| 1 | Pyrite framboids associated with the Mesozoic Jehol Biota in northeastern China:Implications for microenvironment during early fossilization显示文摘Despite of the recent influx of reports describing important fossil specimens from the Mesozoic Jehol Biota, detailed information on the mechanism of fossilization and paleoenvironment in which these fossils were deposited is scanty. We present an analysis of microenvironment based upon scanning electron microscope observations of in situ pyrite framboids and microcrystallines of plant and vertebrate feather fossils in the Jehol Biota. Pyrite microcrystallines and framboids occur extensively inside and on surface of plant fossils.Framboids found on feathers and in sedimentary matrix were in a lower abundance. These framboids have diameters ranging from 6 μm to 31 μm with an average of 20 μm, indicating a dysoxic aqueous condition with free oxygen level less than 30 μmol/L for the microenvironment where these framboids were formed. The outgrowth of framboids inside plant tissues suggests the presence of water molecules and free oxygen at the cellular level during pyritization; the relative timing between tissue decay and framboid formation implies a rapid tissue degradation occurred during the very early stage of fossilization. This line of reasoning is consistent with the observation that cell level structure of plant fossils from these deposits is rarely preserved. | 2 | BIX-01294-enhanced chemosensitivity in nasopharyngeal carcinoma depends on autophagy-induced pyroptosis显示文摘Nasopharyngeal carcinoma(NPC)is a common cancer in southern China and Southeast Asia.Nowadays,radiotherapy is the therapy of choice for NPC patients,and chemotherapy has been found as an alternative treatment for advanced NPC patients.However,finding novel drugs and pharmacologically therapeutic targets for NPC patients is still urgent and beneficial.Our study showed that BIX-01294(BIX)can induce autophagic vacuoles formation and conversion of LC3B-I to LC3B-II in NPC cells in both dose-and time-dependent manners.Notably,the combination of BIX and chemotherapeutic drugs significantly decreased the cell viability and increased the lactate dehydrogenase release.Meanwhile,BIX plus cis-platinum(Cis)treatment induced pyroptosis in NPC cells as featured by cell swelling and bubble blowing from the plasma membrane,the increased frequency of annexin V and propidium iodide(PI)double-positive cells,as well as the cleavage of gasdermin E(GSDME)and caspase-3.Moreover,the deficiency of GSDME completely shifted pyroptosis to apoptosis.Furthermore,the inhibition of autophagy by chloroquine and the knockout of ATG5 gene significantly blocked the BIX-induced autophagy as well as pyroptosis in both in vitro and in vivo studies.Our data demonstrated that BIX-combined chemotherapeutic drugs could induce the Bax/caspase-3/GSDME-mediated pyroptosis through the activation of autophagy to enhance the chemosensitivity in NPC. | Qian Li Min Wang Yan Zhang Liuqian Wang Wei Yu Xiaomin Bao Biyun Zhang Yanghong Xiang Anchun Deng | 2020 | Acta Biochimica et Biophysica Sinica2020,52,10: | 5 |

| 4 | Diversity of Trichoderma in greenhouse soil显示文摘The protected agricultural production has become one of the fast growing and wid espread cultivation technology in the north parts of China. Continuous cultivati on of single crop or less rotation of crops usually resulted in the large amount of accumulation of soilborne pathogens and serious crop diseases in the greenho use. After a few years of investigation of soilborne fungi in the north parts of China, nearly one hundred species of mictosporic fungi have been identified by the authors. Among these fungi 11 species of Trichoderma have been morphol ogically identified, namely T. atroviride, T. aureoviride, T. citrinoviride, T. fertile, T. harzianum, T. inhamatum,T. longibrachiatum, T. parceramosum, T. r eeseii, T. virens and T. viride. Trichoderm is found to be a frequently occurring genus of fungi in greenhouse soil. | 8 | A High-affinity Activator of G551D-CFTR Chloride Channel Identified By High Throughput Screening显示文摘A stably transfected CHO cell line coexpressing G551D-CFTR and iodide-sensitive yellow fluorescent protein mutant EYFP-H148Q-I152L was successfully established and used as assay model to identify small-molecule activators of G551D-CFTR chloride channel from 100000 diverse combinatorial compounds by high throughput screening on a customized Beckman robotic system. A bicyclooctane compound was identified to activate G551D-CFTR chloride channel with high-affinity(K d=1.8 μmol/L). The activity of the bicyclooctane compound is G551D-CFTR-specific, reversible and non-toxic. | 13 | A Sensitive Competitive ELISA for Determination of Biotin in Transformed Yeast Culture Media显示文摘Aim To develop a sensitive competitive ELISA for the determination of biotin in transformed yeast culture media.Methods The ELISA plate was firstly coated with Mycoplasma hyopneumoniae, and then successively incubated with rabbit ami-Mycoplasma hyopneumoniae serum and goat anti-rabbit IgG-biotin to form the solid biotin, which competed with the biotin in the solution (standard or sample) for the limited streptavidin-horse radish peroxidase conjugate. The standard calibration curve for biotin analysis was constructed in the range of 50-2000ng·L^-1. Results The detection limit for biotin was found to be 83 ng·L^-1 , which waa about 1000 times lower than the lowest determination concenlration in the reported ELISA for biotin analysis. The relative standard deviations for the spiked samples at biotin concerarations of 200 ng·L^-1, 500 ng·L^-1 , and 1000 ng·L^-1 were 24.87%, 6.15%, and 7.86%, respectively, with the average recovery of 101.13%. The wild yeast and its sixty-three transformed yeast culture media were applied to the developed ELISA for the determination of biotin. It was found that the biotin concentrations in more than 85 % of the tested samples were enhanced with different increase factors after transformation. | 16 | Synthesis and Characterization of A Small Molecule CFTR Chloride Channel Inhibitor显示文摘A thiazolidinone CFTR inhibitor(CFTR_ inh-172 ) was synthesized by a three-step procedure with trifluromethylaniline as the starting material. The synthesized CFTR inhibitor was characterized structurally by means of 1H NMR and functionally in a CFTR-expressing cell line FRT/hCFTR/EYFP-H148Q by both fluorescent and electrophysiological methods. A large amount(100 g) of high-quality small molecule thiazolidinone CFTR chloride channel inhibitor,CFTR_ inh-172 ,can be produced with this simple three-step synthetic procedure. The structure of the final product 2-thioxo-3-(3-trifluromethylphenyl)-5-[4-carboxyphenyl- methylene]-4-thiazolidinone was confirmed by 1H NMR. The overall yield was 58% with a purity over 99% as analyzed by HPLC. The synthesized CFTR_ inh-172 specifically inhibited CFTR chloride channel function in a cell-based fluorescence assay( K _d≈1.5 μmol/L) and in a Ussing chamber-based short-circuit current assay( K _d≈0.2 μmol/L),indicating better quality than that of the commercial combinatorial compound. The synthesized inhibitor is nontoxic to cultured cells at a high concentration and to mouse at a high dose. The synthetic procedure developed here can be used to produce a large amount of the high-quality CFTR_ inh-172 suitable for antidiarrheal studies and for creation of cystic fibrosis models in large animals.
