2Regulation of FN1 degradation by the p62/SQSTM1-dependent autophagy-lysosome pathway in HNSCC显示文摘Epithelial–mesenchymal transition(EMT)is involved in both physiological and pathological processes.EMT plays an essential role in the invasion,migration and metastasis of tumours.Autophagy has been shown to regulate EMT in a variety of cancers but not in head and neck squamous cell carcinoma(HNSCC).Herein,we investigated whether autophagy also regulates EMT in HNSCC.Analyses of clinical data from three public databases revealed that higher expression of fibronectin-1(FN1)correlated with poorer prognosis and higher tumour pathological grade in HNSCC.Data from SCC-25 cells demonstrated that rapamycin and Earle’s balanced salt solution(EBSS)promoted autophagy,leading to increased FN1 degradation,while 3-methyladenine(3-MA),bafilomycin A1(Baf A1)and chloroquine(CQ)inhibited autophagy,leading to decreased FN1 degradation.On the other hand,autophagic flux was blocked in BECN1 mutant HNSCC Cal-27 cells,and rapamycin did not promote autophagy in Cal-27 cells;also in addition,FN1 degradation was inhibited.Further,we identified FN1 degradation through the lysosome-dependent degradation pathway using the proteasome inhibitor MG132.Data from immunoprecipitation assays also showed that p62/SQSTM1 participated as an autophagy adapter in the autophagy–lysosome pathway of FN1 degradation.Finally,data from immunoprecipitation assays demonstrated that the interaction between p62 and FN1 was abolished in p62 mutant MCF-7 and A2780 cell lines.These results indicate that autophagy significantly promotes the degradation of FN1.Collectively,our findings clearly suggest that FN1,as a marker of EMT,has adverse effects on HNSCC and elucidate the autophagy–lysosome degradation mechanism of FN1.Xinchen Liu Lin Meng Xing Li Daowei Li Qilin Liu Yumeng Chen Xiangwei Li Wenhuan Bu Hongchen Sun 2020International Journal of Oral Science2020,12,4:7
3Molecular basis of CONSTANS oligomerization in FLOWERING LOCUS T activation显示文摘The transcription factor CONSTANS(CO)integrates day-length information to induce the expression of florigen FLOWERING LOCUS T(FT)in Arabidopsis.We recently reported that the C-terminal CCT domain of CO forms a complex with NUCLEAR FACTOR-YB/YC to recognize multiple cis-elements in the FT promoter,and the N-terminal tandem B-box domains form a homomultimeric assembly.However,the mechanism and biological function of CO multimerization remained unclear.Here,we report that CO takes on a head-to-tail oligomeric configuration via its B-boxes to mediate FT activation in long days.The crystal structure of B-boxesCOreveals a closely connected tandem B-box fold forming a continuous head-to-tail assembly through unique CDHH zinc fingers.Mutating the key residues involved in CO oligomerization resulted in a non-functional CO,as evidenced by the inability to rescue co mutants.By contrast,a transgene encoding a human p53-derived tetrameric peptide in place of the B-boxesCOrescued co mutant,emphasizing the essential role of BboxesCO-mediated oligomerization.Furthermore,we found that the four TGTG-bearing cis-elements in FT proximal promoter are required for FT activation in long days.Our results suggest that CO forms a multimer to bind to the four TGTG motifs in the FT promoter to mediate FT activation.Xiaolin Zeng Xinchen Lv Rui Liu Hang He Shiqi Liang Lixian Chen Fan Zhang Liu Chen Yuehui He Jiamu Du 2022Journal of Integrative Plant Biology2022,64,3:2

9Data-driven Based Uncertainty Set Modeling Method for Microgrid Robust Optimization with Correlated Wind Power显示文摘Microgrid is considered an important part of the future zero carbon energy systems.However,the uncertainty caused by renewable energy source brings huge challenges to the scheduling of MG and restricts its ability of carbon emission reduction.In this paper,a novel improved multi-ellipsoidal uncertainty set modeling method is proposed to better depict the uncertainty of wind power and reduce the conservativeness of traditional robust optimization.Probabilistic information from historical data is utilized to capture the temporal correlation of forecast error of wind power,as well as the conditional correlation of forecast error with forecast value,making the uncertainty set more data-adaptive to variation of forecast results and more accurate for uncertainty description.A two-stage robust optimization model of a grid-connected microgrid is established based on the proposed uncertainty set and solved by column and constraint generation algorithm.Simulation results based on actual data illustrate the average unbalanced power of microgrid between day-ahead trading and real-time power exchange with utility grid is dropped by nearly 11.16%compared with a deterministic optimization method,11.86%with traditional box uncertainty set-based robust optimization method,and 2.89%with stochastic optimization method.Xinchen Li Yixin Liu Li Guo Xialin Li Chengshan Wang 2023CSEE Journal of Power and Energy Systems2023,9,2:0
