| 1 | Status and development of high-power laser facilities at the NLHPLP显示文摘In this paper, we review the status of the multifunctional experimental platform at the National Laboratory of High Power Laser and Physics(NLHPLP). The platform, including the SG-II laser facility, SG-II 9th beam, SG-II upgrade(SG-II UP) facility, and SG-II 5 PW facility, is operational and available for interested scientists studying inertial confinement fusion(ICF) and a broad range of high-energy-density physics. These facilities can provide important experimental capabilities by combining different pulse widths of nanosecond, picosecond, and femtosecond scales. In addition, the SG-II UP facility, consisting of a single petawatt system and an eight-beam nanosecond system, is introduced including several laser technologies that have been developed to ensure the performance of the facility. | 4 | Experimental study of separation of ammonia synthesis vent gas by hydrate formation显示文摘Termodynamic data on methane hydrate formation in the presence of ammonia are very important for upgrading of ammonia synthesis vent gas using hydrate formation. This paper is focused on the formation conditions of methane hydrate in the presence of ammonia and the effects of gas-liquid ratio and temperature on the separation of vent gas by hydrate formation. Equilibrium data for methane hydrate within an ammonia mole concentration range from 1% to 5 % were obtained. The experimental results indicated that ammonia has an inhibitive effect on hydrate formation. The higher the ammonia concentration, the higher is the pressure reguired for methane hydrate formation would be. | 6 | Stocking density effects on growth and stress response of juvenile turbot(Scophthalmus maximus) reared in land-based recirculating aquaculture system显示文摘Stocking density is widely recognized as a critical factor in aquaculture and a potential source of long-term stress.The influence of stocking density on growth and stress response of juvenile turbot(Scophthalmus maximus, ~3–75g, initial to final weight) was examined in fish held under low(LD, ~0.21–5.31 kg/m^2, initial to final density),medium(MD, ~0.42–10.81 kg/m^2) and high stocking density(HD, ~0.63–14.27 kg/m^2) for 120 days in a recirculating aquaculture system(RAS). In this trial, the growth curve for weight of juvenile turbot in RAS, all fitted by the Schnute model. No significant difference was found in growth performance among the three densities until at the final sampling(Day 120). The final weight and body weight increase(BWI) in the HD group were significantly lower than in other groups(P<0.05, weight:(75.83±2.49) g,(75.39±2.08) g,(65.72±2.86) g and BWI:(2 436.12±28.10)%,(2 421.29±4.64)%,(2 097.88±20.99)% in LD, MD and HD groups, respectively). Similarly, the specific growth rate(SGR), feed conversion ratio(FCR) and coefficient of variation for weight(CV_w) were adversely affected by high stocking density(P<0.05). However, there was no difference in survival and Fulton's condition factor(K) of turbot among the different groups. Physiological analyses demonstrated a clear increase in the plasma cortisol level and an obvious decrease in growth hormone(GH) concentration in the HD group on Day120(P<0.05). There was no significant effect of stocking density on plasma glucose, Cl– and protein levels. | 8 | The E3 ubiquitin ligase NEDD4-1 protects against acetaminophen-induced liver injury by targeting VDAC1 for degradation显示文摘Acetaminophen(APAP)overdose is a major cause of liver injury.Neural precursor cell expressed developmentally downregulated 4—1(NEDD4-1)is an E3 ubiquitin ligase that has been implicated in the pathogenesis of numerous liver diseases;however,its role in APAP-induced liver injury(AILI)is unclear.Thus,this study aimed to investigate the role of NEDD4-1 in the pathogenesis of AILI.We found that NEDD4-1 was dramatically downregulated in response to APAP treatment in mouse livers and isolated mouse hepatocytes.Hepatocyte-specific NEDD4-1 knockout exacerbated APAP-induced mitochondrial damage and the resultant hepatocyte necrosis and liver injury,while hepatocyte-specific NEDD4-1 overexpression mitigated these pathological events both in vivo and in vitro.Additionally,hepatocyte NEDD4-1 deficiency led to marked accumulation of voltage-dependent anion channel 1(VDAC1)and increased VDAC1 oligomerization.Furthermore,VDAC1 knockdown alleviated AILI and weakened the exacerbation of AILI caused by hepatocyte NEDD4-1 deficiency.Mechanistically,NEDD4-1 was found to interact with the PPTY motif of VDAC1 through its WW domain and regulate K48-linked ubiquitination and degradation of VDAC1.Our present study indicates that NEDD4-1 is a suppressor of AILI and functions by regulating the degradation of VDAC1. | Yiwei Zhu Lin Lei Xinghui Wang Linfang Chen Wei Li Jinxia Li Chenchen Zhao Xiliang Du Yuxiang Song Wenwen Gao Guowen Liu Xinwei Li | 2023 | Acta Pharmaceutica Sinica B2023,13,4: | 1 |

