1Possible early Neoproterozoic magmatism associated with slab window in the Pingshui segment of the Jiangshan-Shaoxing suture zone: Evidence from zircon LA-ICP-MS U-Pb geochronology and geochemistry显示文摘We report here geochemical data, U-Pb zircon ages, and Hf isotopes for the high-Mg diorites (HMDs), Nb-enriched basaltic porphyrys (NEBPs) and plagiogranites (PLAGs) in the Pingshui segment of the Jiangshan-Shaoxing suture zone. The HMDs are characterized by high Mg# (>60), high Na and LREE contents, depletion of HREE and HFSE, and pronounced positive εNd(t) values of 7.0 to 7.7, similar to some adakitic high-Mg andesites. The NEBPs are relatively Na-rich (Na2O/K2O>6) and display high abundances of P2O5 (~1.00%), TiO2 (~3.08%) and HFSE (e.g., Nb=9.53-10.27 ppm). Their Nd isotopic compositions (εNd(t)=6.8-8.0) are comparable to those of the HMDs. The PLAGs are metaluminous (A/CNK=0.84-0.89) and sodic (Na2O/K2O>10). Their depletion in HFSE (e.g., Nb, Ta) is consistent with 'SSZ-type' plagiogranite. Zircon LA-ICP-MS U-Pb dating yields an age of 932±7 Ma for the HMD, 916±6 Ma for the NEBP, and 902±5 Ma for the PLAG, respectively, indicating that they were products of early Neoproterozoic magmatism. The PLAGs exhibit relatively high zircon Hf isotopes and positive εHf(t) values of 11.0 to 16.2, consistent with their Nd isotopic data (εNd(t)=7.5-8.4). Such features are similar to those of oceanic plagiogranites in ophiolites and distinct from those of crust-derived granites. The PLAGs were most likely derived from partial melting of subducted oceanic crust in an active continental margin. Considering these results in the context of the regional geology, we suggest that a slab window in the subducting oceanic crust between the Yangtze Block and Cathaysia Block was possibly the principal cause for the unique arc magmatism in the area. 2SAD2 in Arabidopsis Functions in Trichome Initiation through Mediating GL3 Function and Regulating GL1,TTG1 and GL2 Expression显示文摘Most genes identified that control Arabidopsis trichome initiation and formation are transcription factors or regulatory com- ponents in transcriptional networks and include GLABROUS1 (GL1), GLABRA2 (GL2), GLABRA3 (GL3) and TRANSPARENT TESTA GLABRA1 (TTG1). Herein, we report that an importin β-like protein, SENSITIVE TO ABA AND DROUGHT2 (SAD2), is required for trichome initiation. Mutations in SAD2 disrupted trichome initiation resulting in reduced trichome number, but had no effect on trichome development or root hair number and development. Expression levels of GL1, MYB23, GL2 and TTG1 were reduced in shoots of sad2 mutants while expression levels of GL3 and ENHANCER OF GLABRA3 (EGL3) were enhanced. Overexpression of GL3 increased trichome numbers in wild type but not in sad2 mutants, indicating that the function of the GL3 protein is altered in the sad2 mutants. In contrast, overexpression of GFP-GL1 decreased trichome number in both wild type and sad2. Double mutant analysis of gl1 sad2 and gl3 sad2 indicated that SAD2 functions genetically, at least in part, in the same pathway with these two genes. Co-immunoprecipitation indicated that the sad2 mutation does not disrupt formation of the TTG1-GL3-GL1 complex. Analysis of GFP fusions of GL1, GL2, GL3 and TTG1 suggested that these proteins are most likely not direct cargo of SAD2. 3Quantitative Trait Loci for Panicle Layer Uniformity Identified in Doubled Haploid Lines of Rice in Two Environments显示文摘Uniformity of stem height in rice directly affects crop yield potential and appearance,and has become a vital index for rice improvement.In the present study,a doubled haploid(DH) population,derived from a cross between japonica rice Chunjiang 06 and indica rice TN1 was used to analyze the quantitative trait locus(QTL) for three related traits of paniclelayer-uniformity;that is,the tallest panicle height,the lowest panicle height and panicle layer disuniformity in two locations:Hangzhou(HZ) and Hainan(HN).A total of 16 QTLs for three traits distributed on eight chromosomes were detected in two different environments.Two QTLs,qTPH-4 and qTPH-8 were co-located with the QTLs for qLPH-4 and qLPH-8,which were only significant in the HZ environment,whereas the qTPH-6 and qLPH-6 located at the same interval were only significant in the HN environment.Two QTLs,qPLD-10-1 and qPLD-10-2,were closely linked to qTPH-10,and they might have been at the same locus.One QTL,qPLD-3,was detected in both environments,explaining more than 23% of the phenotypic variations.The CJ06 allele of qPLD-3 could increase the panicle layer disuniformity by 9.23 and 4.74 cm in the HZ and HN environments.Except for qPLD-3,almost all other QTLs for the same trait were detected only in one environment,indicating that these three traits were dramatically affected by environmental factors.The results may be useful for elucidation of the molecular mechanism of panicle-layer-uniformity and marker assisted breeding for super-rice.Liangyong Ma Jinsong Bao Longbiao Guo Dali Zeng Ximing Li Zhijuan Ji Yingwu Xia Changdeng Yang Qian Qian 2009Journal of Integrative Plant Biology2009,51,9:7
