22018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 3Improvement of wheat drought and salt tolerance by expression of a stress- inducible transcription factorGmDREB of soybean (Glycinemax)显示文摘Under stress conditions such as droughthigh-salinity and low-temperature, the transcription factorof DREB (dehydration responsive element binding proteins)improved efficiently stress resistance by regulating the ex-pression of its downstream genes with various environmentastress resistance in plants. GmDREB gene (GenBank Acces-sion No. AF514908) encoding a stress-inducible transcriptionfactor was cloned by screening a cDNA library of Glycinemax cv. Jinong 27 with yeast one-hybrid method. GmDREBgene was 910 bp in length and encoded 174 amino acids con-taining a conserved AP2/EREBP DNA-binding domain of 58amino acids. Two conserved functional amino acids, valineand glutamic acid, were located on the 14th and the 19thamino acid residues in the conserved structural domain. Analkaline amino acid region (KKR) related to a nuclear local-ization signal was at the N-terminal, while an acidic aminoacid region (DDD) related to trans-activation was at theC-terminal. Plant expression vectors were constructed andtransformed into wheat by bombardment. In total, 13 trans-genic plants with Ubi::GmDREB and 11 transgenic plantswith rd29A::GmDREB were identified from 103 regenerationplants by molecular analysis. The drought and salt tolerancesof T1 transgenic lines with Ubi::GmDREB orrd29A::GmDREB were demonstrated to be improved ascompared to wild type. 4ZIKA virus isolated from mosquitoes:a field and laboratory investigation in China,2016显示文摘A field investigation of arboviruses was conducted in Dejiang, Guizhou Province in the summer of 2016. A total of 8,795 mosquitoes, belonging to four species of three genera, and 1,300 midges were collected. The mosquito samples were identified on site according to their morphology, and the pooled samples were ground and centrifuged in the laboratory. The supernatant was incubated with mosquito tissue culture cells(C6/36) and mammalian cells(BHK-21) for virus isolation. The results indicated that 40%(3,540/8,795) were Anopheles sinensis, 30%(2,700/8,795) were Culex pipiens quinquefasciatus, and 29%(2,530/8,795)were Armigeres subbalbeatus. Furthermore, a total of eight virus isolates were obtained, and genome sequencing revealed two Zika viruses(ZIKVs) isolated from Culex pipiens quinquefasciatus and Armigeres subbalbeatus, respectively; three Japanese encephalitis viruses(JEVs) isolated from Culex pipiens quinquefasciatus; two Banna viruses(BAVs) isolated from Culex pipiens quinquefasciatus and Anopheles sinensis, respectively; and one densovirus(DNV) isolated from Culex pipiens quinquefasciatus.The ZIKVs isolated from the Culex pipiens quinquefasciatus and Armigeres subbalbeatus mosquitoes represent the first ZIKV isolates in China's Mainland. 5Cardamonin from a medicinal herb protects against LPS-induced septic shock by suppressing NLRP3 inflammasome显示文摘Aberrant activation of NLRP3 inflammasome has been implicated in the pathogenesis of diverse inflammation-related diseases, and pharmacological molecules targeting NLRP3 inflammasome are of considerable value to identifying potential therapeutic interventions. Cardamonin(CDN), the major active ingredient of the traditional Chinese medicinal herb Alpinia katsumadai, has exerted an excellent anti-inflammatory activity, but the mechanism underlying this role is not fully understood. Here, we show that CDN blocks canonical and noncanonical NLRP3 inflammasome activation triggered by multiple stimuli. Moreover, the suppression of CDN on inflammasome activation is specific to NLRP3, not to NLRC4 or AIM2 inflammasome. Besides, the inhibitory effect is not dependent on the expression of NF-κB-mediated inflammasome precursor proteins. We also demonstrate that CDN suppresses the NLRP3 inflammasome through blocking ASC oligomerization and speckle formation in a dose-dependent manner.Importantly, CDN improves the survival of mice suffering from lethal septic shock and attenuates IL-1βproduction induced by LPS in vivo, which is shown to be NLRP3 dependent. 7Blocking FSH inhibits hepatic cholesterol biosynthesis and reduces serum cholesterol显示文摘Menopause is associated with dyslipidemia and an increased risk of cardio-cerebrovascular disease.The classic view assumes that the underlying mechanism of dyslipidemia is attributed to an insufficiency of estrogen.In addition to a decrease in estrogen, circulating follicle-stimulating hormone (FSH)levels become elevated at menopause.In this study,we find that blocking FSH reduces serum cholesterol via inhibiting hepatic cholesterol biosynthesis.First,epidemiological results show that the serum FSH levels are positively correlated with the serum total cholesterol levels,even after adjustment by considering the effects of serum estrogen,in addition,the prevalence of hypercholesterolemia is significantly higher in peri-menopausal women than that in premenopausal women.Furthermore,we generated a mouse model of FSH elevation by intraperitoneally injecting exogenous FSH into ovariectomized (OVX)mice,in which a normal level of estrogen (E2)was maintained by exogenous supplementation. Consistently,the results indicate that FSH,independent of estrogen,increases the serum cholesterol level in this mouse model. 