| 1 | Comparison of Dryland Climate Change in Observations and CMIP5 Simulations显示文摘A comparison of observations with 20 climate model simulations from the Coupled Model Intercomparison Project, Phase5(CMIP5) revealed that observed dryland expansion amounted to 2.61 × 106km2 during the 58 years from 1948 to 2005,which was four times higher than that in the simulations(0.55 × 106km2). Dryland expansion was accompanied by a decline in aridity index(AI)(drying trend) as a result of decreased precipitation and increased potential evapotranspiration across all dryland subtype areas in the observations, especially in the semi-arid and dry subhumid regions. However, the CMIP5multi-model ensemble(MME) average performed poorly with regard to the decreasing trends of AI and precipitation. By analyzing the factors controlling AI, we found that the overall bias of AI in the simulations, compared with observations, was largely due to limitations in the simulation of precipitation. The simulated precipitation over global drylands was substantially overestimated compared with observations across all subtype areas, and the spatial distribution of precipitation in the MME was largely inconsistent in the African Sahel, East Asia, and eastern Australia, where the semi-arid and dry subhumid regions were mainly located. | JI Mingxia HUANG Jianping XIE Yongkun LIU Jun | 2015 | Advances in Atmospheric Sciences2015,32,11: | 5 |

By analyzing the DPSIR model and its correlation and relation with the evaluation index system, the indexes for geographic national conditions were selected from the aspects of resources, ecology, environment, economy, society, etc to build an evaluation index system for geographic national conditions that consists of three hierarchies: index, criterion, and target levels. | 8 | The human-derived novel gut commensal Luoshenia tenuis regulates body weight and food intake in mice显示文摘Many studies have revealed that gut microbes modulate host metabolism.In this study,we characterized the therapeutic effects of a novel gut commensal Luoshenia tenuis against host metabolic disorders.First,by in silico analysis,we demonstrated that the L.tenuis was prevalent in the gut microbiomes of healthy humans but were depleted specifically in obesity cohorts.Further in vitro cultivation revealed that L.tenuis produced short chain fatty acids that were verified to modulate host metabolism and some other volatile metabolites to benefit hosts by anti-inflammation and anti-tumor.Second,gavage of the L.tenuis significantly decreased the body weight gain and food intake of high-fat diet-feeding C57BL/6J mice,which was in parallel with the changed expression level of genes related to satiety and feeding behavior.We then performed the gavage trial using diet induced obese mice,and it revealed that the administration of L.tenuis alleviated significantly the abnormal glucose and lipid metabolisms and reduced the inflammatory response.In summary,this study revealed a previously-unknown human gut commensal microbe that benefited host metabolism,and set the stage for the development of novel next-generation probiotic applicable for treatment of obesity and related metabolic disorders. | Yu Jiang Mengxuan Du Lisheng Xie Minzhi Jiang Yaokun Zhang Mingxia Bi Chang Liu Hongwei Liu Shuangjiang Liu | 2024 | Food Science and Human Wellness2024,13,2: | 0 |
| 9 | Regulatory Effects of Zuogui Pill on Apoptosis of Follicles in Rats Injured by 60Co-γRays Based on PI3K/Akt/m TOR Signaling Pathway显示文摘[Objectives]To explore the protective effects of Zuogui Pill on ^(60)Co-γ-ray-induced premature aging of rats based on phosphatidylinositol 3-kinase/protein kinase B/mammalian target of rapamycin(PI3K/Akt/mTOR)signaling pathway.[Methods]Sixty sexually mature female SD rats were irradiated with ^(60)Co-γ-ray(6.0 Gy,LD 40)for 24 h at one time.These rats were randomly divided into model group,Progynova group[0.18(g·kg)/d],Progynova[0.09(g·kg)/d]+Zuogui Pill high dose[23.625(g·kg)/d)]group,Zuogui Pill high dose[23.625(g·kg)/d)]group,Zuogui Pill medium dose[9.45(g·kg)/d)]group and Zuogui Pill low dose[4.725(g·kg)/d]group.The administration(once a day)lasted 21 d.The rat serum[follicle-stimulating hormone(FSH),luteinizing hormone(LH)and estradiol(E_(2))]were detected by Enzyme-linked immunosorbent assay(ELISA).The morphological changes of ovary were observed by hematoxylin-eosin(HE)staining.The apoptosis rate of granulosa cells was detected by terminal deoxynucleotidyl transferase(TdT)-mediated dUTP nick-end labeling(TUNEL).The protein expression of phosphorylated(p)-PI3K,p-Akt,p-mTOR,B-cell lymphoma-2(Bcl-2),and Bcl-2-associated X protein(Bax)in ovarian tissues were detected by Western blot.