1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
2Impacts of changing scale on Getis-Ord Gi* hotspots of CPUE:a case study of the neon flying squid (Ommastrephes bartramii)in the northwest Pacific Ocean显示文摘We examined the scale impacts on spatial hot and cold spots of CPUE for Ommastrephes bartramii in the northwest Pacific Ocean. The original fishery data were tessellated to 18 spatial scales from 5′×5′ to 90′×90′ with a scale interval of 5′ to identify the local clusters. The changes in location, boundaries, and statistics regarding the Getis-Ord Gi* hot and cold spots in response to the spatial scales were analyzed in detail. Several statistics including Min, mean, Max, SD, CV, skewness, kurtosis, first quartile(Q1), median, third quartile(Q3), area and centroid were calculated for spatial hot and cold spots. Scaling impacts were examined for the selected statistics using linear, logarithmic, exponential, power law and polynomial functions. Clear scaling relations were identified for Max, SD and kurtosis for both hot and cold spots. For the remaining statistics, either a difference of scale impacts was found between the two clusters, or no clear scaling relation was identified. 3Stereotactic aspiration for hypertensive intracerebral haemorrhage in a Chinese population:a retrospective cohort study显示文摘Objective We aimed to compare the therapeutic effects of stereotactic aspiration and best medical management in patients who developed supratentorial hypertensive intracerebral haemorrhage(HICH)with a volume of haemorrhage between 20 and 40 mL.Methods The clinical data of 220 patients with supratentorial HICH with a volume between 20 and 40 mL were retrospectively analysed.Among them,142 received stereotactic aspiration surgery(stereotactic aspiration group)and 78 received best medical management(conservative group).All were followed up for 6 months.Multivariate logistic regression and Kaplan-Meier survival curves were used to compare the outcome between the two groups.results The rebleeding rate was lower in the group that had stereotactic aspiration when compared with the group with medical treatment(6[4.2%]vs 9[11.5%],χ2=4.364,p=0.037).After 6 months,although the mortality rate did not differ significantly between the two groups(8 cases[5.6%]vs 10 cases[12.8%],χ2=3.461,p=0.063),the rate of a favourable outcome was higher in the group who received stereotactic aspiration(χ2=15.870,p=0.000).Logistic regression identified that medical treatment(OR=1.64,p=0.000)was an independent risk factor for an unfavourable outcome.The Kaplan-Meier curves indicated that the median favourable outcome time in the stereotactic aspiration group was 59.5 days compared with that in the medically treated group(87.0 days).The log-rank test indicated that the prognosis at 6 months was better for those treated with stereotactic haematoma aspiration(χ2=29.866,p=0.000).However,the 6-month survival rate was similar between the two groups(χ2=3.253,p=0.068).Conclusions Stereotactic haematoma aspiration significantly improved the quality of life,although did not effectively reduce the rate of mortality.When selected appropriately,patients with HICH may benefit from this type of surgical intervention.Xuyang Zhang Shaolong Zhou Qiang Zhang Xudong Fu Yuehui Wu Jiasheng Liu Bo Liang Zhuo Yang Xinjun Wang 2019Stroke & Vascular Neurology2019,4,1:19
4Effect of microbial mediated iron plaque reduction on arsenic mobility in paddy soil显示文摘The potential of microbial mediated iron plaque reduction, and associated arsenic (As) mobility were examined by iron reducing bacteria enriched from As contaminated paddy soil. To our knowledge, this is the first time to report the impact of microbial iron plaque reduction on As mobility. Iron reduction occurred during the inoculation of iron reducing enrichment culture in the treatments with iron plaque and ferrihydrite as the electron acceptors, respectively. The Fe(II) concentration with the treatment of anthraquinone-2, 6-disulfonic acid (AQDS) and iron reducing bacteria increased much faster than the control. Arsenic released from iron plaque with the iron reduction, and a significant correlation between Fe(II) and total As in culture was observed. However, compared with control, the increasing rate of As was inhibited by iron reducing bacteria especially in the presence of AQDS. In addition, the concentrations of As(III) and As(V) in abiotic treatments were higher than those in the biotic treatments at day 30. These results indicated that both microbial and chemical reductions of iron plaque caused As release from iron plaque to aqueous phase, however, microbial iron reduction induced the formation of more crystalline iron minerals, leading to As sequestration. In addition, the presence of AQDS in solution can accelerate the iron reduction, the As release from iron plaque and subsequently the As retention in the crystalline iron mineral. 