1Ferritin as a diagnostic, differential diagnostic, and prognostic marker for immune-related adverse events显示文摘Objective:Distinguishing immune-related adverse events(irAEs)caused by immune checkpoint inhibitors(ICIs)from the AEs caused by chemotherapy,targeted therapy,or infection is highly difficult.This study offers new insights into evaluating the diagnosis,differential diagnostic,and prognostic value of ferritin for irAEs induced by ICIs.Methods:From December 1,2018,to April 1,2019,we examined 318 patients with malignant tumors who received serum ferritin monitoring.The cohort comprised 231 patients treated with PD-1 inhibitor or combination with chemotherapy,and 87 patients treated with chemotherapy.Of the 231 patients,90 had irAEs(irAE group),70 had non-irAEs(non-irAE group),67 had no AEs(no irAE-non irAE group),and 4 had unclassified AEs.In the 87 patients,60 had AEs(AE group),and 27 had no AEs(no AE group).Statistical analyses were conducted with nonparametric Mann-Whitney tests.Results:At the onset of AEs in the irAE group,ferritin(normal range,35–150μg/L)rose to a median of 927μg/L(range,117–17,825μg/L)from 86μg/L at baseline(range,29–421μg/L)(P<0.001).Ferritin levels at the onset of AEs in the irAE group were significantly higher than those in the non-irAE group(median,81μg/L;range,32–478μg/L)(P<0.001)and the AE group(median,103μg/L;range,23–712μg/L)(P<0.001).After treatment in the irAE group,ferritin continuously decreased to a normal range in recovered patients,showed no significant changes in stable patients,and continued to rise in patients who died.Conclusions:Ferritin can be used as a diagnostic,differential diagnostic,and prognostic marker for irAEs in patients treated with ICIs.Weihong Zhang Yuan Meng Lin Yang Meng Shen Li Zhou Runmei Li Yang Wang Weijiao Du Yanjuan Xiong Ying Han Xinwei Zhang Liang Liu Xiubao Ren 2021Cancer Biology & Medicine2021,18,4:4
2Joint timing and frequency synchronization in coherent optical OFDM systems显示文摘In this paper,we review our joint timing and frequency synchronization algorithms in coherent optical orthogonal frequency division multiplexing(CO-OFDM)systems.We first present a timing estimation method by designing the pattern of the training symbol,whose timing metric has a sharp and clear peak,to ensure accurate timing offset(TO)estimation performance.Then we provide both data-aided(DA)and blind(BL)approaches to estimate the carrier frequency offset(CFO).For the DA algorithm,we utilize the same training symbol structure as the timing estimation does,while for the BL algorithm,we utilize the zero-subcarrier power(ZSP)to achieve full-range CFO estimation.Note that our joint timing and frequency synchronization approaches require only one OFDM symbol,which ensure not only the data transmission efficiency,but also the TO and CFO estimation performance.A modified BL ZSP algorithm is proposed to further improve the CFO estimation performance by taking the power average over a series of OFDM symbols.The effectiveness of the TO estimation algorithm,and both the DA and BL CFO estimation algorithms are verified and demonstrated in both simulations and experiments.Xinwei DU Pooi-Yuen KAM Changyuan YU 2019Frontiers of Optoelectronics2019,12,1:3
3Al2O3 supported hybrid Pd-CeO2 colloidal spheres and its enhanced catalytic performances for methane combustion显示文摘Fabrication of novel nano-catalyst with improved activity and stability has been a hot topic in heterogeneous catalysis. Herein a novel approach was designed to synthesize Pd-CeO2 colloidal assembled spheres(CASs) in one-pot fashion. The Pd clusters were encompassed by the CeO2 nanocrystals with a high dispersity. Based on this hybrid structure, the Pd/ceria interface area can be greatly improved. After Pd-CeO2 CASs was supported on γ-Al2O3, its catalytic activity for CH4 combustion was evaluated. The result shows that Al2O3 supported Pd-CeO2 CASs exhibits an improved catalytic activity and stability,compared to the Pd/CeO2 catalyst prepared by impregnation method. 4Nuciferine protects against high-fat diet-induced hepatic steatosis and insulin resistance via activating TFEB-mediated autophagy——lysosomal pathway显示文摘Nonalcoholic fatty liver disease(NAFLD) is characterized by hepatic steatosis and insulin resistance and there are currently no approved drugs for its treatment.Hyperactivation of mTOR complex1(mTORCl) and subsequent impairment of the transcription factor EB(TFEB)-mediated autophagy-lysosomal pathway(ALP) are implicated in the development of NAFLD.Accordingly,agents that augment hepatic TFEB transcriptional activity may have therapeutic potential against NAFLD.The objective of this study was to investigate the effects of nuciferine,a major active component from lotus leaf,on NAFLD and its underlying mechanism of action.Here we show that nuciferine activated ALP and alleviated steatosis,insulin resistance in the livers of NAFLD mice and palmitic acid-challenged hepatocytes in a TFEB-dependent manner.Mechanistic investigation revealed that nuciferine interacts with the Ragulator subunit hepatitis B X-interacting protein and impairs the interaction of the Ragulator complex with Rag GTPases,thereby suppressing lysosomal localization and activity of mTORC1,which activates TFEB-mediated ALP and further ameliorates hepatic steatosis and insulin resistance.Our present results indicate that nuciferine may be a potential agent for treating NAFLD and that regulation of the mTORCl-TFEB-ALP axis could represent a novel pharmacological strategy to combat NAFLD.Xiliang Du Chiara Di Malta Zhiyuan Fang Taiyu Shen Xiaodi Niu Meng Chen Bo Jin Hao Yu Lin Lei Wenwen Gao Yuxiang Song Zhe Wang Chuang Xu Zhijun Cao Guowen Liu Xinwei Li 2022Acta Pharmaceutica Sinica B2022,12,6:2
