| 1 | Memory stem T cells generated by Wnt signaling from blood of human renal clear cell carcinoma patients显示文摘Objective: Memory stem T cells(Tscm) have attracted attention because of their enhanced self-renewal, multipotent capacity, and anti-tumor capacities. However, little is known about Tscm in patients with renal clear cell carcinoma(RCC) and the role of Wnt signaling in these cells. We evaluated Tscm from RCC patients concerning their activation of Wnt signaling in vitro and explored the mechanism of preferential survival.Methods: Flow cytometry identified surface markers and cytokines produced from accumulated Tscm in the presence of the glycogen synthase kinase beta inhibitor TWS119. Apoptosis was evaluated after induction using tumor necrosis factor-alpha.Immunofluorescence and Western blot analyses were used to investigate the activation of the nuclear factor-kappa B(NF-КB)pathway.Results: RCC patients had a similar percentage of CD4^+ and CD8^+ Tscm as healthy donors. Activation of Wnt signaling by TWS119 resulted in the accumulation of Tscm in activated T cells, but reversal of differentiated T cells to Tscm was not achieved.Preferential survival of Tscm was associated with increased anti-apoptotic ability mediated downstream of the NF-КB activation pathway.Conclusions: The finding that Tscm can accumulate by Wnt signaling in vitro in blood from RCC patients will help in devising new cancer therapy strategies of Tscm-based adoptive immunotherapy, such as dendritic cell-stimulated Tscm, and T cell receptor or chimeric antigen receptor-engineered Tscm. | Cihui Yan Jingjing Chang Xinmiao Song Fan Yan Wenwen Yu Yang An Feng Wei Lili Yang Xiubao Ren | 2019 | Cancer Biology & Medicine2019,16,1: | 4 |
| 2 | The development of a sensitive fluorescent protein-based transcript reporter for high throughput screening of negative modulators of lncRNAs显示文摘While the human genome is pervasively transcribed,<2%of the human genome is transcribed into protein-coding mRNAs,leaving most of the transcripts as noncoding RNAs,such as microRNAs and long-noncoding RNAs(lncRNAs),which are critical components of epigenetic regulation.lncRNAs are emerging as critical regulators of gene expression and genomic stability.However,it remains largely unknown about how lncRNAs are regulated.Here,we develop a highly sensitive and dynamic reporter that allows us to identify and/or monitor negative modulators of lncRNA transcript levels in a high throughput fashion.Specifically,we engineer a fluorescent fusion protein by fusing three copies of the PEST destruction domain of mouse ornithine decarboxylase(MODC)to the C-terminal end of the codon-optimized bilirubin-inducible fluorescent protein,designated as dBiFP,and show that the dBiFP protein is highly destabilized,compared with the commonly-used eGFP protein.We further demonstrate that the dBiFP signal is effectively down-regulated when the dBiFP and mouse lncRNA H19 chimeric transcript is silenced by mouse H19-specific siRNAs.Therefore,our results strongly suggest that the dBiFP fusion protein may serve as a sensitive and dynamic transcript reporter to monitor the inhibition of lncRNAs by microRNAs,synthetic regulatory RNA molecules,RNA binding proteins,and/or small molecule inhibitors so that novel and efficacious inhibitors targeting the epigenetic circuit can be discovered to treat human diseases such as cancer and other chronic disorders. | Zongyue Zeng Bo Huang Shifeng Huang Ruyi Zhang Shujuan Yan Xinyi Yu Yi Shu Chen Zhao Jiayan Lei Wenwen Zhang Chao Yang Ke Wu Ying Wu Liping An Xiaojuan Ji Cheng Gong Chengfu Yuan Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Rex C.Haydon Hue H.Luu Lan Zhou Russell R.Reid Tong-Chuan He Xingye Wu | 2018 | Genes & Diseases2018,5,1: | 4 |
| 3 | The source and transport of bioaerosols in the air:A review显示文摘Recent pandemic outbreak of the corona-virus disease 2019(COVID-19)has raised widespread concerns about the importance of the bioaerosols.They are atmospheric aerosol particles of biological origins,mainly including bacteria,fungi,viruses,pollen,and cell debris.Bioaerosols can exert a substantial impact on ecosystems,climate change,air quality,and public health.Here,we review several relevant topics on bioaerosols,including sampling and detection techniques,characterization,effects on health and air quality,and control methods.However,very few studies have focused on the source apportionment and transport of bioaerosols.The knowledge of the sources and transport pathways of bioaerosols is essential for a comprehensive understanding of the role microorganisms play in the atmosphere and control the spread of epidemic diseases associated with them.Therefore,this review comprehensively summarizes the up to date progress on the source characteristics,source identification,and diffusion and transport process of bioaerosols.We intercompare three types of diffusion and transport models,with a special emphasis on a widely used mathematical model.This review also highlights the main factors affecting the source emission and transport process,such as biogeographic regions,land-use types,and environmental factors.Finally,this review outlines future perspectives on bioaerosols. | Wenwen Xie Yanpeng Li Wenyan Bai Junli Hou Tianfeng Ma Xuelin Zeng Liyuan Zhang Taicheng An | 2021 | Frontiers of Environmental Science & Engineering2021,15,3: | 2 |
