1OsWRKY03, a rice transcriptional activator that functions in defense signaling pathway upstream of OsNPR1显示文摘WRKY family proteins are a class of plant specific transcription factors that involve in many stress response pathways. It has been shown that one Arabidopsis WRKY protein, AtWRKY29/22, is activated by MAP kinase signaling cascade and confers resistance to both bacterial and fungal pathogens. However, little is known about the biological roles of WRKY proteins in rice. In this study, we investigated the expression patterns of rice AtWRKY29/22 homolog, OsWRKY03, under different conditions, and also its possible role involved in plant defense. Our results showed that OsWRKY03 was up-regulated by several defense signaling molecules or different treatments. Further analysis revealed that the expres- sion of OsWRKY03 was light dependent. Transcriptional activation activity of OsWRKY03 was also demonstrated by yeast functional assay. Transient expression of OsWRKY03-GFP fusion protein in onion epidermis cells showed that OsWRKY03 was a nuclear localized protein. OsNPR1 as well as several other pathogenesis-related genes, such as OsPR1b, phenylalanine ammonia-lyase (ZB8) and peroxidase (POX22.3), were induced in OsWRKY03-overexpressing transgenic plants. 4Deep structure beneath the southwestern section of the Longmenshan fault zone and seimogenetic context of the 4.20 Lushan M_S7.0 earthquake显示文摘Magnetotelluric measurements were carried out along two profiles across the middle and southwestern sections of the Longmenshan fault zone(LMSf)from 2009 to 2011,after the 2008 Wenchuan MW7.9 earthquake.The former profile crosses the Wenchuan event epicenter and the latter one crosses 2013 Lushan MS7.0 event epicenter.The data were analyzed using advanced processing techniques,including phase tensor and two-dimensional inversion methods,in order to obtain reliable 2-D profiles of the electrical structure in the vicinity of the two earthquakes.A comparison of the two profiles indicates both similarities and differences in the deep crustal structure of the LMSf.West of the southwestern section,a crustal high conductivity layer(HCL)is present at about 10 km depth below the Songpan-Garzêblock;this is about 10 km shallower than that under the middle section of the LMSf.A high resistivity body(HRB)is observed beneath the southwestern section,extending from the near surface to the top of upper mantle.It has a smaller size than the HRB observed below the middle section.In the middle section,there is a local area of decreased resistivity within the HRB but there is absence of this area.The 2013 Lushan earthquake occurred close to the eastern boundary of HRB and the Shuangshi-Dachuan fault,of which the seismogenic context has both common and different features in comparison with the 2008 Wenchuan event.On a large scale,the 2013 Lushan earthquake is associated with the HCL and deformation in the crust including HCL of the eastern Tibetan Plateau.In order to assess seismic risk,it is important to consider both the stress state and the detailed crustal structure in different parts of the LMSf.ZHAN Yan ZHAO GuoZe UNSWORTH Martyn WANG LiFeng CHEN XiaoBin LI Tao XIAO QiBin WANG JiJun TANG Ji CAI JunTao WANG YanZhao 2013Chinese Science Bulletin2013,58,28:39
5Early Paleozoic ridge subduction in the Chinese Altai: Insight from the abrupt change in zircon Hf isotopic compositions显示文摘Zircons were separated from granitoids, gneisses, and sedimentary rocks of the Chinese Altai. Those with igneous characteristics yielded U-Pb ages of 280-2800 Ma, recording a long history of magmatic activity in the region. Zircon Hf isotopic compositions show an abrupt change at ~420 Ma, indicating that prior to that time the magmas came from both ancient and juvenile sources, whereas younger magmas were derived mainly from juvenile material. This may imply that the lithosphere was signifi- cantly modified in composition by a rapid addition of melt from the mantle. 6Hydrogen peroxide-induced changes in intracellular pH of guard cells precede stomatal closure显示文摘Epidermal bioassay demonstrated that benzylamine, a membrane-permeable weak base, can mimick hydrogen peroxide (H2O2) to induce stomatal closure, and butyric acid, a membrane-permeable weak acid, can partly abolish the H2O2-induced stomatal closure. Confocal pH mapping with the probe 5-(and-6)- carboxy seminaphthorhodafluor- 1 - acetoxymethylester (SNARF-1-AM) revealed that H2O2 leads to rapid changes in cytoplasmic and vacuolar pH in guard cells of Viola faba L, i. e. alkalinization of cytoplasmic areas occur red in parallel with a decrease of the vacuolar pH, and that butyric acid pretreatment can abolish alkalinization of cytoplasmic areas and acidification of vacuolar areas of guard cells challenged with H2O2. 