1Development and transdifferentiation into inner hair cells require Tbx2显示文摘Atohl is essential for the development of both outer hair cells(OHCs)and inner hair cells(IHCs)in the mammalian co chlea.Whereas Ikzf2 is necessary for OHC development,the key gene re quired for IHC development remains unknown.We found that deletion of Tbx2 in neonatal IHCs led to their transdifferentiation into OHCs by repressing 26.7%of IHC genes and inducing 56.3%of OHC genes,including Ikzf2.More importantly,persistent expression of Tbx2 coupled with transient Atohl expression effe ctively reprogrammed non-sensory supporting cells into new IHCs expressing the functional IHC marker vGlut3.The differentiation status of these new IHCs was considerably more advanced than that previously reported.Thus,Tbx2 is essential for IHC development and co-upregulation of Tbx2 with Atohl in supporting cells represents a new approach for treating deafness related to IHC degeneration.Zhenghong Bi Xiang Li Minhui Ren Yunpeng Gu Tong Zhu Shuting Li Guangqin Wang Suhong Sun Yuwei Sun Zhiyong Liu 2022National Science Review2022,9,12:1
2Asian,regional,and national burdens of respiratory tract cancers and associated risk factors from 1990 to 2019:A systematic analysis for the global burden of disease study 2019显示文摘Background:Respiratory cancer is the leading cause of cancer-related deaths worldwide,but its statistics vary between the East and West.This study aimed to estimate the burdens of tracheal,bronchus,and lung(TBL)cancer and larynx cancer and their attributable risks from 1990 to 2019 in Asia,and at regional and national levels.Methods:This research evaluated the incidence,mortality,years lived with disability,years of life lost,and disability-adjusted life years(DALYs)for respiratory tract cancers using the Global Burden of Diseases,Injuries,and Risk Factors Study(GBD)2019 database.Age-standardized rates were calculated for TBL cancer from 1990 to 2019,adjusted for smoking and socio-demographic index(SDI).Deaths from TBL cancer and larynx cancer attributable to each risk factor were estimated for 33 Asian countries.Results:The age-standardized incidence and death rates for TBL cancer in Asia declined from 2010 to 2019,while the incidence rate of larynx cancer increased.Smoking was the leading specific risk factor for deaths from both TBL and larynx cancers.The burden of TBL cancer in Asian countries was influenced by SDI and smoking,particularly among males in Central Asia.Deaths,DALYs,and incidences of larynx cancer in East Asia had not changed significantly over the past 30 years,but showed slight downward trends in males and both sexes combined,and an upward trend in females in recent years.Conclusions:The past decade saw increases in numbers of incident cases and deaths from TBL cancer and larynx cancer in Asia.SDI and smoking were the main factors influencing the disease burden of TBL cancer in Asian countries.This study highlights the need for tailored cancer control programs to address the burden of respiratory tract cancers in different Asian countries.Ran Zhong Xiuyu Cai Jianfu Li Peiling Chen Runchen Wang Xinting Li Feng Li Shan Xiong Caichen Li Huiting Wang Yang Xiang Shuting Zhan Ziwen Yu Haixuan Wang Chunyan Li Lixuan Lin Wenhai Fu Xin Zheng Shen Zhao Kang Zhang Jianxing He 2023Chinese Medical Journal Pulmonary and Critical Care Medicine2023,1,4:1

4Water-dispersible nano-pollutions reshape microbial metabolism in type-specific manners:A metabolic and bacteriological investigation in Escherichia coli显示文摘Incomplete separation and recycling of nanoparticles are causing undesirable nanopollution and thus raising great concerns with regard to nanosafety.Since microorganisms are important regulator of physiological processes in many organisms,the interaction between nanopollution and microbial metabolomics and the resultant impact on the host’s health are important but unclear.To investigate how typical nanopollution perturbs microbial growth and metabolism,Escherichia coli(E.coli)in vitro was treated with six water-dispersible nanomaterials(nanoplastic,nanosilver,nano-TiO 2,nano-ZnO,semiconductor quantum dots(QDs),carbon dots(CDs))at human-/environment-relevant concentration levels.The nanomaterials exhibited type-specific toxic effects