1Crystal structure of cytotoxin protein suilysin from Streptococcus suis显示文摘Cholesterol-dependent cytolysins(CDC)are pore forming toxins.A prototype of the CDC family members is perfringolysin O(PFO),which directly binds to the cell membrane enriched in cholesterol,causing cell lysis.However,an exception of this general observation is intermedilysin(ILY)of Streptococcus intermedius,which requires human CD59 as a receptor in addition to cholesterol for its hemolytic activity.A possible explanation of this functional difference is the conformational variation between the C-terminal domains of the two toxins,particularly in the highly conserved undecapeptide termed tryptophan rich motif.Here,we present the crystal structure of suilysin,a CDC toxin from the infectious swine pathogen Streptococcus suis.Like PFO,suilysin does not require a host receptor for hemolytic activity;yet the crystal structure of suilysin exhibits a similar conformation in the tryptophan rich motif to ILY.This observation suggests that the current view of the structure-function relationship between CDC proteins and membrane association is far from complete.Lingfeng Xu Bo Huang Huamao Du Xuejun CZhang Jianguo Xu Xuemei Li Zihe Rao 2010Protein & Cell2010,1,1:9
2Effects of sterilization treatments on the analysis of TOC in water samples显示文摘Decomposition experiments conducted with and without microbial processes are commonly used to study the effects of environmental microorganisms on the degradation of organic pollutants.However,the effects of biological pretreatment (sterilization) on organic matter often have a negative impact on such experiments.Based on the principle of water total organic carbon (TOC) analysis,the effects of physical sterilization treatments on determination of TOC and other water quality parameters were investigated.The results revealed that two conventional physical sterilization treatments,autoclaving and 60 Co γ-radiation sterilization,led to the direct decomposition of some organic pollutants,resulting in remarkable errors in the analysis of TOC in water samples.Furthermore,the extent of the errors varied with the intensity and the duration of sterilization treatments.Accordingly,a novel sterilization method for water samples,0.45 μm micro-filtration coupled with ultraviolet radiation (MCUR),was developed in the present study.The results indicated that the MCUR method was capable of exerting a high bactericidal effect on the water sample while significantly decreasing the negative impact on the analysis of TOC and other water quality parameters.Before and after sterilization treatments,the relative errors of TOC determination could be controlled to lower than 3% for water samples with different categories and concentrations of organic pollutants by using MCUR.Yiming Shi Lingfeng Xu Dongqin Gong Jun Lu 2010Journal of Environmental Sciences2010,22,5:3
3Mycorrhizas Affect Polyphyllin Accumulation of Paris polyphylla var.yunnanensis through Promoting PpSE Expression显示文摘Paris polyphylla var.yunnanensis is a traditional Chinese medicinal plant,in which polyphyllin as the main medicinal component is an important secondary metabolite with bioactivity.Arbuscular mycorrhizal fungi(AMF)have multiple positive effects on plants,while it is not clear whether AMF increase the content of medicinal components in medicinal plants.In this study,a total of nine AMF treatments were laid to analyze the mycorrhizal effect on polyphyllin accumulation and PpHMGR and PpSE expression of P.polyphylla var.yunnanensis.AMF increased the content of polyphyllin in the cultivated variety with low relation to the increase of inoculation intensity.Polyphyllin I,II,and VII were identified and partly improved by AMF inoculation,dependent on AMF treatments and culture environments.Similarly,the PpHMGR and PpSE expression was induced by mycorrhization,dependent on AMF species,whilst the induction was more obvious in PpSE than in PpHMGR after mycorrhization.It concluded that the symbiotic relationship between P.polyphylla var.yunnanensis and AMF increased polyphyllin content level in the plant,which was associated with the up-regulation of PpSE transcripts.Hailing Li Lingfeng Xu Zhuowei Li Shunxin Zhao Dongqin Guo Lu Rui Nong Zhou 2021Phyton-International Journal of Experimental Botany2021,90,5:2
