| 1 | Efficacy of Banxia Xiexin decoction in a rat model of chronic atrophic gastritis显示文摘OBJECTIVE:To investigate the effectiveness of Banxia Xiexin decoction(BXD)in a rat model of chronic atrophic gastritis(CAG).METHODS:Sixty 6-week-old healthy Wistar rats(30 males,30 females)were used in the present study.A rat model of CAG was successfully established using the combined active immunization/ethanol/sodium deoxycholate method.BXD was prepared from a mixture of seven Chinese herbs,and was intragastrically administered to CAG rats at three different doses(6,12,and 24 g·kg^-1·d^-1).Af-ter 24 weeks,the rats were euthanized,and gastric tissue specimens were collected.Gastric mucosal specimens were stained with hematoxylin and eosin for histological examination to evaluate the degree of inflammation and morphological changes.Immunohistochemical staining was performed to examine the mucosal expression of proliferating cell nuclear antigen.Serum gastrin levels were measured using radioimmunoassay.The expression of Notch signaling-associated genes was examined by quantitative polymerase chain reaction and Western blot assay.RESULTS:BXD at all three doses significantly reversed the adverse effects generated by CAG in rats.Compared with control rats,the CAG rats who were administered BXD had an accelerated growth rate,reduced inflammatory cell infiltration,improved gastric mucosal morphology,augmented thickness of the gastric mucosa,increased number of gastric glands,enhanced mucosal expression of proliferating cell nuclear antigen,and elevated serum gastrin levels. | Bai Yu Chen Yongcan Chen Yu Yan Maoxiang Wei Hongquan Wang Hengcang Zeng Guoliang Yang Xiyin | 2019 | Journal of Traditional Chinese Medicine2019,39,6: | 14 |

| 4 | Coriander Genomics Database:a genomic,transcriptomic,and metabolic database for coriander显示文摘Coriander(Coriandrum sativum L.),also known as cilantro,is a globally important vegetable and spice crop.Its genome and that of carrot are models for studying the evolution of the Apiaceae family.Here,we developed the Coriander Genomics Database(CGDB,http://gffzzc8ce0bd092c048a1hcpqcu6obxwk56qnb.ffgz.tsg.suse.edu.cn/)to collect,store,and integrate the genomic,transcriptomic,metabolic,functional annotation,and repeat sequence data of coriander and carrot to serve as a central online platform for Apiaceae and other related plants.Using these data sets in the CGDB,we intriguingly found that seven transcription factor(TF)families showed significantly greater numbers of members in the coriander genome than in the carrot genome.The highest ratio of the numbers of MADS TFs between coriander and carrot reached 3.15,followed by those for tubby protein(TUB)and heat shock factors.As a demonstration of CGDB applications,we identified 17 TUB family genes and conducted systematic comparative and evolutionary analyses.RNA-seq data deposited in the CGDB also suggest dose compensation effects of gene expression in coriander.CGDB allows bulk downloading,significance searches,genome browser analyses,and BLAST searches for comparisons between coriander and other plants regarding genomics,gene families,gene collinearity,gene expression,and the metabolome.A detailed user manual and contact information are also available to provide support to the scientific research community and address scientific questions.CGDB will be continuously updated,and new data will be integrated for comparative and functional genomic analysis in Apiaceae and other related plants. | Xiaoming Song Fulei Nie Wei Chen Xiao Ma Ke Gong Qihang Yang Jinpeng Wang Nan Li Pengchuan Sun Qiaoying Pei Tong Yu Jingjing Hu Xinyu Li Tong Wu Shuyan Feng Xiu-Qing Li Xiyin Wang | 2020 | Horticulture Research2020,7,1: | 4 |

| 6 | Epithelial-Mesenchymal Transitions and the Expression of Twist in MCF-7/ADR, Human Multidrug-Resistant Breast Cancer Cells显示文摘0BJECTIVE To study the expression levels of Twist and epithelialmesenchymal transitions in multidrug-resistant MCF-7/ADR breast cancer cells, and to study the relationship between multidrug resistance (MDR) and metastatic potential of the cells. METHODS RT-PCR, immunohistochemical and Western blotting methods were used to examine the changes of expression levels of the transcription factor Twist, E-cadherin and N-cadherin in the MCF-7 breast cancer cell line and its multidrug-resistant variant, MCF-7/ADR. RESULTS In MCF-7 cells, the expression of E-cadherin can be detected, but there is no expression of Twist or N-cadherin. In MCF-7/ADR cells, E-cadherin expression is lost, but the expression of two other genes was significantly positive. | 10 | In Vivo Selection of Phage Sequences and Characterization of Peptide-specific Binding to Breast Cancer Cells显示文摘OBJECTIVE To screen specific polypeptide target binding to breast cancer xenogra s in vivo from a phage-displayed peptide library in order to provide peptide sequences for breast cancer tumor-targeting diagnosis and therapy. METHODS A mouse model for carrying breast cancer xenografts was established using Tientsin Albinao II mice(TA II).A 12-peptide library was biopanned through 4 rounds. Phages were recovered and titrated from tumor xenografts and control tissue(liver).The distribution of phages was detected by immunohistochemical staining. RESULTS Phage homing to breast cancer was enriched through 4 rounds of biopanning,being 14-fold of that recovered from liver tissue.A peptide sequence,ASANPFPTKALL was characterized by randomly picked-up clones which appeared most frequently. Immunohistochemical staining revealed phage localization in cancer xenografts 40 min after injection of the enriched phages. When a specific phage was tested individually,the phage reclaimed from breast cancer xenografts was 14 times as those from control tissues. | 11 | Down-Regulation of CXCR4 Expression by siRNA Inhibits Invasive Ability of Breast Cancer Cells显示文摘OBJECTIVE To investigate the efficiency of gene silencing by CXCR4- siRNAs (small interfering RNA), and to examine the invasive ability and the expression of other metastatic-associated genes in siRNA-treated breast cancer cells. METHODS Three siRNAs were designed and cloned into the pSilenc ? 