| 1 | Bufalin suppresses the migration and invasion of prostate cancer cells through HOTAIR, the sponge of miR-520b显示文摘Bufalin, the major active component of the traditional Chinese medicine ChanSu obtained from the skin and parotid venom glands of toads, has long been known as an anticancer agent. Recent studies show that microRNAs (miRs) are involved in the anticancer activities of bufalin, while long non-coding RNAs (lncRNAs) are known to interact with miRNAs to regulate various biological functions. In this paper, we investigated the possible network related to the antimetastatic effect of bufalin in prostate cancer (PCa) cells. We demonstrated that bufalin (0.05?10 μM) dose-dependently suppressed the proliferation of prostate cancer DU145 and PC3 cells with IC50 values of 0.89 and 1.28 μM, respectively. Furthermore, bufalin treatment significantly suppressed the cell migration and invasion. To explore the role of lncRNAs in the antimetastatic activity of bufalin, we used an lncRNA microarray and found that HOX transcript antisense RNA (HOTAIR) was the most markedly downregulated lncRNA in bufalin-treated PCa cells. Overexpression of HOTAIR counteracted the suppressing effects of bufalin on DU145 and PC3 cells. We then predicted and verified that HOTAIR upregulated FGFR1 expression by sponging miR-520b in PCa cells. In 40 patients with PCa bone metastasis, we used in situ hybridization or immunohistochemical assay to assess the HOTAIR and FGFR1 expression, which revealed that both HOTAIR and FGFR1 expression were significantly higher in bone metastasis tissues than in the primary PCa tissues. In addition, the level of serum HOTAIR was positively associated with the levels of serum bone metabolic markers (CTx, OST, B-ALP and PINP) and may serve as a reasonable biomarker for PCa bone metastasis. | 2 | Associations between individual and environmental factors and habitual physical activity among older Chinese adults:A social–ecological perspective显示文摘Purpose: To examine, within a social–ecological framework, associations between multifaceted individual and environmental factors and habitual physical activity(HPA) among older Chinese adults.Methods: Through a mix of qualitative and quantitative methods, a survey instrument assessing various factors underlying 3 social–ecological dimensions of intrapersonal, interpersonal, and community and environmental resources was developed. Using a cross-sectional design, older adults(n = 1580, aged 67 ± 7 years) recruited from 10 communities in Shandong province completed the social–ecological survey of HPA. | 3 | Potato/Maize intercropping reduces infestation of potato tuber moth, Phthorimaea operculella(Zeller) by the enhancement of natural enemies显示文摘The potato tuber moth(PTM),Phthorimaea operculella(Zeller),is one of the most economically significant insect pests for potato in both field and storage worldwide.To evaluate the infestation,reduction of potato yield and the control efficacy for PTM,field tests were conducted in two seasons by intercropping of potato as the host plant with maize as a non-host plant of PTM.Three intercropping patterns were tested,which were 2 rows of potatoes with either 2,3,or 4 rows of maize(abbreviated 2 P:2 M,2 P:3 M,and 2 P:4 M),and the monocropped potato as the control,2 rows of potatoes,without maize,(abbreviated 2 P:0 M).Results showed that the population and infestation of PTM in the 2 P:3 M intercropping pattern was significantly lower than those in 2 P:2 M,2 P:4 M and the monocropping pattern of 2 P:0 M,due to the enhancement of natural enemies.Cumulative mines and tunneling in potato leaves in 2 P:3 M intercropping were significantly lower than those in 2 P:2 M and 2 P:4 M patterns.The population of parasitoids and the parasitism rate of PTM in intercropping pattern of 2 P:3 M were significantly higher than that in intercropping pattern of 2 P:2 M,2 P:4 M and monocropping pattern of 2 P:0 M.We conclude that the potato intercropped with maize reduced the adult and larva populations,and reduced the damage from PTM by enhancing the number of parasitoids and the level of parasitism.The greatest population density of parasitoids and parasitism rate were in the intercropping pattern of 2 rows of potatoes with 3 rows of maize.These data indicate that the host/non-host intercropping patterns can be used as a biological control tactic against PTM by enhancing the density of natural enemies in the agro-ecosystems. | ZHENG Ya-qiang ZHANG Li-min CHEN Bin YAN Nai-sheng GUI Fu-rong ZAN Qing-an DU Guang-zu HE Shu-qi LI Zheng-yue GAO Yu-lin XIAO Guan-li | 2020 | Journal of Integrative Agriculture2020,19,2: | 8 |

| 7 | Absorption characteristics of elemental mercury in mercury chloride solutions显示文摘Elemental mercury(Hg0) in flue gases can be efficiently captured by mercury chloride(Hg Cl2) solution. However, the absorption behaviors and the influencing effects are still poorly understood. The mechanism of Hg0 absorption by Hg Cl2 and the factors that control the removal were studied in this paper. It was found that when the mole ratio of Cl-to Hg Cl2 is 10:1, the Hg0 removal efficiency is the highest. Among the main mercury chloride species,Hg Cl3-is the most efficient ion for Hg0 removal in the Hg Cl2 absorption system when moderate concentrations of chloride ions exist. The Hg0 absorption reactions in the aqueous phase were investigated computationally using Moller–Plesset perturbation theory. The calculated Gibbs free energies and energy barriers are in excellent agreement with the