| 2 | Water-induced softening behavior of clay-rich sandstone in Lanzhou Water Supply Project,China显示文摘The strength of clay-rich sandstone decreases significantly when in contact with water due to softening effects.This scenario can pose a severe threat to the stability of water diversion tunnels during construction and operation periods.To address the issues related to water-induced softening in clay-rich sandstone zones in a water diversion tunnel of Lanzhou Water Supply Project,the microscopic and micromechanical variations of rocks due to increasing water content in two different zones i.e.zones A and B,were determined by various testing methods,such as X-ray diffraction(XRD),scanning electron microscopy(SEM),thin section microscopy,micro-indentation test,sonic velocity test,and slake durability test.The microscopic analysis confirms the presence of montmorillonite mineral which is the dominant problematic geomaterial in engineering application.The integrity and durability of clay-rich sandstone were determined with sonic velocity and slake durability tests to calibrate the results obtained by the micro-indentation test.It shows that the elastic modulus and hardness of clay-rich sandstone decrease with the increase of saturation time,up to 144 h,which is more significant and rapid during early stage of saturation.After 144 h of saturation,the elastic modulus decreases by 89% and 97%,and the hardness decreases by 89% and 99% for zones A and B sandstones,respectively.The results of slake durability and sonic velocity indicate that zone A sandsto ne remains 56.19% durability after 144 h of saturation,while zone B sandstone loses its durability merely after 72 h of saturation.The clay-rich sandstone starts to dissolve in water when the saturation time exceeds 144 h.The significant decreases in strength and durability of clay-rich sandstone due to water-induced softening are serious threats to tunnel stability.The improvements in the strength of surrounding rock mass by grouting and permeability by installation of drainage galleries can reduce the damage caused by water-induced softening. | Muhammad Usman Azhar Hui Zhou Fanjie Yang Adnan Younis Xinjing Lu Houguo Fang Yijun Geng | 2020 | Journal of Rock Mechanics and Geotechnical Engineering2020,12,3: | 3 |
| 3 | Generation of male-sterile soybean lines with the CRISPR/Cas9 system显示文摘Soybean [Glycine max(L.) Merr.] provides a rich source of plant protein and oil worldwide. The commercial use of transgenic technology in soybean has become a classical example of the application of biotechnology to crop improvement. Although genetically modified soybeans have achieved commercial success,hybrid soybean breeding is also a potential way to increase soybean yield. Soybean cytoplasmic malesterile(CMS) lines have been used in three-line hybrid breeding systems, but their application to exploiting soybean heterosis has been limited by rare germplasm resource of sterile lines. The generation of various genetic diversity male-sterile soybean lines will help to overcome the shortcoming. In this study,we used targeted editing of AMS homologs in soybean by CRISPR/Cas9 technology for the first time to generate stable male-sterile lines. Targeted editing of GmAMS1 resulted in a male-sterile phenotype,while editing of GmAMS2 failed to produce male-sterile lines. | 5 | Geological Conditions and Prospect Forecast of Shale Gas Formation in Qiangtang Basin, Qinghai-Tibet Plateau显示文摘The presence of shale gas has been confirmed in almost every marine shale distribution area in North America. Formation conditions of shale gas in China are the most favorable for marine, organic-rich shale as well. But there has been little research focusing on shale gas in Qiangtang Basin, Qinghai-Tibet Plateau, where a lot of Mesozoic marine shale formations developed. Based on the survey results of petroleum geology and comprehensive test analysis data for Qinghai-Tibet Plateau, for the first time, this paper discusses characteristics of sedimentary development, thickness distribution, geochemistry, reservoir and burial depth of organic-rich shale, and geological conditions for shale gas formation in Qiangtang Basin. There are four sets of marine shale strata in Qiangtang Basin including Upper Triassic Xiaochaka Formation(T3x), Middle Jurassic Buqu Formation(J2b), Xiali Formation(J2x) and Upper Jurassic Suowa Formation(J3s), the sedimentary types of which are mainly bathyal-basin facies, open platform-platform margin slope facies, lagoon and tidal-flat facies, as well as delta facies. By comparing it with the indicators of gas shale in the main U.S. basins, it was found that the four marine shale formations in Qiangtang Basin constitute a multi-layer distribution of organic-rich shale, featuring a high degree of thickness and low abundance of organic matter, high thermal evolution maturity, many kinds of brittle minerals, an equivalent content of quartz and clay minerals, a high content of feldspar and low porosity, which provide