8Antioxidant and immunomodulatory activities of a polysaccharide from Flammulina velutipes显示文摘OBJECTIVE: To evaluate the antioxidant and immunomodulatory activities of a unique polysaccharide from the medicinal fungus Flammulina velutipes in vitro.METHODS: Using water extraction and alcohol precipitation, crude polysaccharides were obtained. After purification by DEAE-cellulose 52 ion exchange chromatography and Sephacryl S-300 HR gel filtration chromatography, High performance liquid chromatography equipped with evaporative light-scattering detector, Infrared radiation and Nuclear magnetic resonance were used to evaluate the structure of the polysaccharide. Its immunomodulatory activity was measured by examining the production of nitric oxide(NO) and cytokine secretion, and via lymphocyte proliferation experiments. Its effects on the scavenging activities of hydroxyl radical, superoxide anion and reducing power were also measured.RESULTS: A water-soluble polysaccharide, Flammulina velutipes polysaccharide I-A(FVP I-A), was obtained with a molecular mass of 8.14×104Da determined by high performance gel permeation chromatography. An in vitro antioxidant assay indicated that FVP I-A could scavenge hydroxyl radical, superoxide anion and possessed reducing power and could largely promote NO production and augment the interleukin-1β, interleukin-6, and tumor necrosis factor-α secretion by RAW264.7 macrophages(P<0.05). 9Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 12Circadian Rhythm of Calling Behavior in the Emei Music Frog(Babina daunchina) is Associated with Habitat Temperature and Relative Humidity显示文摘Generally, the function of vocalizations made by male anurans are to attract females or defend resources. Typically, males vocalize in choruses during one or more periods in a twenty-four-hour cycle, which varies, however, among species. Nevertheless, the causal factors influencing circadian variations of calling patterns in anuran species are not clear. In this study, male chorus vocalizations were monitored in the Emei music frog(Babina daunchina) for 17 consecutive days during the breeding season, while its habitat air temperature and relative humidity in the course of experiments were measured as well. The results revealed that the circadian calling patterns were characterized by two periods of peak vocalization, which were observed from 0500 h to 0700 h and from 1300 h to 2000 h, while the lowest activity period was found from 2100 h to 2200 h. Both calls/h and notes/h were positively correlated with air temperature and negatively with relative humidity. 13Effects of long-term warming on the aboveground biomass and species diversity in an alpine meadow on the Qinghai-Tibetan Plateau of China显示文摘Ecosystems in high-altitude regions are more sensitive and respond more rapidly than other ecosystems to global climate warming.The Qinghai-Tibet Plateau(QTP)of China is an ecologically fragile zone that is sensitive to global climate warming.It is of great importance to study the changes in aboveground biomass and species diversity of alpine meadows on the QTP under predicted future climate warming.In this study,we selected an alpine meadow on the QTP as the study object and used infrared radiators as the warming device for a simulation experiment over eight years(2011-2018).We then analyzed the dynamic changes in aboveground biomass and species diversity of the alpine meadow at different time scales,including an early stage of warming(2011-2013)and a late stage of warming(2016-2018),in order to explore the response of alpine meadows to short-term(three years)and long-term warming(eight years).The results showed that the short-term warming increased air temperature by 0.31℃and decreased relative humidity by 2.54%,resulting in the air being warmer and drier.The long-term warming increased air temperature and relative humidity by 0.19℃and 1.47%,respectively,and the air tended to be warmer and wetter.The short-term warming increased soil temperature by 2.44℃and decreased soil moisture by 12.47%,whereas the long-term warming increased soil temperature by 1.76℃and decreased soil moisture by 9.90%.This caused the shallow soil layer to become warmer and drier under both short-term and long-term warming.Furthermore,the degree of soil drought was alleviated with increased warming duration.Under the long-term warming,the importance value and aboveground biomass of plants in different families changed.The importance values of grasses and sedges decreased by 47.56%and 3.67%,respectively,while the importance value of weeds increased by 1.37%.Aboveground