| 1 | Effects of miR-24 on the expression of vascular endothelial nitric oxide synthase and nitric oxide level in rats显示文摘Objective:To investigate the effects of miR-24 on the expression of vascular endothelial nitric oxide synthase(eNOS)and plasma nitric oxide(NO)level in rats.Methods:The miR-24 over-expression plasmid was constructed.Twenty healthy male Sprague-Dawley rats were randomly divided into two groups:miR-24 group and blank plasmid group.Rats were injected with plasmid DNA in saline via the tail vein.After eight weeks,the rat artery was removed and HE staining was used to observe the morphological changes.The protein expression and activity of eNOS were detected by immunohistochemistry and enzyme-linked immunosorbent assay(ELISA),respectively,and the NO level in plasma was determined by nitrate reduction assay.Results:Compared with the blank plasmid group,the expression and activity of eNOS as well as the NO production in the miR-24 group was significantly decreased(P <0.05 or P <0.01).No significant difference in the morphology of vascular tissue was noted between the two groups.Conclusion:Up-regulation of miR-24 inhibits the expression of eNOS and NO production in rat vascular tissue.It may provide a bio-target for the development of drugs to treat cardiovascular diseases such as hypertension. | Yongxiong Zhong Xuelan Luo Xiaojing Tao Feng Shen Dan Li Yuanyuan Yan Hesheng Ou | 2018 | 广西医科大学学报2018,35,4: | 1 |
| 2 | Crystal structures of main proteases of SARS-CoV-2 variants bound to a benzothiazole-based inhibitor显示文摘Main protease(Mpro)serves as an indispensable factor in the life cycle of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)as well as its constantly emerging variants and is therefore considered an attractive target for antiviral drug development.Benzothiazole-based inhibitors targeting Mpro have recently been investigated by several groups and proven to be promising leads for coronaviral drug development.In the present study,we determine the crystal structures of a benzothiazole-based inhibitor,YH-53,bound to Mpro mutants from SARS-CoV-2 variants of concern(VOCs)or variants of interest(VOIs),including K90R(Beta,B.1.351),G15S(Lambda,C.37),Y54C(Delta,AY.4),M49I(Omicron,BA.5)and P132H(Omicron,B.1.1.529).The structures show that the benzothiazole group in YH-53 forms a C-S covalent bond with the sulfur atom of catalytic residue Cys145 in SARSCoV-2 Mpro mutants.Structural analysis reveals the key molecular determinants necessary for interaction and illustrates the binding mode of YH-53 to these mutant Mpros.In conclusion,structural insights from this study offer more information to develop benzothiazole-based drugs that are broader spectrum,more effective and safer. | Jiqing Luo Weiwei Wang Haihai Jiang Wenwen Li Pei Zeng Jie Wang Xuelan Zhou Xiaofang Zou Shenghui Chen Qisheng Wang Jin Zhang Jian Li | 2023 | Acta Biochimica et Biophysica Sinica2023,55,8: | 0 |
| 3 | Effects and mechanism of miRNA-24 on the proliferation,migration and tube formation of vascular endothelial cells显示文摘Objective:To investigate the effect and mechanism of miRNA-24 on the proliferation,migration and tube formation of vascular endothelial cells.Methods:Human umbilical vein endothelial cells(HUVECs)were assigned into control,microRNA(miR)-24 overexpression and anti-miR-24 groups.The proliferation and migration abilities of HUVECs were detected by methyl thiazolyl tetrazolium(MTT)assay and scratch wound healing assay,respectively.The ability of HUVECs to form tubular structures was evaluated by a tube formation assay.The mRNA and protein expressions of vascular endothelial growth factor(VEGF)and transcription factor Sp1 were determined by RT-PCR,immunocytochemistry and western blotting,respectively.Results:The miR-24 overexpression group exhibited decreased cell proliferation and migration,and expressions of VEGF and Sp1 compared with the control group(P <0.01).No tube-like network structure was formed in the miR-24 overexpression group.However,inhibition of miR-24 in HUVECs markedly increased cell proliferation and migration,enhanced tube formation and expressions of VEGF and Sp1(P<0.05 or P<0.01).Conclusion:MiR-24 suppressed the proliferation,migration and tube formation of HUVECs,and the mechanism might be related to the down-regulation of VEGF expression.Sp1 might participate in this regulation process. | Xiaojing Tao Feng Shen Peng Yang Xuelan Luo Dan Li Yuanyuan Yan Yongxiong Zhong Hesheng Ou | 2018 | 广西医科大学学报2018,35,7: | 0 |
