1ZIKA virus isolated from mosquitoes:a field and laboratory investigation in China,2016显示文摘A field investigation of arboviruses was conducted in Dejiang, Guizhou Province in the summer of 2016. A total of 8,795 mosquitoes, belonging to four species of three genera, and 1,300 midges were collected. The mosquito samples were identified on site according to their morphology, and the pooled samples were ground and centrifuged in the laboratory. The supernatant was incubated with mosquito tissue culture cells(C6/36) and mammalian cells(BHK-21) for virus isolation. The results indicated that 40%(3,540/8,795) were Anopheles sinensis, 30%(2,700/8,795) were Culex pipiens quinquefasciatus, and 29%(2,530/8,795)were Armigeres subbalbeatus. Furthermore, a total of eight virus isolates were obtained, and genome sequencing revealed two Zika viruses(ZIKVs) isolated from Culex pipiens quinquefasciatus and Armigeres subbalbeatus, respectively; three Japanese encephalitis viruses(JEVs) isolated from Culex pipiens quinquefasciatus; two Banna viruses(BAVs) isolated from Culex pipiens quinquefasciatus and Anopheles sinensis, respectively; and one densovirus(DNV) isolated from Culex pipiens quinquefasciatus.The ZIKVs isolated from the Culex pipiens quinquefasciatus and Armigeres subbalbeatus mosquitoes represent the first ZIKV isolates in China's Mainland. 4Biofunctional magnesium coated Ti6Al4V scaffold enhances osteogenesis and angiogenesis in vitro and in vivo for orthopedic application显示文摘The insufficient osteogenesis and osseointegration of porous titanium based scaffold limit its further application.Early angiogenesis is important for scaffold survival.It is necessary to develop a multifunctional surface on titanium scaffold with both osteogenic and angiogenic properties.In this study,a biofunctional magnesium coating is deposited on porous Ti6Al4V scaffold.For osseointegration and osteogenesis analysis,in vitro studies reveal that magnesium-coated Ti6Al4V co-culture with MC3T3-E1 cells can improve cell proliferation,adhesion,extracellular matrix(ECM)mineralization and ALP activity compared with bare Ti6Al4V cocultivation.Additionally,MC3T3-E1 cells cultured with magnesium-coated Ti6Al4V show significantly higher osteogenesisrelated genes expression.In vivo studies including fluorochrome labeling,micro-computerized tomography and histological examination of magnesium-coated Ti6Al4V scaffold reveal that new bone regeneration is significantly increased in rabbits after implantation.For angiogenesis studies,magnesium-coated Ti6Al4V improve HUVECs proliferation,adhesion,tube formation,wound-healing and Transwell abilities.HUVECs cultured with magnesium-coated Ti6Al4V display significantly higher angiogenesis-related genes(HIF-1αand VEGF)expression.Microangiography analysis reveal that magnesium-coated Ti6Al4V scaffold can significantly enhance the blood vessel formation.This study enlarges the application scope of magnesium and provides an optional choice to the conventional porous Ti6Al4V scaffold with enhanced osteogenesis and angiogenesis for further orthopedic applications.Peng Gao Bo Fan Xiaoming Yu Wenwen Liu Jie Wu Lei Shi Di Yang Lili Tan Peng Wan Yulin Hao Shujun Li Wentao Hou Ke Yang Xiaokang Li Zheng Guo 2020Bioactive Materials2020,5,3:15

6Influence of the post-treatment of HZSM-5 zeolite on catalytic performance for alkylation of benzene with methanol显示文摘The porous material HZSM-5 zeolite with micro-mesopore hierarchical porosity was prepared by post-treatment(combined alkali treatment and acid leaching) of parent zeolite and its catalytic performance for benzene alkylation with methanol was investigated. The effect of post-treatment on the textural properties was characterized by various techniques(including ICP-AES, XRD, nitrogen sorption isotherms, SEM, NH3-TPD, Py-IR and TG). The results indicated that the post-treatment could modify the structural and acidic properties of HZSM-5 zeolite. In this procedure, not only additional mesopores were created by selective extraction of silicon but also the acidity was tuned. Consequently, the modified HZSM-5 zeolite showed larger external surface area with less acid sites as compared to the parent zeolite. It was found out that the modified zeolite exhibited a higher benzene conversion and xylene selectivity for alkylation of benzene with methanol as well as excellent life span of the catalyst than conventional ones. This can be explained by the facts that the presence of additional mesopores improved the diffusion property in the reactions. Furthermore, the modified zeolite showed an appropriate Br?nsted acidity for effective suppression of the side reaction of methanol to olefins, thus reduced the accumulation of coke on the HZSM-5 zeolite, which was favorable for the catalyst stability. 