10Report of China's innovation increase and research growth in radiation oncology显示文摘Aims: To investigate the research status of radiation oncology in China through survey of literature in international radiation oncology journals and retrospectively compare the outputs of radiation oncology articles of the three major regions of China—Mainland(ML), Taiwan(TW) and Hong Kong(HK).Methods: Radiation oncology journals were selected from 'oncology' and 'radiology, nuclear & medical image' category from Science Citation Index Expand(SCIE). Articles from the ML, TW and HK were retrieved from MEDLINE. The number of total articles, clinical trials, case reports, impact factors(IF), institutions and articles published in each journals were conducted for quantity and quality comparisons.Results: A total 818 articles from 13 radiation oncology journals were searched, of which 427 are from ML, 259 from TW, and 132 from HK. Ninety-seven clinical trials and 5 case reports are reported in China. Accumulated IF of articles from ML(1,417.11) was much higher than that of TW(1,003.093) and HK(544.711), while the average IF of articles from ML is the lowest.Conclusions: The total number of articles from China especially ML increased significantly in the last decade. The number of articles published from the ML has exceeded those from TW and HK. 11Solid-state preparation of mesoporous Ce-Mn-Co ternary mixed oxide nanoparticles for catalytic degradation of methylene blue显示文摘Mesoporous Ce-Mn-Co ternary metal oxide was fabricated via an efficient oxalate-precursor-based soft reactive grinding route and used to activate H_(2)O_(2) for advanced oxidation of methylene blue in water.In addition,Mn-Co binary oxide and pure Co_(3)O_(4) and Mn_(3)O_(4) were also synthesized as reference catalysts.These catalysts were characterized by X-ray diffraction,N_(2) adsorption-desorption,H_(2)-temperature programmed reduction,transmission electron microscopy,scanning electron microscopy and X-ray photoelectron spectroscopy.The results demonstrate that part of the Ce and Mn can be incorporated into the lattice of Co_(3)O_(4) and cause severe lattice distortion of the unit cell.Compared with the single or binary system,Ce-Mn-Co ternary metal oxide exhibits the best activity in methylene blue removal and nearly 100% decomposition rate and 84% COD removal rate can be achieved in 12 h,and with degradation rate of 93.5% after three rounds.These results are primarily attributed to the synergistic effect of Ce,Mn and Co,which can promote the formation of more lattice defects,higher specific surface area and smaller particle size.Quenching tests show that hydroxyl radicals(·OH) play more dominant role than superoxide radicals(·O_(2)^(-)).Kinetic studies were studied and the activation energies of all the catalysts were calculated.Qingwen Wang Wenyong Deng Xinchen Lin Xigen Huang Ling Wei Lei Gong Changxiang Liu Guangbin Liu Qian Liu 2021Journal of Rare Earths2021,39,7:0
12A novel approach for navigational safety evaluation of inland waterway ships under uncertain environment显示文摘This paper proposes a safety evaluation model of inland waterway ship navigation based on the fuzzy theory and evidential reasoning(ER)approach.The proposed model is a three-level hierarchical system consisting of 18 indicators that are identified and determined through literature review and expert surveys.By using fuzzy theory,the factors of the index level are converted into belief distributions,and the information of indicators within each level is merged by using the ER approach so that the evaluation of the ship’s navigation safety can be realized.This model can deal with multi-source information and both qualitative and quantitative indicators in an integrated framework.Some case studies on the safety status of ships navigating in the middle reaches of the Yangtze River during dry season are conducted to validate the feasibility and practicability of the proposed model.The results show that the risk levels obtained from the proposed model are consistent with the real-life situation to a large extent.The novel model proposed in this study provides a useful reference for maritime safety management and will help managers and decision-makers in the risk prevention and early warning of inland waterway transportation accidents.Jia Liu Xinchen Jiang Weina Huang Yankang He Zhisen Yang 2022Transportation Safety and Environment2022,4,1:0
