1Purifi cation, characterization and hypoglycemic activity of glycoproteins obtained from pea (Pisum sativum L.)显示文摘This study aimed to isolate and characterize the structures of glycoproteins from peas and determine their hypoglycemic activity.The crude pea glycoproteins(PGP)were extracted by hot water and purified by diethylaminoethyl(DEAE)-Sepharose chromatography and Sephadex G-100 size-exclusion chromatography in sequence.Then three main fractions were obtained,namely PGP1,PGP2 and PGP3,with molecular weights of 897615,846740 and 1194692 Da,respectively.The physical and chemical properties of the three fractions were evaluated and compared by Fourier transform infrared spectroscopy(FT-IR),nuclear magnetic resonance(NMR),scanning electron microscope(SEM),high performance liquid chromatography(HPLC)and other analytical techniques.The fraction PGP2 with the highest hypoglycemic activity,was screened using the Caco-2 monolayer cell model.It can inhibit the uptake of glucose in the small intestine,as well as the activities of maltase and sucrase.After simulated gastrointestinal digestion,PGP2 signifi cantly enhanced the inhibitory effect of α-glucosidase,and slightly reduced the inhibitory ability ofα-amylase.In summary,PGP2 possessed strong hypoglycemic activity after digestion.These results indicated that PGP2 has the potential to be developed into a functional food or natural medicine for the treatment of type 2 diabetes mellitus.Gaoyixin Qin Wu Xu Junping Liu Liyan Zhao Guitang Chen 2021Food Science and Human Wellness2021,10,3:8
2Visible and Infrared Video Fusion Using Uniform Discrete Curvelet Transform and Spatial-Temporal Information显示文摘Multiple visual sensor fusion provides an effective way to improve the robustness and accuracy of video surveillance system. Traditional video fusion methods fuse the source videos using static image fusion methods frame-by-frame without considering the information in temporal dimension. The temporal information can't be fully utilized in fusion procedure. Aiming at this problem, a visible and infrared video fusion method based on Uniform discrete curvelet transform(UDCT) and spatial-temporal information is proposed. The source videos are decomposed by using UDCT, and a set of local spatial-temporal energy based fusion rules are designed for decomposition coefficients. In these rules, we consider the current frame's coefficients and the coefficients on temporal dimension which are the coefficients of adjacent frames. 3Ambulatory blood pressure profile and stroke recurrence显示文摘Objectives To establish a new ambulatory blood pressure(ABP)parameter(24-hour ABP profile)and evaluated its performance on stroke outcome in ischaemic stroke(IS)or transient ischaemic attack(TIA)patients.Methods The prospective cohort consisted of 1996 IS/TIA patients enrolled for ABP monitoring and a 3-month follow-up for stroke recurrence as outcome.Profile groups of systolic blood pressure(SBP)were identified via an advanced functional clustering method,and the associations of the profile groups and conventional ABP parameters with stroke recurrence were examined in a Cox proportional hazards model.Results Three discrete profile groups(n=604,781 and 611 in profiles 1,2 and 3,respectively)in 24-hour ambulatory SBP were identified.Profile 1 resembled most to the normal diurnal blood pressure pattern;profile 2 also dropped at night,but climbed earlier and with higher morning surge;while profile 3 had sustained higher nocturnal SBP without significant nocturnal SBP decline.The incidence of stroke recurrence was 2.9%,3.9%and 5.5%in profiles 1,2 and 3,respectively.After adjustment for covariates,profile 3 was significantly associated with higher risk of stroke recurrence with profile 1 as reference(HR 1.76,95%CI:1.00 to 3.09),while no significant difference was observed between profiles 2 and 1(HR 1.22,95%CI:0.66 to 2.25).None of conventional ABP parameters showed significant associations with the outcome.Conclusions Ambulatory 24-hour SBP profile is associated with short-term stroke recurrence.Profiles of ABP may help improve identification of stroke recurrence by capturing the additive effects of individual ABP parameters.Jie Xu Fei Jiang Anxin Wang Hui Zhi Yuan Gao Junping Tian Jinglin Mo Zimo Chen An-Ding Xu Benyan Luo Bo Hu Yuqing Zhang Xingquan Zhao Yilong Wang Hao Li Haipeng Shen Yongjun Wang 2021Stroke & Vascular Neurology2021,6,3:3
4Surface modification of an up-conversion luminescence material by overcoating with SiO_2 and its characterization显示文摘The surface of an up-conversion luminescence material was modified by overcoating with SiO2, which was syn- thesized from a hydrolysis progress of tetraethoxysilane (TEOS) in alkalescent condition. By analyzing the hydrolyzed mechanism of TEOS, it was found that there was not only physical adsorption but also chemical bonding between the up-conversion material and SiO2. At the same time, some adsorption bands at 1100, 475, 950, and 3500 cm?1 were found by FI-IR, which were the characteristic bands of Si?OH and Si?O?Si. By analyzing the surface elements of the coated material by XPS, it was found that its surface only included Si, O, and C elements, and not F and Y. In the picture of XRD, there was no additional peak after surface modification, suggesting that the silica shell was amorphous. The small peak at 2θ = 23° in the X-ray diffraction pattern of the coated material was caused by the amorphous SiO2 shell, and the TEM image also proved that the surface of the material was successfully modified by overcoating with SiO2. 