5Effects of environmental variations on the abundance of western winter-spring cohort of neon flying squid(Ommastrephes bartramii) in the Northwest Pacific Ocean显示文摘During 1995–2011, annual production of winter-spring cohort of Ommastrephes bartramii for Chinese squidjigging fishery has greatly fluctuated, which is closely related to the environmental conditions on the spawning and fishing grounds. To better understand how squid recruitment and abundance were influenced by ocean environmental conditions, biological and physical environmental variables including sea surface temperature(SST), SST anomaly(SSTA), chlorophyll a(Chl a) concentration and the Kuroshio Current were examined during years with the highest(1999), intermediate(2005), and lowest(2009) catches. Catch per unit effort(CPUE) of the squid-jigging vessels was used as an indicator of squid abundance. The results indicated that high SST and Chl a concentration on the spawning ground in 1999 resulted in favorable incubation and feeding conditions for squid recruitment. Whereas the suitable spawning zone(SSZ) in 2009 shifted southward and coincided with low SST and Chl a concentration, resulting in a reduction in the squid recruitment. The small difference of SSZ area in the three years suggested the SSZ provided limited influences on the variability in squid recruitment. Furthermore,high squid abundance in 1999 and 2005 was associated with warm SSTA on the fishing ground. While the cool SSTA on the fishing ground in 2009 contributed to adverse habitat for the squid, leading to extremely low abundance. It was inferred that strengthened intensity of the Kuroshio force generally yielded favorable environmental conditions for O. bartramii. 6Third generation high strength low alloy steels with improved toughness显示文摘On-line thermo mechanical controlled processing(TMCP) was conducted to develop the third generation high strength low alloy(HSLA) steel with high toughness economically.The ultra-low carbon content ensured a high level of upper shelf energy while ultrafine lath martensitic structure transformed from super-thin pancaked austenite during controlled rolling and cooling.The reduction of martensite block size decreased ductile-to-brittle transition temperature(DBTT) and compensated the strength loss due to carbon reduction.Consequently,the excellent balance of strength and toughness values was obtained as 950-1060 MPa for yield strength,180 J for Charpy V-notch impact absorbed energy at 30℃,which is much superior to that of traditional martensitic steel.Two mechanisms for the refinement of lath martensite block were proposed:One is the austenite grain refinement in the direction of thickness,and the other is the reduction in the fraction of sub-block boundaries with small misorientation and the increase in the fraction of block boundaries with large misorientation,possibly due to austenite hardening.SUN XinJun LI ZhaoDong YONG QiLong YANG ZhiGang DONG Han WENG YuQing 2012Science China(Technological Sciences)2012,55,7:7
7Response of winter cohort abundance of Japanese common squid Todarodes pacificus to the ENSO events显示文摘The Japanese common squid Todarodes pacificus is an economically important species with one year lifespan,which is significantly influenced by climatic and environmental variability. According to the fishery data of the winter cohort of T. pacificus from 2003 to 2012, as well as environmental data and the Oceanic Ni?o index(ONI,which was defined by the sea surface temperature(SST) anomaly in the Ni?o 3.4 region), variations in the SST,chlorophyll a(Chl a) concentration, suitable spawning area(SSA) and sea surface height anomaly(SSHA) on the spawning ground of T. pacificus were examined under the El Ni?o and La Ni?a conditions. Their influences on squid abundance(defined by catch per unit effort, CPUE) were further assessed. The results showed that seasonal changes were found in SST, Chl a and SSA on the spawning ground of T. pacificus. Correlation analysis suggested that annual CPUE was significantly positively correlated with Chl a and SSA(p<0.05), but had insignificant relationship with SST(p>0.05). Moreover, the El Ni?o and La Ni?a events tended to dominate the changes of SSA and Chl a concentration in the key area between 25°–29°N and 122.5°–130.5°E, driving the variability of squid abundance. However, this influence varied with the intensity of each anomalous climatic event: the weak El Ni?o event occurred, the spawning ground was occupied by waters with enlarged SSA but with extremely low Chl a concentration, leading to low squid recruitment, the CPUE then decreased; the moderate intensity of El Ni?o event resulted in shrunk SSA but with high Chl a concentration on the spawning ground, the squid recruitment and CPUE increased; the moderate intensity of La Ni?a events yielded elevated SSA and high Chl a concentration on the spawning ground, the squid recruitment and CPUE dramatically increased. 