1Tumor immune checkpoints and their associated inhibitors显示文摘Immunological evasion is one of the defining characteristics of cancers,as the immune modification of an immune checkpoint(IC)confers immune evasion capabilities to tumor cells.Multiple ICs,such as programmed cell death protein-1(PD-1)and cytotoxic T-lymphocyte-associated antigen-4(CTLA-4),can bind to their respective receptors and reduce tumor immunity in a variety of ways,including blocking immune cell activation signals.IC blockade(ICB)therapies targeting these checkpoint molecules have demonstrated significant clinical benefits.This is because antibody-based IC inhibitors and a variety of specific small molecule inhibitors can inhibit key oncogenic signaling pathways and induce durable tumor remission in patients with a variety of cancers.Deciphering the roles and regulatory mechanisms of these IC molecules will provide crucial theoretical guidance for clinical treatment.In this review,we summarize the current knowledge on the functional and regulatory mechanisms of these IC molecules at multiple levels,including epigenetic regulation,transcriptional regulation,and post-translational modifications.In addition,we provide a summary of the medications targeting various nodes in the regulatory pathway,and highlight the potential of newly identified IC molecules,focusing on their potential implications for cancer diagnostics and immunotherapy.Zerui GAO Xingyi LING Chengyu SHI Ying WANG Aifu LIN 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,10:4

3Tandem Molecular Self-Assembly Selectively Inhibits Lung Cancer Cells by Inducing Endoplasmic Reticulum Stress显示文摘The selective formation of nanomaterials in cancer cells and tumors holds great promise for cancer diagnostics and therapy.Until now,most strategies rely on a single trigger to control the formation of nanomaterials in situ.The combination of two or more triggers may provide for more sophisticated means of manipulation.In this study,we rationally designed a molecule(Comp.1)capable of responding to two enzymes,alkaline phosphatase(ALP),and reductase.Since the A549 lung cancer cell line showed elevated levels of extracellular ALP and intracellular reductase,we demonstrated that Comp.1 responded in a stepwise fashion to those two enzymes and displayed a tandem molecular self-assembly behavior.The selective formation of nanofibers in the mitochondria of the lung cancer cells led to the disruption of the mitochondrial membrane,resulting in an increased level of reactive oxygen species(ROS)and the release of cytochrome C(Cyt C).ROS can react with proteins,resulting in endoplasmic reticulum(ER)stress and the unfolded protein response(UPR).This severe ER stress led to disruption of the ER,formation of vacuoles,and ultimately,apoptosis of the A549 cells.Therefore,Comp.1 could selectively inhibit lung cancer cells in vitro and A549 xenograft tumors in vivo.Our study provides a novel strategy for the selective formation of nanomaterials in lung cancer cells,which is powerful and promising for the diagnosis and treatment of lung cancer.Debin Zheng Yumiao Chen Sifan Ai Renshu Zhang Zhengfeng Gao Chunhui Liang Li Cao Yaoxia Chen Zhangyong Hong Yang Shi Ling Wang Xingyi Li Zhimou Yang 2019Research2019,,1:1

6Protective effect of alpha-synuclein knockdown on methamphetamine-induced neurotoxicity in dopaminergic neurons显示文摘The over-expression of α-synuclein is a major factor in the death of dopaminergic neurons in a methamphetamine-induced model of Parkinson's disease. In the present study, α-synuclein knockdown rats were created by injecting α-synuclein-shRNA lentivirus stereotaxically into the right striatum of experimental rats. At 2 weeks post-injection, the rats were injected intraper-itoneally with methamphetamine to establish the model of Parkinson's disease. Expression of α-synuclein mRNA and protein in the right striatum of the injected rats was significantly down-regulated. Food intake and body weight were greater in α-synuclein knockdown rats, and water intake and stereotyped behavior score were lower than in model rats. Striatal dopamine and tyrosine hydroxylase levels were significantly elevated in α-synuclein knockdown rats. Moreover, superoxide dismutase activity was greater in α-synuclein knockdown rat striatum, but the levels of reactive oxygen species, malondialdehyde, nitric oxide synthase and nitrogen monoxide were lower compared with model rats. We also found that α-synuclein knockdown inhibited metham-phetamine-induced neuronal apoptosis. 7Coevolutionary Framework for Generalized Multimodal Multi-Objective Optimization显示文摘Most multimodal multi-objective evolutionary algorithms(MMEAs)aim to find all global Pareto optimal sets(PSs)for a multimodal multi-objective optimization problem(MMOP).However,in real-world problems,decision makers(DMs)may be also interested in local PSs.Also,searching for both global and local PSs is more general in view of dealing with MMOPs,which can be seen as generalized MMOPs.Moreover,most state-of-theart MMEAs exhibit poor convergence on high-dimension MMOPs and are unable to deal with constrained MMOPs.To address the above issues,we present a novel multimodal multiobjective coevolutionary algorithm(Co MMEA)to better produce both global and local PSs,and simultaneously,to improve the convergence performance in dealing with high-dimension MMOPs.Specifically,the Co MMEA introduces two archives to the search process,and coevolves them simultaneously through effective knowledge transfer.The convergence archive assists the Co MMEA to quickly approach the Pareto optimal front.The knowledge of the converged solutions is then transferred to the diversity archive which utilizes the local convergence indicator and the-dominance-based method to obtain global and local PSs effectively.Experimental results show that Co MMEA is competitive compared to seven state-of-the-art MMEAs on fifty-four complex MMOPs.Wenhua Li Xingyi Yao Kaiwen Li Rui Wang Tao Zhang Ling Wang 2023IEEE/CAA Journal of Automatica Sinica2023,10,7:0
8Wetting behavior of AgCu-4.5Ti filler reinforced by carbon nanotubes on C/C composite显示文摘The effect of Carbon Nanotubes(CNTs)content on the wettability of AgCu-4.5Ti+x CNTs(wt%)composite filler alloys on C/C composite was investigated.The results show that the added CNTs reacted with element Ti in the filler and produced the dispersed fine in situ synthesized TiC particles,which increased the consumption of element Ti and provided the nucleus for the growth of Ti-Cu compounds simultaneously.The above effects of introducing CNTs,inhibited the formation of Ti-Cu compounds,also changed the distribution of compounds,which dramatically influenced the interfacial microstructure and characteristics of wetting behavior.The increase of CNTs content refined and dispersed coarse Ti-Cu compounds,decreased the initial spreading temperature,and improved the wettability,but high content of CNTs(more than 0.3 wt%)decreased the wettability of the filler alloy.The wetting interfacial microstructure of corresponding composite filler alloys were analyzed by Scanning Electron Microscope(SEM),Energy Dispersive X-ray Spectrometer(EDS)and Transmission Electron Microscope(TEM),which consisted of TiC,TiCu,TiCu;and TiCu;compound.The typical wetting behavior of AgCu-4.5Ti+0.3wt%CNTs composite filler on C/C composite was divided into four stages.The effect mechanism of CNTs content on the wetting behavior was proposed.Xingyi LI Ling LIU Yanyu SONG Duo LIU Shengpeng HU Xiaoguo SONG Jian CAO 2021Chinese Journal of Aeronautics2021,34,12:0