7Response to paleoatmospheric CO_2 concentration of Solenites vimineus(Phillips)Harris(Ginkgophyta)from the Middle Jurassic of the Yaojie Basin,Gansu Province,China显示文摘Solenites vimineus (Phillips) Harris (Ginkgophyta) specimens with well-preserved cuticles were collected from five different beds in the oil shale member of the Middle Jurassic Yaojie Formation in the Yaojie Basin, Gansu Province, northwestern China. Gross morphology and fine structures of the fossil leaves were studied, and stomatal parameters were analyzed, according to which, the paleoatmospheric CO2 concentration can be deduced as 1512-1896 ppm that would have caused an increase in mean temperature of about 6.5-7.4℃. 11Benzene alkylation with methanol over phosphate modified hierarchical porous ZSM-5 with tailored acidity显示文摘The acidity and acid distribution of hierarchical porous ZSM-5 were tailored via phosphate modification. The catalytic results showed that both benzene conversion and selectivity of toluene and xylene increased with the presence of appropriate amount of phosphorus. Meanwhile, side reactions such as methanol to olefins related with the formation of by-product ethylbenzene formation and isomerization of xylene to meta-xylene were suppressed efficiently. 12Two soybean bHLH factors regulate response to iron deficiency显示文摘Iron is an indispensable micronutrient for plant growth and development. Limited bioavailability of Fe in the soil leads to iron deficiency chlorosis in plants and yield loss. In this study, two soybean basic helix-loophelix transcription factors, Gmb HLH57 and Gmb HLH300,were identified in response to Fe-deficiency. Both transcription factors are expressed in roots and nodules,and are induced by Fe deficiency; these patterns were confirmed in transgenic hairy roots expressing constructs of the endogenous promoters fused to a GUS reporter gene. Bimolecular fluorescence complementation, yeast two-hybrid and coimmunoprecipitation(co-IP) assays indicated a physical interaction between Gmb HLH57 and Gmb HLH300. 13Effect of Cyclocarya paliurus polysaccharides on streptozotocin-induced diabetic nephropathy in rats显示文摘OBJEVTIVE:To investigate the efficacy of Cyclocarya paliurus(C.paliurus)polysaccharides on streptozotocin-induced diabetic nephropathy in rats.METHODS:Rats were divided into 6 groups,including group of normal control,group of diabetic control,group of metformin treatment,low-dose group of C.paliurus polysaccharides treatment,middle-dose group of C.paliurus polysaccharides treatment and high-dose group of C.paliurus polysaccharides treatment.Histological analysis of kidney was analyzed using hematoxilin and eosin.Levels of blood glucose,creatinine,urea,uric acid were determined by spectrophotometry.Anti-oxidative enzymes were measured by real-time polymerase chain reaction(PCR)and enzyme-linked immunosorbent assay(ELISA).Advanced glycation end products(AGEs)and transforming growth factor-β1(TGF-β1)level was measured by ELISA.RESULTS:Abnormal changes were observed in the group of diabetic control characterized by atrophy of the renal glomeruli with hypercellularity,congestion of glomerular tufts,dilation of the renal spaces,and degeneration of renal tubule.Compared with that of normal group,blood glucose,creatinine,urea,uric acid level was significantly increased in the group of diabetic control.Superoxide dismutase,catalase,glutathione peroxidase,glutathione reductase level was significantly decreased,but AGEs and TGF-β1 level was significantly increased.By contrast,administration of C.paliurus polysaccharides and metformin could reverse the above-mentioned results of the group of diabetic control,especially in the high-dose group of C.paliurus polysaccharides.CONCLUSION:Our findings suggest that C.paliurus polysaccharides may play a protecting role for nephropathy of diabetic rats by lowering glucose,creatinine,urea,uric acid level,enhancing the antioxidative ability,and reducing AGEs and TGF-β1 expression.Xia Xichao Mao Dongxue Dai Hongmei Wu Xi Zhang Zuyuan Wang Huaying Zhou Wenwen Dong Yanmei Wang Mengqi Li Yuan Shao Xiangyang Ouyang Jingfeng 2020Journal of Traditional Chinese Medicine2020,40,6:9
14Preparation and properties of tungsten-doped indium oxide thin films显示文摘Tungsten-doped indium oxide (IWO) thin films were deposited on glass substrate by DC reactive magnetron sputtering. The effects of sputtering power and growth temperature on the structure, surface morphology, optical and electrical properties of IWO thin films were investigated. The thickness and surface morphology of the films are both closely dependent on the sputtering power and the substrate temperature. The transparency of the films decreases with the increase of the sputtering power but is not seriously influenced by substrate temperature. All the IWO thin film samples have high transmittance in near-infrared spectral range. With either the sputtering power or the growth temperature increases, the resistivity of the film decreases at the beginning and increases after the optimum parameters. The as-deposited IWO films with minimum resistivity of 6.4 10 4 cm were obtained at a growth temperature of225 C and sputteringpower of 40 W, with carrier mobility of 33.0 cm 2 V 1 s 1 and carrier concentration of 2.8 10 20 cm 3 and the average transmittance of about 81% in near-infrared region and about 87% in visible region.Li, Yuan Wang, Wenwen Zhang, Junying Wang, Rongming 2012Rare Metals2012,31,2:9
