1Cetuximab plus FOLFOX6 or FOLFIRI in metastatic colorectal cancer: CECOG trial显示文摘AIM: To investigate efficacy and safety of cetuximab combined with two chemotherapy regimens in patients with unresectable metastatic colorectal cancer (mCRC). METHODS: Randomized patients received cetuximab with 5-fluorouracil (5-FU), folinic acid (FA) and oxaliplatin (FOLFOX) 6 (arm A, n = 74) or 5-FU, FA and irinotecan (FOLFIRI) (arm B, n = 77). KRAS mutation status was determined retrospectively in a subset of tumors (n = 117). RESULTS: No significant difference was found between treatment arms A and B in the progression-free survival (PFS) rate at 9 mo, 45% vs 34%; median PFS, 8.6 mo vs 8.3 mo [hazard ratio (HR) = 1.06]; overall response rate (ORR) 43% vs 45% [odds ratio (OR) = 0.93] and median overall survival (OS), 17.4 mo vs 18.9 mo (HR = 0.98). Patients with KRAS wild-type tumors demonstrated improved PFS (HR = 0.55, P = 0.0051), OS, (HR = 0.62, P = 0.0296) and ORR (53% vs 36%) and in arm A, improved PFS (HR = 0.49, P = 0.0196), OS (HR = 0.48, P = 0.0201) and ORR (56%vs 30%), compared with patients with KRAS mutated tumors. In arm B no significant differences were found in efficacy by KRAS mutation status. Treatment in arms A and B was generally well tolerated. 2Motility-based label-free detection of parasites in bodily fluids using holographic speckle analysis and deep learning显示文摘Parasitic infections constitute a major global public health issue.Existing screening methods that are based on manual microscopic examination often struggle to provide sufficient volumetric throughput and sensitivity to facilitate early diagnosis.Here,we demonstrate a motility-based label-free computational imaging platform to rapidly detect motile parasites in optically dense bodily fluids by utilizing the locomotion of the parasites as a specific biomarker and endogenous contrast mechanism.Based on this principle,a cost-effective and mobile instrument,which rapidly screens~3.2 mL of fluid sample in three dimensions,was built to automatically detect and count motile microorganisms using their holographic time-lapse speckle patterns.We demonstrate the capabilities of our platform by detecting trypanosomes,which are motile protozoan parasites,with various species that cause deadly diseases affecting millions of people worldwide.Using a holographic speckle analysis algorithm combined with deep learningbased classification,we demonstrate sensitive and label-free detection of trypanosomes within spiked whole blood and artificial cerebrospinal fluid(CSF)samples,achieving a limit of detection of ten trypanosomes per mL of whole blood(~five-fold better than the current state-of-the-art parasitological method)and three trypanosomes per mL of CSF.We further demonstrate that this platform can be applied to detect other motile parasites by imaging Trichomonas vaginalis,the causative agent of trichomoniasis,which affects 275 million people worldwide.With its costeffective,portable design and rapid screening time,this unique platform has the potential to be applied for sensitive and timely diagnosis of neglected tropical diseases caused by motile parasites and other parasitic infections in resource-limited regions.Yibo Zhang Hatice Ceylan Koydemir Michelle M.Shimogawa Sener Yalcin Alexander Guziak Tairan Liu Ilker Oguz Yujia Huang Bijie Bai Yilin Luo Yi Luo Zhensong Wei Hongda Wang Vittorio Bianco Bohan Zhang Rohan Nadkarni Kent Hill Aydogan Ozcan 2018Light(Science & Applications)2018,7,1:6
33mL和5mL利多卡因眼球筋膜囊下麻醉在超声乳化手术中的比较研究(英文)显示文摘目的:分析比较3mL和5mL利多卡因眼球筋膜囊下麻醉对白内障超声乳化手术中眼压,镇痛效果和眼外肌失运动能力的作用效果。眼球筋膜囊下麻醉是白内障超声乳化术中常用的局麻方式。作为一种剂量依赖型的阻滞方式,良好的眼压、镇痛效果以及眼外肌失运动是手术成功需要考虑的重要因素。方法:在获得伦理批准和患者的知情同意后,随机纳入70例患者作为研究对象,分别给予3m L利多卡因(组一)和5m L利多卡因(组二)作眼球筋膜囊下麻醉。麻醉前测量眼压,麻醉后10min评估眼压,镇痛效果和眼外肌失运动能力,并记录球结膜水肿和结膜下出血等并发症。结果:在麻醉管理期间,由于两名患者要求镇静,故从本研究中排除。比较组内及组间患者在麻醉前、后的基本特征,如年龄、体质量、身高、眼轴长度以及ASA生理状态,均无明显差异。与组一相比,组二患者眼外肌失运动能力及镇痛效果均明显较好。组二中有13例患者(38.2%)可达到眼球完全不运动,而组一没有患者可达到该程度。组一中有18例患者(56.3%)术中完全无痛觉,14例患者(43.7%)存在轻微痛觉,组二中有28例患者(82.4%)无痛觉,6例患者(17.6%)术中有轻微痛觉。此外,两组患者在麻醉前与麻醉后10min,其眼压的变化均无统计学差异(P<0.05)。两组患者并发症发生情况无明显差异。结论:3m L利多卡因和5m L利多卡因作眼球筋膜囊下麻醉对眼压均无影响,但在白内障超声乳化手术中给予5m L利多卡因作麻醉,在镇痛及眼外肌失运动能方面效果更好。Muge Coban-Karatas Oya Yalcin Cok Rana Altan-Yaycioglu 2015国际眼科杂志2015,15,11:6
