| 1 | Molecular feature and therapeutic perspectives of immune dysregulation, polyendocrinopathy, enteropathy, X-linked syndrome显示文摘Regulatory T(Treg) cells, a subtype of immunosuppressive CD4^+T cells, are vital for maintaining immune homeostasis in healthy people. Forkhead box protein P3(FOXP3), a member of the forkhead-wingedhelix family, is the pivotal transcriptional factor of Treg cells. The expression, post-translational modifications, and protein complex of FOXP3 present a great impact on the functional stability and immune plasticity of Treg cells in vivo. In particular, the mutation of FOXP3 can result in immune dysregulation,polyendocrinopathy, enteropathy, X-linked(IPEX) syndrome, which is a rare genetic disease mostly diagnosed in early childhood and can soon be fatal. IPEX syndrome is related to several manifestations,including dermatitis, enteropathy, type 1 diabetes, thyroiditis, and so on. Here, we summarize some recent findings on FOXP3 regulation and Treg cell function. | 2 | Curcumenol triggered ferroptosis in lung cancer cells via lncRNA H19/miR-19b-3p/FTH1 axis显示文摘Curcumenol,an effective ingredient of Wenyujin,has been reported that exerted its antitumor potential in a few cancer types.However,the effect and molecular mechanism of curcumenol in lung cancer are largely unknown.Here,we found that curcumenol induced cell death and suppressed cell proliferation in lung cancer cells.Next,we demonstrated that ferroptosis was the predominant method that contributed to curcumenol-induced cell death of lung cancer in vitro and vivo for the first time.Subsequently,using RNA sequencing,we found that the long non-coding RNA H19(lncRNA H19)was significantly downregulated in lung cancer cells treated with curcumenol,when compared to untreated controls.Overexpression of lncRNA H19 eliminated the anticancer effect of curcumenol,while lncRNA H19 knockdown promoted ferroptosis induced by curcumenol treatment.Mechanistically,we showed that lncRNA H19 functioned as a competing endogenous RNA to bind to miR-19b-3p,thereby enhanced the transcription activity of its endogenous target,ferritin heavy chain 1(FTH1),a marker of ferroptosis.In conclusion,our data show that the natural product curcumenol exerted its antitumor effects on lung cancer by triggering ferroptosis,and the lncRNA H19/miR-19b-3p/FTH1 axis plays an essential role in curcumenol-induced ferroptotic cell death.Therefore,our findings will hopefully provide a valuable drug for treating lung cancer patients. | Ruonan Zhang Ting Pan Yu Xiang Mingming Zhang Han Xie Zimao Liang Bi Chen Cong Xu Jing Wang Xingxing Huang Qianru Zhu Ziming Zhao Quan Gao Chengyong Wen Wencheng Liu Weirui Ma Jiao Feng Xueni Sun Ting Duan Elaine Lai-Han Leung Tian Xie Qibiao Wu Xinbing Sui | 2022 | Bioactive Materials2022,7,7: | 8 |

| 10 | Genetic Insights into Glycine’s Protective Role Against CAD—European and East Asia,2015 and 2020显示文摘Introduction:The purpose of this study is to examine the potential causal relationship between levels of circulating glycine and coronary artery disease(CAD)using a two-step Mendelian randomization(MR)analysis.Methods:We analyzed data from genome-wide association studies(GWAS)conducted on European and East Asian populations.To assess the causal effects of circulating glycine levels on the risk of CAD.We used the inverse-variance weighting(IVW),weighted median(WM),MR-Egger,and Mendelian Randomization Pleiotropy RESidual Sum and Outlier(MR-PRESSO)methods.Furthermore,we conducted mediation analysis to investigate the contribution of blood pressure and other cardiovascular disease-related traits.Results:The two-step Mendelian randomization analysis revealed that higher levels of glycine in the blood were associated with a reduced risk of CAD in Europeans[odds ratio(OR)=0.84,95%confidence interval(CI):0.72,−0.98;P=0.029]and East Asians:(OR=0.76,95%CI:0.66,−0.89;P=3.57×10−4).Sensitivity analysis confirmed the robustness of these findings.Additionally,our results suggest that about 6.06%of the observed causal effect is mediated through genetically predicted systolic blood pressure(SBP)in the European population.Discussion:Our results contribute to the current knowledge regarding the involvement of glycine in the progression of CAD,and provide valuable methodological insights for the prevention and treatment of this condition. | Jiaying Yu Zhuolin Zhu Ting Wang Yuanhao Wei Lianjie Huang Qianru Zhang Yuting Zhang Yiran Wang Guiyou Liu Xiang Shu Rennan Feng | 2024 | China CDC weekly2024,6,9: | 0 |

| 16 | A cluster of mutagenesis revealed an osmotic regulatory role of the OsPIP1 genes in enhancing rice salt tolerance显示文摘Aquaporins play important regulatory roles in improving plant abiotic stress tolerance.To better understand whether the Os PIP1 genes collectively dominate the osmotic regulation in rice under salt stress,a cluster editing of the Os PIP1;1,Os PIP1;2 and Os PIP1;3 genes in rice was performed by CRISPR/Cas9 system.Sequencing showed that two mutants with Cas9-free,line 14 and line 18 were successfully edited.Briefly,line 14 deleted a single C base in both the Os PIP1;1 and Os PIP1;3 genes,and inserted a single T base in the Os PIP1;2 gene,respectively.While line 18 demonstrated an insertion of a single A base in the Os PIP1;1gene and a single T base in both the Os PIP1;2 and Os PIP1;3 genes,respectively.Multiplex editing of the Os PIP1 genes significantly inhibited photosynthetic rate and accumulation of compatible metabolites,but increased MDA contents and osmotic potentials in the mutants,thus delaying rice growth under salt stress.Functional loss of the Os PIP1 genes obviously suppressed the expressions of the Os PIP1,Os SOS1,Os CIPK24 and Os CBL4 genes,and increased the influxes of Na+and effluxes of K^(+)/H^(+)in the roots,thus accumulating more Na+in rice mutants under salt stress.This study suggests that the Os PIP1 genes are essential modulators collectively contributing to the enhancement of rice salt stress tolerance,and multiplex editing of the Os PIP1 genes provides insight into the osmotic regulation of the PIP genes. | Leyuan Tao Bing Wang Shichao Xin Wei Li Shengcai Huang Laihua Liu Jing Cui Qianru Zhang Xianguo Cheng | 2023 | The Crop Journal2023,11,4: | 0 |