5Surface modification of up-conversion luminescence material Na[Y_(0.57)Yb_(0.39)Er_(0.04)]F_4 with hydrosulfide group显示文摘A new method was reported for surface modification of an up-conversion luminescence material with hydrosulfide group. The factors that may influence the surface modification,such as reaction time,amount of catalyzer and modifier,and reaction solvent,were investigated. The optimal conditions were that the reaction time,the quantity of the basic catalyzer,the quantity of modifier and the volume of reaction solvent were 40 min,1.0,1.0,and 40 mL,respectively. 6Targeted thrombolysis strategies for neuroprotective effect显示文摘Stroke is usually treated by systemic thrombolytic therapy if the patient presents within an appropriate time window. There is also widespread interest in the development of thrombolytic agents that can be used in cases of delayed presentation. Current agents that can be used in cases of delayed presentation of nerve damage by thrombus. Current systemic thrombolytic therapy is associated with adverse effects such as fibrinogenolysis and bleeding. In an attempt to increase the efficacy, safety, and specificity of thrombolytic therapy, a number of targeted thrombolytic agents have been studied in recent years. 11Tenofovir disoproxil fumarate therapy in patients with chronic hepatitis B and advanced fibrosis or compensated cirrhosis显示文摘Background and aims:Tenofovir disoproxil fumarate(TDF)is the first-line therapy for chronic hepatitis B.This interim analysis presents the efficacy and safety data for TDF at Week 144 in patients with chronic hepatitis B and advanced fibrosis or compensated cirrhosis from China.Methods:Patients were assessed for incidence of newly diagnosed hepatocellular carcinoma(HCC)and disease progression,liver stiffness measurement(LSM),virological suppression(serum hepatitis B virus DNA<20 IU/mL),alanine aminotransferase normalization,hepatitis B e antigen(HBeAg)loss and seroconversion,histological liver fibrosis score,and safety at Week 144.Results:Overall,197 patients were enrolled.At Week 144,the incidence of newly diagnosed HCC was observed in 2.1%patients,and the incidence of disease progression was observed in 3.6%patients.The mean(standard deviation)change in LSM from baseline was5.1(5.85)kPa.Serum hepatitis B virus DNA<20 IU/mL was observed in 94.1%patients,alanine aminotransferase normalization in 33.5%patients,HBeAg loss in 35.6%patients,and HBeAg seroconversion in 14.4%patients.Among patients with stage F3 or F4 fibrosis at baseline by LSM,38.3%patients regressed to stage F0/1,and 22.0%of patients regressed to stage F2 at Week 144.Overall,67.7%patients experienced≥1 adverse events,13.8%patients experienced TDF-related adverse events,and 16.4%patients experienced serious(none were TDF-related).Conclusions:At Week 144 of TDF treatment,low incidence of HCC and disease progression were reported.Virological suppression was observed in 94.1%patients,which was associated with fibrosis regression.No new safety events were identified.Huiying Rao Jia Shang Qing Xie Jianqi Lian Pujun Gao Junping Shi Xinyue Chen Jiefei Wang Min Xu Liaoyun Zhang Yingren Zhao QingMao Maorong Wang Wei Zhao Zong Zhang Jidong Jia Hong Tang Jiming Zhang Xin Zheng Chang Liu Lai Wei 2022iLIVER2022,1,3:1

14CmNF-YB8 affects drought resistance in chrysanthemum by altering stomatal status and leaf cuticle thickness显示文摘Drought is a major abiotic stress that limits plant growth and development.Adaptive mechanisms have evolved to mitigate drought stress,including the capacity to adjust water loss rate and to modify the morphology and structure of the epidermis.Here,we show that the expression of CmNF-YB8,encoding a nuclear factor Y(NF-Y)B-type subunit,is lower under drought conditions in chrysanthemum(Chrysanthemum morifolium).Transgenicchrysanthemumlinesinwhich transcript levels of CmNF-YB8 were reduced by RNA interference(CmNF-YB8-RNAi)exhibited enhanced drought resistance relative to control lines,whereas lines overexpressing CmNF-YB8(CmNF-YB8-OX)were less tolerant to drought.Compared to wild type(WT),CmNF-YB8-RNAi plants showed reduced stomatal opening and a thicker epidermal cuticle that correlated with their water loss rate.We also identified genes involved in stomatal adjustment(CBLinteracting protein kinase 6,CmCIPK6)and cuticle biosynthesis(CmSHN3)that are more highly expressed in CmNF-YB8-RNAi lines than in WT,CmCIPK6 being a direct downstream target of CmNF-YB8.Virus-induced gene silencing of CmCIPK6 or CmSHN3 in the CmNFYB8-RNAi background abolished the effects of CmNF-YB8-RNAi on stomatal closure and cuticle deposition,respectively.CmNF-YB8 thus regulates CmCIPK6 and CmSHN3 expression to alter stomatal movement and cuticle thickness in the leaf epidermis,thereby affecting drought resistance.Tianle Wang Qian Wei Zhiling Wang Wenwen Liu Xin Zhao Chao Ma Junping Gao Yanjie Xu Bo Hong 2022Journal of Integrative Plant Biology2022,64,3:1

