| 1 | Risks of temperature extremes over China under 1.5℃ and 2℃ global warming显示文摘The Paris Agreement aims to keep global warming to well below 2℃ above pre-industrial levels and to pursue efforts to limit it to 1.5℃,recognizing this will reduce the risks of natural disasters significantly.As changes in the risks of temperature extremes are often associated with changes in the temperature probability distribution,further analysis is still needed to improve understanding of the warm extremes over China.In this study,changes in the occurrence probability of temperature extremes and statistic characteristics of the temperature distribution are investigated using the fifth phase of the Coupled Model Intercomparison Project(CMIP5)multimodel simulations from 1861 to 2100.The risks of the once-in-100-year TXx and TNx events are projected to increase by 14.4 and 31.4 times at 1.5℃ warming.Even,the corresponding risks under 2℃ global warming are 23.3 and 50.6,implying that the once-in-100-year TXx and TNx events are expected to occur about every 5 and 2 years over China,respectively.The Tibetan Plateau,Northwest China and south of the Yangtze River are in greater risks suffering hot extremes(both day and night extremes).Changes in the occurrence probability of warm extremes are generally well explained by the combination of the shifts in location and scale parameters in areas with grown variability,i.e.,the Tibetan Plateau for TXx,south of the Yangtze River for both TXx and TNx.The location(scale)parameter leading the risks of once-in-20-year TXx to increase by more than 5(0.25)and 3(0.75)times under 2℃ warming in the Tibetan Plateau and south of the Yangtze River,respectively.The location parameter is more important for regions with decreased variability e.g.,the Tibetan Plateau for TNx,Northwest China for both TXx and TNx,with risks increase by more than 3,6 and 4 times due to changes in location. | SHI Chen JIANG Zhi-Hong ZHU Lian-Hua Xuebin ZHANG YAO Yi-Yi Laurent LI | 2020 | Advances in Climate Change Research2020,11,3: | 3 |

| 3 | The circular RNA circINPP4B acts as a sponge of miR-30a to regulate Th17 cell differentiation during progression of experimental autoimmune encephalomyelitis显示文摘Circular RNAs(circRNAs)regulate gene expression and participate in various biological and pathological processes.However,little is known about the effects of specific circRNAs on T helper cell 17(Th17)differentiation and related autoimmune diseases,such as multiple sclerosis(MS).Here,using transcriptome microarray analysis at different stages of experimental autoimmune encephalomyelitis(EAE),we identified the EAE progression-related circINPP4B,which showed upregulated expression in Th17 cells from mice with EAE and during Th17 differentiation in vitro.Silencing of circINPP4B inhibited Th17 differentiation and alleviated EAE,characterized by less demyelination and Th17 infiltration in the spinal cord.Mechanistically,circINPP4B served as a sponge that directly targeted miR-30a to regulate Th17 differentiation.Furthermore,circINPP4B levels were associated with the developing phases of clinical relapsing-remitting MS patients.Our results indicate that circINPP4B plays an important role in promoting Th17 differentiation and progression of EAE by targeting miR-30a,which provides a potential diagnostic and therapeutic target for Th17-mediated MS. | Jingjing Han Wei Zhuang Wanhua Feng Fuxing Dong Fang Hua Ruiqin Yao Xuebin Qu | 2021 | Cellular & Molecular Immunology2021,18,9: | 2 |

| 12 | Conditional Deletion of Foxg1 Alleviates Demyelination and Facilitates Remyelination via the Wnt Signaling Pathway in Cuprizone-Induced Demyelinated Mice显示文摘The massive loss of oligodendrocytes caused by various pathological factors is a basic feature of many demyelinating diseases of the central nervous system(CNS). Based on a variety of studies, it is now well established that impairment of oligodendrocyte precursor cells(OPCs) to differentiate and remyelinate axons is a vital event in the failed treatment of demyelinating diseases. Recent evidence suggests that Foxg1 is essential for the proliferation of certain precursors and inhibits premature neurogenesis during brain development. To date, very little attention has been paid to the role of Foxg1 in the proliferation and differentiation of OPCs in demyelinating diseases of the CNS. Here, for the first time, we examined the effects of Foxg1 on demyelination and remyelination in the brain using a cuprizone(CPZ)-induced mouse model. In this work, 7-week-old Foxg1 conditional