| 4 | Magnetic nanoparticle drug targeting to patient-specific atherosclerosis: effects of magnetic field intensity and configuration显示文摘Nanoparticle-mediated drug delivery is recognized as a promising option for targeted treatment of atherosclerosis. In this paper, the Eulerian-Lagrangian technique is adopted to simulate the delivery of drug-loaded nanoparticles to patient-specific atherosclerotic plaque with the aid of an external magnetic field. Plaques and vascular walls are introduced as porous media formulated by the Darcy-Forchheimer model in this targeted transport process. The results demonstrate that the delivery efficiency of particles to atherosclerosis depends on the external magnetic field, such as configuration and intensity, in which the configuration angle of the current wire is a key factor and the double current wires have advantages over the single current wire. Meanwhile, the delivery efficiency gradually decreases as the distance between the plaque cap and the current wire increases. Further, although augmenting the current or magnetic susceptibility can generally improve the delivery efficiency of nanoparticles, this increase is not apparent when small-sized nanoparticles are employed as drug transport particles. | 5 | Effects of miR-21 antisense oligonucleotides on proliferation,migration and autophagy of human umbilical vein endothelial cells显示文摘Objective:To investigate the effects of microRNA-21 antisense nucleotide(AS-miR-21)on the proliferation,migration and autophagy of human umbilical vein endothelial cells(HUVECs).Methods:HUVECs were treated with1,000 nmol/L rapamycin for 6 h(rapamycin group)or ASmiR-21 transfection followed by 1,000 nmol/L rapamycin for6 h(AS-miR-21+rapamycin group).HUVECs without any treatment were defined as control group.The proliferation and migration abilities of HUVECs were detected by methyl thiazolyl tetrazolium(MTT)assay,scratch wound healing assay and transwell test,respectively.The expressions of microtubule-associated protein light chain 3 Ⅱ/Ⅰ(LC3 Ⅱ/Ⅰ)and Becline-1 were determined by western blotting.Results:The rapamycin group showed decreased OD value and migration rate,an increased ratio of LC3 Ⅱ/Ⅰ and up-regulated expression of Beclin-1 compared with the control group(P<0.05).The AS-miR-21+rapamycin group demonstrated lower OD value,migration rate,the number of migrated cells,and significantly higher ratio of LC3 Ⅱ/Ⅰ and Beclin-1 protein expression level than the control group and the rapamycin group(P<0.05).Conclusion:AS-miR-21 suppressed the autophagy,proliferation and migration in the HUVECs model of autophagy induced by rapamycin. | Lyu Dongning Luo Xuelan Yang Ruixia Wang Guangyao Zhou Dong Gan Na Ou Hesheng | 2018 | 广西医科大学学报2018,35,8: | 0 |
| 6 | Effect of Different Processing Methods on Content ofβ-Asarone in the Zhuang Medicine Rhizoma Acori Tatarinowii显示文摘[Objectives]This paper aims to establish a method to simultaneously determine the content ofβ-asarone in Rhizoma Acori Tatarinowii dried by three different methods.[Methods]Reversed-phase high-performance liquid chromatography was used,and the chromatographic conditions were as follows:column,Thermo SCIENTIFIC Hypersil GOLD Dim.(mm);mobile phase,methanol-0.1%phosphoric acid(63∶37);column temperature,30℃;flow rate,1mL/min;detection wavelength,257 nm;sample size,10μL.[Results]The linear range of the injection volume ofβ-asarone was 49.28-246.40μg/mL(R=0.9993);the limit of quantification was 0.85 ng and the detection limit was 0.34 ng;the RSD values of precision,stability and reproducibility tests were all less than 3%;and the sample recovery rate was 98.53%-98.97%(RSD<3.00).The results show that the content ofβ-asarone was highest in shade-dried Rhizoma Acori Tatarinowii.The order ofβ-asarone content was as follows:Rhizoma Acori Tatarinowii dried in shade>Rhizoma Acori Tatarinowii dried at 55℃>Rhizoma Acori Tatarinowii dried at 60℃.[Conclusions]This method is sensitive,reliable,and reproducible.It can be used to simultaneously determine the content ofβ-asarone in Rhizoma Acori Tatarinowii dried by three different methods. | Peng YANG Jiangcun WEI Wen ZHONG Xiumei MA Xuelan LUO Peitao XIE Jiabao MA Qian HAN | 2020 | Medicinal Plant2020,11,5: | 0 |