| 12 | Interaction of AcMADS68 with transcription factors regulates anthocyanin biosynthesis in red-fleshed kiwifruit显示文摘In red-fleshed kiwifruit,anthocyanin pigmentation is a crucial commercial trait.The MYB-bHLH-WD40(MBW)complex and other transcription factors regulate its accumulation.Herein,a new SEP gene,AcMADS68,was identified as a regulatory candidate for anthocyanin biosynthesis in the kiwifruit by transcriptome data and bioinformatic analyses.AcMADS68 alone could not induce the accumulation of anthocyanin both in Actinidia arguta fruit and tobacco leaves.However,in combination with AcMYBF110,AcMYB123,and AcbHLH1,AcMADS68 co-overexpression increased anthocyanin biosynthesis,whereas its silencing reduced anthocyanin accumulation.The results of the dual-luciferase reporter,firefly luciferase complementation,yeast two-hybrid and co-immunoprecipitation assays showed that AcMADS68 could interact with both AcMYBF110 and AcMYB123 but not with AcbHLH1,thereby co-regulating anthocyanin biosynthesis by promoting the activation of the target genes,including AcANS,AcF3GT1,and AcGST1.Moreover,AcMADS68 also could activate the promoter of AcbHLH1 surported by dual-luciferase reporter and yeast one-hybrid assays,thereby further amplifying the regulation signals from the MBW complex,thus resulting in enhanced anthocyanin accumulation in the kiwifruit.These findings may facilitate better elucidation of various regulatory mechanisms underlying anthocyanin accumulation and contribute to the quality enhancement of red-fleshed kiwifruit. | Yanfei Liu Guowen Lv Yaqi Yang Kangxun Ma Xiaolin Ren Mingjun Li Zhande Liu | 2023 | Horticulture Research2023,10,2: | 0 |
| 13 | Molecular mechanisms of seasonal photoperiod effects of the pineal gland on the hippocampus in rats显示文摘Background:Based on the theory of“five Zang-organs corresponding to the seasons”in traditional Chinese medicine(TCM),physiological functions including emotions vary with the seasons.We aimed to investigate the seasonal photoperiod effects of melatonin(MT)released from the pineal gland on the MT receptor(MTR)-Gs/Gi-cyclic adenosine monophosphate(cAMP)-protein kinase A(PKA)-cAMP response element-binding protein(CREB)signaling pathway in the hippocampus.Methods:Rats were divided into three groups:control,operation(surgery with pineal gland removal),and pseudo-operation groups(same surgery as operation group but without removing pineal gland),and fed at specific time across the four seasons.The levels of MTR,adenylate cyclase(AC),cAMP,PKA,and CREB in the hippocampus were analyzed using ELISA.The concentrations of Gs and Gi were analyzed using Western blot.The expression of CREB mRNA was detected by PCR.Results:For intragroup comparisons,compared with spring,the levels of Gs/Gi in the control group were higher in summer,autumn,and winter(P=.009 in summer;P<.001 in autumn and winter);the levels of MTR,cAMP,PKA,and CREB in the control group were significantly higher in autumn and winter than in spring(all P<.001).The levels of MTR,cAMP,PKA,and CREB in each season were significantly lower in the operation group than in the control group(all P<.05).Significant differences were noted in Gs/Gi levels between the operation group and control group in spring,autumn,and winter(all P<.05).Conclusion:Our findings suggest that MTR-Gs/Gi-cAMP-PKA-CREB signaling pathway is involved in the seasonal photoperiod effects of the pineal gland on the hippocampus and may underpin seasonal changes in emotions.It can support the prevention and treatment of the seasonal onset of mental disorders,and enrich the theory of“five Zang-organs corresponding to the seasons”. | Qi Han Guowen Du Leilei Liu Lepeng Wang Wenna Li Hairong Zhang Yike Sun Pei Zhu Ruisen Hao Shuran Ma | 2021 | Journal of Traditional Chinese Medical Sciences2021,8,2: | 0 |

