| 1 | Optimization of Agrobacterium tumefaciens-Mediated Transformation Systems in Tea Plant(Camellia sinensis)显示文摘In this study, an efficient plant regeneration protocol in vitro and transformation by Agrobacterium-mediated method of Camellia sinensis was achieved, which would lay the foundation for genetic improvement of tea plant by genetic engineering technology. The cotyledon callus of C.sinensis were used as the receptors for transformation by Agrobacterium tumefaciens EHA105 containing PS1aG-3. Some factors which affected the result of Agrobacterium-mediated transformation of C. sinensis were studied on the basis of GUS transient expression system. The optimum system of Agrobacterium-mediated transformation was that the cotyledon callus were pre-cultured for 3 d, and then infected by EHA105 for 15 min followed by 3 d co-culture in the dark on the YEB medium containing 150 μmol·L^(-1) acetosyringone(AS). The transient expression rate of GUS gene was 62.6%. After being delayed selective culture for 3 d, infected callus were transferred into the differentiation medium and the root induction medium both of which were supplemented with 100 mg·L^(-1) spectinomycin, and then resistant seedlings of C. sinensis were obtained. | 2 | Molecular feature and therapeutic perspectives of immune dysregulation, polyendocrinopathy, enteropathy, X-linked syndrome显示文摘Regulatory T(Treg) cells, a subtype of immunosuppressive CD4^+T cells, are vital for maintaining immune homeostasis in healthy people. Forkhead box protein P3(FOXP3), a member of the forkhead-wingedhelix family, is the pivotal transcriptional factor of Treg cells. The expression, post-translational modifications, and protein complex of FOXP3 present a great impact on the functional stability and immune plasticity of Treg cells in vivo. In particular, the mutation of FOXP3 can result in immune dysregulation,polyendocrinopathy, enteropathy, X-linked(IPEX) syndrome, which is a rare genetic disease mostly diagnosed in early childhood and can soon be fatal. IPEX syndrome is related to several manifestations,including dermatitis, enteropathy, type 1 diabetes, thyroiditis, and so on. Here, we summarize some recent findings on FOXP3 regulation and Treg cell function. | 3 | Curcumenol triggered ferroptosis in lung cancer cells via lncRNA H19/miR-19b-3p/FTH1 axis显示文摘Curcumenol,an effective ingredient of Wenyujin,has been reported that exerted its antitumor potential in a few cancer types.However,the effect and molecular mechanism of curcumenol in lung cancer are largely unknown.Here,we found that curcumenol induced cell death and suppressed cell proliferation in lung cancer cells.Next,we demonstrated that ferroptosis was the predominant method that contributed to curcumenol-induced cell death of lung cancer in vitro and vivo for the first time.Subsequently,using RNA sequencing,we found that the long non-coding RNA H19(lncRNA H19)was significantly downregulated in lung cancer cells treated with curcumenol,when compared to untreated controls.Overexpression of lncRNA H19 eliminated the anticancer effect of curcumenol,while lncRNA H19 knockdown promoted ferroptosis induced by curcumenol treatment.Mechanistically,we showed that lncRNA H19 functioned as a competing endogenous RNA to bind to miR-19b-3p,thereby enhanced the transcription activity of its endogenous target,ferritin heavy chain 1(FTH1),a marker of ferroptosis.In conclusion,our data show that the natural product curcumenol exerted its antitumor effects on lung cancer by triggering ferroptosis,and the lncRNA H19/miR-19b-3p/FTH1 axis plays an essential role in curcumenol-induced ferroptotic cell death.Therefore,our findings will hopefully provide a valuable drug for treating lung cancer patients. | Ruonan Zhang Ting Pan Yu Xiang Mingming Zhang Han Xie Zimao Liang Bi Chen Cong Xu Jing Wang Xingxing Huang Qianru Zhu Ziming Zhao Quan Gao Chengyong Wen Wencheng Liu Weirui Ma Jiao Feng Xueni Sun Ting Duan Elaine Lai-Han Leung Tian Xie Qibiao Wu Xinbing Sui | 2022 | Bioactive Materials2022,7,7: | 8 |

| 5 | Do exercise therapies restore the deficits of joint position sense in patients with chronic ankle instability? A systematic review and meta-analysis显示文摘To determine whether existing exercise therapies can restore the joint position sense(JPS)deficits of patients with chronic ankle instability(CAI)when compared with controlled non-training patients.Seven databases were searched using ankle,injury,proprioception,and exercise-therapy-related terms.Peer-reviewed human studies in English that used the absolute errors score of joint position reproduction(JPR)test to compare the JPS of injured ankles in CAI patients before and after exercise therapy and non-training controls were included and analyzed.Demographic information,sample size,description of exercise therapies,methodological details of the JPR test,and absolute error scores were extracted by two researchers independently.Meta-analysis of the differences in JPS changes(i.e.,absolute errors after treatment minus the baseline)between the exercise therapies and non-training controls was performed with the weighted mean difference(WMD)and 95%confidence interval(CI).Seven studies were finally included.Meta-analyses revealed significantly higher improvements in passive JPS during inversion with,WMD?1.54and eversion,of,WMD?1.80,after exercise therapies when compared with non-training controls.However,no significant changes in the impaired side active JPS were observed with regard to inversion and eversion.Existing exercise therapies may have a positive effect on passive JPS during inversion and eversion,but do not restore the active JPS deficits of injured ankles in patients with CAI when compared with non-training controls.Updated exercise components with a longer duration that focus on active JPS with longer duration are needed to supplement the existing content of exercise therapies. | Xiao'ao Xue Weichu Tao Xiaoyun Xu Zhengbiao Jin Qianru Li Yiran Wang Xicheng Gu Ru Wang Yinghui Hua | 2023 | Sports Medicine and Health Science2023,5,1: | 0 |
| 6 | Eu-based anolytes for high-voltage and long-lifetime aqueous flow batteries显示文摘Aqueous flow batteries(AFBs) are among the most promising electrochemical energy storage solutions for the massive-scale adoption of renewable electricity because of decoupled energy and power, design flexibility, improved safety and low cost. The development of high-voltage AFB is, however, limited by the lack of stable anolytes that have low redox potential. Here we report Eu-based anolytes for high-voltage p H-neutral AFB applications. Eu^(3+) has a reduction potential of -0.39 V vs. SHE, which can be dramatically lowered when forming stable complex with inexpensive organic chelates. A typical complex, Eu DTPA,features a low redox potential of -1.09 V vs. SHE, fast redox kinetics, and a high water solubility of 1.5 M. When paired with ferrocyanide, the battery had an open-circuit voltage of 1.56 V and demonstrated stable cell cycling performance, including a capacity retention rate of 99.997% per cycle over500 cycles at 40 m A cm^(-2), a current efficiency of >99.9%, and an energy efficiency of >83.3%.