12The Dihedral Angle and Intersection Processes of a Conjugate Strike-Slip Fault System in the Tarim Basin, NW China显示文摘Recent studies, focused on dihedral angles and intersection processes, have increased understandings of conjugate fault mechanisms. We present new 3-D seismic data and microstructural core analysis in a case study of a large conjugate strike-slip fault system from the intracratonic Tarim Basin, NW China. Within our study area, 'X' type NE and NW trending faults occur within CambrianOrdovician carbonates. The dihedral angles of these conjugate faults have narrow ranges, 19° to 62° in the Cambrian and 26° to 51° in the Ordovician, and their modes are 42° and 44° respectively. These data are significantly different from the ~60° predicted by the Coulomb fracture criterion. It is concluded that:(1) The dihedral angles of the conjugate faults were not controlled by confining pressure, which was low and associated with shallow burial;(2) As dihedral angles were not controlled by pressure they can be used to determine the shortening direction during faulting;(3) Sequential slip may have played an important role in forming conjugate fault intersections;(4) The conjugate fault system of the Tarim basin initiated as rhombic joints; these subsequently developed into sequentially active 'X' type conjugate faults; followed by preferential development of the NW-trending faults; then reactivation of the NE trending faults. 13Survival of transplanted neurotrophin-3 expressing human neural stem cells and motor function in a rat model of spinal cord injury显示文摘<正>BACKGROUND:Many methods have been attempted to repair nerves following spinal cord injury, including peripheral nerve transplantation,Schwann cell transplantation,olfactory ensheathing cell transplantation,and embryonic neural tissue transplantation.However,there is a need for improved outcomes. OBJECTIVE:To investigate the repair feasibility for rat spinal cord injury using human neural stem cells(hNSCs) genetically modified by lentivirus to express neurotrophin-3. DESIGN,TIME AND SETTING:In vitro cell biological experiment and in vivo randomized,controlled, genetic engineering experiment were performed at the Third Military Medical University of Chinese PLA and First People's Hospital of Yibin,China from March 2006 to December 2007. MATERIALS:A total of 64 adult,female,Wistar rats were used for the in vivo study.Of them,48 rats were used to establish models of spinal cord hemisection,and were subsequently equally and randomly assigned to model,genetically modified hNSC,and normal hNSC groups.The remaining 16 rats served as normal controls. METHODS:hNSCs were in vitro genetically modified by lentivirus to secrete both green fluorescence protein and neurotrophin-3.Neurotrophin-3 expression was measured by Western blot. Genetically modified hNSC or normal hNSC suspension(5×10~5) was injected into the rat spinal cord following T_(10) spinal cord hemisection.A total of 5μL Dulbecco's-modified Eagle's medium was infused into the rat spinal cord in the model group.Transgene expression and survival of transplanted hNSCs were determined by immunohistochemistry.Motor function was evaluated using the Basso,Beattie,and Bresnahan(BBB) scale. MAIN OUTCOME MEASURES:The following parameters were measured:expression of neurotrophin-3 produced by genetically modified hNSCs,transgene expression and survival of hNSCs in rats,motor function in rats. RESULTS:hNSCs were successfully genetically modified by lentivirus to stably express neurotrophin-3.The transplanted hNSCs primarily gathered at,or around,the injection site two weeks following transplantation,and gradually migrated towards the surrounding tissue. Transplanted hNSCs were observed 7.0-8.0 mm away from the injection site.In addition,hNSCs were observed 10 weeks after transplantation.At week 4,BBB locomotor scores were significantly greater in the genetically modified hNSC and normal hNSC groups,compared with the model group (P<0.05),and scores were significantly greater in the genetically modified hNSC group compared with the normal hNSC group(P<0.05). 15Generation of a monkey with MECP2 mutations by TALEN-based gene targeting显示文摘Gene editing in model organisms has provided critical insights into brain development and diseases. Here, we report the generation of a cynomolgus monkey(Macaca fascicularis) carrying MECP2 mutations using transcription activator-like effector nucleases(TALENs)-mediated gene targeting. After injecting TALENs mRNA into monkey zygotes achieved by in vitro fertilization and embryo transplantation into surrogate monkeys, we obtained one male newborn monkey with an MECP2 deletion caused by frameshifting mutation in various tissues. The monkey carrying the MECP2 mutation failed to survive after birth, due to either the toxicity of TALENs or the critical requirement of MECP2 for neural development. The level of MeCP2 protein was essentially depleted in the monkey's brain. 17Scutellarin protects oxygen/glucose-deprived astrocytes and reduces focal cerebral ischemic injury显示文摘Scutellarin, a bioactive flavone isolated from Scutellaria baicalensis, has anti-inflammatory, anti-neurotoxic, anti-apoptotic and anti-oxidative effects and has been used to treat cardiovascular and cerebrovascular diseases in China. However, the mechanisms by which scutellarin mediates neuroprotection in cerebral ischemia remain unclear. The interaction between scutellarin and nicotinamide adenine dinucleotide phosphate oxidase 2(NOX2) was assessed by molecular docking study, which showed that scutellarin selectively binds to NOX2 with high affinity. Cultures of primary astrocytes isolated from the cerebral cortex of neonatal Sprague-Dawley rats were pretreated with 2, 10 or 50 μM scutellarin for 30 