1Activation of STAT3 stimulates AHSP expression in K562 cells显示文摘Studies on the chaperone proteinα-hemoglobin stabilizing protein(AHSP)reveal that abundant AHSP in erythroid cells enhance the cells’tolerance to oxidative stress imposed by excessα-hemoglobin in pathological conditions.However,the potential intracellular modulation of AHSP expression itself in response to oxidative stress is still unknown.The present study examined the effect and molecular mechanism of STAT3,an oxidative regulator,on the expression of AHSP.AHSP expression increased in K562 cells upon cytokine IL-6-induced STAT3 activation and decreased in STAT3 knock-down K562 cells.Regulation of AHSP in oxidative circumstance was then examined inα-globin-overloaded K562 cells,and real-time PCR showed strengthened expression of both AHSP and STAT3.ChIP analysis showed binding of STAT3 to AHSP promoter and binding was significantly augmented with IL6 stimulation and uponα-globin overexpression.Dual luciferase reporter assays of the wildtype and mutated SB3 element,an IL-6RE site,in the AHSP promoter in K562 cells highlighted the direct regulatory effect of STAT3 on AHSP gene.Finally,direct binding of STAT3 to SB3 site of AHSP promoter was confirmed with EMSA assays.Our work reveals an adaptive AHSP regulation mediated by the redox-sensitive STAT3 signaling pathway,and provides clues to the therapeutic strategy for AHSP enhancement.CAO Cong ZHAO GuoWei YU Wei XIE XueMin WANG WenTian YANG RuiFeng LV Xiang LIU DePei 2014Science China(Life Sciences)2014,57,5:5
2Protein tyrosine phosphatases in skeletal development and diseases显示文摘Skeletal development and homeostasis in mammals are modulated by finely coordinated processes of migration, proliferation,differentiation, and death of skeletogenic cells originating from the mesoderm and neural crest. Numerous molecular mechanisms are involved in these regulatory processes, one of which is protein posttranslational modifications, particularly protein tyrosine phosphorylation(PYP). PYP occurs mainly through the action of protein tyrosine kinases(PTKs), modifying protein enzymatic activity, changing its cellular localization, and aiding in the assembly or disassembly of protein signaling complexes. Under physiological conditions, PYP is balanced by the coordinated action of PTKs and protein tyrosine phosphatases(PTPs).Dysregulation of PYP can cause genetic, metabolic, developmental, and oncogenic skeletal diseases. Although PYP is a reversible biochemical process, in contrast to PTKs, little is known about how this equilibrium is modulated by PTPs in the skeletal system.Whole-genome sequencing has revealed a large and diverse superfamily of PTP genes(over 100 members) in humans, which can be further divided into cysteine(Cys)-, aspartic acid(Asp)-, and histidine(His)-based PTPs. Here, we review current knowledge about the functions and regulatory mechanisms of 28 PTPs involved in skeletal development and diseases;27 of them belong to class Ⅰ and Ⅱ Cys-based PTPs, and the other is an Asp-based PTP. Recent progress in analyzing animal models that harbor various mutations in these PTPs and future research directions are also discussed. 3SHP2 regulates skeletal cell fate by modifying SOX9 expression and transcriptional activity显示文摘Chondrocytes and osteoblasts differentiate from a common mesenchymal precursor, the osteochondroprogenitor(OCP), and help build the vertebrate skeleton. The signaling pathways that control lineage commitment for OCPs are incompletely understood. We asked whether the ubiquitously expressed protein-tyrosine phosphatase SHP2(encoded by Ptpn11) affects skeletal lineage commitment by conditionally deleting Ptpn11 in mouse limb and head mesenchyme using 'Cre-lox P'-mediated gene excision.SHP2-deficient mice have increased cartilage mass and deficient ossification, suggesting that SHP2-deficient OCPs become chondrocytes and not osteoblasts. Consistent with these observations, the expression of the master chondrogenic transcription factor SOX9 and its target genes Acan, Col2a1, and Col10a1 were increased in SHP2-deficient chondrocytes, as revealed by gene expression arrays, q RT-PCR, in situ hybridization, and immunostaining. Mechanistic studies demonstrate that SHP2 regulates OCP fate determination via the phosphorylation and SUMOylation of SOX9, mediated at least in part via the PKA signaling pathway. 