4Costunolide covalently targets NACHT domain of NLRP3 to inhibit inflammasome activation and alleviate NLRP3-driven inflammatory diseases显示文摘The NLRP3 inflammasome’s core and most specific protein,NLRP3,has a variety of functions in inflammation-driven diseases.Costunolide(COS)is the major active ingredient of the traditional Chinese medicinal herb Saussurea lappa and has anti-inflammatory activity,but the principal mechanism and molecular target of COS remain unclear.Here,we show that COS covalently binds to cysteine 598 in NACHT domain of NLRP3,altering the ATPase activity and assembly of NLRP3 inflammasome.We declare COS’s great anti-inflammasome efficacy in macrophages and disease models of gouty arthritis and ulcerative colitis via inhibiting NLRP3 inflammasome activation.We also reveal that theα-methylene-γ-butyrolactone motif in sesquiterpene lactone is the certain active group in inhibiting NLRP3 activation.Taken together,NLRP3 is identified as a direct target of COS for its anti-inflammasome activity.COS,especially theα-methylene-γ-butyrolactone motif in COS structure,might be used to design and produce novel NLRP3 inhibitors as a lead compound.Haowen Xu Jiahao Chen Pan Chen Weifeng Li Jingjing Shao Shanshan Hong Yi Wang Lingfeng Chen Wu Luo Guang Liang 2023Acta Pharmaceutica Sinica B2023,13,2:2

6Cy5.5-MSA-G250 nanoparticles(CMGNPs)induce M1 polarity of RAW264. 7 macrophage cells via TLR4-dependent manner显示文摘Cy5.5-MSA-G250 nanoparticles(CMGNPs)had been proved to have unique advantages for cancer treatment,including excellent photothermal performance,tumor cell-selective cytotoxicity,direct visualization,and good biocompatibility.However,to cellular systems,the CMGNPs are considered as fo reign invaders,and the effect of CMGNPs on immunity system is still unknown.Therefore,more efforts are needed to understand the role of CMGNPs on the immunity system.In this study,we attempted to screen the pro-inflammatory responses on RAW264.7 macrophages after treated with the CMGNPs.In vitro experiments clearly showed that CMGNPs not only enhances phagocytosis capacity of RAW264.7 cells,but also promotes Ml polarization,associated with changes in cell morphology and increased expression of inflammatory cytokines.This ability to induce Ml polarization may be beneficial to CMGNPs to achieve better anticancer effects in clinical trials.Moreover,the observed Ml macrophages’ polarization triggered by CMGNPs can be abolished after adding TLR4 inhibitor,CLI095,suggesting that TLR4 is involved in CMGNP-induced inflammation.Zhuoxuan Lu Lingfeng Xu Nongyue He Fengying Huang Tiefeng Xu Li Li Yanwei Zhang Liming Zhang 2019Chinese Chemical Letters2019,30,6:2

9来源于人膀胱癌细胞株EJ的HSP70负载的DC所激发的特异性CTL细胞的体外应答效应(英文)显示文摘Objective: To investigate whether human dendritic cells (DC) derived from peripheral blood mononuclear cells (PBMC), which were pulsed by heat shock protein 70 (HSP70) isolated from human bladder tumor cell lines of EJ, were able to induce peptide specific cytotoxic T-lymphocytes (CTL) response in vitro and give the experimental foundation for the future clinical trials of immunotherapy in bladder tumor. Methods: The EJ-derived HSP70 co-cultured with DC from the healthy volunteers' PBMC, along with the crude lysate (the supernatant before HSP70 purification) from EJ cells were used as the experimental groups and DC not pulsed by any tumor cells antigen were the blank control. The autologous T-lymphocytes were added into the above various DC groups, and after incubation, the stimulation indexes (SI) and interferon-γ (IFN-γ) were detected to evaluate the immune activities of various DC groups. The killing effects of CTL to target cells, EJ and Hela cells, were determined with 51Cr releasing test. Results: Both DC/HSP70 and DC/the crude lysate could effectively activate CTL in vitro and kill target cells EJ. The killing effect of DC/HSP70 to EJ was much stronger than DC/the crude lysate (the supernatant before HSP70 purification) (P < 0.05). DC without any tumor cell antigens had a lower killing power to EJ. Meanwhile, DC/ HSP70 had little killing power to Hela non-relevant to bladder tumor histopathologically as compared with EJ cells (P < 0.05). 