4Precancerous lesions of oral mucosa显示文摘Precancerous lesions of oral mucosa, known as potentially malignant disorders in recent years, are consists of a group of diseases, which should be diagnosed in the early stage. Oral leukoplakia, oral submucous fibrosis, and oral erythroplakia are the most common oral mucosal diseases that have a very high malignant transformation rate. Oral lichen planus is one of the potentially malignant disorders that may be seen in six different subtypes including papular, reticular, plaquelike, atrophic, erosive, and bullous type, clinically. Atrophic and erosive subtypes have the greater increased malignant transformation risk compared to another subtypes. Although there are various etiological studies, the etiology of almost all these diseases is not fully understood. Geographically, etiologic factors may vary.The most frequently reported possible factors are tobacco use, alcohol drinking, chewing of betel quid containing areca nut, and solar rays. Early diagnosis is very important and can be lifesaving, because in late stages, they may be progressed to severe dysplasia and even carcinoma in situ and/or squamous cell carcinoma. For most diseases, treatment results are not satisfactory in spite of miscellaneous therapies. 5Effects of resveratrol on hydrogen peroxide-induced oxidative stress in embryonic neural stem cells显示文摘Resveratrol,a natural phenolic compound,has been shown to prevent cardiovascular diseases and cancer and exhibit neuroprotective effects.In this study,we examined the neuroprotective and antioxidant effects of resveratrol against hydrogen peroxide in embryonic neural stem cells.Hydrogen peroxide treatment alone increased catalase and glutathione peroxidase activities but did not change superoxide dismutase levels compared with hydrogen peroxide + resveratrol treatment.Nitric oxide synthase activity and concomitant nitric oxide levels increased in response to hydrogen peroxide treatment.Conversely,resveratrol treatment decreased nitric oxide synthase activity and nitric oxide levels.Resveratrol also attenuated hydrogen peroxide-induced nuclear or mitochondrial DNA damage.We propose that resveratrol may be a promising agent for protecting embryonic neural stem cells because of its potential to decrease oxidative stress by inducing higher activity of antioxidant enzymes,decreasing nitric oxide production and nitric oxide synthase activity,and alleviating both nuclear and mitochondrial DNA damage.Sibel Konyalioglu Guliz Armagan Ayfer Yalcin Cigdem Atalayin Taner Dagci 2013Neural Regeneration Research2013,8,6:5
6Reflection and Transmission of Regular Waves from/Through Single and Double Perforated Thin Walls显示文摘In this paper, reflection and transmission coefficients of regular waves from/through perforated thin walls are investigated. Small scale laboratory tests have been performed in a wave flume firstly with single perforated thin Plexiglas plates of various porosities. The plate is placed perpendicular to the flume with the height from the flume bottom to the position above water surface. With this thin wall in the flume wave overtopping is prohibited and incident waves are able to transmit. The porosities of the walls are achieved by perforating the plates with circular holes.Model settings with double perforated walls parallel to each other forming so called chamber system, have been also examined. Several parameters have been used for correlating the laboratory tests' results. Experimental data are also compared with results from the numerical model by applying the multi-domain boundary element method(MDBEM)with linear wave theory. Wave energy dissipation due to the perforations of the thin wall has been represented by a simple yet effective porosity parameter in the model. 8Correlation of neutrophil/lymphocyte and platelet/lymphocyte ratio with visual acuity and macular thickness in age-related macular degeneration显示文摘AIM:To investigate the place of neutrophil-to-lymphocyte ratio(NLR) and platelet-to-lymphocyte ratio(PLR) in the diagnosis of and prognosis for neovascular age-related macular degeneration(AMD). METHODS:One hundred AMD patients and 100 healthy controls were included in the study. Blood samples were obtained from the venous blood, which is used for routine analysis, and these samples were subjected to complete blood count. NLR was defined as the neutrophil count divided by the number of lymphocytes, and PLR was defined as the platelet count divided by the number of lymphocytes. RESULTS:No statistically significant difference was observed between the two groups under consideration in terms of demographic features(P>0.05). The average NLR in the patient group was found to be significantly higher than that in the healthy control group(P<0.05). The average PLR was significantly higher in the patient group as compared to the control group(P<0.05). As best corrected visual acuity(BCVA) increased, both NLR and PLR decreased(significant negative correlations at 49.8% and 63.0%, respectively), whereas as central macular thickness(CMT) increased, both NLR and PLR increased(significant positive correlations at 59.3% and 70.0%, respectively).CONCLUSION:NLR and PLR levels are higher among neovascular AMD patients as compared to healthy control group. 