1Embryonic stem cells generated by nuclear transfer of human somatic nuclei into rabbit oocytes显示文摘To solve the problem of immune incompatibility, nuclear transplantation has been envisaged as a means to produce cells or tissues for human autologous transplantation. Here we have derived embryonic stem cells by the transfer of human somatic nuclei into rabbit oocytes. The number of blastocysts that developed from the fused nuclear transfer was comparable among nuclear donors at ages of 5, 42, 52 and 60 years, and nuclear transfer (NT) embryonic stem cells (ntES cells) were subsequently derived from each of the four age groups. These results suggest that human somatic nuclei can form ntES cells independent of the age of the donor. The derived ntES cells are human based on karyotype, isogenicity, in situ hybridization, PGR and immunocytochemistry with probes that distinguish between the various species. The ntES cells maintain the capability of sustained growth in an undifferentiated state, and form embryoid bodies, which, on further induction, give rise to cell types such as neuron and muscle, as well as mixed cell populations that express markers representative of all three germ layers. 2Multi-UAV reconnaissance task allocation for heterogeneous targets using an opposition-based genetic algorithm with double-chromosome encoding显示文摘This paper presents a novel multiple Unmanned Aerial Vehicles(UAVs) reconnaissance task allocation model for heterogeneous targets and an effective genetic algorithm to optimize UAVs' task sequence. Heterogeneous targets are classified into point targets, line targets and area targets according to features of target geometry and sensor's field of view. Each UAV is regarded as a Dubins vehicle to consider the kinematic constraints. And the objective of task allocation is to minimize the task execution time and UAVs' total consumptions. Then, multi-UAV reconnaissance task allocation is formulated as an extended Multiple Dubins Travelling Salesmen Problem(MDTSP), where visit paths to the heterogeneous targets must meet specific constraints due to the targets' feature. As a complex combinatorial optimization problem, the dimensions of MDTSP are further increased due to the heterogeneity of targets. To efficiently solve this computationally expensive problem, the Opposition-based Genetic Algorithm using Double-chromosomes Encoding and Multiple Mutation Operators(OGA-DEMMO) is developed to improve the population variety for enhancing the global exploration capability. 3Novel resonant pressure sensor based on piezoresistive detection and symmetrical in-plane mode vibration显示文摘In this paper,a novel resonant pressure sensor is developed based on electrostatic excitation and piezoresistive detection.The measured pressure applied to the diaphragm will cause the resonant frequency shift of the resonator.The working mode stress–frequency theory of a double-ended tuning fork with an enhanced coupling beam is proposed,which is compatible with the simulation and experiment.A unique piezoresistive detection method based on small axially deformed beams with a resonant status is proposed,and other adjacent mode outputs are easily shielded.According to the structure design,high-vacuum wafer-level packaging with different doping in the anodic bonding interface is fabricated to ensure the high quality of the resonator.The pressure sensor chip is fabricated by dry/wet etching,high-temperature silicon bonding,ion implantation,and wafer-level anodic bonding.The results show that the fabricated sensor has a measuring sensitivity of~19 Hz/kPa and a nonlinearity of 0.02%full scale in the pressure range of 0–200 kPa at a full temperature range of−40 to 80°C.The sensor also shows a good quality factor>25,000,which demonstrates the good vacuum performance.Thus,the feasibility of the design is a commendable solution for high-accuracy pressure measurements.Xiangguang Han Qi Mao Libo Zhao Xuejiao Li Li Wang Ping Yang Dejiang Lu Yonglu Wang Xin Yan Songli Wang Nan Zhu Zhuangde Jiang 2020Microsystems & Nanoengineering2020,6,1:4
4Transcriptome Analysis Reveals Clues into leaf-like flower mutant in Chinese orchid Cymbidium ensifolium显示文摘The floral morphology of Cymbidium ensifolium,a well-known orchid in China,has increasingly attracted horticultural and commercial attention.However,the molecular mechanisms that regulate flower development defects in C.ensifolium mutants are poorly understood.In this work,we examined a domesticated variety of C.ensifolium named‘CuiYuMuDan',or leaf-like flower mutant,which lacks typical characteristics of orchid floral organs but continues to produce sepal-to leaf-like structures along the inflorescence.We used comparative transcriptome analysis to identify 6234 genes that are differentially expressed between mutant and wild-type flowers.The majority of these differentially expre ssed genes are involved in membrane-building,anabolism regulation,and plant hormone signal transduction,implying that in the leaf-like mutant these processes play roles in the development of flower defects.In addition,we identified 152 differentially expre ssed transcription factors,including the bHLH,MYB,MIKC,and WRKY gene families.Moreover,we found 20 differentially expressed genes that are commonly involved in flower development,including MADS-box genes,CLAVATA3(CLV3),WUSCHEL(WUS),and PERIANTHIA(PAN).Among them,floral homeotic genes were further investigated by phylogenetic analysis and expression validation,which displayed distinctive spatial expression patterns and significant changes between the wild type and the mutant.This is the first report on the C.ensifolium leaf-like flower mutant transcriptome.Our results shed light on the molecular regulation of orchid flower development,and may improve our understanding of floral patterning regulation and advance molecular breeding of Chinese orchids.Yonglu Wei Jianpeng Jin Xiani Yao Chuqiao Lu Genfa Zhu Fengxi Yang 2020Plant Diversity2020,42,2:4
5Amur tiger stripes:individual identification based on deep convolutional neural network显示文摘The automatic individual identification of Amur tigers(Panthera tigris altaica)is important for population monitoring and making effective conservation strategies.Most existing research primarily relies on manual identifi-cation,which does not scale well to large datasets.In this paper,the deep convolution neural networks algorithm is constructed to implement the automatic individual identification for large numbers of Amur tiger images.The experimental data were obtained from 40 Amur tigers in Tieling Guaipo Tiger Park,China.The number of images collected from each tiger was approximately 200,and a total of 8277 images were obtained.The experiments were carried out on both the left and right side of body.Our results suggested that the recognition accuracy rate of left and right sides are 90.48%and 93.5%,respectively.The accuracy of our network has achieved the similar level compared to other state of the art networks like LeNet,ResNet34,and ZF_Net.The running time is much shorter than that of other networks.Consequently,this study can provide a new approach on automatic individual identification technology in the case of the Amur tiger.Chunmei SHI Dan LIU Yonglu CUI Jiajun XIE Nathan James ROBERTS Guangshun JIANG 2020Integrative Zoology2020,15,6:2
