| 1 | Space Debris Reentry Analysis Methods and Tools显示文摘The reentry of uncontrolled spacecraft may be broken into many pieces of debris at an altitude in the range of 75-85 km. The surviving fragments could pose great hazard and risk to ground and people. In recent years, methods and tools for predicting and analyzing debris reentry and ground risk assessment have been studied and developed in National Aeronautics and Space Ad-ministration (NASA), European Space Agency (ESA) and other organizations, including the group of the present authors. This paper reviews the current progress on this topic of debris reentry briefly. We outline the Monte Carlo method for uncertainty analysis, breakup prediction, and parameters affecting survivability of debris. The existing analysis tools can be classified into two categories, i.e. the object-oriented and the spacecraft-oriented methods, the latter being more accurate than the first one. The past object-oriented tools include objects of only simple shapes. For more realistic simulation, here we present an object-oriented tool debris reentry and ablation prediction system (DRAPS) developed by the present authors, which introduces new object shapes to 15 types, as well as 51 predefined motions and relevant aerodynamic and aerothermal models. | 2 | Shake table tests of suspended ceilings to simulate the observed damage in the M_s 7.0 Lushan earthquake, China显示文摘Severe damage to suspended ceilings of metal grids and lay-in panels was observed in public buildings during the 2013 Ms7.0 Lushan earthquake in China. Over the past several years, suspended ceilings have been widely used practice in public buildings throughout China, including government offices, schools and hospitals. To investigate the damage mechanism of suspended ceilings, a series of three-dimensional shake table tests was conducted to reproduce the observed damage. A full-scale reinforced concrete frame was constructed as the testing frame for the ceiling, which was single-story and infilled with brick masonry walls to represent the local construction of low-rise buildings. In general, the ceiling in the tests exhibited similar damage phenomena as the field observations, such as higher vulnerability of perimeter elements and extensive damage to the cross runners. However, it exhibited lower fragility in terms of peak ground/roof accelerations at the initiation of damage. | 7 | Flame-retardant concentrated electrolyte enabling a Li F-rich solid electrolyte interface to improve cycle performance of wide-temperature lithium–sulfur batteries显示文摘Lithium–sulfur batteries have been regarded as the most promising high-energy electrochemical energy storage device owing to the high energy density, low cost and environmental friendliness. However, traditional lithium–sulfur batteries using ether-based electrolytes often suffer from severe safety risks(i.e. combustion). Herein, we demonstrated a novel kind of flame-retardant concentrated electrolyte(6.5 M lithium bis(trifluoromethylsulphonyl)imide/fluoroethylene carbonate) for highly-safe and widetemperature lithium–sulfur batteries. It was found that such concentrated electrolyte showed superior flame retardancy, high lithium-ion transference number(0.69) and steady lithium plating/stripping behavior(2.5 m Ah cm^(-2) over 3000 h). Moreover, lithium–sulfur batteries using this flame-retardant concentrated electrolyte delivered outstanding cycle performance in a wide range of temperatures(-10 °C, 25 °C and 90 °C). This superior battery performance is mainly attributed to the LiF-rich solid electrolyte interphase formed on lithium metal anode, which can effectively suppress the continuous growth of lithium dendrites. | 10 | AtKP1, a kinesin-like protein, mainly localizes to mitochondria in Arabidopsis thaliana显示文摘Kinesins and kinesin-like proteins (KLPs) constitute a large family of microtubule-based motors that play important roles in many fundamental cellular and developmental processes. To date, a number of kinesins or KLPs have been identified in plants including Arabidopsis thaliana. Here, a polyclonal antibody against AtKP1 (kinesin-like protein 1 in A.thaliana) was raised by injection the expressed AtKP1 specific C-terminal polypeptides in rabbits, and immunoblot analysis was conducted with the affinity-purified anti-AtKP1 antibody. The results indicated that this antibody recognized the AtKP1 fusion proteins expressed in E. coli and proteins of ~125 kDa in the soluble fractions of Arabidopsis extracts. The molecular weight was consistent with the calculated molecular weight based on deduced amino acids sequence of AtKP1. To acquire the subcellular localization