| 1 | CCL5 deficiency promotes liver repair by improving inflammation resolution and liver regeneration through M2 macrophage polarization显示文摘Despite the diverse etiologies of drug-induced liver injury(DILI),innate immunity activation is a common feature involved in DILI progression.However,the involvement of innate immunity regulation in inflammation resolution and liver regeneration in DILI remains obscure.Herein,we identified the chemokine CCL5 as a central mediator of innate immunity regulation in the pathogenesis of DILI.First,we showed that serum and hepatic CCL5 levels are elevated in both DILI patients and an APAP-induced liver injury(AILI)mouse model.Interestingly,both nonparenchymal cells and stressed hepatocytes are cell sources of CCL5 induction in response to liver injury.Functional experiments showed that CCL5 deficiency has no effect on the early phase of AILI but promotes liver repair in the late phase mainly by promoting inflammation resolution and liver regeneration,which are associated with an increased number of hepatic M2 macrophages.Mechanistically,CCL5 can directly activate M1 polarization and impede M2 polarization through the CCR1-and CCR5-mediated activation of the MAPK and NF-κB pathways.We then showed that CCL5 inhibition mediated by either a CCL5-neutralizing antibody or the antagonist Met-CCL5 can greatly alleviate liver injury and improve survival in an AILI mouse model.Our data demonstrate CCL5 induction during DILI,identify CCL5 as a novel innate immunity regulator in macrophage polarization,and suggest that CCL5 blockage is a promising therapeutic strategy for the treatment of DILI. | Meng Li Xuehua Sun Jie Zhao Lei Xia Jichang Li Min Xu Bingrui Wang Han Guo Chang Yu Yueqiu Gao Hailong Wu Xiaoni Kong Qiang Xia | 2020 | Cellular & Molecular Immunology2020,17,7: | 13 |

| 9 | A general law of human mobility显示文摘The intrinsic factors that drive human mobility have remained unclear for decades. Our observations from both intra-urban and inter-urban trips demonstrate a general law of human mobility. Specifically, the probability that a trip will occur is inversely proportional to the size of population located inside a circle with radius equal to the travel distance centered at the trip origin. A simple parameterless rank-based model is presented; this model can predict human flows with a convincing fidelity. Moreover, existing models can be implemented as special cases of our model, suggesting that our model is stable at more spatial scales. | 14 | Emerging roles of DJ-1 in liver diseases through regulation of oxidative stress and immune response显示文摘Reactive oxygen species(ROS)and immune response play critical roles in the progression of liver diseases.DJ-1,also known as Parkinson disease 7(Park7),is extensively expressed in cells and tissues,where it governs numerous biological functions including chaperone activity,protease function,transcriptional and mitochondrial regulation,and ROS modulation.Moreover,we have established that DJ-1 plays a critical role in initiating an inflammatory response by modulating ROS generation.Therefore,DJ-1 may play an important role in the progression of liver diseases by modulating ROS and the immune response.Recently,we have shown that DJ-1 deficiency negatively regulates proliferation of hepatic progenitor cells(HPCs)by impairing the formation of HPC-associated fibrosis and inflammatory niches.Deficiency of DJ-1 ameliorates liver fibrosis by inhibiting hepatic ROS production and inflammation;moreover,in a classic diethylnitrosamine(DEN)-mediated hepatocellular carcinoma(HCC)mouse model,deletion of DJ-1 ameliorates tumorigenesis and HCC cell proliferation by regulating hepatic inflammation and reducing the activity of the interleukin 6/signal transducer and activator of transcription 3(IL-6/STAT3)signaling pathway.Taken together,these data suggest a critical function for,and therapeutic value of,DJ-1 in treatment of liver diseases.The aim of this review is to summarize these functions and the underlying molecular mechanisms of DJ-1 in liver diseases,and to highlight the potential therapeutic value and future research direction of DJ-1 in liver diseases. | Min Xu Lili Chen Jichang Li Hailong Wu Qiang Xia Xiaoni Kong | 2018 | Liver Research2018,2,2: | 0 |
| 15 | Polar Sea Ice Identification and Classification Based on HY-2A/SCAT Data显示文摘In this paper,a Bayesian sea ice detection algorithm is first used based on the HY-2A/SCAT data,and a backpropagation(BP)neural network is used to classify the Arctic sea ice type.During the implementation of the Bayesian sea ice detection algorithm,linear sea ice model parameters and the backscatter variance suitable for HY-2A/SCAT were proposed.The sea ice extent obtained by the Bayesian sea ice detection algorithm was projected on a 12.5 km grid sea ice map and validated by the Advanced Microwave Scanning Radiometer 2(AMSR2)15%sea ice concentration data.The sea ice extent obtained by the Bayesian sea ice detection al-gorithm was found to be in good agreement with that of the AMSR2 during the ice growth season.Meanwhile,the Bayesian sea ice detection algorithm gave a wider ice edge than the AMSR2 during the ice melting season.For the sea ice type classification,the BP neural network was used to classify the Arctic sea ice type(multi-year and first-year ice)from January to May and October to De-cember in 2014.Comparison results between the HY-2A/SCAT sea ice type and Equal-Area Scalable Earth Grid(EASE-Grid)sea ice age data showed that the HY-2A/SCAT multi-year ice extent variation had the same trend as the EASE-Grid data.Classification errors,defined as the ratio of the mismatched sea ice type points between HY-2A/SCAT and EASE-Grid to the total sea ice points,were less than 12%,and the average classification error was 8.6%for the study period,which indicated that the BP neural network classification was a feasible algorithm for HY-2A/SCAT sea ice type classification. | XU Rui ZHAO Chaofang ZHAI Xiaochun ZHAO Ke SHEN Jichang CHEN Ge | 2022 | Journal of Ocean University of China2022,21,2: | 0 |

| 19 | Study on ELISA for detection of sulfamethazine residues显示文摘In this study,various factors of ELISA for detection of sulfamethazine residues were explored,the coating antigen was diluted to 1:400,the best coating condition was at 4℃overnight,the working concentration of HRP-IgG enzyme conjugate was 1:7 000.The pre-incubation time and incubation time was 30 min and 120 min,respectively,the substrate solution working time was 20 min.Two moL·L^(-1) H_2SO_4 was used to stop the reaction and checked.A standard curve of direct competitive ELISA had been established to detect the sulfamethazine residues in milk.The detection limit of this method was 1.97 ng·mL^(-1).The mean concentration of sulfamethazine required to inhibit 30% antibody was 7.1 ng·mL^(-1).The linear range of the detection was 5-200 ng·mL^(-1).