8Modeling Monthly Spatial Distribution of Ommastrephes bartramii CPUE in the Northwest Pacific and Its Spatially Nonstationary Relationships with the Marine Environment显示文摘There are substantial spatial variations in the relationships between catch-per-unit-effort(CPUE) and oceanographic conditions with respect to pelagic species. This study examines the monthly spatiotemporal distribution of CPUE of the neon flying squid, Ommastrephes bartramii, in the Northwest Pacific from July to November during 2004–2013, and analyzes the relationships with oceanographic conditions using a generalized additive model(GAM) and geographically weighted regression(GWR) model. The results show that most of the squids were harvested in waters with sea surface temperature(SST) between 7.6 and 24.6℃, chlorophyll-a(Chl-a) concentration below 1.0 mgm^(-3), sea surface salinity(SSS) between 32.7 and 34.6, and sea surface height(SSH) between-12.8 and 28.4 cm. The monthly spatial distribution patterns of O. bartramii predicted using GAM and GWR models are similar to observed patterns for all months. There are notable variations in the local coefficients of GWR, indicating the presence of spatial non-stationarity in the relationship between O. bartramii CPUE and oceanographic conditions. The statistical results show that there were nearly equal positive and negative coefficients for Chl-a, more positive than negative coefficients for SST, and more negative than positive coefficients for SSS and SSH. The overall accuracies of the hot spots predicted by GWR exceed 60%(except for October), indicating a good performance of this model and its improvement over GAM. 9A β-glucosidase-producing M-2 strain: Isolation from cow dung and fermentation parameter optimization for flaxseed cake显示文摘Flaxseed cake contains cyanogenic glucosides, which can be metabolized into hydrocyanic acid in an animal's body, leading to asphyxia poisoning in cells. Beta-glucosidase is highly efficient in degrading cyanogenic glucosides. The Cattle may have b-glucosidase-producing strains in the intestinal tract after eating small amounts of flaxseed cake for a long time. This study aimed to isolate of a strain from cow dung that produces b-glucosidase with high activity and can significantly reduce the amount of cyanogenic glucosides. We used cow dung as the microflora source and an esculin agar as the selective medium. After screening with 0.05% esculin and 0.01% ferric citrate, we isolated 5 strains producing high amounts of b-glucosidase. In vitro flaxseed cake fermentation was fermented by these 5 strains, in which the strain M-2 exerted the best effect(P < 0.05). The strain M-2 was identified as Lichtheimia ramosa and used as the fermentation strain to optimize the fermentation parameters by a single factor analysis and orthogonal experimental design. The optimum condition was as follows: inoculum size3%, water content 60%, time 144 h, and temperature 32℃. Under this condition, the removal rate of cyanogenic glucosides reached 89%, and crude protein increment reached 44%. 10A comprehensive review of bio-oil, bio-binder and bio-asphalt materials: Their source, composition, preparation and performance显示文摘To promote the application of bio-materials and provide a direction for their further researches,this paper comprehensively summarizes the research progress of biomaterials in recent years.The review results show that bio-oil is a mixture obtained from different biomasses through pyrolysis,alcoholysis,acidolysis,high liquefaction,etc.,and those biomasses mainly include wood fiber type,waste oil type and animal manure type.Biobinder refers to the product of bio-oil processed by distillation,extraction oxidation and polymer modification,and it can be served as a modifier,diluent or substitute material of asphalt;the main chemical components of bio-oil include ethers,esters,aldehydes,ketones,phenols,organic acids,alcohols and sugars.Bio-asphalt is obtained by adding biobinder into the petroleum asphalt for modification or blending,and the shear temperature and shear rate of bio-asphalt derived from wood fiber type and waste oil type are usually higher than that of bio-asphalt derived from swine manure.Compared with petroleum asphalt,bio-asphalt binder usually shows lower high-temperature performance as well as higher low-temperature performance and aging resistance.Also,bio-asphalt mixture generally exhibits lower high-temperature stability,higher low-temperature crack resistance and water stability than petroleum asphalt mixture.Future studies should be conducted combining with the source,composition,preparation,properties and oil production rate of bio-oil.First,how to raise the bio-binder content in bio-asphalt as much as possible while ensuring the sufficient performance becomes the focus of future researches.Second,the micro reaction mechanism between bio-binder and petroleum asphalt should be illustrated in depth.Moreover,developing a complete and unifying technical standard and application specification of bio-asphalt technology is necessary for future researches.Furthermore,determining the optimum bio-binder potentially used as the substitute of petroleum asphalt is also an interesting topic.Zhengqi Zhang Ying Fang Jianhua Yang Xinjun Li 2022Journal of Traffic and Transportation Engineering(English Edition)2022,9,2:5