biomass of grasses decreased by 36.55%,while those of sedges and weeds increased by 8.09%and 15.24%,respectively.The increase in temperature had a non-significant effect on species diversity.The species diversity indices increased at the early stage of warming and decreased at the late stage of warming,but none of them reached significant levels(P>0.05).Species diversity had no significant correlation with soil temperature and soil moisture under both short-term and long-term warming.Soil temperature and aboveground biomass were positively correlated in the control plots(P=0.014),but negatively correlated under the long-term warming(P=0.013).Therefore,eight years of warming aggravated drought in the shallow soil layer,which is beneficial for the growth of weeds but not for the growth of grasses.Warming changed the structure of alpine meadow communities and had a certain impact on the community species diversity.Our studies have great significance for the protection and effective utilization of alpine vegetation,as well as for the prevention of grassland degradation or desertification in high-altitude regions.WEN Jing QIN Ruimin ZHANG Shixiong YANG Xiaoyan XU Manhou 2020Journal of Arid Land2020,12,2:11
14Response of sediment yield to vegetation restoration at a large spatial scale in the Loess Plateau显示文摘The impact of vegetation coverage on erosion and sediment yield in the Loess Plateau has been extensively studied,but the research has been primarily based on observations from slope runoff plots or secondary forest regions;the scaling method remains unresolved when it is applied at a large spatial scale,and it is difficult to apply to regions with severe soil and water loss given the predominance of herbs and shrubs.To date,there is little data on the quantitative impact of changes to vegetation on sediment concentration at a large spatial scale.This paper is based on vegetation information from remote sensing images,measured rainfall and sediment data over nearly 60 years,and results from previous runoff and sediment variation research on the Yellow River.We introduce the concepts of a sediment yield coefficient and the percentage of effective vegetation and erodible area,analyze the impact of different vegetation conditions on the flood sediment concentration and sediment yield,and evaluate the effect of rainfall intensity on sediment yield under different vegetation conditions at the watershed scale.We propose models to evaluate the impact of vegetation on sediment yield in the loess gully hilly region,which are based on remote sensing data and support an application at a large spatial scale.The models can be used to assess sediment reduction that results from the current significant improvement of vegetation in the Loess Plateau.LIU XiaoYan YANG ShengTian DANG SuZhen LUO Ya LI XiaoYu ZHOU Xu 2014Science China(Technological Sciences)2014,57,8:11
15Major advances in studies of the physical geography and living environment of China during the past 70 years and future prospects显示文摘The natural environment provides material essentials for human survival and development. The characteristics,processes, regional differentiation and forcing mechanisms of the elements of the natural environment(e.g. geomorphology,climate, hydrology, soil, etc.) are the main objects of research in physical geography. China has a complex natural environment and huge regional differentiation and therefore it provides outstanding reserach opportunities in physical geography. This review summarizes the most important developments and the main contributions of research in the physical geography and human living environment in China during the past 70 years. The major topics addressed are the uplift of the Tibetan Plateau and the evolution of its cryosphere, the development of fluvial systems, the acidification of the vast arid region of the Asian interior, variations in the monsoon and westerly climate systems on multiple timescales, the development of lakes and wetlands, the watershed system model, soil erosion, past human-environment interactions, biogeography, and physical geographic zonality. After briefly introducing international research developments, we review the history of research in physical geography in China, focusing on the major achievements and major academic debates, and finally we summarize the status of current research and the future prospects. 