of the protein, AtKP1 in Arabidopsis root cells was observed by indirect immunofluorescence microscopy. AtKP1 was localized to particle-like organelles in interphase or dividing cells, but not to mitotic microtubule arrays. Relatively more AtKP1 was found in isolated mitochondria fraction on immunoblot of the subcellular fractions. | 12 | Lessons from the seismic behavior of a steel grid roof structure heavily damaged in Lushan earthquake显示文摘The seismic behavior of a school gymnasium, whose steel grid roof was heavily damaged during the Mw6.6 Lushan earthquake in 2013, is simulated through nonlinear dynamic analysis. The simulated damage is compared with field observations to validate the numerical model, based on which a parametric study was performed to provide insight into the failure process and damage patterns of steel grids. The results suggest that the grid damage is strongly related to roofsubstructure interactions. These include not only the substructure's amplification of the vibration, but the uncoordinated displacement of the substructure's columns which support the grid also play an equally important role. In particular, the latter effect may significantly alter the internal force distribution in the steel grid and lead to unexpected buckling of members that are proportioned as tension-only members. While such interactions are generally not accounted for in the design practice for grid structures in China, similar seismic damage may be expected for other existing grid roofs in future earthquakes. | 17 | Evaluation of the fishbone model in simulating the seismic response of multistory reinforced concrete moment-resisting frames显示文摘The fishbone model is a simplified numerical model for moment-resisting frames that is capable of modelling the effects of column-beam strength and stiffness ratios. The applicability of the fishbone model in simulating the seismic responses of reinforced concrete moment-resisting frames of different sets of column-beam strength and stiffness ratios are evaluated through nonlinear static, dynamic and incremental dynamic analysis on six prototype buildings of 4-, 8-and 12-stories. The results show that the fishbone model is practically accurate enough for reinforced concrete frames, although the assumption of equal joint rotation does not hold in all cases. In addition to the ground motion characteristics and the number of stories in the structures, the accuracy of the model also varies with the column-beam stiffness and strength ratios. The model performs better for strong column-weak beam frames, in which the lateral drift patterns are better controlled by the continuous stiffness provided by the strong columns. When the inelastic deformation is large, the accuracy of the model may be subjected to large record-to-record variability. | 18 | Re-expression of DIRAS3 and P53 induces apoptosis and impaired autophagy in head and neck squamous cell carcinoma显示文摘Background:p53 and DIRAS3 are tumor suppressors that are frequently silenced in tumors.In this study,we sought to determine whether the concurrent re-expression of p53 and DIRAS3 could effectively induce head and neck squamous cell carcinoma(HNSCC)cell death.Methods:CAL-27 and SCC-25 cells were treated with Ad-DIRAS3 and rAd-p53 to induce re-expression of DIRAS3 and p53 respectively.The effects of DIRAS3 and p53 re-expression on the growth and apoptosis of HNSCC cells were examined by TUNEL assay,flow cytometric analysis and MTT.The effects of DIRAS3 and p53 re-expression on Akt phosphorylation,oncogene expression,and the interaction of 4 E-BP1 with eIF4 E were determined by real-time PCR,Western blotting and immunoprecipitation analysis.The ability of DIRAS3 and p53 re-expression to induce autophagy was evaluated by transmission electron microscopy,LC3 fluorescence microscopy and Western blotting.The effects of DIRAS3 and p53 re-expression on HNSCC growth were evaluated by using an orthotopic xenograft mouse model.Results:TUNEL assay and flow cytometric analysis showed that the concurrent re-expression of DIRAS3 and p53 significantly induced apoptosis(P<0.001).MTT and flow cytometric analysis revealed that DIRAS3 and p53 reexpression significantly inhibited proliferation and induced cell cycle arrest(P<0.001).Mechanistically,the concurrent re-expression of DIRAS3 and p53 down-regulated signal transducer and activation of transcription 3(STAT3)and upregulated p21WAF1/CIP1 and Bax(P<0.001).DIRAS3 and p53 re-expression also inhibited Akt phosphorylation,increased the interaction of eIF4 E with 4 E-BP1,and reduced the expression of c-Myc,cyclin D1,vascular endothelial growth factor(VEGF),fibroblast growth factor(FGF),epidermal growth factor receptor(EGFR)and Bcl-2(P<0.001).Moreover,the concurrent re-expression of DIRAS3 and p53 increased the percentage of cells with GFP-LC3 puncta compared with that in cells treated with control adenovirus(50.00%±4.55%vs.4.67%±1.25%,P<0.001).LC3 fluorescence microscopy and Western blotting further showed that DIRAS3 and p53 re-expression significantly promoted autophagic activity but also inhibited autophagic flux,resulting in overall impaired autophagy.Finally,the concurrent re-expression of DIRAS3 and p53 significantly decreased the tumor volume compared with the control group in a HNSCC xenograft mouse model[(3.12±0.75)mm^(3) vs.