| 1 | Origin and phylogenetic analysis of Tibetan Mastiff based on the mitochondrial DNA sequence显示文摘At present,the Tibetan Mastiff is the oldest and most ferocious dog in the world. However,the origin of the Tibetan Mastiff and its phylogenetic relationship with other large breed dogs such as Saint Bernard are unclear. In this study,the primers were designed accord-ing to the mitochondrial genome sequence of the domestic dog,and the 2,525 bp mitochondrial sequence,containing the whole sequence of Cytochrome b,tRNA-Thr,tRNA-Pro,and control region of the Tibetan Mastiff,was obtained. Using grey wolves and coyotes as out-groups,the Tibetan Mastiff and 12 breeds of domestic dogs were analyzed in phylogenesis. Tibetan Mastiff,domestic dog breeds,and grey wolves were clustered into a group and coyotes were clustered in a group separately. This indicated that the Tibetan Mastiff and the other domestic dogs originated from the grey wolf,and the Tibetan Mastiff belonged to Carnivora,Canidae,Canis,Canis lupus,Canis lupus familiaris on the animal taxonomy. In domestic dogs,the middle and small breed dogs were clustered at first; German Sheepdog,Swedish Elkhound,and Black Russian Terrier were clustered into one group,and the Tibetan Mastiff,Old English Sheepdog,Leonberger,and Saint Bernard were clustered in another group. This confirmed the viewpoint that many of the famous large breed dogs worldwide such as Saint Bernard possibly had the blood lineage of the Tibetan Mastiff,based on the molecular data. | 2 | Comparison of basic features and origins of oolitic shoal reservoirs between carbonate platform interior and platform margin locations in the Lower Triassic Feixianguan Formation of the Sichuan Basin,southwest China显示文摘The oolitic shoal reservoirs of the Lower Triassic Feixianguan Formation carbonates in the Sichuan Basin of southwest China are an important target for gas exploration in the basin.Their occurrence,like other cases worldwide,can be divided into two locations in general,i.e.,platform interior and platform margin locations.Their differences of reservoir features and origins,however,have not been investigated comprehensively due to different exploration degrees.This issue is addressed in this paper,to provide basic data and information for the basin's hydrocarbon exploration and for the study of carbonate platform sedimentology and reservoir geology worldwide.We compared the features of these two types of reservoirs in detail,including the depositional and diagenetic features,pore types and petrophysical features.Based on the comparison,the origin of the reservoirs was further discussed.It is shown that the reservoirs in platform interior and platform margin locations differ significantly.The interior carbonates were deposited in moderate to high energy settings and the dominant lithologic type was limestone,which was weakly compacted and intensely cemented and has undergone meteoric dissolution.Pore types include intragranular dissolution and moldic pores,with low porosities(<6%) and low permeabilities(<0.1 mD).By contrast,the platform margin carbonates were deposited in relatively high energy settings and mainly consisted of dolostones with some limestones.The rocks were strongly compacted but incompletely cemented.As a result,some primary intergranular pores were preserved.Both meteoric solution and burial solution have taken place.There are various types of pore spaces including intergranular and intercrystalline solution pores and residual intergranular pores.This type of reservoir generally has better petrophysical properties(>9% porosity and >0.1 mD permeability) and pore-throat structures than the interior reservoirs.These differences were influenced by both primary depositional features and secondary diagenesis.For the interior carbonate reservoirs,early meteoric dissolution,weak compaction and strong cementation are important controlling factors.By contrast,the factors controlling the formation of the margin carbonate reservoirs mainly include dolomitization,preservation of primary pores and burial dissolution. | Tan Xiucheng Zhao Luzi Luo Bing Jiang Xingfu Cao Jian Liu Hong Li Ling Wu Xingbo Nie Yong | 2012 | Petroleum Science2012,9,4: | 9 |
