1Adsorption of 17β-estradiol from aqueous solution by raw and direct/pre/post-KOH treated lotus seedpod biochar显示文摘Five biochars derived from lotus seedpod(LSP)were applied to examine and compare the adsorption capacity of 17β-estradiol(E2)from aqueous solution.The effect of KOH activation and the order of activation steps on material properties were discussed.The effect of contact time,initial concentration,p H,ionic strength and humic acid on E2 adsorption were investigated in a batch adsorption process.Experimental results demonstrated that the pseudo second-order model fitted the experimental data best and that adsorption equilibrium was reached within 20 hr.The efficiency of E2 removal increased with increasing E2 concentration and decreased with the increase of ionic strength.E2 adsorption on LSP-derived biochar(BCs)was influenced little by humic acid,and slightly affected by the solution p H when its value ranged from 4.0 to 9.0,but considerably affected at p H 10.0.Low environmental temperature is favorable for E2 adsorption.Chemisorption,π–πinteractions,monolayer adsorption and electrostatic interaction are the possible adsorption mechanisms.Comparative studies indicated that KOH activation and the order of activation steps had significant impacts on the material.Post-treated biochar exhibited better adsorption capacity for E2 than direct treated,pretreated,and raw LSP biochar.Pyrolyzed biochar at higher temperature improved E2 removal.The excellent performance of BCs in removing E2 suggested that BCs have potential in E2 treatment and that the biochar directly treated by KOH would be a good choice for the treatment of E2 in aqueous solution,with its advantages of good efficiency and simple technology.Ni Liu Yunguo Liu Guangming Zeng Jilai Gong Xiaofei Tan Jun Wen Shaobo Liu Luhua Jiang Meifang Li Zhihong Yin 2020Journal of Environmental Sciences2020,32,1:4
2Glucocorticoid receptor modulates myeloid-derived suppressor cell function via mitochondrial metabolism in immune thrombocytopenia显示文摘Myeloid-derived suppressor cells(MDSCs)are a heterogeneous population of immature cells and natural inhibitors of adaptive immunity.Intracellular metabolic changes in MDSCs exert a direct immunological influence on their suppressive activity.Our previous study demonstrated that high-dose dexamethasone(HD-DXM)corrected the functional impairment of MDSCs in immune thrombocytopenia(ITP);however,the MDSC population was not restored in nonresponders,and the mechanism remained unclear.In this study,altered mitochondrial physiology and reduced mitochondrial gene transcription were detected in MDSCs from HD-DXM nonresponders,accompanied by decreased levels of carnitine palmitoyltransferase-1(CPT-1),a rate-limiting enzyme in fatty acid oxidation(FAO).Blockade of FAO with a CPT-1 inhibitor abolished the immunosuppressive function of MDSCs in HD-DXM responders.We also report that MDSCs from ITP patients had lower expression of the glucocorticoid receptor(GR),which can translocate into mitochondria to regulate the transcription of mitochondrial DNA(mtDNA)as well as the level of oxidative phosphorylation.It was confirmed that the expression of CPT-1 and mtDNA-encoded genes was downregulated in GR-siRNA-treated murine MDSCs.Finally,by establishing murine models of active and passive ITP via adoptive transfer of DXM-modulated MDSCs,we confirmed that GR-silenced MDSCs failed to alleviate thrombocytopenia in mice with ITP.In conclusion,our study indicated that impaired aerobic metabolism in MDSCs participates in the pathogenesis of glucocorticoid resistance in ITP and that intact control of MDSC metabolism by GR contributes to the homeostatic regulation of immunosuppressive cell function.Yu Hou Jie Xie Shuwen Wang Daqi Li Lingjun Wang Haoyi Wang Xiaofei Ni Shaoqiu Leng Guosheng Li Ming Hou Jun Peng 2022Cellular & Molecular Immunology2022,19,7:4
3Hyaluronic acid-based dual-responsive nanomicelles mediated mutually synergistic photothermal and molecular targeting therapies显示文摘Precise clinical treatment of triple-negative breast cancer(TNBC)is an obstacle in clinic.Nanotechnology-assisted photothermal therapy(PTT)is a superior treatment modality for TNBC in terms of precision.However,thermoresistance arising from PTT and insufficient drug release from nanocarriers decrease the efficacy of PTT.AT13387 is a novel HSP90 inhibitor that can weaken thermoresistance and undergoing clinic II phase study,showing satisfactory antitumour activity through molecularly targeted therapy(MTT).Whereas,it has poor solubility.Hence hyaluronic acid and stearic acid were connected by hydrazone bonds and disulfide bonds,forming an amphipathic copolymer that could self-assembled into nanomicelles,followed by encapsulating Cypate and AT13387.These nanomicelles exhibited great features,including achieving mutually synergistic PTT/MTT for overcoming thermoresistance and promoting translocation of drugs,increasing the solubility of Cypate and AT13387,showing a pH/redox dual response that contributes to drug release,and having the ability of active targeting.Thus,the nanomicelles developed in this study may be a promising strategy for the precise treatment of TNBC.Liangliang Cai Ronghua Ni Xiaofei Ma Rongrong Huang Zhiyuan Tang Jinqiu Xu Yong Han Yuehua Guo Zhifeng Gu 2022Nano Research2022,15,7:1