on E.coli growth.Global metabolite profiling was used to characterize metabolic disruption patterns in the model microorganism exposed to different nanopollutants.The percentage of significant metabolites(p<0.05,VIP>1)accounted for 6%–38%of the total 293 identified metabolites in each of the nanomaterial-contaminated bacterial groups.Metabolic results also exhibited significant differences between different nanopollutants and dose levels,revealing type-specific and untypical concentration-dependent metabolic responses.Key metabolites responsive to nanopollution exposures were mainly involved in amino acid and purine metabolisms,where 5,4,and 7 significant metabolic features were included in arginine and proline metabolism,phenylalanine metabolism,and purine metabolism,respectively.In conclusion,this study horizontally compared and demonstrated how typical nanopollution perturbs microbial growth and metabolomics in a type-specific manner,which broadens our understanding of the ecotoxicity of nanopollutants on microorganisms.Shuqin Liu Rui Wu Xi Yang Shuting Fang Zhangmin Xiang Shenghong Yang Gangfeng Ouyang 2022Frontiers of Environmental Science & Engineering2022,16,9:0
5Kinetics and capacities of non-reactive phosphorus (NRP) sorption to crushed autoclaved aerated concrete (CAAC)显示文摘With growing interest in resource recovery and/or reuse,wastematerials have been considered a promising alternative for phosphorus(P)adsorption because they are low-cost and easily accessible.Crushed autoclaved aerated concrete(CAAC),as representative construction waste,has been extensively studied for P removal in ecological technologies such as treatment wetlands.However,most of the previous studies focused on the adsorption of orthophosphate,namely reactive phosphorus,and lacked attention to non-reactive phosphorus(NRP)which is widely present in sewage.This study presents the first investigation on the potential and mechanism of CAAC removing four model NRP compounds.Adsorption isotherm and kinetics of NRP onto CAAC indicate that the removal of NRP was a chemisorption process and also involved a two-step pore diffusion process.The desorption experiment shows that different NRP species showed varying degrees of desorption.Most NRP was irreversibly adsorbed on CAAC.Among the model compounds considered in this study,the adsorption capacity and hydrolysis rate of organophosphorus were much less than that of inorganic phosphorus.Moreover,the adsorption of different NRP species by CAAC in the mesocosm study was different from the results of laboratory adsorption experiments,and the possible biodegradation was essential for the conversion and removal of NRP.The findings confirmed the validity of CAAC for NRP removal and the potential advantages of CAAC in terms of costs and environmental impact.This study will contribute to a better understanding of NRP conversion and environmental fate and that can be the basis for a refined risk assessment.Shuting Shen Xiang Li Zhuofan Geng Xiwu Lu 2023Journal of Environmental Sciences2023,,5:0

7Fgf8^(P2A-3×GFP/+):A New Genetic Mouse Model for Specifically Labeling and Sorting Cochlear Inner Hair Cells显示文摘The cochlear auditory epithelium contains two types of sound receptors,inner hair cells(IHCs)and outer hair cells(OHCs).Mouse models for labelling juvenile and adult IHCs or OHCs exist;however,labelling for embryonic and perinatal IHCs or OHCs are lacking.Here,we generated a new knock-in Fgf8^(P2A-3×GFP/+)(Fgf8^(GFP)/+)strain,in which the expression of a series of three GFP fragments is controlled by endogenous Fgf8 cis-regulatory elements.After confirming that GFP expression accurately reflects the expression of Fgf8,we successfully obtained both embryonic and neonatal IHCs with high purity,highlighting the power of Fgf8^(GFP)/+.Furthermore,our fate-mapping analysis revealed,unexpectedly,that IHCs are also derived from inner ear progenitors expressing Insm1,which is currently regarded as an OHC marker.Thus,besides serving as a highly favorable tool for sorting early IHCs,Fgf8^(GFP)/+will facilitate the isolation of pure early OHCs by excluding IHCs from the entire hair cell pool.Yi Pan Shuting Li Shunji He Guangqin Wang Chao Li Zhiyong Liu Mingliang Xiang 2023Neuroscience Bulletin2023,39,12:0