5The E3 ubiquitin ligase NEDD4-1 protects against acetaminophen-induced liver injury by targeting VDAC1 for degradation显示文摘Acetaminophen(APAP)overdose is a major cause of liver injury.Neural precursor cell expressed developmentally downregulated 4—1(NEDD4-1)is an E3 ubiquitin ligase that has been implicated in the pathogenesis of numerous liver diseases;however,its role in APAP-induced liver injury(AILI)is unclear.Thus,this study aimed to investigate the role of NEDD4-1 in the pathogenesis of AILI.We found that NEDD4-1 was dramatically downregulated in response to APAP treatment in mouse livers and isolated mouse hepatocytes.Hepatocyte-specific NEDD4-1 knockout exacerbated APAP-induced mitochondrial damage and the resultant hepatocyte necrosis and liver injury,while hepatocyte-specific NEDD4-1 overexpression mitigated these pathological events both in vivo and in vitro.Additionally,hepatocyte NEDD4-1 deficiency led to marked accumulation of voltage-dependent anion channel 1(VDAC1)and increased VDAC1 oligomerization.Furthermore,VDAC1 knockdown alleviated AILI and weakened the exacerbation of AILI caused by hepatocyte NEDD4-1 deficiency.Mechanistically,NEDD4-1 was found to interact with the PPTY motif of VDAC1 through its WW domain and regulate K48-linked ubiquitination and degradation of VDAC1.Our present study indicates that NEDD4-1 is a suppressor of AILI and functions by regulating the degradation of VDAC1.Yiwei Zhu Lin Lei Xinghui Wang Linfang Chen Wei Li Jinxia Li Chenchen Zhao Xiliang Du Yuxiang Song Wenwen Gao Guowen Liu Xinwei Li 2023Acta Pharmaceutica Sinica B2023,13,4:1

7Enzymatic synthesis of indigo derivatives by tuning P450 BM3 peroxygenases显示文摘Indigoids,a class of bis-indoles,have long been applied in dyeing,food,and pharmaceutical industries.Recently,interest in these‘old’molecules has been renewed in the field of organic semiconductors as functional building blocks for organic electronics due to their excellent chemical and physical properties.However,these indigo derivatives are difficult to access through chemical synthesis.In this study,we engineer cytochrome P450 BM3 from an NADPH-dependent monooxygenase to peroxygenases through directed evolution.A select number of P450 BM3 variants are used for the selective oxidation of indole derivatives to form different indigoid pigments with a spectrum of colors.Among the prepared indigoid organic photocatalysts,a majority of indigoids demonstrate a reduced band gap than indigo due to the increased light capture and improved charge separation,making them promising candidates for the development of new organic electronic devices.Thus,we present a useful enzymatic approach with broad substrate scope and cost-effectiveness by using low-cost H2O2 as a cofactor for the preparation of diversified indigoids,offering versatility in designing and manufacturing new dyestuff and electronic/sensor components.Li Ma Tianjian Sun Yunjie Liu Yue Zhao Xiaohui Liu Yuxuan Li Xinwei Chen Lin Cao Qianqian Kang Jiawei Guo Lei Du Wei Wang Shengying Li 2023Synthetic and Systems Biotechnology2023,8,3:0

12Proton Conductivity Improvement Effect of Cellulose on SPEEKK Based PEM显示文摘The proton exchange membranes(PEMs) were prepared through the solution mixing method of sulfonated poly(etlier ether ketone ketoneXsPEEKK) and cellulose. Cellulose was dissolved by 1-ally 1-3-methylimidazolium chioride(AMIMC1) and then mixed with sPEEKK solution. sPEEKK/cellulose(SC) composite membranes were prepared by solution casting. The membranes have high flexibility and transparency, which meant the compounding in molecular level. Meanwhile, the composite membranes showed excellent mechanical properties and high proton conductivity. The mechanical property reached 29 MPa, and the proton conductivity was as high as 0.32 S/cm. 13Fault diagnosis for lithium-ion batteries in electric vehicles based on signal decomposition and two-dimensional feature clustering显示文摘Battery fault diagnosis is essential for ensuring the reliability and safety of electric vehicles(EVs).The existing battery fault diagnosis methods are difficult to detect faults at an early stage based on the real-world vehicle data since lithium-ion battery systems are usually accompanied by inconsistencies,which are difficult to distinguish from faults.A fault diagnosis method based on signal decomposition and two-dimensional feature clustering is introduced in this paper.Symplectic geometry mode decomposition(SGMD)is introduced to obtain the components characterizing battery states,and distance-based similarity measures with the normalized extended average voltage and dynamic time warping distances are established to evaluate the state of batteries.The 2-dimensional feature clustering based on DBSCAN is developed to reduce the number of feature thresholds and differentiate flaw cells from the battery pack with only one parameter under a wide range of values.The proposed method can achieve fault diagnosis and voltage anomaly identification as early as 43 days ahead of the thermal runaway.And the results of four electric vehicles and the comparison with other traditional methods validated the proposed method with strong robustness,high reliability,and long time scale warning,and the method is easy to implement online.Shuowei Li Caiping Zhang Jingcai Du Xinwei Cong Linjing Zhang Yan Jiang Leyi Wang 2022Green Energy and Intelligent Transportation2022,1,1:0
