1A detailed procedure for CRISPR/Cas9-mediated gene editing in Arabidopsis thaliana显示文摘The newly developed CRISPR(Clustered Regularly Interspaced Short Palindromic Repeats)/Cas(CRISPR-associated) system has emerged as an efficient tool for genome-editing, providing an alternative to classical mutagenesis and transgenic methods to study gene function and improve crop traits. CRISPR/Cas facilitates targeted gene editing through RNA-guided DNA cleavage followed by cellular DNA repair mechanisms that introduce sequence changes at the site of cleavage. Here we describe a detailed procedure for our previously developed and highly efficient CRISPR/Cas9 method that allows the generation of heritable-targeted gene mutations andcorrections in Arabidopsis. This protocol describes the strategies and steps for the selection of targets, design of single-guide RNA(sg RNA), vector construction and analysis of transgenic lines. We also offer a method to target two loci simultaneously using vectors containing two different sg RNAs. 3Critical function of DNA methyltransferase 1 in tomato development and regulation of the DNA methylome and transcriptome显示文摘DNA methylation confers epigenetic regulation on gene expression and thereby on various biological processes.Tomato has emerged as an excellent system to study the function of DNA methylation in plant development.To date,regulation and function of DNA methylation maintenance remains unclear in tomato plants.Here,we report the critical function of tomato(Solanum lycopersicum)Methyltransferase 1(Sl MET1)in plant development and DNA methylome and transcriptome regulation.Using CRISPR-Cas9 gene editing,we generated slmet1 mutants and observed severe developmental defects with a frame-shift mutation,including small and curly leaves,defective inflorescence,and parthenocarpy.In leaf tissues,mutations in Sl MET1 caused CG hypomethylation and CHH hypermethylationon a whole-genome scale,leading to a disturbed transcriptome including ectopic expression of many RIN target genes such as ACC2 in leaf tissues,which are normally expressed in fruits.Neither the CG hypomethylation nor CHH hypermethylation in the slmet1 mutants is related to tissue culture.Meanwhile,tissue culture induces non-CG hypomethylation,which occurs more frequently at gene regions than at TE regions.Our results depict Sl MET1-and tissue culture-dependent tomato DNA methylomes,and that Sl MET1 is required for maintaining a normal transcriptome and normal development of tomato.Yu Yang Kai Tang Tatsiana U Datsenka Wenshan Liu Suhui Lv Zhaobo Lang Xingang Wang Jinghui Gao Wei Wang Wenfeng Nie Zhaoqing Chu Heng Zhang Avtar K HANDa Jian-Kang Zhu Huiming Zhang 2019Journal of Integrative Plant Biology2019,61,12:3
4北斗卫星自主完好性监测及在轨性能评估显示文摘With the development of satellite navigation technology,the user demands for the integrity of Global Navigation Satellite System(GNSS)have increased more and more.A ground-based monitoring system can hardly report an alarm message to GNSS users during the valid alarming period due to the satellite-Earth propagation delay.It is beneficial to monitor abnormal events and report the corresponding alarms from orbit.Adopting this approach,which is an important feature for future GNSS integrity monitoring,the time needed to provide an alarm is shorter and the system integrity capability is strengthened.The BeiDou Navigation Satellite System(BDS)new generation satellites have the capabilities of satellite autonomous integrity monitoring(SAIM).This paper presents the technical scheme of SAIM,and proposes the integrity monitoring method of both navigation signals and the clocks onboard.The proposed method was verified through the onboard test on the BDS satellites.In addition,we analyzed the integrity telemetry data from the new generation of BDS satellite,including signal delay,power,carrier phase measurement,correlation peak,consistency of pseudo-code and carrier phase,clock phase and frequency step.The analysis results indicated that the quality of the data on orbit met the requirements,and SAIM could monitor effectively any abnormal change of satellite clocks and navigation signal,generate rapidly an alarm message,and transmit it to the user.The alarm time was less than 6 s through the message,and 2 s through non-standard code(NSC).Finally,we present future opportunities for improving the SAIM technology of BDS.BIAN Lang LIU Xiao LIU Wenshan YAN Tao LEI Wenying JIA Yizhe MENG Yansong ZHANG Lixin 2020Aerospace China2020,21,4:2

11Gradient-based method for the identification of multi-nodes in sugarcane显示文摘In view of the obvious changes in color between the upper and lower leaf scar in sugarcane nodes,a method of simultaneous multi-nodes identification on a single sugarcane stem was proposed based on the analysis of gradient characteristics of sugarcane images.In combination with image processing and machine vision recognition technology,two cameras were used to acquire different parts of sugarcane images,and the two images were integrated into a complete image of sugarcane by image mosaicking.The Sobel operator is used to calculate the gradient of the sugarcane image in a horizontal direction,and the gradient image is obtained.The sugarcane gradient image was scanned by a rectangular template with a width of 14 pixels and a step length of 12 pixels.The features of average gradient and variance gradient were used to identify sugarcane nodes for the first time.The experimental results showed that the recognition accuracy was 96.8952%,and there were fewer false detected sugarcane segments.The detection efficiency could be improved by detecting multi-nodes on a single sugarcane stem at the same time.Rui Yang Jie Li Qi Liu Wenshan Huang Kai Yin Xi Qiao Yiqi Huang 2020Information Processing in Agriculture2020,7,4:1

19A simple and sensitive assay for apurinic/apyrimidinic endonuclease 1activity based on host-guest interaction of β-cyclodextrin polymer and pyrene显示文摘Apurinic/apyrimidinic endonuclease 1(APE1) plays an important role in protecting the fidelity of genetic material and is also a promising biomarker of cancers. Herein, a simple and sensitive fluorescent method for APE1 activity detection was designed based on host-guest interaction between β-cyclodextrin polymer(β-CDP) and pyrene. In this method, pyrene-labelled DNA probes(AP-S1 S2) and β-CDP functioned as fluorescent signal producer and enhancer, respectively. When APE1 was absent, pyrene on the AP-S1 S2 could not enter the cavity of β-CDP because of steric hindrance, leading to a weak fluorescence intensity/anisotropy. When APE1 was present, it would recognize and cleave AP site in APS1 S2, thus producing a dissociated pyrene-labelled oligonucleotide segment. Pyrene labelled at the 50 end of oligonucleotide segment could be easily trapped into the cavity of β-CDP via host-guest interaction, leading to a significant enhancement of fluorescence intensity/anisotropy. This assay offered a dynamic range of 0.05 U/m L–5 U/m L and an estimated detection limit of 0.05 U/m L. 20Effects of frost action on soil physical properties of plough pan显示文摘Undisturb ed soil cores of loamy clay plough pan were subjected to the treatments of freezing and thawing in laboratory. The effects of frost action on its bulk density, porosity, and hydraulic conductivity were investigated. The results indicate that freezing and thawing significantly reduced the bulk density of plough pan and increased its porosity and hydraulic conductivity.