Methods: The blood samples were taken from peripheral veins of 70 patients with NSCLC and 18 patients with benign diseases at 3 intervals during the surgery. The transcription of carcinoembryonic antigen messenger ribonucleic acid (CEA mRNA) was assayed by means of nested reverse transcriptase polymerase chain reaction (RT-PCR) and micro-fluid chip. Results: The CEA mRNA positive rates at each of the 3 time spots were as follows: 50% at beginning of the surgery (group 1), 62.8% in the samples collected when ligating the pulmonary vein (group 2) and 57.1% in samples collected 1 h after ligation (group 3). A significant difference was found between groups 1 and 2 (χ2 = 7.114, P < 0.05). | 19 | The effects of pH, surfactant, ion concentration,coformer, and molecular arrangement on the solubility behavior of myricetin cocrystals显示文摘Pharmaceutical cocrystals are a promising technology that can be used to improve the solubility of poor aqueous compounds. The objective of this study was to systematically investigate the solubility of myricetin(MYR) cocrystals, including their kinetic solubility, thermodynamic solubility, and intrinsic dissolution rate(IDR). The effects of pH, surfactant, ion concentration, and coformers on the cocrystal solubility were evaluated. Furthermore, single crystal structures of MYR, myricetin–isonicotinamide(MYR–INM) and myricetin–caffeine(MYR–CAF) cocrystals were analyzed to discuss the possible reasons for the enhancement of cocrystal solubility from the perspective of the spatial structure.The results indicated that the kinetic solubility of MYR cocrystals was modulated by pH and cocrystal coformer(CCF) ionization in buffer solution, while it primarily depended on the CCF solubility in pure water. In addition, the solubility of MYR cocrystals was increased in a concentration dependent fashion by the surfactant or ion concentration. The thermodynamic solubility of MYR–INM(1:3) cocrystals decreased with the increases of the pH value of the dissolution media. The IDR of MYR cocrystals was faster than that of MYR in the same medium and extremely fast in pH 4.5 buffer. The improved solubility of MYR cocrystals was probably related to the alternate arrangements of MYR and INM/CAF molecules and increased intermolecular distance. | 20 | Real-world effectiveness and safety of goserelin 10.8-mg depot in Chinese patients with localized or locally advanced prostate cancer显示文摘Objective:Real-word data on long-acting luteinizing hormone-releasing hormone(LHRH)agonists in Chinese patients with prostate cancer are limited.This study aimed to determine the real-world effectiveness and safety of the LHRH agonist,goserelin,particularly the long-acting 10.8-mg depot formulation,and the follow-up patterns among Chinese prostate cancer patients.Methods:This was a multicenter,prospective,observational study in hormone treatment-na?ve patients with localized or locally advanced prostate cancer who were prescribed goserelin 10.8-mg depot every 12 weeks or 3.6-mg depot every 4 weeks with or without an anti-androgen.The patients had follow-up evaluations for 26 weeks.The primary outcome was the effectiveness of goserelin in reducing serum testosterone and prostate-specific antigen(PSA)levels.The secondary outcomes included testosterone and PSA levels,attainment of chemical castration(serum testosterone<50 ng/d L),and goserelin safety.The exploratory outcome was the monitoring pattern for serum testosterone and PSA.All analyses were descriptive.Results:Between September 2017 and December 2019,a total of 294 eligible patients received≥1 dose of goserelin;287 patients(97.6%)were treated with goserelin 10.8-mg depot.At week 24±2,the changes from baseline[standard deviation(95%confidence interval)]in serum testosterone(n=99)and PSA(n=131)were-401.0 ng/d L[308.4 ng/d L(-462.5,-339.5 ng/d L)]and-35.4 ng/m L[104.4 ng/m L(-53.5,-17.4 ng/m L)],respectively.Of 112 evaluable patients,100(90.2%)achieved a serum testosterone level<50 ng/d L.Treatment-emergent adverse events(TEAEs)and severe TEAEs occurred in 37.1%and 10.2%of patients,respectively.The mean testing frequency(standard deviation)was 1.6(1.5)for testosterone and 2.2(1.6)for PSA.Conclusions:Goserelin 10.8-mg depot effectively achieved and maintained castration and was well-tolerated in Chinese patients with localized and locally advanced prostate cancer. | Nanhui Chen Zengjun Wang Ming Chen Qi Ma Yi He Yujie Wang Xin Li Mingxing Qiu Lei Shi Shaoxing Zhu Qun Xie Xiuheng Liu Benkang Shi Guowen Lin Weizhong Yang Yongbin Liao Haibin Zhang Shusheng Wang Jiexian Li Shaogang Wang Lijun Dong Hui Chen Jiaju Lu YongyiCheng Xiaoping Zhang Lulin Ma Liqun Zhou He Wang Shen Li Dingwei Ye | 2023 | Cancer Biology & Medicine2023,20,12: | 0 |
