5Quantitative angiographic haemodynamic evaluation of bypasses for complex aneurysms: a preliminary study显示文摘Objective Open microsurgery,often with bypass techniques,is indispensable for complex aneurysms.To date,it remains unknown whether arterial anatomy or quantitative blood flow measurements can predict insufficient flow-related stroke(IRS).The present study aimed to evaluate the risk factors for IRS in patients treated with open microsurgery with bypass procedures for complex internal carotid artery aneurysms.Methods Patients with complex aneurysms undergoing bypass surgery were retrospectively reviewed.The recipient/donor flow index(RDFI)was preoperatively evaluated using colour-coding angiography.RDFI was defined as the ratio of the cerebral blood volume of the recipient and donor arteries.The sizes of the recipient and donor arteries were measured.The recipient/donor diameter index(RDDI)was then calculated.IRS was defined as the presence of new postoperative neurological deficits and infarction on postoperative CT scans.We assessed the association between RDFI and other variables and the IRS.Results Twenty patients(38±12 years)were analysed.IRS was observed in 12 patients(60%).Patients with postoperative IRS had a higher RDFI than those without postoperative IRS(p<0.001).RDDI was not significantly different between patients with and without IRS(p=0.905).Patients with RDFI>2.3 were more likely to develop IRS(p<0.001).Conclusion Quantitative digital subtraction angiography enables preoperative evaluation of cerebral blood volume.RDFI>2.3,rather than RDDI,was significantly associated with postoperative IRS.This preoperative evaluation allows appropriate decisions regarding the treatment strategy for preventing postoperative IRS.Junlin Lu Chao Xue Xulin Hu Yuanli Zhao Dong Zhang Xiaolin Chen Ji Zong Zhao 2022Stroke & Vascular Neurology2022,7,1:1

11The Establishment and Validation of the Human U937 Cell Line as a Cellular Model to Screen Immunomodulatory Agents Regulating Cytokine Release Induced by Influenza Virus Infection显示文摘Severe influenza infections are often associated with the excessive induction of pro-inflammatory cytokines,which is also referred to as'cytokine storms'.Several studies have shown that cytokine storms are directly associated with influenzainduced fatal acute lung injury and acute respiratory distress syndrome.Due to the narrow administration window,current antiviral therapies are often inadequate.The efforts to use immunomodulatory agents alone or in combination with antiviral agents in the treatment of influenza in animal models have resulted in the achievement of protective effects accompanied with reduced cytokine production.Currently,there are no immunomodulatory drugs for influenza available for clinical use.Animal models,despite being ideal to study the anti-inflammatory responses to influenza virus infection,are very costly and time-consuming.Therefore,there is an urgent need to establish fast and economical screening methods using cellbased models to screen and develop novel immunomodulatory agents.In this study,we screened seven human cell lines and found that the human monocytic cell U937 supports the replication of different subtypes of influenza viruses as well as the production of the important pro-inflammatory cytokines and was selected to develop the cell-based model.The U937 cell model was validated by testing a panel of known antiviral and immunomodulatory agents and screening a drug library consisting of 1280 compounds comprised mostly of FDA-approved drugs.We demonstrated that the U937 cell model is robust and suitable for the high-throughput screening of immunomodulators and antivirals against influenza infection.Ge Liu Si Chen Ao Hu Li Zhang Wenyu Sun Jungang Chen Wei Tang Haiwei Zhang Chunlan Liu Chang Ke Xulin Chen 2019Virologica Sinica2019,34,6:0
12SARS-CoV-2 N protein enhances the anti-apoptotic activity of MCL-1 to promote viral replication显示文摘Viral infection in respiratory tract usually leads to cell death,impairing respiratory function to cause severe disease.However,the diversity of clinical manifestations of SARS-CoV-2 infection increases the complexity and difficulty of viral infection prevention,and especially the high-frequency asymptomatic infection increases the risk of virus transmission.Studying how SARS-CoV-2 affects apoptotic pathway may help to understand the pathological process of its infection.Here,we uncovered SARS-CoV-2 imployed a distinct anti-apoptotic mechanism via its N protein.We found SARS-CoV-2 virus-like particles(trVLP)suppressed cell apoptosis,but the trVLP lacking N protein didn’t.Further study verified that N protein repressed cell apoptosis in cultured cells,human lung organoids and mice.Mechanistically,N protein specifically interacted with anti-apoptotic protein MCL-1,and recruited a deubiquitinating enzyme USP15 to remove the K63-linked ubiquitination of MCL-1,which stabilized this protein and promoted it to hijack Bak in mitochondria.Importantly,N protein promoted the replications of IAV,DENV and ZIKV,and exacerbated death of IAV-infected mice,all of which could be blocked by a MCL-1 specific inhibitor,S63845.Altogether,we identifed a distinct anti-apoptotic function of the N protein,through which it promoted viral replication.These may explain how SARS-CoV-2 effectively replicates in asymptomatic individuals without cuasing respiratory dysfunction,and indicate a risk of enhanced coinfection with other viruses.We anticipate that abrogating the N/MCL-1-dominated apoptosis repression is conducive to the treatments of SARS-CoV-2 infection as well as coinfections with other viruses.Pan Pan Weiwei Ge Zhiwei Lei Wei luo Yuqing Liu Zhanwen Guan Lumiao Chen Zhenyang Yu Miaomiao Shen Dingwen Hu Qi Xiang Wenbiao Wang Pin Wan Mingfu Tian Yang Yu Zhen Luo Xulin Chen Heng Xiao Qiwei Zhang Xujing Liang Xin Chen Yongkui Li Jianguo Wu 2023Signal Transduction and Targeted Therapy2023,8,6:0
