1Optimization of Agrobacterium tumefaciens-Mediated Transformation Systems in Tea Plant(Camellia sinensis)显示文摘In this study, an efficient plant regeneration protocol in vitro and transformation by Agrobacterium-mediated method of Camellia sinensis was achieved, which would lay the foundation for genetic improvement of tea plant by genetic engineering technology. The cotyledon callus of C.sinensis were used as the receptors for transformation by Agrobacterium tumefaciens EHA105 containing PS1aG-3. Some factors which affected the result of Agrobacterium-mediated transformation of C. sinensis were studied on the basis of GUS transient expression system. The optimum system of Agrobacterium-mediated transformation was that the cotyledon callus were pre-cultured for 3 d, and then infected by EHA105 for 15 min followed by 3 d co-culture in the dark on the YEB medium containing 150 μmol·L^(-1) acetosyringone(AS). The transient expression rate of GUS gene was 62.6%. After being delayed selective culture for 3 d, infected callus were transferred into the differentiation medium and the root induction medium both of which were supplemented with 100 mg·L^(-1) spectinomycin, and then resistant seedlings of C. sinensis were obtained. 2Optimization of Protein Extraction and Decoloration Conditions for Tea Residues显示文摘To optimize alkaline method for extracting proteins from tea residue(TR), the effect of extraction conditions on tea protein extraction rate(TPER) was investigated. Single factor experiment showed the extraction temperature 80 °C, extraction time 100 min, p H value 13 and liquid–solid ratio 40:1 as the optimal extraction conditions. The orthogonal test revealed that the maximum TPER reached 29.71% under the following optimal combination of conditions: extraction temperature 70 °C, extraction time 60 min, p H 12 and liquid–solid ratio 50:1. For optimizing the purification of tea residue proteins, isoelectric point precipitation(p I), ammonium sulfate precipitation(a S) and isoelectric point plus ammonium sulfate precipitation(i PAS) were compared. The result showed that the highest protein precipitation rate(PPR) was 89.70% which was generated by using i PAS. 3Transcriptomic analyses identify albino-associated genes of a novel albino tea germplasm ‘Huabai 1’显示文摘Albinism in shoots of tea plants is a common phenotypic expression which gives the tea infusion a pleasant umami taste.A novel natural albino mutant tea germplasm containing high amino acids content was found and named as‘Huabai 1’.‘Huabai 1’has white jade tender shoots under low temperature and turns green with increased temperature.In order to understand the molecular mechanism of color change in leaf of‘Huabai 1’,transcriptome analysis was performed to identify albino-associated differentially expressed genes(DEGs).A total of 483 DEGs were identified from white shoots of‘Huabai 1’compared to its green shoots.There were 15 DEGs identified to be involved in phenylpropanoid biosynthesis,which account for the majority of characterized DEGs.The metabolites related to phenylpropanoid biosynthesis revealed similar expression pattern of DEGs.Furthermore,metabolic pathways such as ubiquonone,tyrosine,and flavonoid biosynthesis associated with phenylpropanoid biosynthesis could also contribute to the color change in‘Huabai 1’tender shoots.Protein–protein interaction analysis revealed a hub protein NEDD8(CSA009575)which interacted with many regulated genes in spliceosome,nitrogen metabolism,phenylpropanoid biosynthesis,and other pathways.In conclusion,the findings in this study indicate that the color change of‘Huabai 1’tender shoots is a combined effect of phenylpropanoid biosynthesis pathway and other metabolic pathways including flavonoid biosynthesis in tea plants.Chlorophyll biosynthesis-related genes LHCII and SGR may also play some roles in color change of‘Huabai 1’.Qingping Ma Huan Li Zhongwei Zou Emmanuel Arkorful Qianru Lv Qiongqiong Zhou Xuan Chen Kang Sun Xinghui Li 2018Horticulture Research2018,5,1:7