4Targeted Ptpn11 deletion in mice reveals the essential role of SHP2 in osteoblast differentiation and skeletal homeostasis显示文摘The maturation and function of osteoblasts(OBs)rely heavily on the reversible phosphorylation of signaling proteins.To date,most of the work in OBs has focused on phosphorylation by tyrosyl kinases,but little has been revealed about dephosphorylation by protein tyrosine phosphatases(PTPases).SHP2(encoded by PTPN11)is a ubiquitously expressed PTPase.PTPN11 mutations are associated with both bone and cartilage manifestations in patients with Noonan syndrome(NS)and metachondromatosis(MC),although the underlying mechanisms remain elusive.Here,we report that SHP2 deletion in bone gamma-carboxyglutamate protein-expressing(Bglap+)bone cells leads to massive osteopenia in both trabecular and cortical bones due to the failure of bone cell maturation and enhanced osteoclast activity,and its deletion in Bglap+chondrocytes results in the onset of enchondroma and osteochondroma in aged mice with increased tubular bone length.Mechanistically,SHP2 was found to be required for osteoblastic differentiation by promoting RUNX2/OSTERIX signaling and for the suppression of osteoclastogenesis by inhibiting STAT3-mediated RANKL production by osteoblasts and osteocytes.These findings are likely to explain the compromised skeletal system in NS and MC patients and to inform the development of novel therapeutics to combat skeletal disorders.Lijun Wang Huiliang Yang Jiahui Huang Shaopeng Pei Liyun Wang Jian Q.Feng Dian Jing Hu Zhao Henry M.Kronenberg Douglas C.Moore Wentian Yang 2021Bone Research2021,9,1:3
5Granitoid emplacement during syn-convergent transtension:An example from the Huamenlou pluton in North Qinling,central China显示文摘The Huamenlou pluton,is an elongated granite intrusion with high aspect ratio,emplaced within the southern margin of the North Qinling(central China).Here we investigate this pluton through multiple techniques including the fabric study,microstructural observation and zircon geochronology.Our zircon U-Pb data confirm that the granite crystallized at ca.462 Ma which is consistent with the ages of other linear plutons in North Qinling.Microstructural observations of the Huamenlou granites illustrate that the pluton has undergone superimposed deformation during its emplacement,from magmatic to hightemperature solid state conditions.The internal fabric obtained by anisotropy of magnetic susceptibility(AMS)and shape preferred orientation(SPO)show similar results.The fabrics are relatively concordant and generally vary from NE-SW to NEE-SWW which are roughly oblique to the trend of the pluton elongation and the regional structures.Meanwhile,scalar parameters reflect two completely different strain regimes for the pluton and its host rocks,i.e.,the fabrics within host rocks are mainly oblate while the central part of the intrusion displays mainly prolate fabrics.It is inferred that the structural pattern recorded in this pluton was caused by local dextral transtension in consequence of oblique convergence between the South and North China Blocks.We propose that the local transtension in convergence setting probably evolved from vertical extrusion tectonics that provided room for the magma emplacement and imparted prolate fabrics in the Huamenlou pluton.Yang Li Wentian Liang Guowei Zhang Yazhou Ran Qi Shen Jingli Wang Chunsheng Jin 2018Geoscience Frontiers2018,9,1:1
64-6 Measurement of the Ratio of C3+ and O4+ Ions Produced by ECR Source to Prepare a Laser Cooling Experiment at Storage Rings显示文摘The cooling of heavy ions can provide high-quality beams that are especially important for high-precisionexperimental nuclear and atomic physics. The laser cooling of relativistic C3+ ion beams at the experimental coolerstorage ring (CSRe) is being currently prepared at Institute of Modern Physics (IMP) in Lanzhou. An electroncyclotron resonance ion source (ECRIS) will be used to produce C3+ ion beams. Meanwhile, O4+ ions could alsobe produced due to residual gas because of the same mass-to-charge ratio. Therefore, both C3+ and O4+ ion beamswill be injected and circulate in a storage ring during the laser cooling experiment at the same time. 74-16 Triple Coincidence (e, γ2e) Experiment for Ionization-excitation of Helium*显示文摘Electron induced ionization-excitation (IE) of helium is a basic four-body Coulomb problem in which all thefour charged particles are actively involved. It is much more challenging to both experiment and theory in contrastto direct ionization with the residual He+ ion in the ground state. The 2s or 2p separated TDCS data, especially atlow incident energy range where the high order effects are expected to play a significant role, would offer the moststringent test to theoretical models. 9Highly Sensitive and Portable Gas Sensing System Based on Reduced Graphene Oxide显示文摘Graphene has been widely used in gas-sensing applications due to its large specific surface area and strong adsorption ability. Among different forms of graphene used as gas-sensing materials, reduced graphene oxide is one of the most convenient and economical materials to integrate with Si-based electronics, which is very important to graphene-based gas sensors. In addition, the stacking structure of graphene oxide flakes facilitates absorption and detection of gas molecules. Based on reduced graphene oxide, a highly sensitive and portable gas-sensing system was demonstrated here. Solution-based graphene oxide was cast on a chip like a TF memory card and then reduced thermally. A signal acquisition system was designed to monitor resistance variation as a sign of gas concentration. This miniature graphene-based gas sensor array demonstrates a new path for the use of graphene in gas-detection technologies.
