| 1 | Do land utilization patterns affect methanotrophic communities in a Chinese upland red soil?显示文摘Soil samples were collected from three plots under different land utilization patterns including degradation,farming,and restoration.The abundances of methanotrophs were quantified using real-time polymerase chain reaction (PCR) based on the pmoA and 16S rRNA genes,and the community fingerprint was analyzed using denaturing gradient gel electrophoresis (DGGE) aiming at pmoA gene.Significantly lower 16S rRNA and pmoA genes copies were found in the degradation treatment than in farming and restoration.Higher abundances of Type I than those of Type II methanotrophs were detected in all treatments.The treatment of farming was clearly separated from degradation and restoration according to the DGGE profile by cluster analysis.The lowest diversity indices were observed in the F (farming plot),suggesting that the community structure was strongly affected by farming activities.There were significantly positive correlations between the copy numbers of pmoA also Type II-related 16S rRNA genes and soil available K content.Strong negative and positive correlations were found between Type I and soil pH,and available P content,respectively.We concluded that the vegetation cover or not,soil characteristics including pH and nutrients of P and K as a result of anthropogenic disturbance may be key factors affecting methanotrophic communities in upland soil. | Yong Zheng Xinzhan Liu Limei Zhang Zhifeng Zhou Jizheng He | 2010 | Journal of Environmental Sciences2010,22,12: | 4 |
| 2 | A multiplex microsatellite PCR method for evaluating genetic diversity in grass carp(Ctenopharyngodon idellus)显示文摘Grass carp is one of the most important cultured fishes all over the world.The genetic diversity of grass carp plays an important role whatever in selective breeding progress or ecological conservation purposes.However,some genetic diversity researches were not accuracy and cannot be compared with each other due to different molecular markers,sample size and detection methods.In this study,we constructed five multiplex PCR assays contained 20 microsatellites with highly polymorphism and heterozygosity for evaluating genetic information of grass carp.We used nine cultured populations consisting of 507 individuals to detect stability of the five multiplex PCR assays.The results showed that the number of alleles(Na),effective number of alleles(Ne),observed heterozygosity(Ho),expected heterozygosity(He)and polymorphism information content(PIC)of the 20 microsatellite loci were relative high compared with the genetic diversity parameters of microsatellite loci developed by other researchers.Six loci were significantly deviated from Hardy-Weinberg equilibrium(P<0.01).And with exception of the Shaoguan,Indian and Nepal cultured population,all other cultured populations showed very high genetic diversity.Through the test of grass carp populations,we developed an effective and accurate multiplex SSR-PCR assays that can be as statistical powerful tool for detecting genetic information of grass carp. | Da Li Shentong Wang Yubang Shen Xinzhan Meng Xiaoyan Xu Rongquan Wang Jiale Li | 2018 | Aquaculture and Fisheries2018,3,6: | 4 |

| 4 | Experimental and Theoretical Study on the Flexural Behavior of Recycled Concrete Beams Reinforced with GFRP Bars显示文摘This paper experimentally investigated the flexural behavior of reinforced recycled aggregate concrete(RAC)beams reinforced with glass fiber-reinforced polymer(GFRP)bars.A total of twelve beams were built and tested up to failure under four-point bending.The main parameters were reinforcement ratio(0.38%,0.60%,and 1.17%),recycled aggregate replacement ratio(R=0,50%,and 100%)and longitudinal reinforcement types(GFRP and steel).The flexural capacity,failure modes,flexibility deformation,reinforcement strains and crack distribution of the tested beams were investigated and compared with the calculation models of American code ACI 440.1-R-15,Canadian code CSA S806-12 and ISIS-M03-07.The tested results indicated that the reinforcement ratio has great influence on the ultimate load,crack width and deflection of GFRP-RAC beams,the recycled aggregate replacement ratio has little influence on it.However,it was found that the reinforcement ratio has no obvious influence on the cracking load which was only related to the recycled aggregate replacement ratio.The average cracking load decreased by 5%and 15%as the recycled aggregate replacement ratio increased from 0 to 50%and 100%.For the steel-RAC beams,the ultimate load was found to be about 1/2 of the ultimate load of GFRP-RAC beam under the same condition and the trend of strain,deflection and crack width were different from GFRP-RAC beams.This is due to the different material properties of GFRP bars and steel rebar.On the other hand,the calculation results showed that ACI 440.1-R-15 and CSA S806-12 underestimated the ultimate load of GFRP-RAC beams.Moreover,the deflection prediction of GFRP-RAC beams by CSA S806-12 is relatively accurate compared with ACI 440.1-R-15 and ISIS-M03-07.As for the prediction of crack width,the results of ACI 440.1-R-15 prediction were in good agreement with the experimental results at the ultimate load,with the average value of 1.09±0.28. | Xinzhan Chen Xiangqing Kong Ying Fu Wanting Sun Renguo Guan | 2021 | Journal of Renewable Materials2021,9,6: | 0 |