basic conditions for an accumulation of shale gas resources. Xiaochaka Formation shale is widely distributed, with big thickness and the best gas generating indicators. It is the main gas source layer. Xiali Formation shale is of intermediate thickness and coverage area, with relatively good gas generating indicators and moderate gas formation potential. Buqu Formation shale and Suowa Formation shale are of relatively large thickness, and covering a small area, with poor gas generating indicators, and limited gas formation potential. The shale gas geological resources and technically recoverable resources were estimated by using geologic analogy method, and the prospective areas and potentially favorable areas for Mesozoic marine shale gas in Qiangtang Basin are forecast and analyzed. It is relatively favorable in a tectonic setting and indication of oil and gas, shale maturity, sedimentary thickness and gypsumsalt beds, and in terms of mineral association for shale gas accumulation. | 6 | 树突状细胞疫苗对人化荷人膀胱癌SCID鼠循环微转移转归的影响(英文)显示文摘Objective: To study the effect of dendritic cells loaded with whole tumor antigen on hematogenous micrometastasis of bladder cancer model in hu-PBL-SCID mice. Methods: T24-3 cell subset was selected from human bladder transitional cell carcinoma T24 cell line by Boyden chamber system. The SCID mice intraperitoneally injected with 4 × 107 hu-PBL and subcutaneously injected with 3 × 106 T24-3 cells were named hu-PBL-T24-3-SCID model. Human IgG level in the blood plasma of mice was detected by ELISA, and human CD3+, CD4+, CD8+ T cells in blood and spleen cells of mice were detected by FCM analysis for human immune reconstruction study. Human CK20 mRNA expression in mice peripheral blood was detected by RT-PCR to investigate metastasis of tumor cells. The PBMCs were isolated from human peripheral blood, and were induced into DCs by co-culture with rhGM-CSF and rhIL-4 in vitro. The DC vaccines were produced by co-culturing with whole tumor antigen which was purified through freezing and melting T24-3 cell subset. After T24-3 cells injected into SCID mice for 5 weeks, the mice were treated with DC vaccines. Results: All mice were initially treated at 5th week. The expression of CK20 mRNA in peripheral blood of DC vaccines treated mice was the lowest. There was 2 mice showing CK20 mRNA expression and 3 mice with metastasis tumor in PBS group. MMP-7 mRNA expression in tumor tissues of DC vaccines treated mice was statistically lower than that of PBS group (P < 0.01). | 7 | Molecular-level effects of acupuncture on depression: a genome-wide transcriptome analysis of pituitary gland in rats exposed to chronic restraint stress显示文摘OBJECTIVE: To investigate the antidepressant mechanism of acupuncture at the molecular level in the pituitary gland of rats exposed to chronic restraint stress(CRS).METHODS: Forty-eight rats were randomly divided into a control group(C), model group(M), fluoxetine group(F), and acupuncture group(A). CRS was induced in the model group by restraining rats for 28 days. Groups F and A were given fluoxetine administration and acupuncture intervention, respectively, 1 h before the CRS procedures were implemented. On the 29 th day, rat pituitary glands were harvested for RNA extraction and RNA-sequencing analysis.RESULTS: Differentially expressed gene analysis identified 148, 87, and 179 response genes differen-tially expressed in group C vs group M, group F vs group M, and group A vs group M, respectively. | 8 | Mitigate noisy data for smart IoT via GAN based machine unlearning显示文摘With the development of IoT applications,machine learning dramatically improves the utility of variable IoT systems such as autonomous driving.Although the pretrain-finetune framework can cope well with data heterogeneity in complex IoT scenarios,the data collected by sensors often contain unexpected noisy data,e.g.,out-of-distribution(OOD)data,which leads to the reduced performance of fine-tuned models.To resolve the problem,this paper proposes Mu GAN,a method that can mitigate the side-effect of OOD data via the generative adversarial network(GAN)-based machine unlearning.Mu GAN follows a straightforward but effective idea to mitigate the performance loss caused by OOD data,i.e.,“flashbacking”the model to the condition where OOD data are excluded from model training.To achieve the goal,we design an adversarial game,where a discriminator is trained to identify whether a sample belongs to the training set by observing the confidence score.Meanwhile,a generator(i.e.,the target model)is updated to fool the discriminator into believing that the OOD data are not included in the training set but others do.The experimental results show that benefiting from the high unlearning rate(more than 90%)and retention rate(99%),Mu GAN succeeds in lowering the model performance degradation caused by OOD data from 5.88%to 0.8%. | Zhuo MA Yilong YANG Yang LIU Xinjing LIU Jianfeng MA | 2024 | Science China(Information Sciences)2024,67,3: | 0 |
