1The Interactions among DWARF10,Auxin and Cytokinin Underlie Lateral Bud Outgrowth in Rice显示文摘Previous studies have shown that DWARF10(D10) is a rice ortholog of MAX4/RMS1/DAD1,encoding a carotenoid cleavage dioxygenase and functioning in strigolactones/strigolactone-derivatives(SL) biosynthesis.Here we use D10-RNA interference(RNAi) transgenic plants similar to d10 mutant in phenotypes to investigate the interactions among D10,auxin and cytokinin in regulating rice shoot branching.Auxin levels in node 1 of both decapitated D10-RNAi and wild type plants decreased significantly,showing that decapitation does reduce endogenous auxin concentration,but decapitation has no clear effects on auxin levels in node 2 of the same plants.This implies that node 1 may be the location where a possible interaction between auxin and D10 gene would be detected.D10 expression in node 1 is inhibited by decapitation,and this inhibition can be restored by exogenous auxin application,indicating that D10 may play an important role in auxin regulation of SL.The decreased expression of most OsPINs in shoot nodes of D10-RNAi plants may cause a reduced auxin transport capacity.Furthermore,effects of auxin treatment of decapitated plants on the expression of cytokinin biosynthetic genes suggest that D10 promotes cytokinin biosynthesis by reducing auxin levels.Besides,in D10-RNAi plants,decreased storage cytokinin levels in the shoot node may partly account for the increased active cytokinin contents,resulting in more tillering phenotypes.Shuying Zhang Gang Li Jun Fang Weiqi Chen Haipai Jiang Junhuang Zou Xue Liu Xianfeng Zhao Xiaobing Li Chengcai Chu Qi Xie Xiangning Jiang Lihuang Zhu 2010Journal of Integrative Plant Biology2010,52,7:14

3Quantitative Analysis of Cytokinins in Plants by High Performance Liquid Chromatography:Electronspray Ionization Ion Trap Mass Spectrometry显示文摘The present paper introduces a highly sensitive and selective method for simultaneous quantification of 12 cytokinins(free form and their conjugates).The method includes a protocol of extraction with methanol/water/formic acid(15/4/1,v/v/v) to the micro-scale samples,pre-purification with solid phase extraction(SPE) cartridges of the extracts,separation with a high performance liquid chromatography(HPLC) and detection by an electrospray ionization ion trap mass spectrometry(ESI-Ion trap-MS) system in a consecutive ion monitoring(CRM) mode at the three stage fragmentation of mass spectrometry(MS 3).The lowest detection level of the cytokinins of the method reaches 0.1-2.0 pg with a very wide range of linear regression from 1-512 pg,at the coefficient factors of 0.98-0.99.The feasibility of this method has been proven in the application of the method to the analysis of the trace-amount contents of cytokinins in the micro-scale samples of various types of plant materials,such as aerial parts of rice and poplar leaves etc.12 endogenous cytokinins had been identified and quantified in the plant tissues,with an acceptable relatively higher recovery rate from 40% to 70%.Weiqi Chen Ying Gai Shichang Liu Renxiao Wang Xiangning Jiang 2010Journal of Integrative Plant Biology2010,52,10:4

5Construction and analysis of a plant transformation binary vector pBDGG harboring a bi-directional promoter fusing dual visible reporter genes显示文摘The constitutive promoter of cauliflower mosaic virus 35S (CaMV 35S) is a polar unidirectional promoter and is widely used in plant genetic engineering. In the present study, the unidirectional CaMV 35S promoter has been modified to a bi-directional promoter by fusing its minimal promoter element to the 5′end of CaMV 35S promoter in the opposite orientation. To qualitatively and quantitatively esti-mate its bi-directional transcriptional function and activity, two visible reporter genes, gusA (β-glucuronidase, GUS) and gfp (green fluo-rescent protein, GFP), were fused to the two ends of the promoter in bi-orientations ending with NOS terminator sequences, respectively. Stable expression of gusA and gfp genes in transgenic tobacco (Nicotiana tabacum L.) was visulized by histochemically staining for GUS and fluorescence microscopic observation under UV for GFP in transgenic plants. The expression of two reporter genes showed that the constructed bi-directional promoter did have the bi-directional transcriptional function in both expected orientations. The quantitative estimation of GUS and GFP were determined on a HITACHI F1000 Fluorescence Spectrophotometer with various wavelengths of excita-tion and emission. The GUS activity varied from 8 to 250 pmol 4-MU / min / mg protein and the GFP content varied from 0.9 to 1.8 μg / mg protein in various lines of transgenic tobacco plants. 