| 2 | Biofunctional magnesium coated Ti6Al4V scaffold enhances osteogenesis and angiogenesis in vitro and in vivo for orthopedic application显示文摘The insufficient osteogenesis and osseointegration of porous titanium based scaffold limit its further application.Early angiogenesis is important for scaffold survival.It is necessary to develop a multifunctional surface on titanium scaffold with both osteogenic and angiogenic properties.In this study,a biofunctional magnesium coating is deposited on porous Ti6Al4V scaffold.For osseointegration and osteogenesis analysis,in vitro studies reveal that magnesium-coated Ti6Al4V co-culture with MC3T3-E1 cells can improve cell proliferation,adhesion,extracellular matrix(ECM)mineralization and ALP activity compared with bare Ti6Al4V cocultivation.Additionally,MC3T3-E1 cells cultured with magnesium-coated Ti6Al4V show significantly higher osteogenesisrelated genes expression.In vivo studies including fluorochrome labeling,micro-computerized tomography and histological examination of magnesium-coated Ti6Al4V scaffold reveal that new bone regeneration is significantly increased in rabbits after implantation.For angiogenesis studies,magnesium-coated Ti6Al4V improve HUVECs proliferation,adhesion,tube formation,wound-healing and Transwell abilities.HUVECs cultured with magnesium-coated Ti6Al4V display significantly higher angiogenesis-related genes(HIF-1αand VEGF)expression.Microangiography analysis reveal that magnesium-coated Ti6Al4V scaffold can significantly enhance the blood vessel formation.This study enlarges the application scope of magnesium and provides an optional choice to the conventional porous Ti6Al4V scaffold with enhanced osteogenesis and angiogenesis for further orthopedic applications. | Peng Gao Bo Fan Xiaoming Yu Wenwen Liu Jie Wu Lei Shi Di Yang Lili Tan Peng Wan Yulin Hao Shujun Li Wentao Hou Ke Yang Xiaokang Li Zheng Guo | 2020 | Bioactive Materials2020,5,3: | 15 |

| 5 | Benefit of everolimus as a monotherapy for a refractory breast cancer patient bearing multiple genetic mutations in the PI3K/AKT/mTOR signaling pathway显示文摘A postmenopausal patient with a diagnosis of estrogen receptor(ER)(+), progesterone receptor(PR)(+), and human epidermal growth factor receptor-2(HER2)(-) breast cancer was reported. The patient refused surgery and was resistant to conventional chemotherapy regimens. Computed tomography and the circulating tumor cell test indicated that the patient's tumor burden increased rapidly even after several chemotherapy sessions. Multiple genetic aberrances in the phosphatidylinositol3-kinases(PI3 K) signaling pathway were detected via next-generation sequencing(NGS)-based liquid biopsy, including a p. G1007 R missense mutation in exon 21 of PIK3 CA(33.61%), a p.L70 fs frameshift mutation in exon 3 of phosphatase and tension homolog deleted on chromosome ten(PTEN)(49.14%), and a p. D1542 Y missense mutation in exon 32 of mammalian target of rapamycin(m TOR)(1.66%). Therefore, only the m TOR inhibitor everolimus was administered to the patient. Partial remission(PR) was observed after 2 months, and sustained stable disease(SD) was observed after a year and a half. Subsequent sequencing showed that the mutation ratio of PIK3 CA decreased to 4.17%, and that the PTEN and m TOR mutations disappeared, which revealed the significant curative effect of everolimus. We report the first case of successful monotherapy treatment using everolimus in a patient with advanced breast cancer bearing mutations in genes involved in the PI3 K/ARK/m TOR signaling pathway. | 9 | Memory stem T cells generated by Wnt signaling from blood of human renal clear cell carcinoma patients显示文摘Objective: Memory stem T cells(Tscm) have attracted attention because of their enhanced self-renewal, multipotent capacity, and anti-tumor capacities. However, little is known about Tscm in patients with renal clear cell carcinoma(RCC) and the role of Wnt signaling in these cells. We evaluated Tscm from RCC patients concerning their activation of Wnt signaling in vitro and explored the mechanism of preferential survival.Methods: Flow cytometry identified surface markers and cytokines produced from accumulated Tscm in the presence of