1Current perspectives on in vitro maturation and its effects on oocyte genetic and epigenetic profiles显示文摘In vitro maturation(IVM), the maturation in culture of immature oocytes, has been used in clinic for more than 20 years.Although IVM has the specific advantages of low cost and minor side effects over controlled ovarian stimulation, the prevalence of IVM is less than 1% of routine in vitro fertilization and embryo transfer techniques in many reproductive centers. In this review, we searched the MEDLINE database for all full texts and/or abstract articles published in English with content related to oocyte IVM mainly between 2000 and 2016. Many different aspects of the IVM method may influence oocyte potential,including priming, gonadotrophin, growth factors, and culture times. The culture conditions of IVM result in alterations in the oocyte or cumulus cell transcriptome that are not observed under in vivo culture conditions. Additionally, epigenetic modifications, such as DNA methylation or acetylation, are also different between in vitro and in vivo cultured oocytes. In sum,current IVM technique is still not popular and requires more systematic and intensive research to improve its effects and applications. 2Prioritizing natural-selection signals from the deep-sequencing genomic data suggests multi-variant adaptation in Tibetan highlanders显示文摘Human genetic adaptation to high altitudes(>2500 m)has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified,largely owing to the lack of deep-genome sequencing data available to previous studies.Here,w e build a list of putative adaptive variants,including 63 missense,7 loss-of-function;1,298 evolutionarily conserved variants and 509 expression quantitative traits loci.Notably,the top signal of selection is located in TMEM 247,a transmembrane protein-coding gene.The Tibetan version of TMEM 247 harbors one high-frequency(76.3%)missense variant,rsl 16983452(c.248C>T;p.Ala83Val);with the T allele derived from archaic ancestry and carried by>94%of Tibetans but absent or in low frequencies(<3%)in non-Tibetan populations.The rsl 16983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p=5.78×10^-5),red blood cell count(p=5.72×10^-7)and hematocrit(p=2.57×10^-6).In particular,TMEM247-rs 116983452 shows greater effect size and better predicts the phenotypic outcome than any E P A S1 variants in association with adaptive traits in Tibetans.Modeling the interaction between TM£M247-rsl 16983452 and EPAS1 variants indicates weak but statistically significant epistatic effects.Our results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism.Lian Deng Chao Zhang Kai Yuan Yang Gao Yuwen Pan Xueling Ge Yaoxi He Yuan Yuan Yan Lu Xiaoxi Zhang Hao Chen Haiyi Lou Xiaoji Wang Dongsheng Lu Jiaojiao Liu Lei Tian Qidi Feng Asifullah Khan Yajun Yang Zi-Bing Jin Jian Yang Fan Lu Jia Qu Longli Kang Bing Su Shuhua Xu 2019National Science Review2019,6,6:9

4Seed Setting and Its Spatial Characteristics in Tartary Buckwheat(Fagopyrum tataricum)显示文摘A low seed-setting rate is the main limiting factor influencing Tartary buckwheat production under high-yield cultivation conditions.To investigate the seed setting and its spatial characteristics,two Tartary buckwheat cultivars(high seed-setting rate cultivar Qianku 3;low seed-setting rate cultivar Liuku 3)were compared by a two-year field trial in 2017 and 2018.The results showed that the Tartary buckwheat underwent simultaneous flowering and fruiting.Flowers,generated from branch,were still blooming during the mature stage of grains on stem,which resulting in a greater number of flowers and grains on the branch than those on the stem at the low part of plant.The seed-setting characteristics significantly differed between two cultivars.The high seed-setting rate of Qianku 3 was 26.6%and 33.2%higher than Liuku 3 in 2017 and 2018,respectively.Meanwhile,Qianku 3 showed a higher filled grain number(157.8%)and seed-setting rate(66.4%)on branch than Liuku 3.A network analysis showed that the whole-plant seed-setting rate was positively correlated with grain number,which was closely correlated with flower number at the same position of plant.The path-coefficient analysis revealed that grains number on branches was the most dominant component(Path coefficient(P)=2.19)of the seed-setting rate,followed by grains number on stem(P=0.60).The grains number on branches showed the greatest positive direct effect with significant correlation(r=0.76 and P<0.01)on the seed-setting rate.Overall,the present study indicated that the grain number of branches may play a vital role in improving the seed-setting rate in Tartary buckwheat.Dabing Xiang Yue Song Chao Song Yan Wan Xueling Ye Changying Liu Chenggang Liang Gang Zhao 2022Phyton-International Journal of Experimental Botany2022,91,8:0