(189.02±17.54)mm^(3),P<0.001].Conclusions:The concurrent re-expression of DIRAS3 and p53 is a more effective approach to HNSCC treatment than current treatment strategies. | Zhe Liu Douglas R.Hurst Xing Qu Li-Guang Lu Chen-Zhou Wu Yu-Yu Li Yi Li | 2021 | Military Medical Research2021,8,2: | 3 |
| 19 | On civil engineering disasters and their mitigation显示文摘Civil engineering works such as buildings and infrastructure are the carriers of human civilization. They are, however, also the origins of various types of disasters, which are referred to in this paper as civil engineering disasters. This paper presents the concept of civil engineering disasters, their characteristics, classification, causes, and mitigation technologies. Civil engineering disasters are caused primarily by civil engineering defects, which are usually attributed to improper selection of construction site, hazard assessment, design and construction, occupancy, and maintenance. From this viewpoint, many so-called natural disasters such as earthquakes, strong winds, floods, landslides, and debris flows are substantially due to civil engineering defects rather than the actual natural hazards. Civil engineering disasters occur frequently and globally and are the most closely related to human beings among all disasters. | 20 | Melatonin combined with chondroitin sulfate ABC promotes nerve regeneration after root-avulsion brachial plexus injury显示文摘After nerve-root avulsion injury of the brachial plexus, oxidative damage, inflammatory reaction, and glial scar formation can affect nerve regeneration and functional recovery. Melatonin(MT) has been shown to have good anti-inflammatory, antioxidant, and neuroprotective effects. Chondroitin sulfate ABC(ChABC) has been shown to metabolize chondroitin sulfate proteoglycans and can reduce colloidal scar formation. However, the effect of any of these drugs alone in the recovery of nerve function after injury is not completely satisfactory. Therefore, this experiment aimed to explore the effect and mechanism of combined application of melatonin and chondroitin sulfate ABC on nerve regeneration and functional recovery after nerve-root avulsion of the brachial plexus. Fifty-two Sprague-Dawley rats were selected and their C5–7 nerve roots were avulsed. Then, the C6 nerve roots were replanted to construct the brachial plexus nerve-root avulsion model. After successful modeling, the injured rats were randomly divided into four groups. The first group(injury) did not receive any drug treatment, but was treated with a pure gel-sponge carrier nerve-root implantation and an ethanol-saline solution via intraperitoneal(i.p.) injection. The second group(melatonin) was treated with melatonin via i.p. injection. The third group(chondroitin sulfate ABC) was treated with chondroitin sulfate ABC through local administration. The fourth group(melatonin + chondroitin sulfate ABC) was treated with melatonin through i.p. injection and chondroitin sulfate ABC through local administration. The upper limb Terzis grooming test was used 2–6 weeks after injury to evaluate motor function. Inflammation and oxidative damage within 24 hours of injury were evaluated by spectrophotometry. Immunofluorescence and neuroelectrophysiology were used to evaluate glial scar, neuronal protection, and nerve regeneration. The results showed that the Terzis grooming-test scores of the three groups that received treatment were better than those of the injury only group. Additionally, these three groups showed lower levels of C5–7 intramedullary peroxidase and malondialdehyde. Further, glial scar tissue in the C6 spinal segment was smaller and the number of motor neurons was greater. The endplate area of the biceps muscle was larger and the structure was clear. The latency of the compound potential of the myocutaneous nerve-biceps muscle was shorter. All these indexes were even greater in the melatonin + chondroitin sulfate ABC group than in the melatonin only or chondroitin sulfate ABC only groups.