results obtained from experiments. In the presence of SO32-and SO2, Hg2+reduction occurred and Hg0 removal efficiency decreased. The reduced Hg0 removal can be controlled through increased chloride concentration to some degree. Low p H value in Hg Cl2 solution enhanced the Hg0 removal efficiency, and the effect was more significant in dilute Hg Cl2 solutions. | 8 | Controlled release of hydrogen by implantation of magnesium induces P53-mediated tumor cells apoptosis显示文摘Hydrogen has been used to suppress tumor growth with considerable efficacy.Inhalation of hydrogen gas and oral ingestion of hydrogen-rich saline are two common systemic routes of hydrogen administration.We have developed a topical delivery method of hydrogen at targeted sites through the degradation of magnesium-based biomaterials.However,the underlying mechanism of hydrogen’s role in cancer treatment remains ambiguous.Here,we investigate the mechanism of tumor cell apoptosis triggered by the hydrogen released from magnesium-based biomaterials.We find that the localized release of hydrogen increases the expression level of P53 tumor suppressor proteins,as demonstrated by the in vitro RNA sequencing and protein expression analysis.Then,the P53 proteins disrupt the membrane potential of mitochondria,activate autophagy,suppress the reactive oxygen species in cancer cells,and finally result in tumor suppression.The anti-tumor efficacy of magnesium-based biomaterials is further validated in vivo by inserting magnesium wire into the subcutaneous tumor in a mouse.We also discovered that the minimal hydrogen concentration from magnesium wires to trigger substantial tumor apoptosis is 91.2μL/mm^(3)per day,which is much lower than that required for hydrogen inhalation.Taken together,these findings reveal the release of H2 from magnesium-based biomaterial exerts its anti-tumoral activity by activating the P53-mediated lysosome-mitochondria apoptosis signaling pathway,which strengthens the therapeutic potential of this biomaterial as localized anti-tumor treatment. | Rui Zan Hao Wang Weijie Cai Jiahua Ni Berengere J.C.Luthringer-Feyerabend Wenhui Wang Hongzhou Peng Weiping Ji Jun Yan Jiazeng Xia Yang Song Xiaonong Zhang | 2022 | Bioactive Materials2022,7,3: | 3 |
| 9 | High expression of human serum albumin in milk of trans-genie mice directed by the goat β-casein gene promoter region显示文摘We have constructed a mammary gland expression vector that contained the goat β-casein gene pro-moter, 5’upstream regulatory region, exons 1, 2, intron 1 as well as the human serum albumin (hALB) mini-gene (including the full-long sequences of hALB cDNA and its intron 1). Injection of the vector into mouse tail veins showed that the recombinant construct was expressed only in mammary glands. The vector was microinjected into the mouse fertilized eggs, followed by transferring the eggs into the foster mice. 33 F0 mice were obtained. Of the 33, 8 mice (5 , 3 ) were transgenic with hALB gene integration identified by PCR as well as Southern blot hybridization. The integration rate was 24.2% (8/33). Western blot analysis showed that 3 female transgenic mice had hALB expression in their milk. | 19 | Coupling model and optimal combination scheme of water,fertilizer,dissolved oxygen and temperature in greenhouse tomato under drip irrigation显示文摘Water-fertilizer coupling technology has been widely used in the world.Poor soil aeration,low temperature or high temperature can affect the rate of nutrient uptake by crop roots.Aiming at the interaction between water,fertilizer,dissolved oxygen and temperature(WFOT)coupling model and irrigation flux of tomato in greenhouse,using these four factors with a five-level uniform-precision rotatable central composite design,a mathematical model was established among the four factors affecting tomato yield in a greenhouse,and the optimal combination scheme of WFOT was obtained.Within the test range,tomato yields increased with increasing irrigation quotas(X_(1)),fertilization amount(X_(2)),dissolved oxygen(X_(3))and geothermal pipe water temperature(X_(4)).The magnitude of the effect of each factor of WFOT on tomato yield was in the following order:X_(1),X_(2),X_(4),X_(3)(spring and summer),and X_(1),X_(3),X_(2),X_(4)(autumn and winter).The interaction between high water-low heat and low water-high heat were beneficial for yield increase(spring and summer),the high fertilizer-low heat and low fertilizer-high heat interactions were beneficial to yield increase(autumn and winter).If WFOT agronomic measures were adopted according to the 95%confidence interval,there was a 95%probability that the spring-summer tomato yield will be higher than 89902 kg/hm^(2).The WFOT coupling scheme was X_(1)of 4808-5091 m3/hm^(2),X_(2)(N-P_(2)O_(5)-K_(2)O)of 171-57-84 to 186-62-89 kg/hm^(2),X_(3)of 7.9-8.2 mg/L,and X_(4)of 34.9°C-37.0°C.There was a 95%probability of tomato yield higher than 85209 kg/hm^(2)in autumn and winter,and the WFOT coupling scheme was X_(1)of 5270-5416 m3/hm^(2),X_(2)(N-P_(2)O_(5)-K_(2)O)of 151-50-76 to 167-56-82 kg/hm^(2),X_(3)of 8.0-8.2 mg/L,and X_(4)of 34.1°C-36.2°C.Overall,and the model had a very good simulation effect,with application value.The relative error between spring-summer and autumn-winter yields ranged from 1.12%to 25.34%.The results of the study can provide a theoretical basis for improving the quality and efficiency of greenhouse tomatoes. | Zan Ouyang Juncang Tian Ce Zhao Xinfang Yan | 2021 | International Journal of Agricultural and Biological Engineering2021,14,6: | 1 |