4Structural basis of ALMT1-mediated aluminum resistance in Arabidopsis显示文摘The plant aluminum(Al)-activated malate transporter ALMT1 mediates the efflux of malate to chelate the Al in acidic soils and underlies the plant Al resistance.Here we present cryo-electron microscopy(cryo-EM)structures of Arabidopsis thaliana ALMT1(AtALMT1)in the apo,malate-bound,and Al-bound states at neutral and/or acidic pH at up to 3.0Åresolution.The AtALMT1 dimer assembles an anion channel and each subunit contains six transmembrane helices(TMs)and six cytosolicα-helices.Two pairs of Arg residues are located in the center of the channel pore and contribute to malate recognition.Al binds at the extracellular side of AtALMT1 and induces conformational changes of the TM1-2 loop and the TM5-6 loop,resulting in the opening of the extracellular gate.These structures,along with electrophysiological measurements,molecular dynamic simulations,and mutagenesis study in Arabidopsis,elucidate the structural basis for Al-activated malate transport by ALMT1.Jiangqin Wang Xiafei Yu Zhong Jie Ding Xiaokang Zhang Yanping Luo Ximing Xu Yuan Xie Xiaoxiao Li Tian Yuan Shao Jian Zheng Wei Yang Jiangtao Guo 2022Cell Research2022,32,1:3

7Neuroprotection of phenolic alkaloids from Menispermum dauricum versus melatonin against oxygen-glucose-serum-deprivation-mediated injury显示文摘BACKGROUND: Recent studies have demonstrated that phenolic alkaloids from Menispermum dauricum (PAMD) can protect the heart and brain from ischemia/reperfusion injury, and promote neuron survival by inhibiting neuronal Bax and upregulating Bcl-2 expression following ische- mia/reperfusion. OBJECTIVE: To investigate the neuroprotective effects of PAMD versus exogenous melatonin against ischemia/reperfusion injury. DESIGN, TIME AND SETTING: Observation and comparison experiments at a cellular level were performed at the Department of Biochemistry and Molecular Biology, Tongji Medical College of Huazhong University of Science and Technology between February 2007 and February 2008. MATERIALS: PAMD (95% purity) was provided by Kunming Institute of Botany, Chinese Academy of Sciences; melatonin was provided by Sigma, USA. METHODS: N2a mouse neuroblastoma cells were cultured in vitro deprived of glucose, serum and oxygen for 90 minutes, then cultured in normal medium containing different concentrations of PAMD (0.1, 1.0, 10 mg/L) or melatonin (1, 10, and 100 μmol/L). Cells cultured in normal conditions served as a control. MAIN OUTCOME MEASURES: The culture solution was collected to determine the content of ex- citatory neurotransmitters such as glutamic acid and aspartic acid; cell viability was detected by MTT methods; reactive oxygen species production was determined by fluorescence spectroscopy; mito- chondrial transmembrane potential (?Ψm) was detected by laser confocal scanning; cytochrome C was measured by western blotting; and caspase-3 activity was determined by visible spectropho- tometry. RESULTS: Melatonin and PAMD both promoted oxygen-glucose-serum deprivation-mediated N2a cell survival (P < 0.01) and inhibited glutamic acid release (P < 0.01), but melatonin did not inhibit aspartic acid production. The protective effects were the strongest using melatonin 100 μmol/L and PAMD 10 mg/L, so subsequent experiments were the performed at those doses. Although PAMD could no longer maintain mitochondrial transmembrane potential 6 hours after reperfusion, its in- hibitory effects on cytochrome C release from mitochondria and scavengers of reactive oxygen species were stronger than those of melatonin (P < 0.01). However, its inhibitory effect on caspase-3 activity was weaker than that of melatonin: PAMD could inhibit caspase-3 activity 12 hours after reperfusion (P < 0.01), but melatonin inhibited caspase-3 activity 28 hours after reperfusion (P < 0.01). CONCLUSION: The results show that melatonin and PAMD have neuroprotective effects, but that the mechanisms are varied.