5The effect of different cancer therapies on clinical outcomes of assisted reproductive technology in breast cancer patients显示文摘Objective To evaluate the clinical outcomes of assisted reproductive technology(ART)on fertility preservation and infertility treatment in breast cancer patients who had undergone different cancer therapies.Methods 20 infertile females who had undergone breast cancer treatments during 2011–2018 were studied retrospectively.The patients were divided into two groups based on their cancer treatment methods and their needs of fertility preservation:the combined treatment group,who had both breast cancer surgery combining with any of the three treatments(adjuvant endocrine therapy,radiotherapy or chemotherapy),and the surgery only group.A group of infertile females without breast cancer history were used as a control group.An aromatase inhibitor Letrozole-based ovarian micro-stimulation protocol was used in females from the three groups for in vitro fertilization and embryo transfer.The ART clinical outcomes were evaluated by using the parameters of antral follicle count(AFC),the ratio of FSH/LH,oocyte retrieval number,2 pronucleus(2 PN)fertilization rate,high-quality embryo rate,clinical pregnancy rate,and delivery outcome.Results The surgery only group had significantly lower ratio of FSH/LH than the combined treatment group and the control group.No significant difference on the ART clinical outcomes,evaluated by the aforementioned criteria,were found between the three groups.Conclusions Breast cancer surgery with adjuvant therapies,but not surgery alone,may damage ovarian function.The best time-limited window to preserve fertility for breast cancer patients is after surgery but before the initiation of adjuvant therapies.Importantly,the clinical outcomes of aromatase inhibitor-based ovary micro-stimulation in breast cancer patients are similar to that in non-breast cancer controls.Mengmeng Zhang Xueling Song Caihong Ma Xiumei Zhen Hongbin Chi Xiaowei Zhang Rong Li Jie Yan Jie Qiao 2021Gynecology and Obstetrics Clinical Medicine2021,1,1:0

7Accounting for tastes:do low-income populations have a higher preference for spicy foods?显示文摘Based on the Theory of Rational Addiction(TORA),this paper uses the China Health and Nutrition Survey(CHNS)data to identify the correlation between income and preference for spicy foods.Results show that low-income individuals have a higher preference for spicy foods compared to high-income people,even in the same geographic area.Males and young people prefer spicy foods more than females and the elderly.Instrumental variable(IV)regression results also support that low-income individuals have a higher preference for spicy foods.The effect-channel results show that income affects the preference of spicy foods through health behaviors and health awareness.However,there is no significant evidence to support arguments about health capital stock and food selection channels.Chao Ma Ze Song Xueling Yan Guangchuan Zhao 2018The Journal of Chinese Sociology2018,5,1:0
8Polysaccharide Elicitor from the Endophyte Bionectria sp.Fat6 Improves Growth of Tartary Buckwheat under Drought Stress显示文摘Drought can limit the growth and reduce the yield of crops,but the safe and effective bio-approach to improve the drought resistance of crops is very little.We conducted an experiment in which we monitored the effects of polysaccharide from the endophyte Bionectria sp.Fat6 on the growth of Tartary buckwheat(Fagopyrum tataricum(L.)Gaertn)seedlings under control and drought-stressed conditions by determining gas exchange,photosynthesis parameters,photosynthetic pigment contents,and metabolite accumulation.Results indicated that the polysaccharide from endophyte stimulated plant growth and increased the aboveground biomass,root mass,and root/shoot ratio of Tartary buckwheat.Application of the polysaccharide to drought-stressed plants resulted in a significant increase in the net photosynthetic rate,stomatal conductance,and transpiration rate of Tartary buckwheat and decreased the intercellular CO_(2) concentration.The contents of chlorophyll a,chlorophyll b,chlorophyll a+b,and carotenoids in leaves were higher in polysaccharide-treated seedlings than that in control.Polysaccharide notably increased the soluble protein and proline content and decreased the malondialdehyde content in Tartary buckwheat leaves.The endophytic polysaccharide may protect Tartary buckwheat against drought by improving leaf gas exchange and photosynthetic capacity,and altering concentrations of protective metabolites.Together,these changes may compensate for the negative impacts of drought stress on the growth of Tartary buckwheat.Thus,the polysaccharide from the endophyte Bionectria sp.Fat6 may be an effective biotic elicitor and a promising bio-approach to improve Tartary buckwheat production worldwide.Dabing Xiang Wei Wei Yan Wan Xiaoyong Wu Xueling Ye Lianxin Peng Linyun Zhong Qi Wu Liang Zou Gang Zhao Jianglin Zhao 2021Phyton-International Journal of Experimental Botany2021,90,2:0