13Trajectory Design for UAV-Enabled Maritime Secure Communications:A Reinforcement Learning Approach显示文摘This paper investigates an unmanned aerial vehicle(UAV)-enabled maritime secure communication network,where the UAV aims to provide the communication service to a legitimate mobile vessel in the presence of multiple eavesdroppers.In this maritime communication networks(MCNs),it is challenging for the UAV to determine its trajectory on the ocean,since it cannot land or replenish energy on the sea surface,the trajectory should be pre-designed before the UAV takes off.Furthermore,the take-off location of the UAV and the sea lane of the vessel may be random,which leads to a highly dynamic environment.To address these issues,we propose two reinforcement learning schemes,Q-learning and deep deterministic policy gradient(DDPG)algorithms,to solve the discrete and continuous UAV trajectory design problem,respectively.Simulation results are provided to validate the effectiveness and superior performance of the proposed reinforcement learning schemes versus the existing schemes in the literature.Additionally,the proposed DDPG algorithm converges faster and achieves higher utilities for the UAV,compared to the Q-learning algorithm.Jintao Liu Feng Zeng Wei Wang Zhichao Sheng Xinchen Wei Kanapathippillai Cumanan 2022China Communications2022,19,9:0
14Land use/cover and landscape pattern changes in Manas River Basin based on remote sensing显示文摘Large-scale water and soil development in inland river basins in arid areas has made changes in landscape composition and structure,threatening the ecological balance.In order to study the trend of land use/cover and landscape pattern change and its relationship with water resources utilization in Manas River Basin,the land-use data of five periods in 1976,1990,2000,2010 and 2015 were analyzed.The results showed that:(1)During 1976-2015,farmland and construction land continuously increased,forest land and grassland continuously decreased,the water area initially reduced and then increased,the area of saline-alkali land initially increased and then reduced,the overall trend of unused land and the sandy area was decreasing and the area of different time periods is floating.The areas of land-use types either increased or decreased.This indicates that the landscape pattern of the basin changes dramatically,and human activities are the main reasons for this phenomenon;(2)The oasis area increased from 3480.2 km2 in 1976 to 7982.0 km2 in 2015,with an obvious increasing trend.The oasis area clearly increased during 1976-1990,the growth rate was 40.6%,the growth rate of the oasis area was 129.4%.In the last 40 years,the degree of desertification fluctuated and decreased.The increase of the oasis area directly leads to the increase of water resources utilization and aggravates the degree of water resources shortage;(3)The pattern of land-use types showed a non-equilibrium trend.In the region with increasing landscape heterogeneity,the overall landscape pattern was increasingly controlled by the majority of patches.The intensive land management model and drip irrigation under mulch have improved the utilization efficiency of water resources and saved water resources from engineering renovation and irrigation management.Xiaolong Li Xinlin He Guang Yang Hongguang Liu Aihua Long Fulong Chen Bing Liu Xinchen Gu 2020International Journal of Agricultural and Biological Engineering2020,13,5:0
15Identification and verification of aging-related lncRNAs for prognosis prediction and immune microenvironment in patients with head and neck squamous carcinoma显示文摘Aging is highly associated with tumor formation and progression.However,little research has explored the association of aging-related lncRNAs(ARLs)with the prognosis and tumor immune microenvironment(TIME)of head and neck squamous cell carcinoma(HNSCC).RNA sequences and clinicopathological data of HNSCC patients and normal subjects were downloaded from The Cancer Genome Atlas.In the training group,we used Pearson correlation,univariate Cox regression,least absolute shrinkage/selection operator regression analyses,and multivariate Cox regression to build a prognostic model.In the test group,we evaluated the model.Multivariate Cox regression was done to screen out