3Development of a PDRA Method for Detection of the D614G Mutation in COVID-19 Virus—Worldwide,2021显示文摘ABSTRACT Background:COVID-19 infection is a major public health problem worldwide,and the D614G mutation enhances the infectivity of COVID-19.Methods:A probe-directed recombinase amplification(PDRA)assay was discussed to detect the D614G mutation at 39℃for 30 min.The sensitivity,specificity,and reproducibility of the PDRA were evaluated by D614 and G614 recombinant plasmids.The clinical performance of PDRA assay was validated by testing of 53 previously confirmed COVID-19 positive RNAs and 10 negative samples.Direct sequencing was carried out in parallel for comparison.Result:With good reproducibility and specificity,the PDRA assay worked well with the concentration in the range of 103–107 copies/reaction.Compared with direct sequencing as a reference,the recombinase-aided amplification(RAA)assay obtained 100%sensitivity and 100%specificity using clinical samples.Ziwei Chen Xinxin Shen Ji Wang Xiang Zhao Yuan Gao Ruiqin Zhang Jinrong Wang Leping Liu Xinmin Nie Xuejun Ma 2021China CDC weekly2021,3,21:1
4Spatial distribution of health risks for residents located close to solvent-consuming industrial VOC emission sources显示文摘Emissions derived from the consumption of organic solvents have been proven to be the primary industrial source of volatile organic compounds(VOCs).In conjunction with epidemiologic studies, water-based paints(WBPs) and solvent-based paints(SBPs) were selected as representatives of newly developed solvents and traditional solvents, respectively,to simulate the effects of consuming solvents emitted during industrial production.And non-carcinogenic and carcinogenic risks to residents near emission sources were studied in detail.The results showed that the spatial distribution of health risks varied with meteorological conditions and type of emission source, and the prevailing wind direction strongly affected the distribution range and shape of the influenced area.The areas of influence maximized on heavy-polluting days for both WBP and SBP emission sources with the total span reaching 804 m and 16 km, respectively;meanwhile, the areas of influence for carcinogenic risk resulting from WBP emission sources were 1.2 and 2.3 times greater than those measured on fine and rainy days, respectively, and 1.8 and 2.9 times greater for SBP emission sources.Compared with WBPs, the total spans of negatively influenced regions resulting from SBP emission sources were 10.4, 12.5 and 19.9 times greater on fine, rainy and heavypolluting days, respectively.Therefore, carcinogenic risk was the dominant health threat for populations residing close to solvent-consuming industrial emission sources.The findings suggest that newly developed solvents are capable of significantly reducing consequent health threats, nevertheless, they could still pose occasional threats to nearby residents under specific meteorological conditions.Di Wang Xuan Li Xinmin Zhang Wenjuan Zhao Weiqi Zhang Shuaifeng Wu Xia Shao Lei Nie 2021Journal of Environmental Sciences2021,33,9:1
5支气管软镜下冷冻消融后冻切法即刻治疗中央气道阻塞(英文)显示文摘Objective:In order to achieve immediate relief of central airway obstruction caused by malignant tumor after interventional therapy, we observed the efficacy and safety of cryorecanalization after cryosurgery via flexible bronchoscope. Methods:Atotal of 64 cases of patients in all suffered from central airway obstruction were observed and treated by applying cryorecanalization after cryosurgery via flexible bronchoscope. Divide the operation into two steps, including cryosurgery and cryorecanalization. Evaluate the therapeutic effect immediately after the treatment, wherein the evaluating indicator includes preoperative and postoperative dyspnea indexes, quality of life score, bronchoscopy, etc. The surgical safety assessment is mainly by observing the risk of bleeding in the surgery. Results:Forty cases (62.5%) has postoperative dyspnea score improved at least one level compared with preoperative dyspnea score, 24 patients (37.5%) have no significant improvement on dyspnea score. Quality of life score (Karnofsky score):preoperative 52±18.7 points, postoperative 70±9.2 points. Bronchoscopy review:complete suecess in 12 cases (18.8%), partial success in 45 cases (70.3%), no success in 7 cases (12%). 89.1% overall clinical efficiency was achieved. In the surgery, the control to bleeding was satisfactory since no severe bleeding, moderate bleeding in 14 cases (21.9%) and mild bleeding in 50 cases (78.1%) were found as well as rigid bronchoscope was free.