1310 Gb/s classical secure key distribution based on temporal steganography and private chaotic phase scrambling显示文摘Secure distribution of high-speed digital encryption/decryption keys over a classical fiber channel is strongly pursued for realizing perfect secrecy communication systems.However,it is still challenging to achieve a secret key rate in the order of tens of gigabits per second to be comparable with the bit rate of commercial fiber-optic systems.In this paper,we propose and experimentally demonstrate a novel solution for high-speed secure key distribution based on temporal steganography and private chaotic phase scrambling in the classical physical layer.The encryption key is temporally concealed into the background noise in the time domain and randomly phase scrambled bit-by-bit by a private chaotic signal,which provides two layers of enhanced security to guarantee the privacy of key distribution while providing a high secret key rate.We experimentally achieved a record classical secret key rate of 10 Gb/s with a bit error rate lower than the hard-decision forward error correction(HD-FEC)over a 40 km standard single mode fiber.The proposed solution holds great promise for achieving high-speed key distribution in the classical fiber channel by combining steganographic transmission and chaotic scrambling.ZHENSEN GAO ZHITAO DENG LIHONG ZHANG XULIN GAO YUEHUA AN ANBANG WANG SONGNIAN FU ZHAOHUI LI YUNCAI WANG YUWEN QIN 2024Photonics Research2024,12,2:0
14Novel 3D printed shape-memory PLLA-TMC/GA-TMC scaffolds for bone tissue engineering with the improved mechanical properties and degradability显示文摘The biodegradable substitution materials for bone tissue engineering have been a research hotspot.As is known to all,the biodegradability,biocompatibility,mechanical properties and plasticity of the substitution materials are the important indicators for the application of implantation materials.In this article,we reported a novel binary substitution material by blending the poly(lactic-acid)-co-(trimethylenecarbonate)and poly(glycolic-acid)-co-(trimethylene-carbonate),which are both biodegradable polymers with the same segment of flexible trimethylene-carbonate in order to accelerate the degradation rate of poly(lactic-acid)-co-(trimethylene carbonate)substrate and improve its mechanical properties.Besides,we further fabricate the porous poly(lactic-acid)-co-(trimethylene-carbonate)/poly(glycolic-acid)-co-(trimethylene-carbonate)scaffolds with uniform microstructure by the 3D extrusion printing technology in a mild printing condition.The physicochemical properties of the poly(lactic-acid)-co-(trimethylenecarbonate)/poly(glycolic-acid)-co-(trimethylene-carbonate)and the 3D printing scaffolds were investigated by universal tensile dynamometer,fourier transform infrared reflection(FTIR),scanning electron microscope(SEM)and differential scanning calorimeter(DSC).Meanwhile,the degradability of the PLLATMC/GA-TMC was performed in vitro degradation assays.Compared with PLLA-TMC group,PLLA-TMC/GATMC groups maintained the decreasing Tg,higher degradation rate and initial mechanical performance.Furthermore,the PLLA-TMC/GA-TMC 3D printing scaffolds provided shape-memory ability at 37℃.In summary,the PLLA-TMC/GA-TMC can be regarded as an alternative substitution material for bone tissue engineering.Xulin Hu Weiming Zhao Zhen Zhang Jianping Xie Jian He Jianfei Cao Qing Li Yajing Yan Chengdong Xiong Kainan Li 2023Chinese Chemical Letters2023,34,1:0
15CRITICAL PROCESSES AND MAJOR FACTORS THAT DRIVE NITROGEN TRANSPORT FROM FARMLAND TO SURFACE WATER BODIES显示文摘Agricultural non-point source pollution is increasingly an important issue affecting surface water quality.Currently,the majority of the studies on nitrogen loss have focused on the agricultural field scale,however,the response of surface water quality at the watershed scale into the nitrogen loss at the field scale is poorly understood.The present study systematically reviewed the critical processes and major factors that nitrogen transport from farm fields to surface water bodies.The critical processes of farmland nitrogen entering surface water bodies involve the processes of nitrogen transport from farmland to ditches and the transformation processes of nitrogen during migration in ditches/rivers.Nitrogen transport from farmland to ditches is one of the prerequisites and critical processes for farmland nitrogen transport to surface water bodies.The transformation of nitrogen forms