10Mass Selection for Growth Improvement in Black Shell Line of Pacific Oyster Crassostrea gigas显示文摘The Pacific oyster,Crassostrea gigas,naturally distributing along the coast of northwest Pacific,is one of the most important bivalve species due to its high economic value and fecundity.In China,we have initiated a selective breeding program on both shell color and growth rate of C.gigas since 2010.A black shell line was obtained through four-generation family selection.In this study,mass selection for growth improvement was conducted in the sixth generation and seventh generation of black shell lines.To assess the progress of potential genetic improvement,the progeny of two generations of black shell lines were selected to evaluate their shell heights via a 450-day farming experiment.As the results,after growing for 450 days,the sixth generation and seventh generation of selected lines were 9.03% and 11.42% larger than the control lines,respectively.During the grow-out stage,the genetic gain of two generations was 8.82%±0.18% and 11.54%±0.43%,respectively;and the corresponding realized heritability was 0.45±0.04 and 0.41±0.04,respectively.These results indicated that the mass selection for shell height achieved steady progress in the two generations of C.gigas.XU Chengxun LI Qi CHONG Jindou LIU Shikai KONG Lingfeng 2019Journal of Ocean University of China2019,18,6:1

12A novel therapeutic vaccine based on graphene oxide nanocomposite for tumor immunotherapy显示文摘With an intensive understanding of the mechanism of immune system,developing a therapeutic tumor vaccine is one of the most perspective strategy of cancer immunotherapy.In this study,we report a facile approach to prepare graphene oxide(GO)-based therapeutic cancer-nanovaccine.The model antigen(ovalbumin,OVA)and adjuvant(CpG ODN),are conjugated with GO-PEI nanosheet through electrostatic interaction.The addition of PEG can improve biocompatibility and prevent nanoparticle aggregation.The prepared GO-based nanovaccine,GO-PEI-OVA-PEG-CpG,exhibits good biocompatibility and low toxicity both in vivo and in vitro.More importantly,it can efficiently induce the maturation of dendritic cells(DCs),the enhancement of antigen cross-presentation ability,and the amplification of cytokine production of immune cells.Impressively,this nanovaccine shows a remarkable therapeutic effect against preestablished B16-OVA-melanoma tumors,which can significantly inhibit tumor growth and prolong the survival time of the OVA-expressed tumor-bearing mice.Moreover,combining GO-PEI-OVA-PEG-CpG with NLG919,an IDO-1(indoleamine-2,3-dioxygenase)inhibitor which can regulate the tumor microenvironment,displays a synergistic therapeutic effect.These findings indicate the GO-PEI-OVA-PEG-CpG nanovaccine actively induces an antigen-specific antitumor immune response and it combined with NLG919 could achieve better therapeutic outcomes.Liming Zhang Lingfeng Xu Yi Wang Jieyu Liu Guanghong Tan Fengying Huang Nongyue He Zhuoxuan Lu 2022Chinese Chemical Letters2022,33,8:1

17Mycorrhizal Fungal Effects on Growth,Antioxidant Capacity,and Medicine Quality of Paris polyphylla var.yunnanensis显示文摘A field experiment was conducted to determine the effects of two commercial strains composed of mulple arbuscular mycorrhizal fungi(AMF)species on plant growth,antioxidant capacity,and medicine quality of Paris polyphylla var.yunnanensis in three subtropical soils from Wanzhou,Anshun and Baoshan in fields.The results showed that AMF inoculation enhanced the fungal colonization rate and activities of both succinate dehydrogenase and alkaline phosphatase,thereby,enhancing the mycorrhizal viability of P.polyphylla var.yunnanensis.The concentrations of photosynthetic pigments(chlorophyll a,b,a+b and carotenoids),soluble sugar,soluble protein and photosynthetic capacity were higher in AMF-inoculated plants than in non-AMF-treated plants in field.AMFtreated plants recorded higher activities of catalase,peroxidase and superoxide dismutase,and caused the reduction in malondialdehyde content,indicating lower oxidative damage,compared with non-AMF plants.Polyphyllin I,Polyphyllin II,Polyphyllin III,Polyphyllin IV and total polyphyllin contents were increased by AMF treatment.In conclusion,AMF improved the plant growth,antioxidant capacity and medicinal quality of P.polyphylla var.yunnanensis seedlings.Hereinto,AMF effects on the soil from Wanzhou was relatively greater than on other soils.Nong Zhou Lingfeng Xu Min Yang Dongqin Guo Qiuxia Gan Jingjing Zhao 2021Phyton-International Journal of Experimental Botany2021,90,3:1