17Enhancing mineral liberation of a Canadian rare earth are with microwave pretreatment显示文摘Liberation. as an attribute of mineralogy characteristic. whose impacts on finely disseminated Canadian rare earth ore was studied with microwave pretreatment. Samples of a light rare earth ore along with mostly ankerite and biotite as dominant gangue minerals as well as bastnaesite, strontianite and goethite as dominated minerals were exposed to further comminution by ball mill and microwave pretreatment fragmentation. Mineralogical characteristics were analyzed by using a mineral liberation analyzer(MLA).The results indicated that tight association mutually penetrates among dominated minerals in the range size of -300 + 212 μm and 212 + 150 μm and gangue minerals in the form of adjacent type. fine vein type. shell type and packing type. Temperature in the ore samples pre-treated by microwave can rapidly rise to 250 ℃ with microwave power of 0-1.5 kW and microwave time of 0-2 min. Applying the microwave pretreatment merely reduces the hardness of the ore causing the fracture of rare earth ore, but this does not transform or change the original mineralogy characteristics of the ore samples. On the basis of above study. the liberation value of bastnaesite, strontianite and goethite with microwave pretreatment is greater than with conventional comminution when the liberation class is above 75%. The distribution of particle size of rare earth ore samples is better with microwave pretreatment than with conventional comminution for particle size of 7.4 × 10^(-5) m. 19miR-188-5p regulates proliferation and invasion via PI3K/Akt/MMP-2/9 signaling in keloids显示文摘Keloids(KDs)and hypertrophic scars(HSs),two forms of pathological scars,seriously affect the physical and psychological health of patients.Despite many similarities with HSs,KDs are characterized by invasion and a high rate of recurrence after surgery,features they share in common with tumors.The underlying molecular mechanisms of this phenomenon have not been fully elucidated.In this study,we used microRNA(miRNA)array analysis to search for invasion-associated miRNAs in KDs.The expression of miR-188-5p in KDs,HSs,normal skin(NS)tissues,and cell lines was measured by quantitative real-time polymerase chain reaction.Furthermore,cell proliferation,migration,and invasion were detected in KD fibroblasts(KFs)and HS fibroblasts(HSFs),and interrelated proteins were ascertained by western blot analysis.It was found that miR-188-5p was significantly decreased in KD tissue compared with HS and NS tissues.Upregulated expression of miR-188-5p suppressed KF proliferation,migration,and invasion;and decreased expression of miR-188-5p also promoted HSF proliferation,migration,and invasion.The protein levels of MMP-2,MMP-9,PI3K,and p-Akt in miR-188-5p mimic-transfected KFs were repressed.In contrast,after transfection with miR-188-5p inhibitor,the protein levels of MMP-2,MMP-9,PI3K,and p-Akt were higher than the control in HSFs.Treatment with PI3K/Akt inhibitor LY294002 in KFs with miR-188-5p inhibitor did not further reduce their proliferation,migration,and invasion.The upregulation of MMP-2 and MMP-9 by miR-188-5p inhibitor could be abolished by LY294002.These findings together demonstrate a tumor-suppressive role of miR-188-5p in KD proliferation and invasion via PI3K/Akt/MMP-2/9 signaling,indicating that miR-188-5p may be a potential prognostic marker and therapeutic target for KDs.Wenyan Zhu Xiaoyan Wu Bo Yang Xiaodong Yao Xiaomei Cui Pan Xu Xiaodong Chen 2019Acta Biochimica et Biophysica Sinica2019,51,2:9
20Bavachin enhances NLRP3 inflammasome activation induced by ATP or nigericin and causes idiosyncratic hepatotoxicity显示文摘Psoraleae Fructus(PF)is a well-known traditional herbal medicine in China,and it is widely used for osteoporosis,vitiligo,and other diseases in clinical settings.However,liver injury caused by PF and its preparations has been frequently reported in recent years.Our previous studies have demonstrated that PF could cause idiosyncratic drug-induced liver injury(IDILI),but the mechanism underlying its hepatotoxicity remains unclear.This paper reports that bavachin isolated from PF enhances the specific stimuli-induced activation of the NLRP3 inflammasome and leads to hepatotoxicity.Bavachin boosts the secretion of IL-ip and caspase-1 caused by ATP or nigericin but not those induced by poly(I:C),monosodium urate crystal,or intracellular lipopolysaccharide.Bavachin does not affect AIM2 or NLRC4 inflammasome activation.Mechanistically,bavachin specifically increases the production of nigericin-induced mitochondrial reactive oxygen species among the most important upstream events in the activation of the NLRP3 inflammasome.Bavachin increases the levels of aspartate transaminase and alanine aminotransferase in serum and hepatocyte injury accompanied by the secretion of IL-ip via a mouse model of lipopolysaccharide-mediated susceptibility to IDILI.These results suggest that bavachin specifically enhances the ATP-or nigericin-induced activation of the NLRP3 inflammasome.Bavachin also potentially contributes to PF-induced idiosyncratic hepatotoxicity.Moreover,bavachin and PF should be evaded among patients with diseases linked to the ATP-or nigericin-mediated activation of the NLRP3 inflammasome,which may be a dangerous factor for liver injury.Nan Qin Guang Xu Yan Wang Xiaoyan Zhan Yuan Gao Zhilei Wang Shubin Fu Wei Shi Xiaorong Hou Chunyu Wang Ruisheng Li Yan Liu Jiabo Wang Haiping Zhao Xiaohe Xiao Zhaofang Bai 2021Frontiers of Medicine2021,15,4:9