[Results]Compared with the normal group,the model group showed significant increase in the serum FSH(P<0.01),significant decrease in serum E_(2)(P<0.05),and decrease in the number of early follicles and luteum in the ovary(P<0.01).Besides,the apoptosis rate of granulosa cells increased significantly(P<0.01);the expression of p-PI3K,p-Akt,p-mTOR and Bcl-2 in ovarian tissue decreased significantly,while the expression of Bax increased significantly(P<0.01).Compared with the model group,the number of early follicles in the ovary increased and the apoptosis rate of granulosa cells decreased after intervention in each administration group.In addition,the protein expressions of p-PI3K,p-Akt,p-mTOR and Bcl-2 increased,while the expression of Bax decreased,especially in Progynova+Zuogui Pill high dose group,the differences were statistically significant(P<0.05,P<0.01).[Conclusions]Zuogui Pill may protect the radiation-injured ovary through activating the expression of PI3K/Akt/mTOR protein in ovarian tissue,increasing the amount of Bcl-2 protein and inhibiting the expression of Bax protein. | Fenqin ZHAO Mingxia AN Xiaonan DING Jieying LIU Yan ZHAO Zhihui XIE Shuping LI | 2022 | Medicinal Plant2022,13,5: | 0 |
| 10 | Low glucose metabolite 3-phosphoglycerate switches PHGDH from serine synthesis to p53 activation to control cell fate显示文摘Glycolytic intermediary metabolites such as fructose-1,6-bisphosphate can serve as signals,controlling metabolic states beyond energy metabolism.However,whether glycolytic metabolites also play a role in controlling cell fate remains unexplored.Here,we find that low levels of glycolytic metabolite 3-phosphoglycerate(3-PGA)can switch phosphoglycerate dehydrogenase(PHGDH)from cataplerosis serine synthesis to pro-apoptotic activation of p53.PHGDH is a p53-binding protein,and when unoccupied by 3-PGA interacts with the scaffold protein AXIN in complex with the kinase HIPK2,both of which are also p53-binding proteins.This leads to the formation of a multivalent p53-binding complex that allows HIPK2 to specifically phosphorylate p53-Ser46 and thereby promote apoptosis.Furthermore,we show that PHGDH mutants(R135W and V261M)that are constitutively bound to 3-PGA abolish p53 activation even under low glucose conditions,while the mutants(T57A and T78A)unable to bind 3-PGA cause constitutive p53 activation and apoptosis in hepatocellular carcinoma(HCC)cells,even in the presence of high glucose.In vivo,PHGDH-T57A induces apoptosis and inhibits the growth of diethylnitrosamine-induced mouse HCC,whereas PHGDH-R135W prevents apoptosis and promotes HCC growth,and knockout of Trp53 abolishes these effects above.Importantly,caloric restriction that lowers whole-body glucose levels can impede HCC growth dependent on PHGDH.Together,these results unveil a mechanism by which glucose availability autonomously controls p53 activity,providing a new paradigm of cell fate control by metabolic substrate availability. | Yu-Qing Wu Chen-Song Zhang Jinye Xiong Dong-Qi Cai Chen-Zhe Wang Yu Wang Yan-Hui Liu Yu Wang Yiming Li Jian Wu Jianfeng Wu Bin Lan Xuefeng Wang Siwei Chen Xianglei Cao Xiaoyan Wei Hui-Hui Hu Huiling Guo Yaxin Yu Abdul Ghafoor Changchuan Xie Yaying Wu Zheni Xu Cixiong Zhang Mingxia Zhu Xi Huang Xiufeng Sun Shu-Yong Lin Hai-Long Piao Jianyin Zhou Sheng-Cai Lin | 2023 | Cell Research2023,33,11: | 0 |
| 11 | Generation and Developmental Characteristics of Porcine Tetraploid Embryos and Tetraploid/diploid Chimeric Embryos显示文摘The aim of this study was to optimize electrofusion conditions for generating porcine tetraploid(4n)embryos and produce tetraploid/diploid(4n/2n)chimeric embryos.Different electric feld intensities were tested and 2 direct current(DC)pulses of 0.9 kV/cm for 30 ls was selected as the optimum condition for electrofusion of 2-cell embryos to produce 4n embryos.The fusion rate of 2-cell embryos and the development rate to blastocyst of presumably 4n embryos,reached85.4%and 28.5%,respectively.68.18%of the fused embryos were found to be 4n as demonstrated by fluorescent in situ hybridization(FISH).Although the number of blastomeres in 4n blastocysts was signifcantly lower than in 2n blastocysts(P<0.05),there was no signifcant difference in developmental rates of blastocysts between 2n and 4n embryos(P>0.05),suggesting that the blastocyst forming capacity in 4n embryos is similar to those in 2n embryos.Moreover,4n/2n chimeric embryos were obtained by aggregation of 4n and 2n embryos.We found that the developmental rate and cell number of blastocysts of 4-cell(4n)/4-cell(2n)chimeric embryos were signifcantly higher than those of 2-cell(4n)/4-cell(2n),4-cell(4n)/8-cell(2n),4-cell(4n)/2-cell(2n)chimeric embryos(P<0.05).Consistent with mouse chimeras,the majority of 4n cells contribute to the trophectoderm(TE),while the 2n cells are mainly present in the inner cell mass(ICM)of porcine4n/2n chimeric embryos.Our study established a feasible and effcient approach to produce porcine4n embryos and 4n/2n chimeric embryos. | Wenteng He Qingran Kong Yongqian Shi Bingteng Xie Mingxia Jiao Tianqing Huang Shimeng Guo Kui Hu Zhonghua Liu | 2013 | Genomics, Proteomics & Bioinformatics2013,11,5: | 0 |