12Aquaporin 4 expression and ultrastructure of the blood-brain barrier following cerebral contusion injury显示文摘This study aimed to investigate aquaporin 4 expression and the ultrastructure of the blood-brain barrier at 2-72 hours following cerebral contusion injury,and correlate these changes to the formation of brain edema.Results revealed that at 2 hours after cerebral contusion and laceration injury,aquaporin 4 expression significantly increased,brain water content and blood-brain barrier permeability increased,and the number of pinocytotic vesicles in cerebral microvascular endothelial cells increased.In addition,the mitochondrial accumulation was observed.As contusion and laceration injury became aggravated,aquaporin 4 expression continued to increase,brain water content and blood-brain barrier permeability gradually increased,brain capillary endothelial cells and astrocytes swelled,and capillary basement membrane injury gradually increased.The above changes were most apparent at 12 hours after injury,after which they gradually attenuated.Aquaporin 4 expression positively correlated with brain water content and the blood-brain barrier index.Our experimental findings indicate that increasing aquaporin 4 expression and blood-brain barrier permeability after cerebral contusion and laceration injury in humans is involved in the formation of brain edema.Xinjun Li Yangyun Han Hong Xu Zhongshu Sun Zengjun Zhou Xiaodong Long Yumin Yang Linbo Zou 2013Neural Regeneration Research2013,8,4:2
13Cell cycle-related genes p57kip2, Cdk5 and Spin in the pathogenesis of neural tube defects显示文摘In the field of developmental neurobiology, accurate and ordered regulation of the cell cycle and apoptosis are crucial factors contributing to the normal formation of the neural tube. Preliminary studies identified several genes involved in the development of neural tube defects. In this study, we established a model of developmental neural tube defects by administration of retinoic acid to pregnant rats. Gene chip hybridization analysis showed that genes related to the cell cycle and apoptosis, signal transduction, transcription and translation regulation, energy and metabolism, heat shock, and matrix and cytoskeletal proteins were all involved in the formation of developmental neural tube defects. Among these, cell cycle-related genes were predominant. Retinoic acid treat-ment caused differential expression of three cell cycle-related genes p57kip2, Cdk5 and Spin, the expression levels of which were downregulated by retinoic acid and upregulated during normal neural tube formation. The results of this study indicate that cell cycle-related genes play an im-portant role in the formation of neural tube defects. 18Ausforming effects on anisotropy of mechanical properties in HSLA martensitic steel显示文摘In order to clarify effects of prior pancaked austenitic structure on microstructure and mechanical properties of transformed martensite in ausformed steel,a super-thin pancaked austenite was processed by multi-pass rolling in a 0.03-2.6Mn0.06Nb-0.01Ti(wt%) low alloy steel.The evolution of prior pancaked austenite grain during multi-pass rolling was studied using Ni-30Fe model alloy.Related with the structure and texture in the prior super-thin pancaked austenite in Ni-30Fe alloy,the texture and anisotropy of mechanical properties of transformed martensite in the studied ausformed steel were focused on.There were mainly three kinds of rolling texture components in the super-thin pancaked austenite:Goss {110} 001,copper {112} 111 and brass {110} 112.They were further transformed into the weak {001} 110 and strong {112} 110,{111} 112 texture components in the martensitic structure.The orientation relationship(OR) of lath martensite transformation from pancaked austenite in the ausformed steel deviated larger from the exact Kurdjumov-Sachs(K-S) OR than in the case of equiaxed austenite without deformation.The tensile and yield strengths of the ausformed martensitic steel first decreased and then increased as the angle between tension direction and rolling direction increased.The main reason for the anisotropy of strength was considered as the texture component {112} 110 in martensite.However,the anisotropy of impact toughness was more complex and the main reasons for it are unknown.LI ZhaoDong SUN XinJun CAO WenQuan YONG QiLong YANG ZhiGang DONG Han WENG YuQing 2012Science China(Technological Sciences)2012,55,7:1