in ditches/rivers is an intermediate process in the migration of nitrogen from farmland to surface water bodies.Nitrogen loss from farmland is related to soil storage and exogenous inputs.Therefore,nitrogen input management should not only consider the current input,but also the contribution of soil storage due to the historical surpluses.Ditches/rivers have a strong retention capacity for nitrogen,which will significantly affect the process of farmland nitrogen entering surface water bodies.The factors affecting nitrogen transformation in river/ditches can be placed in four categories:(1)factors affecting hydraulic retention time,(2)factors affecting contact area,(3)factors affecting biological activity,and(4)forms and amount of nitrogen loading to river/ditches.Ditch systems are more biologically(including plants and microbes)active than rivers with biological factors having a greater influence on nitrogen transformation.When developing pollution prevention and control strategies,ecological ditches can be constructed to increase biological activity and reduce the amount of surplus nitrogen entering the water body.The present research should be valuable for the evaluation of environment impacts of nitrogen loss and the non-point source pollution control.Wenchao LI Wen XU Gaofei YIN Xulin ZHANG Zihan ZHANG Bin XI Qiuliang LEI Limei ZHAI Qiang ZHANG Linzhang YANG Hongbin LIU 2023Frontiers of Agricultural Science and Engineering2023,10,4:0
16Nomogram for Predicting the Severity of Coronary Artery Disease in Young Adults≤45 Years of Age with Acute Coronary Syndrome显示文摘Background:A non-invasive predictive model has not been established to identify the severity of coronary lesions in young adults with acute coronary syndrome(ACS).Methods:In this retrospective study,1088 young adults(≤45 years of age)first diagnosed with ACS who underwent coronary angiography were enrolled and randomized 7:3 into training or testing datasets.To build the nomogram,we determined optimal predictors of coronary lesion severity with the Least Absolute Shrinkage and Selection Operator and Random Forest algorithm.The predictive accuracy of the nomogram was assessed with calibration plots,and performance was assessed with the receiver operating characteristic curve,decision curve analysis and the clinical impact curve.Results:Seven predictors were identified and integrated into the nomogram:age,hypertension,diabetes,body mass index,low-density lipoprotein cholesterol,mean platelet volume and C-reactive protein.Receiver operating characteristic analyses demonstrated the nomogram’s good discriminatory performance in predicting severe coronary artery disease in young patients with ACS in the training(area under the curve 0.683,95%confidence interval[0.645–0.721])and testing(area under the curve 0.670,95%confidence interval[0.611–0.729])datasets.The nomogram was also well-calibrated in both the training(P=0.961)and testing(P=0.302)datasets.Decision curve analysis and the clinical impact curve indicated the model’s good clinical utility.Conclusion:A simple and practical nomogram for predicting coronary artery disease severity in young adults≤45 years of age with ACS was established and validated.Xulin Hong Duanbin Li Xinrui Yang Guosheng Fu Chenyang Jiang Wenbin Zhang 2022Cardiovascular Innovations and Applications2022,7,3:0
17Inhibition of aryl hydrocarbon receptor signaling promotes the terminal differentiation of human erythroblasts显示文摘The aryl hydrocarbon receptor(AHR)plays an important role during mammalian embryo development.Inhibition of AHR signaling promotes the development of hematopoietic stem/progenitor cells.AHR also regulates the functional maturation of blood cells,such as T cells and megakaryocytes.However,little is known about the role of AHR modulation during the development of erythroid cells.In this study,we used the AHR antagonist StemRegenin 1(SR1)and the AHR agonist 2,3,7,8-tetrachlorodibenzo-p-dioxin during different stages of human erythropoiesis to elucidate the function of AHR.We found that antagonizing AHR signaling improved the production of human embryonic stem cell derived erythrocytes and enhanced erythroid terminal differentiation.RNA sequencing showed that SR1 treatment of proerythroblasts upregulated the expression of erythrocyte differentiation-related genes and downregulated actin organization-associated genes.We found that SR1 accelerated F-actin remodeling in terminally differentiated erythrocytes,favoring their maturation of the cytoskeleton and enucleation.We demonstrated that the effects of AHR inhibition on erythroid maturation were associated with F-actin remodeling.Our findings help uncover the mechanism for AHRmediated human erythroid cell differentiation.We also provide a new approach toward the large-scale production of functionally mature human pluripotent stem cell-derived erythrocytes for use in translational applications.Yijin Chen Yong Dong Xulin Lu Wanjing Li Yimeng Zhang Bin Mao Xu Pan Xiaohong Li Ya Zhou Quanming An Fangxin Xie Shihui Wang Yuan Xue Xinping Cai Mowen Lai Qiongxiu Zhou Yan Yan Ruohan Fu Hong Wang Tatsutoshi Nakahata Xiuli An Lihong Shi Yonggang Zhang Feng Ma 2022Journal of Molecular Cell Biology2022,14,2:0
