| 3 | Expression analysis,single nucleotide polymorphisms within SIRT4 and SIRT7 genes and their association with body size and meat quality traits in Qinchuan cattle显示文摘Silent information regulator 2(Sir2) proteins, or sirtuins, are nicotine adenine dinucleotide(NAD)-dependent deacetylases that connect metabolism with longevity in lower organisms. In mammals, there are seven Sir2 homologs, namely, silent information regulators(SIRT1–7). SIRT4 and SIRT7 genes play a crucial role in regulating lipid metabolism, cellular growth and metabolism. This suggests that they are potential candidate genes for affecting body size and meat quality traits in animals. Hence, this study aimed to detect genetic variations of both SIRT4 and SIRT7 bovine genes in Qinchuan cattle, and to evaluate the effect of these variations on economically important body size and meat quality traits. Expression analysis using quantitative real-time PCR(q PCR) indicated that SIRT4 and SIRT7 were broadly expressed in all thirteen studied tissues. The expression of SIRT4 was higher in liver, muscle, and in subcutaneous fat tissue. In the case of SIRT7, the expression was higher in lung, abomasum, and subcutaneous fat. Using DNA sequencing, a total of three single nucleotide polymorphisms(SNPs) were identified within SIRT4 and SIRT7 genes in 468 Qinchuan cattle. These included one novel SNP within 3′ untranslated regions(UTR) of SIRT4(SNP1: g.13915A>G) and two novel synonymous substitutions in SIRT7(SNP2: g.3587C>T and SNP3: g.3793T>C). Statistical analyses indicated that all three SNPs could significantly influence some body size and meat quality traits in Qinchuan cattle. | 4 | A modified scar model with controlled tension on secondary wound healing in mice显示文摘Pathological scars might cause a distorted appearance and restricted mobility,and the study of scar pathophysiology has been hindered by the absence of a reliable model.In this study,we introduce a model with a modified device to induce controlled tension on a wound healing by secondary intention to overcome the shortcomings of the model generated by Aarabi et al.We investigated and recommend an induction of 0.1 N/mm^(2) tension on day 7 for 14 days to mimic the characteristics of human scars.A 3.5-fold increase in scar tissue and a 2-fold increase in collagen production were induced by the modified model.Histologically,the modified method increased scar thickness.However,no significant difference was found in cell density between the two groups.This modified procedure significantly increased scar tissue,which could be used for further cellular and biomolecular research.The mechanical force applied to the wound became measurable and controllable.This method is more convenient for researchers to observe in realtime and for providing timely adjustments of the tension used in this modified model. | Zi Wang Xin Huang Tao Zan Qingfeng Li Haizhou Li | 2020 | Burns & Trauma2020,8,1: | 2 |
| 5 | Plant species coexistence alleviates the impacts of lead on Zea mays L.显示文摘Whether plant coexistence can reduce the impacts of lead (Pb) on crops in agroecosystems has not been well understood. We conducted a factorial experiment to investigate the effects of weeds coexisting with maize (Zea mays L.) on Pb accumulation in maize and soil microbes at two Pb levels (ambient and 300 mg/kg). Elevated Pb tended to increase the Pb concentration in maize and decreased soil microbial activity (indicated by the average well color development, AWCD), functional group diversity, as well as arbuscular mycorrhizal (AM) colonization and vesicle number of maize. Compared to the monoculture, weeds coexisting with maize reduced the Pb concentrations in the root, leaf, sheath and stem of maize at both seedling and mature stages. In maize-weed mixtures, soil microbial activity and functional group diversity tended to increase for both Pb treatments relative to the monoculture. Furthermore, principal component analysis revealed that the soil microbial community structure changed with the introduction of weeds. The highest Pb accumulation in weeds occurred for the elevated Pb treatment in a three species mixture. | 11 | Local delivery of superagonist gene based on polymer nanoparticles for cancer immunotherapy显示文摘Cancer is the leading cause that threatens human life expectancy due to the lack of effective therapies.Cancer immunotherapy has been explored to improve the body's