3.1-H1 neo vector. The reconstructed plasmids were purified and trans- fected into the T47D breast cancer cell line, which highly expressed CXCR4. The amount of CXCR4 expression in the transfected cells was measured by flow cytometry and Real-time PCR. Cell invasive ability was evaluated us- ing 24-well Matrigel invasion chambers. In addition, the expression of other metastatic-associated genes, such as E-cad, IGFBP-5, FN and MMP-2, was assessed by Real-time PCR. RESULTS The suppression rates of CXCR4 mRNA expression reached 95.7%, 85.9% and 98.3%compared with control-siRNA cells in the 3 CXCR4- siRNA T47D cells respectively. FCM assays for CXCR4 protein expression showed a similar inhibitory effect. The invasion indexes of these CXCR4- siRNA cells were 0.037, 0.290 and 0.188 respectively compared with control- siRNA cells. After treatment of the cells with CXCR4-siRNA, the expression of E-cad showed an upward tendency and that of IGFBP-5 had a downward trend, while alteration in expression of FN and MMP2 varied without a con- sistant effect. CONCLUSION CXCR4 plays an important role in modulating migra- tion of human breast cancer cells. Small interfering RNA can significantly silence the CXCR4 gene in the human T47D breast cancer cell line. | 12 | The Effect of Twist Expression on Angiogenesis in Hepatocellular Carcinoma显示文摘OBJECTIVE Hepatocellular carcinoma (HCC) is a hypervascular tumor for which angiogenesis plays an important role in its progression. The aim of this study was to investigate the expression of TWIST and VEGF and determine their roles in angiogenesis of HCC. METHODS Expression Twist and VEGF mRNA was determined by real-time RT-PCR in 30 pairs of hepatocellular carcinoma and matched non-cancerous tissues. Immunohistochemistry was carried out to analyze the protein expression of Twist and VEGF in 40 hepatocellular carcinoma cases. Staining of endothelial cells for CD34 was used to evaluate the microvessel density (MVD). RESULTS We found that the HCC specimens showing positive Twist expression in tumor cells had a higher microvessel density than those without Twist expression. Furthermore, we found that overexpression of the Twist protein positively correlated with up-regulation of VEGF in the HCC tissues (r=0.479, P=0.002). | 13 | Proteasome Inhibitors Sensitize Hepatocellular Carcinoma Cells to TRAIL显示文摘OBJECTIVE To investigate the effect of proteasome inhibition on the sensitivity of carcinoma cells to TRAIL-inducing apoptosis, and to study the mechanism of the response. METHODS Human hepatocellular carcinoma cells, pretreated with the proteasome inhibitor, MG132, were cotreated with TRAIL. Western blot assays, immunoprecipitation and RT-PCR were performed to test the expression of the Bcl-2 family proteins and Bax mRNA. RESULTS We found that (i) proteasome inhibition sensitized the human hepatocellular carcinoma cells to TRAIL; and (ii) resulted in Bax accumulation before release of cytochrome C and induction of apoptosis. These results were associated with the ability of proteasome inhibitors to overcome Bcl-2-mediated antiapoptotic function; (iii) Bax is regulated by an ubiquitin/proteasome-dependent degradation pathway. | 14 | Study on the atmospheric heat engine efficiency and heat source characteristics of the Qinghai-Tibet Plateau in summer显示文摘There are many types of atmospheric heat engines in land-air systems.The accurate definition,calculation and interpretation of the efficiency of atmospheric heat engines are key to understanding energy transfer and transformation of landair systems.The atmosphere over the Qinghai-Tibet Plateau(QTP)in summer can be regarded as a positive heat engine.The study of the heat engine efficiency is helpful to better understand land-air interaction and thermal-dynamic processes on the QTP.It also provides a new perspective to explain the impact of the QTP on the climate of China,East Asia and even the world.In this paper,we used MOD08 and ERA5 reanalysis data to calculate the atmospheric heat engine efficiency,surface heat source and atmospheric heat source on the QTP in summer(May to September)from 2000 to 2020.The average atmospheric heat engine efficiency on the QTP in summer from 2000 to 2020 varies between 1.2%and 1.5%,which is less than 1.6%;the heat engine efficiency in summer is higher than that in June,July and August;the Qaidam Basin is the region with the highest atmospheric heat engine efficiency,followed by the western QTP.The mean surface heat source on the QTP in summer from 2000 to 2020 is 96.0 W m^(−2),the atmospheric heat source is 90.7 W m^(−2),and the release of precipitation condensation latent heat is the most important component of the atmospheric heat source on the QTP in summer.There is a strong and significant positive correlation between the atmospheric heat engine efficiency and the surface heat source on the QTP in summer.The precipitation condensation latent heat is the most important component of the atmospheric heat source in summer and can reflect the precipitation process.There is a strong and significant negative correlation between the atmospheric heat engine efficiency and the atmospheric heat source on the QTP in summer. | Yujie LI Xiaoqing GAO Yaoming MA Zeyong HU Zhenchao LI Liwei YANG Xiao JIN Xiyin ZHOU | 2024 | Science China Earth Sciences2024,67,1: | 0 |