minutes. The astrocytes were then subjected to oxygen/glucose deprivation by incubation for 2 hours in glucose-free Dulbecco's modified Eagle's medium in a 95% N2/5% CO_2 incubator, followed by simulated reperfusion for 22 hours. Cell viability was assessed by cell counting kit-8 assay. Expression levels of NOX2, connexin 43 and caspase-3 were assessed by western blot assay. Reactive oxygen species were measured spectrophotometrically. Pretreatment with 10 or 50 μM scutellarin substantially increased viability, reduced the expression of NOX2 and caspase-3, increased the expression of connexin 43, and diminished the levels of reactive oxygen species in astrocytes subjected to ischemia-reperfusion. We also assessed the effects of scutellarin in vivo in the rat transient middle cerebral artery occlusion model of cerebral ischemia-reperfusion injury. Rats were given intraperitoneal injection of 100 mg/kg scutellarin 2 hours before surgery. The Bederson scale was used to assess neurological deficit, and 2,3,5-triphenyltetrazolium chloride staining was used to measure infarct size. Western blot assay was used to assess expression of NOX2 and connexin 43 in brain tissue. Enzyme-linked immunosorbent assay was used to detect 8-hydroxydeoxyguanosine(8-OHd G), 4-hydroxy-2-nonenal(4-HNE) and 3-nitrotyrosin(3-NT) in brain tissue. Immunofluorescence double staining was used to determine the co-expression of caspase-3 and Neu N. Pretreatment with scutellarin improved the neurological function of rats with focal cerebral ischemia, reduced infarct size, diminished the expression of NOX2, reduced levels of 8-OHd G, 4-HNE and 3-NT, and reduced the number of cells co-expressing caspase-3 and Neu N in the injured brain tissue. Furthermore, we examined the effect of the NOX2 inhibitor apocynin. Apocynin substantially increased connexin 43 expression in vivo and in vitro. 18Relationship of Inflammation and Endothelial Dysfunction with Risks to Cardiovascular Disease among People in Inner Mongolia of China显示文摘Objective To explore the relationship of inflammation and endothelial dysfunction with risks to cardiovascular disease(CVD).Methods Blood pressure,body weight,body height,waist circumference and lifestyle risk factors were measured and studied among 2589 participants in Inner Mongolia of China,and biomarkers of inflammation and endothelial dysfunction including high-sensitivity C-reactive protein(hsCRP),soluble inter-cellular adhesion molecule-1(sICAM-1),soluble E-selectin(sE-selectin),and angiotensin II were investigated.Results Subjects with metabolic risk factors for CVD had higher levels of hsCRP,sE-selectin and sICAM-1 than those without such risk factors(all P<0.05).Levels of all biomarkers positively and significantly increased with aggregation of the metabolic risk factors among the subjects(all P for trend<0.001).Data from the multivariate analysis showed that participants with high levels of hsCRP [odds ratio(OR): 1.96,95% confidence interval(CI): 1.52-2.53],sE-selectin(OR: 1.35,95% CI: 1.05-1.72),and angiotensin II(OR: 1.81,95% CI: 1.40-2.33) were more likely to develop hypertension; participants with high levels of hsCRP(OR: 2.33,95% CI: 1.85-2.94),sE-selectin(OR: 1.24,95% CI: 1.00-1.54),and sICAM-1(OR: 1.70,95% CI: 1.30-2.22) were more likely to develop dyslipidemia,and those with high levels of hsCRP(OR: 2.95,95% CI: 2.27-3.83) and sICAM-1(OR: 2.80,95% CI: 2.06-3.80) were more likely to develop hyperglycemia.Conclusion Biomarkers of inflammation and endothelial dysfunction were separately associated with relevant metabolic risk factors for CVD.And appropriate measures should be taken to control inflammation and improve endothelial function among individuals with different metabolic risk factors for CVD.PENG Hao HAN Shu Hai LIU Hai Ying Vasisht CHANDNI CAI Xiao Qing ZHANG Yong Hong 2013Biomedical and Environmental Sciences2013,26,10:15

20Constrained large-eddy simulation and detached eddy simulation of flow past a commercial aircraft at 14 degrees angle of attack显示文摘With the development of computational power and numerical algorithms,computational fluid dynamics(CFD) has become an important strategy for the design of aircraft,which significantly reduces the reliance on wind-tunnel and flight tests.In this paper,we conducted a numerical investigation on the flow past a full commercial aircraft at Mach number 0.2 and 14 degrees angle of attack by means of Reynolds-averaged Navier-Stokes(RANS),detached-eddy simulation(DES) and our newly developed constrained large-eddy simulation(CLES).The objective of this paper is to study the capability of these models in simulating turbulent flows.To our knowledge,this is the first large-eddy simulation method for full commercial aircraft simulation.The results show that the CLES can predict the mean statistical quantities well,qualitatively consistent with traditional methods,and can capture more small-scale structures near the surface of the aircraft with massive separations.Our study demonstrates that CLES is a promising alternative for simulating real engineering turbulent flows.CHEN ShiYi CHEN YingChun XIA ZhenHua QU Kun SHI YiPeng XIAO ZuoLi LIU QiuHong CAI QingDong LIU Feng LEE Cunbiao ZHANG RiKui CAI JinSheng 2013Science China(Physics,Mechanics & Astronomy)2013,56,2:14
