeting treatment on T-ALL and the underlying molecular mechanism.Methods:Primary T-ALL cell lines Jurkat and DU528 were treated with the LDH inhibitor oxamate.MTT,colony formation,apoptosis,and cell cycle assays were performed to investigate the effects of oxamate on T-ALL cells.Quantitative real-time PCR(qPCR)and Western blotting analyses were applied to determine the related signaling pathways.A mitochondrial reactive oxygen species(ROS)assay was performed to evaluate ROS production after T-ALL cells were treated with oxamate.A T-ALL transgenic zebrafish model with LDHA gene knockdown was established using CRISPR/Cas9 gene-editing technology,and then TUNEL,Western blotting,and T-ALL tumor progression analyses were conducted to investigate the effects of LDHA gene knockdown on T-ALL transgenic zebrafish.Results:Oxamate significantly inhibited proliferation and induced apoptosis of Jurkat and DU528 cells.It also arrested Jurkat and DU528 cells in G0/G1 phase and stimulated ROS production(all P<0.001).Blocking LDHA significantly decreased the gene and protein expression of c-Myc,as well as the levels of phosphorylated serine/threonine kinase(AKT)and glycogen synthase kinase 3 beta(GSK-3β)in the phosphatidylinositol 3′-kinase(PI3K)signaling pathway.LDHA gene knockdown delayed disease progression and down-regulated c-Myc mRNA and protein expression in T-ALL transgenic zebrafish.Conclusion:Targeting LDHA exerted an antileukemic effect on T-ALL,representing a potential strategy for T-ALL treatment. | Haizhi Yu Yafei Yin Yifang Yi Zhao Cheng Wenyong Kuang Ruijuan Li Haiying Zhong Yajuan Cui Lingli Yuan Fanjie Gong Zhihua Wang Heng Li Hongling Peng Guangsen Zhang | 2020 | Cancer Communications2020,40,10: | 2 |