15Genetic diversity,population structure,and relationships of apricot(Prunus)based on restriction site-associated DNA sequencing显示文摘Single-nucleotide polymorphisms(SNPs)are the most abundant form of genomic polymorphisms and are widely used in population genetics research.Here,high-throughput sequencing was used to examine the genome-level diversity,population structure,and relationships of apricot,which are important for germplasm conservation and molecular breeding.Restriction site-associated DNA sequencing(RAD-seq)was adopted to sequence 168 Prunus spp.accessions distributed in five ecological groups,including 74 accessions of cultivated Prunus armeniaca L.and 94 accessions of wild apricots(P.armeniaca L.and Prunus sibirica L.),which generated 417,961 high-quality SNPs.We used cluster,genetic structure,and principal component analyses to examine the genetic diversities and genetic relationships of the 168 accessions.The Dzhungar-Ili ecological group accessions showed the highest genetic diversity in terms of private allele number,observed heterozygosity,and nucleotide diversity.We speculate that the Central Asian ecological group accessions were domesticated from the Dzhungar-Ili ecological group accessions.The population structure and gene flow of the North China and European ecological group accessions suggested a genetic background of P.sibirica.We argue that the two groups should be considered hybrid swarms connected to P.sibirica by continuous and extensive gene flow.P.armeniaca originated in Northwest China(Ili Valley),subsequently spread throughout Central Asia,and eventually spread to Europe.In addition,selective sweep signatures in P.armeniaca during domestication from wild to cultivated apricots,combined with differentially expressed genes,underlie distinct fruit traits,including sugars,aromas,organic acids,and carotenoids.This study provides substantive and valuable genomic resources that will significantly advance apricot improvement and effective utilization.Wenwen Li Liqiang Liu Yanan Wang Qiuping Zhang Guoquan Fan Shikui Zhang Yatong Wang Kang Liao 2020Horticulture Research2020,7,1:9

17Genome sequencing and phylogenetic analysis of Banna virus(genus Seadornavirus,family Reoviridae)isolated from Culicoides显示文摘In an investigation of blood-sucking insects and arboviruses, a virus(YN12243) was isolated from Culicoides samples collected in the Sino-Burmese border region of Yunnan Province, China. The virus caused cytopathic effect(CPE) in C6/36 cells and passaged stably. Polyacrylamide gel analysis showed that the genome of YN12243 was composed of 12 segments of double-stranded RNA(dsRNA), with a distribution pattern of 6-6. The nucleotide and amino acid sequences of the coding region(1.12 segments) were17,803 bp and 5,925 amino acids in length, respectively. The phylogenetic analysis of VP1 protein(RdRp) revealed that YN12243 belonged to genus Seadornavirus of family Reoviridae, and further analysis indicated that YN12243 belongs to the Banna virus(BAV) genotype A2. Additionally, YN12243 was located in the same evolutionary cluster as BAV strains isolated from different mosquito species, suggesting that the BAV isolated from Culicoides does not have species barriers. These results indicate that Culicoides can also be a vector for BAV. 18BubR1 phosphorylates CENP-E as a switch enabling the transition from lateral association to end-on capture of spindle microtubules显示文摘Error-free mitosis depends on accurate chromosome attachment to spindle microtubules, powered congression of those chromosomes, their segregation in anaphase, and assembly of a spindle midzone at mitotic exit. The centromere-associated kinesin motor CENP-E, whose binding partner is BubR1, has been implicated in congression of misaligned chromosomes and the transition from lateral kinetochore-microtubule association to end-on capture. Although previously proposed to be a pseudokinase, here we report the structure of the kinase domain of Drosophila melanogaster BubR1, revealing its folding into a conformation predicted to be catalytically active. BubR1 is shown to be a bona fide kinase whose phosphorylation of CENP-E switches it from a laterally attached microtubule motor to a plus-end microtubule tip tracker. Computational modeling is used to identify bubristatin as a selective BubR1 kinase antagonist that targets the αN1 helix of N-terminal extension and αC helix of the BubR1 kinase domain. Inhibition of CENP-E phosphorylation is shown to prevent proper microtubule capture at kinetochores and, surprisingly, proper assembly of the central spindle at mitotic exit. Thus, BubR1-mediated CENP-E phosphorylation produces a temporal switch that enables transition from lateral to end-on microtubule capture and organization of microtubules into stable midzone arrays.Yuejia Huang Lin Lin Xing Liu Sheng Ye Phil Y.