7CD13 inhibition augments DR4-induced tumor cell death in a p-ERK1/2-independent manner显示文摘Objective:Death receptor 4(DR4;TRAIL-R1)critically mediates extrinsic apoptosis cascades via binding to TNF-related apoptosis-inducing ligand(TRAIL).However,intrinsic and/or acquired resistance are observed in the clinical application of TRAIL.The aim of this study was to investigate the function and molecular mechanism of CD13 in the TRAIL/DR4 pathway against tumor cells,and provide a new strategy for improving therapeutic efficacy or overcoming TRAIL-resistance.Methods:TRAIL protein was expressed as a secretory protein in a Pichia pastoris expression system and was isolated and purified by affinity chromatography.The cell viability and apoptosis were evaluated with MTT(thiazolyl blue tetrazolium bromide)assays and annexin V-FITC/PI staining with flow cytometry analysis,respectively.Western blot analysis was used to detect the levels of the indicated proteins in tumor cells.DR4 degradation or stability was examined with cycloheximide chase assays,and cell surface DR4 was assessed with flow cytometric analysis after staining with a FITC-conjugated antibody.The effects of cell migration were determined with Transwell and gelatin zymography assays.A xenograft nude mouse model was used to detect the anti-tumor effect in vivo,and the proliferation in tumor tissues was examined with immunohistochemical staining.Results:CD13 inhibition potently sensitized tumor cells to TRAIL-induced killing,including proliferation inhibition,increased apoptosis,and migration suppression.In addition,the inhibition of CD13 elevated both total cellular expression and cell surface DR4 through stabilizing DR4 by suppressing its degradation.DR4 si RNA attenuated the enhanced anti-tumor effects of TRAIL plus CD13 inhibition.Interestingly,these phenomena were p-ERK1/2 independent,although p-ERK1/2 down-regulation was tightly correlated with the cooperation of TRAIL and CD13 inhibition.Moreover,a synergistic decrease in tumor growth was surprisingly achieved in the xenograft model by treatment of TRAIL with a CD13 inhibitor(**P<0.01,CDI=0.47).Conclusions:CD13 inhibition cooperates with TRAIL in enhancing DR4-mediated cell death,through the up-regulation and stabilization of DR4 in a p-ERK1/2-independent manner.Thus CD13 inhibition has emerged as an effective strategy for TRAIL/DR4-based therapy.Jun Ni Xiaofei Wang Yue Shang Yi Li Shuzhen Chen 2021Cancer Biology & Medicine2021,18,2:1

12APPLICATION OF LAPAROSCOPIC ULTRASONOGRAPHY IN ADNEXAL OPERATIONS显示文摘Objective:To explore the clinical value on application of laparoscopic ultrasonography (Lap US) in the adnexal operation. 14SPATIOTEMPORAL VARIATION OF WATER QUALITY AND ALGAL BIOMASS IN ERHAI LAKE AND ITS ENVIRONMENTAL MANAGEMENT IMPLICATIONS显示文摘Elucidating the spatiotemporal pattern of water quality and algal biomass is crucial for accurately tracing pollution sources and reducing the risk of algal blooms in lake systems.This study analyzed the spatiotemporal variability of water quality and algal biomass in Erhai Lake from 1994 to 2021 using water quality index(WQi),Mann-Kendall test and Sen's slope combined methods.The potential causes of water quality deterioration and algal biomass dynamics were also elucidated.The results showed that the historical changes in the water environment of Erhai Lake mainly had three stages:satisfactory(1994-2001),deteriorating(2002-2010)and improving(2011-2021).The changes in water quality and algal biomass were primarily affected by total nitrogen,total phosphorus and chemical oxygen demand in different stages.The water environment of Erhai Lake is currently improving significantly,starting in the southern area that is furthest from the sources of agricultural pollution,especially in summer and autumn.This is attributed to the implementation of control measures resulting in lower pollutant loads at particular times and places.Therefore,it is necessary to continue to promote standardized livestock farming,to strengthen rural wastewater collection and to investigate measures such as the interruption of the endogenous cycle.Xiaofei LIU Yue WU Zhaokui NI Shengrui WANG 2023Frontiers of Agricultural Science and Engineering2023,10,4:0
15An Evaluation Method of Human Gut Microbial Homeostasis by Testing Specific Fecal Microbiota显示文摘Research on microecology has been carried out with broad perspectives in recent decades,which has enabled a better understanding of the gut microbiota and its roles in human health and disease.It is of great significance to routinely acquire the status of the human gut microbiota;however,there is no method to evaluate the gut microbiome through small amounts of fecal microbes.In this study,we found ten predominant groups of gut bacteria that characterized the whole microbiome in the human gut from a large-sample Chinese cohort,constructed a real-time quantitative polymerase chain reaction(qPCR)method and developed a set of analytical approaches to detect these ten groups of predominant gut bacterial species with great maneuverability,efficiency,and quantitative features.Reference ranges for the ten predominant gut bacterial groups were established,and we found that the concentration and pairwise ratios of the ten predominant gut bacterial groups varied with age,indicating gut microbial dysbiosis.By comparing the detection results of liver cirrhosis(LC)patients with those of healthy control subjects,differences were then analyzed,and a classification model for the two groups was built by machine learning.Among the six established classification models,the model established by using the random forest algorithm achieved the highest area under the curve(AUC)value and sensitivity for predicting LC.This research enables easy,rapid,stable,and reliable testing and evaluation of the balance of the gut microbiota in the human body,which may contribute to clinical work.Zhongwen Wu Xiaxia Pan Yin Yuan Pengcheng Lou Lorina Gordejeva Shuo Ni Xiaofei Zhu Bowen Liu Lingyun Wu Lanjuan Li Bo Li 2023Engineering2023,,10:0