9Growth,ROS Markers,Antioxidant Enzymes,Osmotic Regulators and Metabolic Changes in Tartary Buckwheat Subjected to Short Drought显示文摘Tartary buckwheat(Fagopyrum tataricum)is an important pseudocereal feed crop with medicinal and nutritional value.Drought is one of the main causes of reduced growth and yield in these plants.We investigated the growth,physiological,and metabolic responses of the widely promoted Tartary buckwheat variety Chuan Qiao No.1 to polyethylene glycol(PEG)-mediated drought stress.Drought significantly decreased shoot length,shoot biomass and relative water content.Root length,malondialdehyde content,electrolyte leakage,activities of superoxide dismutase,peroxidase,catalase and amylase,and contents of soluble sugar,soluble protein and proline were increased by PEG-mediated drought.Untargeted metabolomics analysis identified 32 core metabolites in seedlings subjected to PEG-mediated drought,16 of which increased—including quercetin,isovitexin,cyanidin 3-O-beta-D-glucoside,L-arginine,and glycerophosphocholine,while the other 16 decreased—including 3-methoxytyramine,2,6-diaminopimelic acid,citric acid,UDP-alpha-D-glucose,adenosine,keto-D-fructose.The 32 core metabolites were enriched in 29 metabolic pathways,including lysine biosynthesis,citrate(TCA)cycle,anthocyanin biosynthesis,and aminoacyl-tRNA biosynthesis.Among them,taurine and hypotaurine metabolism,flavor and flavor biosynthesis,indole alkaline biosynthesis,and alanine,aspartate and glutamate metabolism were the four main metabolic pathways affected by drought.Our findings provide new insights into the physiological and metabolic response mechanisms of Tartary buckwheat to drought stress.Yan Wan Yuan Liang Xuxiao Gong Jianyong Ouyang Jingwei Huang Xiaoyong Wu Qi Wu Changying Liu Xueling Ye Xiaoning Cao Gang Zhao Liang Zou Dabing Xiang 2023Phyton-International Journal of Experimental Botany2023,92,1:0

11Overexpression of Toll-like receptor 4 contributes to the internalization and elimination of Escherichia coli in sheep by enhancing caveolae-dependent endocytosis显示文摘Background:Gram-negative bacterial infections have a major economic impact on both the livestock industry and public health.Toll-like receptor 4(TLR4)plays a crucial role in host defence against Gram-negative bacteria.Exploring the defence mechanism regulated by TLR4 may provide new targets for treatment of inflammation and control of bacterial infections.In a previous study,we generated transgenic sheep overexpressing TLR4 by microinjection to improve disease resistance.The defence mechanism through which TLR4 overexpression protected these sheep against pathogens is still not fully understood.Results:In the present study,we used Escherichia coli to infect monocytes isolated from peripheral blood of the animal model.The overexpression of TLR4 strongly enhanced the percentage of endocytosis and capacity of elimination in monocytes during the early stages of infection.This phenomenon was mainly due to overexpression of TLR4 promoting caveolae-mediated endocytosis.Pretreatment of the transgenic sheep monocytes with inhibitors of TLR4,Src signalling,or the caveolae-mediated endocytosis pathway reduced the internalization of bacteria,weakened the ability of the monocytes to eliminate the bacteria,and increased the pH of the endosomes.Conclusion:Together,our results reveal the effects of TLR4 on the control of E.coli infection in the innate immunity of sheep and provide crucial evidence of the caveolae-mediated endocytosis pathway required for host resistance to invading bacteria in a large animal model,providing theoretical support for breeding disease resistance in the future.Furthermore,Src and caveolin 1(CAV1)could be potentially valuable targets for the control of infectious diseases.Yao Li Yue Zhao Xueling Xu Rui Zhang Jinlong Zhang Xiaosheng Zhang Yan Li Shoulong Deng Zhengxing Lian 2021Journal of Animal Science and Biotechnology2021,12,4:0