independent prognostic factors,with which we constructed a nomogram.Afterward,we demonstrated the predictive value of the risk scores based on the model and the nomogram using time-dependent receiver operating characteristics.Gene set enrichment analysis,immune correlation analysis,and half-maximal inhibitory concentration were also performed to reveal the different landscapes of TIME between risk groups and to predict immuno-and chemo-therapeutic responses.The most important LINC00861 in the model was examined in HNE1,CNE1,and CNE2 nasopharyngeal carcinoma cell lines and transfected into the cell lines CNE1 and CNE2 using the LINC00861-pcDNA3.1 construct plasmid.In addition,CCK-8,Edu,and SA-β-gal staining assays were conducted to test the biofunction of LINC00861 in the CNE1 and CNE2 cells.The signature based on nine ARLs has a good predictive value in survival time,immune infiltration,immune checkpoint expression,and sensitivity to multiple drugs.LINC00861 expression in CNE2 was significantly lower than in the HNE1 and CNE1 cells,and LINC00861 overexpression significantly inhibited the proliferation and increased the senescence of nasopharyngeal carcinoma cell lines.This work built and verified a new prognostic model for HNSCC based on ARLs and mapped the immune landscape in HNSCC.LINC00861 is a protective factor for the development of HNSCC.QING GAO YUJING SHI YUANYUAN SUN SHU ZHOU ZEYUAN LIU XINCHEN SUN XIAOKE DI 2023Oncology Research2023,31,1:0
16A metal-organic framework-based redox homeostasis disruptor selectively potentiate the cytotoxicity of dihydroartemisinin for cancer therapy显示文摘Artemisinin and its derivatives have emerged as promising therapeutic agents for cancer therapy by endogenous iron-mediated generation of free radicals.However,the enhanced antioxidant defense systems in cancer cells provide them with resistance to oxidative damage,greatly antagonizing the therapeutic efficacy that relies on inducing oxidative stress.Herein,a metal-organic framework(MOF)-based nanoplatform(CMD)is constructed to disrupt the cellular redox homeostasis and selectively potentiate the cytotoxicity of dihydroartemisinin for cancer therapy.In cancer cells,the copper(II)sites in the MOF nanocarrier of CMD can efficiently weaken the cellular antioxidant capacity by depleting the overexpressed glutathione,simultaneously leading to the decomposition of the framework structure and the release of the encapsulated dihydroartemisinin.As a result,the damaged antioxidant defense system of cancer cells reduces its effect on oxidative stress alleviation and strengthens the therapeutic efficacy of dihydroartemisinin.On contrast,the low concentration of cellular glutathione in normal cells protects them from dihydroartemisinin-induced cytotoxicity by decelerating the drug release.In vivo results demonstrate that CMD could completely suppress the tumor growth in mice and show no evidence of toxicity,providing an effective strategy for the practical usage of dihydroartemisinin in cancer therapy.Jiawen Fan Xinchen Liu Qishun Wang Huan Wang Hao Liu Deming Han Jinsong Ren 2023Nano Research2023,16,5:0
17Amorphous NH_(2)-MIL-68 as an efficient electro-and photo-catalyst for CO_(2)conversion reactions显示文摘To produce metal-organic framework(MOF)catalysts with both high activity and durability is interesting but challenging.We report an amorphous MOF(NH_(2)-MIL-68),which combines the advantages of(1)a large number of open metal sites,(2)the basic building blocks and connectivity of crystalline NH_(2)-MIL-68,and(3)hierarchically meso-and microporous structure.It exhibits high performances in electrocatalytic reduction of CO_(2)and photochemical cycloaddition of CO_(2)under mild conditions.For the former reaction,the maximum Faradaic efficiency for product formic acid(FEHCOOH)reaches 93.3%with a current density of 34.2 mA·cm^(−2)at^(−2).05 V vs.Ag/Ag+catalyzed by amorphous NH_(2)-MIL-68,while the crystalline NH_(2)-MIL-68 shows FEHCOOH of 67.7%with considerable productions of CO and H_(2)at the same experimental conditions.For the photochemical cycloaddition of CO_(2)with styrene oxide,the yield by amorphous NH_(2)-MIL-68 can reach 94.1%at 12 h,which is higher than the reported value under similar conditions.The structure–efficiency relationship of the catalyst for the two reactions was investigated.This work opens up new possibility for designing high-performance MOF and MOF-based catalysts.Lifei Liu Jianling Zhang Xiuyan Cheng Mingzhao Xu Xinchen Kang Qiang Wan Buxing Han Ningning Wu Lirong Zheng Chenyan Ma 2023Nano Research2023,16,1:0