the glycogen synthase kinase beta inhibitor TWS119. Apoptosis was evaluated after induction using tumor necrosis factor-alpha.Immunofluorescence and Western blot analyses were used to investigate the activation of the nuclear factor-kappa B(NF-КB)pathway.Results: RCC patients had a similar percentage of CD4^+ and CD8^+ Tscm as healthy donors. | 14 | Sediment transport in the Luanhe River delta:grain size trend analysis显示文摘Sediment grain size in the deltaic environment of the Luanhe River(LR),Liaoning,China,contains sediment transport pathway information useful in elucidating the shoreline change and fluvialmarine interaction.In this study,we utilized numerical partitioning of the sedimentary components and geostatistical grain size trend analysis(GSTA)to define the sediment transport pattern in the Luanhe River delta(LRD)and interpolated the sediment transport pattern using content changes of end numbers(EM).EM1(the mean grain size 7.12Ф,fine silt),EM2(2.37Ф,fine sand),and EM3(1.27Ф,medium sand)components were identified by the numerical partitioning by GSTA.Kriging interpolation method was used to interpolate the parameters of the grain size for the regular grid,and the interpolation radius was 0.015 decimal degree.We chose 0.09 decimal degree as the characteristic distance for GSTA in the semivariogram model using the geostatistical method.The FB(-)case(finer,better sorted and more negatively skewed)was adopted in GSTA for its satisfaction in the Global Moran’s I test.The result of the GSTA shows that the sediments in the south barriers(SBs)were transported to the southwest of the study area.The sediments in the north,in the SE direction of sediment transport trend from the river mouth,indicated that the sediments in the north of the study area were transported from the LR to the northern beaches,and to the south and east of the study area.The sediment transport trend that simplified by GSTA as the FB(-)case was approved by the content changes of sedimentary components(i.e.EM1,EM2,and EM3).In addition,the turbulent jet diffusion pattern indicated that the coarse sediments(EM3)were delivered by LR during the flood season,and the EM2 and EM1 were from wave and tide,respectively. | YU Xiaoxiao LI Tiegang GU Dongqi FENG Aiping LIU Shihao LI Ping XU Guoqiang YAN Wenwen ZHANG Zhiwei ZHU Zhengtao | 2019 | Journal of Oceanology and Limnology2019,37,3: | 3 |

| 18 | Newly self-administered two-step tool for screening cognitive function in an ageing Chinese population:an exploratory cross-sectional study显示文摘Background Early screening of cognitive function is critical to dementia treatment and care.However,traditional tests require face-to-face administration and are often limited by implementation costs and biases.Aims This study aimed to assess whether the Thoven Cognitive Self-Assessment(TCSA),a novel,innovative two-step touchscreen-based cognition assessment tool,could identify early cognitive impairment due to dementia in older adults.Methods The TCSA was administered to 61 healthy controls(HCs),46 participants with mild cognitive impairment(MCI)and 44 participants diagnosed with dementia recruited from Shanghai.Two outcome measures were generated from the TCSA test:the TCSA_(primary task)score and the TCSA_(secondary task)score.Results The total average scores in the control group for the TCSA_(primary task)and TCSA_(secondary task)were significantly higher than those in the MCI and dementia groups(TCSA_(primary task):HCs vs MCI group vs dementia group,8.58±1.76 vs 5.40±2.67 vs 2.74±2.11,F=75.40,p<0.001;TCSA_(secondary task):HCs vs MCI group vs dementia group,23.02±3.31 vs 17.95±4.93 vs 11.93±5.50,F=76.46,p<0.001).Moreover,receiver operating characteristic analysis showed that a score below 7.5 for the TCSA_(primary task)and a score below 22.5 for the TCSA_(secondary task)were indicators of MCI.Conclusions The TCSA appears to be efficacious for the detection of cognitive impairment in older adults.It demonstrates the potential for large-scale cognition screening in community service settings. | Jing Nie Yang Yang Yining Gao Wenwen Jiang Aisikeer Aidina Fei Sun Lucas R Prieto Jie Yu Kang Ju Lisheng Song Xia Li | 2023 | General Psychiatry2023,36,1: | 2 |