knockout and wild-type(WT) mice were fed a diet containing 0.2% CPZ w/w for 5 weeks, after which CPZ was withdrawn to enable remyelination. Our results demonstrated that, compared with WT mice, Foxg1-knockout mice exhibited not only alleviated demyelination but also accelerated remyelination of the demyelinated corpus callosum. Furthermore, we found that Foxg1 knockout decreased the proliferation of OPCs and accelerated their differentiation into mature oligodendrocytes both in vivo and in vitro. Wnt signaling plays a critical role in development and in a variety of diseases. GSK-3 b, a key regulatory kinase in the Wnt pathway, regulates the ability of b-catenin to enter nuclei, where it activates the expression of Wnt target genes. We then used SB216763,a selective inhibitor of GSK-3 b activity, to further demonstrate the regulatory mechanism by which Foxg1 affects OPCs in vitro. The results showed that SB216763 clearly inhibited the expression of GSK-3 b, which abolished the effect of the proliferation and differentiation of OPCs caused by the knockdown of Foxg1. These results suggest that Foxg1 is involved in the proliferation and differentiation of OPCs through the Wnt signaling pathway. | 13 | OpNAC1 transcription factor regulates the biosynthesis of the anticancer drug camptothecin by targeting loganic acid O-methyltransferase in Ophiorrhiza pumila显示文摘Camptothecin(CPT) is an anticancer pentacyclic quinoline alkaloid widely used to treat cancer patients worldwide. However, the biosynthetic pathway and transcriptional regulation of camptothecin are largely unknown. Ophiorrhiza pumila, the herbaceous plant from the Rubiaceae family, has emerged as a model plant for studying camptothecin biosynthesis and regulation. In this study, a high-quality reference genome of O. pumila with estimated size of ~456.90Mb was reported, and the accumulation level of camptothecin in roots was higher than that in stems and leaves. Based on its spatial distribution in the plant, we examined gene functions and expression by combining genomics with transcriptomic analysis.Two loganic acid O-methyltransferase(OpLAMTs)were identified in strictosidine-producing plant O.pumila, and enzyme catalysis assays showed that OpLAMT1 and not OpLAMT2 could convert loganic acid into loganin. Further knock-out of OpL AMT1expression led to the elimination of loganin and camptothecin accumulation in O. pumila hairy roots.Four key residues were identified in OpLAMT1 protein crucial for the catalytic activity of loganic acid to loganin. By co-expression network, we identified a NAC transcription factor, OpNAC1, as a candidate gene for regulating camptothecin biosynthesis.Transgenic hairy roots and biochemical assays demonstrated that OpNAC1 suppressed OpLAMT1 expression. | 16 | VAMP726 from maize and Arabidopsis confers pollen resistance to heat and UV radiation by influencing lignin content of sporopollenin显示文摘Sporopollenin in the pollen cell wall protects male gametophytes from stresses.Phenylpropanoid derivatives,including guaiacyl(G)lignin units,are known to be structural components of sporopollenin,but the exact composition of sporopollenin remains to be fully resolved.We analyzed the phenylpropanoid derivatives in sporopollenin from maize and Arabidopsis by thioacidolysis coupled with nuclear magnetic resonance(NMR)and gas chromatography–mass spectrometry(GC–MS).The NMR and GC–MS results confirmed the presence of p-hydroxyphenyl(H),G,and syringyl(S)lignin units in sporopollenin from maize and Arabidopsis.Strikingly,H units account for the majority of lignin monomers in sporopollenin from these species.We next performed a genome-wide association study to explore the genetic basis of maize sporopollenin composition and identified a vesicle-associated membrane protein(ZmVAMP726)that is strongly associated with lignin monomer composition of maize sporopollenin.Genetic manipulation of VAMP726 affected not only lignin monomer composition in sporopollenin but also pollen resistance to heat and UV radiation in maize and Arabidopsis,indicating that VAMP726 is functionally conserved in monocot and dicot plants.Our work provides new insight into the lignin monomers that serve as structural components of sporopollenin and characterizes VAMP726,which affects sporopollenin composition and stress resistance in pollen. | Wenqi Yang Dongdong Yao Haiyang Duan Junli Zhang Yaling Cai Chen Lan Bing Zhao Yong Mei Yan Zheng Erbing Yang Xiaoduo Lu Xuehai Zhang Jihua Tang Ke Yu Xuebin Zhang | 2023 | Plant Communications2023,4,6: | 0 |