11BRAF mutation in colorectal carcinomas with signet ring cell component显示文摘Objective:Signet ring cell carcinoma is a rare subtype of colorectal carcinoma(CRC)with an associated BRAFV600E mutation.We investigated frequencies of BRAF mutation in 28 CRCs containing variable signet ring cell component and their relation with clinicopathologic parameters.Methods:According to the presence of signet ring cell component,tumors were categorized into groups as follows:0%–9%,10%–24%,25%–49%,and>50%.Genomic DNA was isolated and analyzed for BRAF V600E gene mutation by polymerase chain reaction-restriction fragment length polymorphism.Eleven of 28 cases(39.3%)showed BRAFV600E mutation,which was also confirmed by Sanger sequencing.To elucidate the importance of existence of signet ring cell component at the molecular level,we separated cases into two groups with cut-off levels of 10%and 50%,which pertain to percentages of signet ring cells.Results:Seven of 19 cases(36.8%)under the threshold of 50%and four of nine cases(44.4%)over this threshold value demonstrated BRAF mutation.Three of 7 cases(42.8%)featuring<10%signet ring cell component and eight out of 21 cases(38.1%)showing>10%were BRAF mutated.Conclusions:BRAF mutation must be closely associated with the presence of malignant signet ring cells regardless of their percentages.Serap Yalcin Onder Onguru 2017Cancer Biology & Medicine2017,14,3:2
12A micromirror array with annular partitioning for high-speed random-access axial focusing显示文摘Dynamic axial focusing functionality has recently experienced widespread incorporation in microscopy,augmented/virtual reality(AR/VR),adaptive optics and material processing.However,the limitations of existing varifocal tools continue to beset the performance capabilities and operating overhead of the optical systems that mobilize such functionality.The varifocal tools that are the least burdensome to operate(e.g.liquid crystal,elastomeric or optofluidic lenses)suffer from low(≈100 Hz)refresh rates.Conversely,the fastest devices sacrifice either critical capabilities such as their dwelling capacity(e.g.acoustic gradient lenses or monolithic micromechanical mirrors)or low operating overhead(e.g.deformable mirrors).Here,we present a general-purpose random-access axial focusing device that bridges these previously conflicting features of high speed,dwelling capacity and lightweight drive by employing lowrigidity micromirrors that exploit the robustness of defocusing phase profiles.Geometrically,the device consists of an 8.2mm diameter array of piston-motion and 48-μm-pitch micromirror pixels that provide 2πphase shifting for wavelengths shorter than 1100 nm with 10-90% settling in 64.8μs(i.e.,15.44 kHz refresh rate).The pixels are electrically partitioned into 32 rings for a driving scheme that enables phase-wrapped operation with circular symmetry and requires<30 V per channel.Optical experiments demonstrated the array’s wide focusing range with a measured ability to target 29 distinct resolvable depth planes.Overall,the features of the proposed array offer the potential for compact,straightforward methods of tackling bottlenecked applications,including high-throughput single-cell targeting in neurobiology and the delivery of dense 3D visual information in AR/VR.Nathan Tessema Ersumo Cem Yalcin Nick Antipa Nicolas Pegard Laura Waller Daniel Lopez Rikky Muller 2020Light(Science & Applications)2020,9,1:2
13The cross section calculation of 112Sn(α,γ)116Te reaction with different nuclear models at the astrophysical energy range显示文摘The theoretical cross section calculations for the astrophysical p process are needed because most of the related reactions are technically very difficult to be measured in the laboratory. Even if the reaction was measured,most of the measured reactions have been carried out at the higher energy range from the astrophysical energies.Therefore, almost all cross sections needed for p process simulation have to be theoretically calculated or extrapolated to the astrophysical energies.^(112)Sn(α,γ)^(116)Te is an important reaction for the p process nucleosynthesis. The theoretical cross section of ^(112)Sn(α,γ)^(116)Te reaction was investigated for different global optical model potentials,level density, and strength function models at the astrophysically interested energies. Astrophysical S factors were calculated and compared with experimental data available in the EXFOR database. The calculation with the optical model potential of the dispersive model by Demetriou et al., and the back-shifted Fermi gas level density model and Brink-Axel Lorentzian strength function model best served to reproduce experimental results at an astrophysically relevant energy region. The reaction rates were calculated with these model parameters at the p process temperature and compared with the current version of the reaction rate library Reaclib and Starlib.C.