6THEMIS observations of two substorms on February 26,2008显示文摘Two substorms occurred at ~04:05 and ~04:55 UT on February 26,2008 are studied with the in-situ observations of THEMIS satellites and ground-based aurora and magnetic field measurements.Angelopoulos et al.have made a comprehensive study of the 04:55 UT event.We showed detailed features of the two substorms with much attention to the first event and to the rela-tionship between mid-tail magnetic reconnection(MR) and substorm activities.It was found that in the earlier stage of each substorm,a first auroral intensification occurred 2-3 min soon after the start of mid-tail MR,followed by a slow and very lim-ited expansion.The auroral arcs were weak,short-lived,and localized,characterizing all features of a pseudobreakup.We re-garded the first auroral brightening as the initial onset of the substorms.A few minutes later,a second stronger auroral intensification appeared,followed by quick and extensive expansions.It was interesting to note that the second brightening and related poleward expansion happened almost simultaneously(within a couple of minutes) with the onset of earthward flow and dipolarization in the near-Earth tail and other phenomenon of the substorm expansion phase.We thus regarded the second auroral brightening as the major onset of the substorms.Furthermore,it was seen that during the growth phase of the two substorms,the polar cap open flux Ψ kept increasing,while it quickly reduced during the substorm expansion and recovery phase.These variations of Ψ implied that the evolution of the two substorm expansion phases were closely related to MR of tail lobe open field lines.Analysis of substorm activities revealed that the two events studied were small substorms;while estimate of MR rate indicated that the MR processes in the two substorms were weak.The aforementioned observations suggested that mid-tail MR initiated the pseudobreakup first;the earthward flow generated by MR transported magnetic flux and energy to the near-Earth tail to cause the formation of SCW and CD,which induced near-Earth dipolarization and major auroral brightening,and eventually led to the onset of the substorm expansion phase.These results were clearly consistent with the picture of NENL and RCS models and supported the two step initiation scenario of substorms.CHU XiangNing PU ZuYin CAO Xin WANG Jue V.MISHIN V.ANGELOPOULOS LIU Jiang WEI Yong K.H.GLASSMEIER J.MCFADDEN D.LARSON S.MENDE H.FREY C.T.RUSSELL I.MANN D.SIBECK ZONG QiuGang FU SuiYan XIE Lun T.I.SAIFUDINOVA M.V.TOLOCHKO L.A.SAPRONOVA H.REME E.LUCEK 2010Science China(Technological Sciences)2010,53,5:3
7Molecular and physiological characterization of the effects of auxin-enriched rootstock on grafting显示文摘Grafting is a highly useful technique,and its success largely depends on graft union formation.In this study,we found that root-specific expression of the auxin biosynthetic gene iaaM in tobacco,when used as rootstock,resulted in more rapid callus formation and faster graft healing.However,overexpression of the auxin-inactivating iaaL gene in rootstocks delayed graft healing.We observed increased endogenous auxin levels and auxin-responsive DR5::GUS expression in scions of WT/iaaM grafts compared with those found in WT/WT grafts,which suggested that auxin is transported upward from rootstock to scion tissues.A transcriptome analysis showed that auxin enhanced graft union formation through increases in the expression of genes involved in graft healing in both rootstock and scion tissues.We also observed that the ethylene biosynthetic gene ACS1 and the ethylene-responsive gene ERF5 were upregulated in both scions and rootstocks of the WT/iaaM grafts.Furthermore,exogenous applications of the ethylene precursor ACC to the junction of WT/WT grafts promoted graft union formation,whereas application of the ethylene biosynthesis inhibitor AVG delayed graft healing in WT/WT grafts,and the observed delay was less pronounced in the WT/iaaM grafts.These results demonstrated that elevated auxin levels in the iaaM rootstock in combination with the increased auxin levels in scions caused by upward transport/diffusion enhanced graft union formation and that ethylene was partially responsible for the effects of auxin on grafting.Our findings showed that grafting success can be enhanced by increasing the auxin levels in rootstocks using transgenic or gene-editing techniques.Longmei Zhai Xiaomin Wang Dan Tang Qi Qi Huseyin Yer Xiangning Jiang Zhenhai Han Richard McAvoy Wei Li Yi Li 2021Horticulture Research2021,8,1:3
