| 1 | Pooled Plasmid Sequencing Reveals the Relationship Between Mobile Genetic Elements and Antimicrobial Resistance Genes in Clinically Isolated Klebsiella pneumoniae显示文摘Plasmids remain important microbial components mediating the horizontal gene transfer(HGT)and dissemination of antimicrobial resistance.To systematically explore the relationship between mobile genetic elements(MGEs)and antimicrobial resistance genes(ARGs),a novel strategy using single-molecule real-time(SMRT)sequencing was developed.This approach was applied to pooled conjugative plasmids from clinically isolated multidrug-resistant(MDR)Klebsiella pneumoniae from a tertiary referral hospital over a 9-month period.The conjugative plasmid pool was obtained from transconjugants that acquired antimicrobial resistance after plasmid conjugation with 53 clinical isolates.The plasmid pool was then subjected to SMRT sequencing,and 82 assembled plasmid fragments were obtained.In total,124 ARGs(responsible for resistance to b-lactam,fluoroquinolone,and aminoglycoside,among others)and 317 MGEs[including transposons(Tns),insertion sequences(ISs),and integrons]were derived from these fragments.Most of these ARGs were linked to MGEs,allowing for the establishment of a relationship network between MGEs and/or ARGs that can be used to describe the dissemination of resistance by mobile elements.Key elements involved in resistance transposition were identified,including IS26,Tn3,IS903 B,ISEcp1,and ISKpn19.As the most predominant IS in the network,a typical IS26-mediated multi-copy composite transposition event was illustrated by tracing its flanking 8-bp target site duplications(TSDs).The landscape of the pooled plasmid sequences highlights the diversity and complexity of the relationship between MGEs and ARGs,underpinning the clinical value of dominant HGT profiles. | Yan Jiang Yanfei Wang Xiaoting Hua Yue Qu Anton Y.Peleg Yunsong Yu | 2020 | Genomics, Proteomics & Bioinformatics2020,18,5: | 1 |
| 2 | Polyoxometalate-Based Ionic Frameworks for Highly Selective CO_(2) Capture and Separation显示文摘During the utilization of structural and functional advantages of polyoxometalates(POMs)for enhanced applications,a suitable assembly of these clusters in framework materials to act as binding nodes represents a promising approach.In contrast to well-developed coordination/covalent combinations,we have developed a convenient strategy to build porous structures of POMs with smaller-sized counterions as bridging ligands via ionic interactions to reinforce their capability in gas adsorption in parallel to metal-organic frameworks(MOFs)/covalent organic frameworks(COFs).With this goal,a series of POMs-based ionic frameworks(IFs)were constructed with triol-ligand modified Anderson-Evans-type clusters as building blocks,and their sodium counter-cations were used as linkers.The three-dimensional(3D)open-frameworks obtained displayed unusually selective CO_(2) capturing capability and efficient separation from their N_(2),H_(2),and CH_(4) mixtures under low pressure at room temperature.Among the synthesized IFs,the cobalt-centered cluster exhibited the best performance for the uptake and selective separation of CO_(2) over N_(2) and CH_(4) in a range of 0-1 bar,while the nickel-centered cluster displayed the highest selectivity over H_(2) at 1 bar.Breakthrough experiments based on real binary gas mixtures demonstrated that the cobalt-containing framework illustrated high performance in the actual gas separation and sustained stability against a simulated operating environment. | Fengxue Duan Xiaoting Liu Di Qu Bao Li Lixin Wu | 2021 | CCS Chemistry2021,3,11: | 1 |

| 4 | Deep learning model improves radiologists'performance in detection and classification of breast lesions显示文摘Objective:Computer-aided diagnosis using deep learning algorithms has been initially applied in the field of mammography,but there is no large-scale clinical application.Methods:This study proposed to develop and verify an artificial intelligence model based on mammography.Firstly,mammograms retrospectively collected from six centers were randomized to a training dataset and a validation dataset for establishing the model.Secondly,the model was tested by comparing 12 radiologists’performance with and without it.Finally,prospectively enrolled women with mammograms from six centers were diagnosed by radiologists with the model.The detection and diagnostic capabilities were evaluated using the freeresponse receiver operating characteristic(FROC)curve and ROC curve.Results:The sensitivity of model for detecting lesions after matching was 0.908 for false positive rate of 0.25 in unilateral images.The area under ROC curve(AUC)to distinguish the benign lesions from malignant lesions was0.855[95%confidence interval(95%CI):0.830,0.880].The performance of 12 radiologists with the model was higher than that of radiologists alone(AUC:0.852 vs.0.805,P=0.005).The mean reading time of with the model was shorter than that of reading alone(80.18 s vs.62.28 s,P=0.032).In prospective application,the sensitivity of detection reached 0.887 at false positive rate of 0.25;the AUC of radiologists with the model was 0.983(95%CI:0.978,0.988),with sensitivity,specificity,positive predictive value(PPV),and negative predictive value(NPV)of94.36%,98.07%,87.76%,and 99.09%,respectively.Conclusions:The artificial intelligence model exhibits high accuracy for detecting and diagnosing breast lesions,improves diagnostic accuracy and saves time. | Yingshi Sun Yuhong Qu Dong Wang Yi Li Lin Ye Jingbo Du Bing Xu Baoqing Li Xiaoting Li Kexin Zhang Yanjie Shi Ruijia Sun Yichuan Wang Rong Long Dengbo Chen Haijiao Li Liwei Wang Min Cao | 2021 | Chinese Journal of Cancer Research2021,33,6: | 1 |

| 6 | Changes in corneal curvature and aberrations after cataract surgery显示文摘Background:Surgically induced astigmatism(SIA)and corneal high-order aberrations(HOAs)are the two main causes of poor visual quality after cataract surgery.Changes in the parameters of corneal HOAs after cataract surgery and their effects on and relationships with changes in corneal curvature have not yet been reported.This study aimed to explore changes in anterior,posterior and total corneal curvature,astigmatism and HOAs after microincision cataract surgery.Methods:Sixty-one age-related cataract patients(61 eyes)were included in this prospective study.The total,anterior and posterior corneal astigmatism and corneal HOAs were analyzed by anterior segment optical coherence tomography(AS-OCT)and iTrace before,one day,one week and three months after 2.2 mm temporal microincision coaxial phacoemulsification to evaluate the changes in anterior,posterior and total corneal curvature,astigmatism and corneal HOAs.Results:The mean J0 and J45 values of anterior,posterior and total corneal curvature obtained by ASOCT showed no statistically significant difference between preoperatively and any postoperative followup.SIA occurred on the anterior,posterior and total corneal surfaces and showed no statistically significant difference at any postoperative follow-up.No significant changes in 3rd-order oblique trefoil,vertical coma or 4th-order spherical aberrations were observed after surgery except for a significant increase in horizontal coma at postoperative day 1(POD1).Conclusions:There were no significant changes in corneal curvature after 2.2 mm temporal microincision coaxial phacoemulsification,and the corneal HOAs were not changed significantly except for the increase in horizontal coma at POD1,which may be one of the main reasons of poor visual quality at POD1 in some cataract patients who have good uncorrected or corrected distance vision. | Ye Dai Xiaoting Ruan Wei Wang Xiaoyun Chen Guangming Jin Lanhua Wang Xiaoxun Gu Bo Qu Jianping Liu Xuhua Tan Enen Zhang Jun Fu Lixia Luo Zhenzhen Liu Yizhi Liu | 2022 | Annals of Eye Science2022,,3: | 0 |
