3Cancer-educated neutrophils promote lung cancer progression via PARP-1-ALOX5-mediated MMP-9 expression显示文摘Objective:Neutrophils are one of the most predominant infiltrating leukocytes in lung cancer tissues and are associated with lung cancer progression.How neutrophils promote lung cancer progression,however,has not been established.Methods:Kaplan–Meier plotter online analysis and tissue immunohistochemistry were used to determine the relationship between neutrophils and overall survival in lung cancer patients.The effect of neutrophils on lung cancer was determined using the Transwell migration assay,a proliferation assay,and a murine tumor model.Gene knockdown was used to determine poly ADPribose polymerase(PARP)-1 function in lung cancer-educated neutrophils.Western blot analysis and gelatin zymography were used to demonstrate the correlation between PARP-1 and matrix metallopeptidase 9(MMP-9).Immunoprecipitation coupled to mass spectrometry(IP/MS)was used to identify the proteins interacting with PARP-1.Co-immunoprecipitation(Co-IP)was used to confirm that PARP-1 interacts with arachidonate 5-lipooxygenase(ALOX5).Neutrophil PARP-1 blockage by AG14361 rescued neutrophil-promoted lung cancer progression.Results:An increased number of infiltrating neutrophils was negatively associated with overall survival in lung cancer patients(P<0.001).Neutrophil activation promoted lung cancer cell invasion,migration,and proliferation in vitro,and murine lung cancer growth in vivo.Mechanistically,PARP-1 was shown to be involved in lung cancer cell-induced neutrophil activation to increase MMP-9 expression through interacting and stabilizing ALOX5 by post-translational protein modification(PARylation).Blocking PARP-1 by gene knockdown or AG14361 significantly decreased ALOX5 expression and MMP-9 production,and eliminated neutrophil-mediated lung cancer cell invasion and in vivo tumor growth.Conclusion:We identified a novel mechanism by which PARP-1 mediates lung cancer cell-induced neutrophil activation and PARylates ALOX5 to regulate MMP-9 expression,which exacerbates lung cancer progression.Lulu Han Yuxin Chen Nan Huang Xiaowan Zhou Yanfang Lv Huizhong Li Dafei Chai Junnian Zheng Gang Wang 2024Cancer Biology & Medicine2024,21,2:0
4Characterization and expression analysis of genes encoding Taxol biosynthetic enzymes in Taxus spp.显示文摘Taxol(Paclitaxel),an important anticancer drug,is derived at very low yields from Taxus(yew)species that grow very slowly.In the present study,thirteen genes that encode enzymes involved in Taxol biosynthesis in Taxus spp.were analyzed with bioinformatics methods,and their expression levels in different tissues and after cold and hormone treatments were also analyzed.The results indicated that many cis-elements related to abiotic stresses and hormones were found in the promoter sequences of the 8 genes involved in Taxol biosynthesis.Moreover,the 13 enzymes encoded by the target genes were located in different organelles and had many phosphorylation sites in the response proteins.The 13 genes were expressed highly either in roots or in stems,with lower transcripts in needles,and they were highly expressed after treatment with cold,gibberellin,methyl jasmonate or coronatine,consistent with predictions based on the bioinformatics analysis.These results suggest that the factors such as hormones and abiotic stresses stimulate taxane biosynthesis in yews,providing an important way to sustainably generate taxanes from yew trees or their cell cultures to improve Taxol yields.Luyuan Jiang Kaikai Zhang Xing Lv Liying Yang Shuai Wang Duanfen Chen Yanfang Yang Deyou Qiu 2021Journal of Forestry Research2021,32,6:0
5Measurement and Characteristics of the Integration of China’s Trade in Services into Digital Global Value Chain显示文摘In this paper,the characteristics of digital trade in services of China are measured and revealed from the perspective of digitalization of trade in services.A digital global value chain is constructed and comprehensively analyzed at the three levels of path decomposition,two-way digital connection and bilateral connection.The study finds that the digital added value of China’s services driven by domestic demand outweighs that driven by foreign demand,and most countries engage in digital trade in services with China through simple participation;the role of China’s services in the digital global value chain is shifting from“digital value input”to“digital value output”,but its relative position is still low;China’s dependence on the import of digital intermediate products from developed countries has been significantly reduced,and the country has become the main source of digital intermediate imports for most countries,acting as a“hub”in the digital global value chain.This study comprehensively evaluates the strategic positioning and paths of integrating China’s services into the digital global value chain in multiple dimensions under the unifi ed accounting framework.It provides the reference for further improving the statistics accounting framework of China’s digital trade and promoting the high-quality development of the digital economy.Yanfang Lv Ruonan Fang Dong Wang 2021China Finance and Economic Review2021,10,1:0