8Whole-Brain Connectome of GABAergic Neurons in the Mouse Zona Incerta显示文摘The zona incerta(ZI)is involved in various functions and may serve as an integrative node of the circuits for global behavioral modulation.However,the long-range connectivity of different sectors in the mouse ZI has not been comprehensively mapped.Here,we obtained whole-brain images of the input and output connections via fluorescence micro-optical sectioning tomography and viral tracing.The principal regions in the input-output circuits of ZI GABAergic neurons were topologically organized.The 3D distribution of cortical inputs showed rostro-caudal correspondence with different ZI sectors,while the projection fibers from ZI sectors were longitudinally organized in the superior colliculus.Clustering results show that the medial and lateral ZI are two different major functional compartments,and they can be further divided into more subdomains based on projection and input connectivity.This study provides a comprehensive anatomical foundation for understanding how the ZI is involved in integrating different information,conveying motivational states,and modulating global behaviors.Yang Yang Tao Jiang Xueyan Jia Jing Yuan Xiangning Li Hui Gong 2022Neuroscience Bulletin2022,38,11:3

12Pressure gradient evolution in the near-Earth magnetotail at the arrival of BBFs显示文摘Using in situ observations from THEMIS A, D and E during the 2008–2011 tail season, we present a statistical study of the evolution of pressure gradients in the near-Earth tail during bursty bulk flow(BBF) convection.We identified 138 substorm BBFs and 2,197 non-substorm BBFs for this study. We found that both the pressure and the BZcomponent of the magnetic field were enhanced at the arrival of BBFs at the spacecraft locations. We suggest that the increase of BZduring non-substorm BBFs is associated with flux pile-up. However, the much stronger enhancement of BZduring substorm BBFs implies the occurrence of magnetic field dipolarization which is caused by both the flux pile-up process and near-Earth current disruption. Furthermore, a bow-wave-like high pressure appears to be formed at the arrival of substorm BBFs,which is responsible for the formation of region-1-sense FACs. The azimuthal pressure gradient associated with the arrival of substorm BBFs lasts for about 5 min. The enhanced pressure gradient associated with the bow waveis caused by the braking and diversion of the Earthward flow in the inner plasma sheet. 13Whole-Brain Direct Inputs to and Axonal Projections from Excitatory and Inhibitory Neurons in the Mouse Primary Auditory Area显示文摘Neurons in the primary auditory area(AUDp)innervate multiple brain regions with long-range projections while receiving informative inputs for diverse functions.However,the brain-wide connections of these neurons have not been comprehensively investigated.Here,we simultaneously applied virus-based anterograde and retrograde tracing,labeled the connections of excitatory and inhibitory neurons in the mouse AUDp,and acquired whole-brain information using a dual-channel fuorescence micro-optical sectioning tomography system.Quantified results showed that the two types of neurons received inputs with similar patterns but sent heterogeneous projections to downstream regions.In the isocortex,functionally different areas consistently sent feedback-dominated projections to these neurons,with concomitant laterallydominated projections from the sensory and limbic cortices to inhibitory neurons.In subcortical regions,the dorsal and medial parts of the non-lemniscal auditory thalamus(AT)were reciprocally connected to the AUDp,while the ventral part contained the most fibers of passage from the excitatory neurons and barely sent projections back,indicating the regional heterogeneity of the AUDp-AT circuit.Our results reveal details of the whole-brain network and provide new insights for further physiological and functional studies of the AUDp.Mengting Zhao Miao Ren Tao Jiang Xueyan Jia Xiaojun Wang Anan Li Xiangning Li Qingming Luo Hui Gong 2022Neuroscience Bulletin2022,38,6:1