| 3 | The phylogenetic status of typical Chinese native pigs: analyzed by Asian and European pig mitochondrial genome sequences显示文摘China is one of the most diverse countries, which have developed 88 indigenous pig breeds. Several studies showed that pigs were independently domesticated in multiple regions of the world. The purpose of this study was to investigate the origin and evolution of Chinese pigs using complete mitochondrial genomic sequences (mtDNA) from Asian and European domestic pigs and wild boars. Thirty primer pairs were designed to determine the mtDNA sequences of Xiang pig, Large White, Lantang, Jinhua and Pietrain. The phylogenetic status of Chinese native pigs was investigated by comparing the mtDNA sequences of complete coding regions and D-loop regions respectively amongst Asian breeds, European breeds and wild boars. The analyzed results by two cluster methods contributed to the same conclusion that all pigs were classified into two major groups, European clade and Asian clade. It revealed that Chinese pigs were only recently diverged from each other and distinctly different from European pigs. Berkshire was clustered with Asian pigs and Chinese pigs were involved in the development of Berkshire breeding. The Malaysian wild boar had distant genetic relationship with European and Asian pigs. Jinhua and Lanyu pigs had more nucleotide diversity with Chinese pigs although they all belonged to the Asian major clade. | 4 | Mitochondrial DNA T7719G in tRNA-Lys gene affects litter size in Small-tailed Han sheep显示文摘Background: In farm animals, mitochondrial DNA(mtDNA) effect on economic performance remains hot-topic for breeding and genetic selection. Here, 53 maternal lineages of Small-tailed Han sheep were used to investigate the association of mitochondrial DNA variations and the lambing litter size.Results: Sequence sweeping of the mitochondrial coding regions discovered 31 non-synonymous mutations, and the association study revealed that T7719G in mt DNA t RNA-Lys gene was associated with litter size(P < 0.05),manifesting 0.29 lambs per litter between the G and T carriers. Furthermore, using the mixed linear model, we assayed the potential association of the ovine litter size and haplogroups and multiple-level mtDNA haplotypes,including general haplotypes, assembled haplotypes of electron transport chain contained sequences(H-ETC),mitochondrial respiratory complex contained sequences(H-MRC) and mitochondrial genes(H-gene, including polypeptide-coding genes, rRNA genes and tRNA genes). The strategy for assembled mitochondrial haplotypes was proposed for the first time in mtDNA association analyses on economic traits, although none of the significant relations could be concluded(P > 0.05). The continuous 9 polymorphic sites were checked by PCR-RFLP, the results indicated that Erhualian had extraordinary gene frequency, presented most significant difference by x2 test compared with Landrace, Largewhite, Neijiang and Minpig respectively, significant level compared with Meishan; and Meishan also had significant difference compared with Landrace and Minpig respectively. | 7 | Removal of roosters alters the domestic phenotype and microbial and genetic profile of hens显示文摘Hens are raised apart from roosters in modern poultry production, a substantial change from their natural social structure. We compared productivity, injuries, behavior, physiology, microbiome and transcriptome of hens housed with(R+) or without(R–)roosters to quantify the effects of this change in social structure. Hens were raised free-range from 70 to 280 days when 30 birds per treatment were assigned to battery cages until Day 315(R+C vs. R–C), while 30 birds per treatment remained in free-range pens(R+F vs. R–F). Response to a novel environment and object, behavioral time budgets, cecum microbiome, blood composition and transcriptomic sequencing of thigh muscle and spleen were analyzed. Hens housed without roosters showed better survival, consumed less food, produced more eggs and had better feed conversion. R+F hens clustered around the rooster and were less mobile in the novel environment and object tests. R+F hens displayed the richest microbiome, and the presence of roosters resulted in differentially expressed genes related to muscle development, cellular processes, environmental information processing and immune function. | 15 | Molecular authentication of the traditional Chinese medicine Tongren Dahuoluo Wan and its alternative formulation显示文摘Tongren Dahuoluo Wan has been a popular traditional Chinese medicine in international pharmaceutical markets for hundreds of years. Leopard bone powder is the key element in its formulation. However, the leopard has been listed for wildlife conservation, which limits the use of the leopard bone supplies. Therefore, an alternative formulation which substitutes leopard bone with zokor bone in the formula of Tongren Dahuoluo Wan is now manufactured. To develop a simple and reliable molecular method for authenticating the two patent medicines,mitochondrial nucleotide polymorphic sites of 12 S rRNA,COI and Cytb genes were screened in leopard and zokor bones, and nine pairs of species-specific primers were verified for discriminating the two species. For the patent medicine authentication, we set up a molecular diagnostic assay to resolve the difficulties of low concentration of target DNAs and presence of PCR-inhibitory substances in this complex medicine, and successfully confirmed leopard or zokor content using the nine pairs of species-specific primers.