4Effectiveness of ZiYin Xiehuo granules and Zishen Qinggan granules on partial precocious puberty in girls: a multicenter, randomized, single-blind, controlled trial显示文摘OBJECTIVE: To evaluate the effect of ZiYin Xiehuo granules(ZYXH) and Zishen Qinggan granules(ZSQG) on partial precocious puberty(PPP).METHODS: This was a multicenter, randomized, single-blind, positive-controlled trial. A total of 143 pa-tients were assigned to either the ZYXH group or the ZSQG group using a random number table. The ZYXH group received ZYXH three times daily for 6 months and the ZSQG group received ZSQG three times daily for 6 months. Mammary nucleus diameter; the results of uterus, ovarian, and maximum follicle measures; and Chinese medicine symptom pattern scores were compared at baseline and after 3 months and 6 months of treatment.RESULTS: After 3 months' treatment, there were no significant differences between the two groups in mammary nucleus index changes(left 3.4 ± 3.1 vs3.5 ± 3.1, P = 0.790; right 3.0 ± 2.9 vs 3.6 ± 3.0, P =0.719). The uterine volume in the ZYXH group was smaller than that in the ZSQG group(2.1 ± 1.6 vs2.6 ± 2.2, P = 0.006). There were no significant between-group differences in ovarian volume and maximum follicular diameter on either side(ovarian volume: left 1.2 ± 0.7 vs 1.3 ± 0.6, P = 0.8; right1.2 ± 0.7 vs 1.4 ± 1.1, P = 0.984; maximum follicular diameter: left 3.9 ± 1.7 vs 3.5 ± 2.2, P = 0.158; right3.5 ± 1.7 vs 3.9 ± 2.1, P = 0.314).CONCLUSION: ZYXH granules and ZSQG granules both affected the size of the mammary nucleus in girls with PPP, and improved Chinese medicine symptom patterns. 10Estimation of seawater movement based on reflectors from a seismic profile显示文摘Seismic oceanography is a new cross-discipline of reflection seismology and physical oceanography. The biggest difference between seismic oceanography and traditional reflection seismology is its research object of time-varied seawater. How to estimate the temporal variations of reflectors in water structure and make some corrections in seismic data are basic problems in seismic oceanography research. Here a method of estimation for seawater movement is provided based on the reflectors. The application results of this method to the simulated and field seismic data turn out to be acceptable. 12The E3 ubiquitin ligase NEDD4-1 protects against acetaminophen-induced liver injury by targeting VDAC1 for degradation显示文摘Acetaminophen(APAP)overdose is a major cause of liver injury.Neural precursor cell expressed developmentally downregulated 4—1(NEDD4-1)is an E3 ubiquitin ligase that has been implicated in the pathogenesis of numerous liver diseases;however,its role in APAP-induced liver injury(AILI)is unclear.Thus,this study aimed to investigate the role of NEDD4-1 in the pathogenesis of AILI.We found that NEDD4-1 was dramatically downregulated in response to APAP treatment in mouse livers and isolated mouse hepatocytes.Hepatocyte-specific NEDD4-1 knockout exacerbated APAP-induced mitochondrial damage and the resultant hepatocyte necrosis and liver injury,while hepatocyte-specific NEDD4-1 overexpression mitigated these pathological events both in vivo and in vitro.Additionally,hepatocyte NEDD4-1 deficiency led to marked accumulation of voltage-dependent anion channel 1(VDAC1)and increased VDAC1 oligomerization.Furthermore,VDAC1 knockdown alleviated AILI and weakened the exacerbation of AILI caused by hepatocyte NEDD4-1 deficiency.Mechanistically,NEDD4-1 was found to interact with the PPTY motif of VDAC1 through its WW domain and regulate K48-linked ubiquitination and degradation of VDAC1.Our present study indicates that NEDD4-1 is a suppressor of AILI and functions by regulating the degradation of VDAC1.Yiwei Zhu Lin Lei Xinghui Wang Linfang Chen Wei Li Jinxia Li Chenchen Zhao Xiliang Du Yuxiang Song Wenwen Gao Guowen Liu Xinwei Li 2023Acta Pharmaceutica Sinica B2023,13,4:1