16The modification of titanium in mesoporous silica for Co-based Fischer–Tropsch catalysts显示文摘Ordered SBA-15 mesoporous silica with incorporated titanium was successfully synthesized via a onepot hydrothermal crystallization method.The characterization including powder X-ray diffraction,Brunauer–Emmett–Teller,transmission electron microscope,temperatureprogrammed reduction,temperature-programmed desorption,Fourier transform infrared and ultraviolet-visible-near infrared spectrometer was performed to explore the physical and chemical structures of both the supports and the catalysts.The results showed that titanium was successfully incorporated into the mesoporous silica framework with a limited amount of titanium(Si/Ti>20),and the mesoporous structure was retained.However,the increased titanium content inevitably resulted in the formation of anatase TiO2 particles on the support surface.The increased incorporated titanium strengthened the interactions between cobalt species and supports,which was favorable for the cobalt species dispersion,despite the limited cobalt oxide reducibility.The enhanced metalsupport interactions were beneficial for the CO/H2 ratio at the active cobalt sites,which facilitated the formation of more C5+hydrocarbons.This study provides a promising method for support modification with incorporatedheteroatoms for the rational development of Fischer–Tropsch catalysts.Xin Li Meng Su Yao Chen Mehar UNisa Ning Zhao Xiangning Jiang Zhenhua Li 2022Frontiers of Chemical Science and Engineering2022,16,8:0
17Quantitative assessment of rhodamine spectra显示文摘With the development of single-molecule detection and super-resolution fluorescence imaging,rhodamine dyes gain new life.Through the modification of the N-substituents a nd the replacement of the oxygen atom in xanthene,the wavelength and brightness can be effectively changed.However,the spectra of rhodamine,especially due to the balance between ring-closed non-fluorescent lactone and ring-opened fluorescent zwitterion/cation,are sensitive to interference from various environmental facto rs,In this way,the spectral data of various rhodamines repo rted by different re search groups under different test conditions lacked comparability,sometimes even lacked accuracy.In order to meet the requirements for the accuracy and uniformity of spectral data in the research of single molecule imaging and dye structure-fluorescence relationship study,we have tested the spectra of fifteen rhodamine dyes that cover the visible and near-infrared regions under exactly the same conditions.By studying the dependence of the spectra on dye concentrations,it was confirmed that 1 μmol/L was ideal fo r detection less from the interference of dye molecule aggregation.We provide compre hensive and reliable spectral data of these fifteen dyes,which are expected to be used as references for future research.And the direct comparison of different rhodamine spectra would help to understand the structure-fluorescence relationship of rhodamines.Wei Zhou Xiangning Fang Qinglong Qiao Wenchao Jiang Yue Zhang Zhaochao Xu 2021Chinese Chemical Letters2021,32,2:0
18Cloning and characterization of geranylgeranyl diphosphate synthetase from Pinus massoniana and its correlation with resin productivity显示文摘Geranylgeranyl pyrophosphate synthetase(GGPPS) has gained increasing attention as a key enzyme in terpene analysis.We designed specific primers based on plant GGPPS homologs and used reverse transcription polymerase chain reaction(RT-PCR) to obtain and identify Pin GGPPS,a GGPPS gene sequence from Pinus massoniana,using bioinformatics tools.Quantitative PCR analysis of Pin GGPPS expression levels in roots,pine needles,immature stems,and semilignified stems from 6-month-old P.massoniana showed that expression levels of Pin GGPPS were highest in pine needles,followed by immature stems and semilignified stems,and lowest in roots.When we examined the correlation between Pin GGPPS gene expression levels and resin productivity in 20 adult plants for 28 successive days,Pin GGPPS expression levels presented a substantially linear distribution when plotted against their corresponding resin yields.In summary,we characterized the gene Pin GGPPS for the first time in P.massoniana,and established a correlation between Pin GGPPS gene expression levels and resin productivity,suggesting the importance of theory and production practice for P.massoniana.Bowen Chen Yufei Xiao Junji Li Hailong Liu Hu Chen Jie Jia Nan Chao Ying Gai Xiangning Jiang 2018Journal of Forestry Research2018,29,2:0