immune system against cancer and accompanied by promising results in recent years.Interleukin 15(IL-15),a pleiotropic immunomodulator,is critical for immune cells development and displays great anti-tumor potential in both preclinical and clinical trials.In this study,superagonist IL-15 plasmid(psIL-15)consisting of IL-15Rα-sushi-linker-IL-15 was constructed in order to secret superagonist IL-15(sIL-15)in tumor site.A gene delivery system through self-assembly by methylated polyethylene glycol-b-polylactic acid-b-methylated polyethylene glycol(m PEG-PLA-m PEG)and 1,2-dioleoyl-3-trimethylammonium-propane(DOTAP),named DMAM,was designed to deliver psIL-15.Further study showed that DMAM/psIL-15 could successfully deliver psIL-15 to tumor cells and the supernatants of the tumor cells could further stimulate lymphocytes proliferation as well as activation in vitro.Local delivery of DMAM/psIL-15 in animal models demonstrated significant tumor inhibition through enhancing immune cells responses,reducing angiogenesis,promoting tumor cell apoptosis and inhibiting proliferation,with no evidence of system toxicities.These results indicate that DMAM/psIL-15 may be a promising strategy for cancer immunotherapy. | Zehua Hong Xin Zan Ting Yu Yuzhu Hu Hongfeng Gou Songping Zheng Xiang Gao Peizhi Zhou | 2023 | Chinese Chemical Letters2023,34,3: | 1 |

| 18 | Theoretical Investigation on Structures,Stabilities,and Hydrolysis Reactions of Small RuO_(2)Nanoclusters显示文摘The geometric structures and relative stabilities of both neutral and ionic(RuO_(2))_(n)(n=1-5)nanoclusters have been systematically investigated using density functional theory calculations.Differently from the crystal structure,ruthenium tends to adopt a 4-fold coordination with either terminal Ru=O or bridging Ru-O bonds in gas phase clusters.Our calculations show that three-dimensional compact structures are more preferred with the increase of the cluster size.For the low-lying(RuO_(2))_(n)clusters,the cluster formation energy per RuO_(2)unit and the electron af-finity are found to increase whereas the ionization potential is computed to decrease with the cluster size.Based on the optimized lower-energy structures of neutral and cationic(RuO_(2))_(n)(n=1-5),the hydrolysis reactions of these clusters with water molecules have been further explored.The calculated results indicate that the adsorption of wa-ter molecules to the cationic species is much easier than the neutral ones.The overall hydrolysis reaction energies are more exothermic for the cationic clusters than for the neutral clusters.Our calculations indicate that thermody-namically,H_(2)O can be more easily split on the cationic ruthenium oxide clusters than on the neutral clusters. | Hao Dong Zhonghua Zan Xin Zhou | 2014 | Chinese Journal of Chemistry2014,32,6: | 0 |

| 20 | pIL-12 delivered by polymer based nanovector for anti-tumor genetherapy显示文摘Finding more effective and safe non-viral vectors to transfer genes into cancer cells has become the key of immune gene therapy for cancer.Herein a triblock compound MPEG_(2000)-PDLLA_(4000)-MPEG_(2000) modified by cationic liposome DOTAP was used as a non-viral vector DOTAP/MPEG_(2000)-PDLLA_(4000)-MPEG_(2000)(DMPM)to effectively transfer interleukin(IL)-12 plasmid(pIL-12)into tumor tissue.IL-12 produced by transfected tumor cells successfully inducing lymphocyte proliferation and promoting interferon-γ(IFN-γ)secretion,which resulted in tumor cells death.The ability of DMPM to transfer pIL-12 and the immune effect induced by IL-12 in cells had been explored.The anti-tumor effect,mechanism and safety of pIL-12/DMPM in mice cancer model were investigated in this study.Our results showed that the pIL-12 transferred by DMPM was highly expressed both in CT26 cells and B16-F10 cells.IL-12 expressed in the culture supernatant of transfected tumor cells stimulated lymphocyte proliferation and promoted IFN-γsecretion.The experimental result confirmed that pIL-12/DMPM therapy significantly reduced tumor growth in mice model.We designed the nanocomposite DMPM to deliver pIL-12 for cancer treatment and explored its therapeutic efficacy and the underlying anti-tumor mechanism.Our study suggested pIL-12 loaded by DMPM complex would be an effective strategy for cancer treatment. | Lianbin Wen Xin Zan Qidi Pang Yuzhu Hu Songping Zheng Mengni Ran Xiang Gao Xiang Wang Bilan Wang | 2023 | Chinese Chemical Letters2023,34,11: | 0 |
