| 1 | Retardant effect of dihydroartemisinin on ulcerative colitis in a JAK2/STAT3-dependent manner显示文摘Dihydroartemisinin(DHA)is a semi-synthetic derivative and the main active metabolite of artemisinin.The purpose of this study was to investigate the effect of DHA on the ulcerative colitis(UC)in both in vivo and in vitro models.Weight,survival rate,colon length,and Disease Activity Index score were used to evaluate the severity of colitis.Reverse transcription quantitative polymerase chain reaction and enzyme-linked immunosorbent assay were used to detect the expressions of cytokines interleukin(IL)-1,IL-1β,IL-4,IL-6,IL-10,IL-12,and tumor necrosis factor-α(TNF-α).The expressions of Janus kinase 2(JAK2)and signal transducer and activator of transcription 3(STAT3),and the phosphorylation of JAK2(p-JAK2)and STAT3(p-STAT3),were measured by western blot analysis.Western blot analysis and immunohistochemistry were used to detect the expressions of tight junction proteins.We found that the weights and colon lengths of mice in dextran sodium sulfate(DSS)+DHA group were significantly lower and longer than those in the DSS group,respectively.Compared with those in the DSS group,the expressions of IL-1β,IL-6,IL-17,and TNF-αin the DSS+DHA and DSS+5-aminosalicylic acid(5-ASA)groups were decreased,while the expressions of IL-4 and IL-10 were significantly upregulated.DHA largely increased the expressions of zonula occludens-1 and occludin.Western blot analysis and/or immunohistochemical staining analysis showed that the expressions of JAK2,STAT3,p-JAK2,and p-STAT3 in DSS+DHA and DSS+5-ASA groups were significantly lower than those in DSS group.DHA has a specific therapeutic effect on UC.The anti-inflammatory mechanism of DHA is related to the blockage of the JAK2/STAT3 signaling pathway.These findings provide evidence that DHA may be a useful drug and is expected to become a promising new treatment for human UC. | Mingrui Jiang Guangjun Zhong Yichao Zhu Liuhua Wang Yuzhe He Qiannan Sun Xiaoqing Wu Xiaolan You Sujun Gao Dong Tang Daorong Wang | 2021 | Acta Biochimica et Biophysica Sinica2021,53,9: | 6 |
| 2 | Virtual population pharmacokinetic using physiologically based pharmacokinetic model for evaluating bioequivalence of oral lacidipine formulations in dogs显示文摘The aim of the present study was to investigate virtual population pharmacokinetic using physiologically based pharmacokinetic(PBPK) model for evaluating bioequivalence of oral lacidipine formulations in dogs. The dissolution behaviors of three lacidipine formulations including one commercial product and two self-made amorphous solid dispersions(ASDs)capsules were determined in 0.07% Tween 80 media. A randomized 3-period crossover design in 6 healthy beagle dogs after oral administration of the three formulations at a single dose of 4 mg was conducted. The PBPK modeling was utilized for the virtual bioequivalence study.In vitro dissolution experiment showed that the dissolution behaviors of lacidipine amorphous solid dispersions(ASDs) capsules, which was respectively prepared by HPMC-E5 or Soluplus, as polymer displayed similar curves compared with the reference formulation in 0.07% Tween 80 media. In vivo pharmacokinetics experiments showed that three formulations had comparable maximum plasma drug concentration(Cmax), and the time(Tmax) to reach Cmax of lacidipine tablet, which was prepared by Soluplus, as polymer was slower than other two formulations in consistency with the in vitro dissolution rate. The 90% confidence interval(CI) for the Cmax, AUC0–24 h and AUC0–∞ of the ratio of the test drug to the reference drug exceeded the acceptable bioequivalence(BE) limits(0.80–1.25). However, the 90% CI of the AUC0–24 h, AUC0–∞ and Cmax of the ratio of test to reference drug were within the BE limit,calculated using PBPK modeling when the virtual subjects reached 24 dogs. | 4 | Quasi-two-dimensionalβ-Ga2O3 field effect transistors with large drain current density and low contact resistance via controlled formation of interfacial oxygen vacancies显示文摘Quasi-two-dimensi on al(2D)β-Ga2O3 is a rediscovered metal-oxide semiconductor with an ultra-wide bandgap of 4.6-4.9eV.It has been reported to be a promising material for next-generation power and radio frequency electronics.Field effect transistors(FETs)that can switch at high voltage are key compone nts in power and radio frequency devices,and reliable Ohmic con tacts are essential for high FET performance.However,obtaining low contact resistance onβ-Ga2O3 FETs is difficult since reactions betweenβ-Ga2O3 and metal contacts are not fully understood.Herein,we experimentally demonstrate the importanee of reactions at the metal/β-Ga2O3 interface and the corresponding effects of these reactions on FET performance.When Ti is employed as the metal contact,annealing ofβ-Ga2O3 FETs in argon can effectively transform Schottky contacts into Ohmic contacts and permit a large drain current density of-3.1 mA//μm.The contact resistance(Rcontact)between the Ti electrodes andβ-Ga2O3 decreased from^430 to^0.387Ω·mm after annealing.X-ray photoelectron spectroscopy(XPS)confirmed the formation of oxygen vacancies at the Ti/β-Ga2O3 interface after annealing,which is believed to cause the improved FET performance.The results of this study pave the way for greater application ofβ-Ga2O3 in electronics. | Zhen Li Yihang Liu Anyi Zhang Qingzhou Liu Chenfei Shen Fanqi Wu Chi Xu Mingrui Chen Hongyu Fu Chongwu Zhou | 2019 | Nano Research2019,12,1: | 2 |