17Cascade biocatalysis for production of enantiopure(S)-2-hydroxybutyric acid using recombinant Escherichia coli with a tunable multi-enzyme-coordinate expression system显示文摘Racemize 2-hydroxybutyric acid is usually synthesized by organic methods and needs additional deracemization to obtain optically pure enantiomers for industrial application.Here we present a cascade biocatalysis system in Escherichia coli BL21 which employed L-threonine deaminase(TD),NAD-dependent L-lactate dehydrogenase(LDH)and alcohol dehydrogenase(ADH)for producing optically pure(S)-2-hydroxybutyric acid((S)-2-HBA)from bulk chemical L-threonine.To solve the mismatch in the conversion rate and the consumption rate of intermediate 2-oxobutyric acid(2-OBA)formed in the multi-enzyme catalysis reaction,ribosome binding site regulation strategy was explored to control TD expression levels,achieving an eightfold alteration in the conversion rate of 2-OBA.With the optimized activity ratio of the three enzymes and using ADH for NADH regeneration,the recombinant strain ADH-r53 showed increased production of(S)-2-HBA with the highest titer of 129 g/L and molar yield of 93%within 24 h,which is approximately 1.65 times that of the highest yield reported so far.Moreover,(S)-2-HBA could easily be purified by distillation,making it have great potential for industrial application.Additionally,our results indicated that constructing a tunable multi-enzyme-coordinate expression system in single cell had great significance in biocatalysis of hydroxyl acids.Lingzhi Tian Junping Zhou Taowei Yang Xian Zhang Meijuan Xu Zhiming Rao 2021Systems Microbiology and Biomanufacturing2021,1,2:1
18Construction, detection and microarray analysis on Shigella dysenteriae A1 IroN, ShuA single, double mutants显示文摘In this study, we constructed single mutants MTS-1, MTS-2 of IroN and ShuA gene and double mutant MTS of them in Shigella dysenteriae A1 strain 51197 by insert and absence. The functional detection of every mutant was performed at the level of culture medium and cell experiment. The gene expression profiles of the mutants and the wild-type strains under iron- enriched and iron-limited conditions were analyzed by the SD51197 whole genomic microarray. The results showed that all the mutants grew obviously less well than the wild-type strains in L broth appending iron chelator DIP. The addition of iron to the cultures can stimulate the growth of mutants back to wild-type levels. In either the experiments on the ability of intracellular multiplication or the cell-to-cell spread in HeLa and U937 cell lines, mutants showed no obvious change in virulence compared with the parental strain SD51197. However when DIP was added to the cultured HeLa cells, the ability of intracellular multiplication of MTS-1, MTS-2, MTS has reduced about 23.4%, 25.2%, 43.6% respectively. The analysis of expression profiles under the iron-limited condition showed that the mutants were more sensitive for the changes of iron deficiency than the wild-type strains, many genes have been altered. Up-regulated genes mainly involved genes of transcription, coenzyme metabolism, amino acid transport and metabolism, and unknown functional genes, while down-regulated genes mainly involved genes of energy and carbohydrate metabolism and unknown function genes; the expression levels of known iron-transport associated genes generally showed up-regulated. 19Using Micro-Level Data to Evaluate Infrastructure of the Walking Environment Around Bus Rapid Transit Stations:A Case Study of Xiamen,China显示文摘China has experienced rapid car-oriented suburbanization during the past decades and is facing emerging challenges such as traffic congestion,air pollution,and even health threats.Learning from North American cities,Chinese cities have started adopting and implementing a Transit-Oriented Development(TOD) strategy,which encourages the transit ridership and walking trips by creating a pedestrian-friendly urban environment.Although much research has examined the walking environment in various neighborhoods or communities,few existing studies have examined the walking environment around transit stations,especially using micro-level data,such as walking infrastructure variables.This research chooses several infrastructure variables that are critical for pedestrians to investigate the walking environment around stations of the Bus Rapid Transit(BRT) No.1 Line in Xiamen City,China.This study defines a half mile buffer centered on each station as the spatial unit of analysis.The Geographic Information System(GIS) and field audits are employed to measure the characteristics of the infrastructure for pedestrians in each spatial unit of analysis.The results of this analysis can help assess the existing conditions of the infrastructure for pedestrians in each station area,which might need to be added or improved.The findings will not only provide empirical support for improving TODs in Xiamen,but also offer a lesson on how areas around transit stations could be retrofitted to improve the pedestrian environment.Xu Junping Wang Peng Kenneth Joh Bi Bo 2016China City Planning Review2016,25,2:1
