1High quality reference genome of drumstick tree(Moringa oleifera Lam.), a potential perennial crop显示文摘The drumstick tree(Moringa oleifera Lam.) is a perennial crop that has gained popularity in certain developing countries for its high-nutrition content and adaptability to arid and semi-arid environments. Here we report a high-quality draft genome sequence of M. oleifera. This assembly represents 91.78% of the estimated genome size and contains 19,465 protein-coding genes. Comparative genomic analysis between M. oleifera and related woody plant genomes helps clarify the general evolution of this species, while the identification of several species-specific gene families and positively selected genes in M. oleifera may help identify genes related to M. oleifera's high protein content, fast-growth, heat and stress tolerance. 2TAZ inhibits osteoclastogenesis by attenuating TAK1/NF-κB signaling显示文摘Osteoporosis is an osteolytic disorder commonly associated with excessive osteoclast formation.Transcriptional coactivator with PDZ-binding motif(TAZ)is a key downstream effector of the Hippo signaling pathway;it was suggested to be involved in the regulation of bone homeostasis.However,the exact role of TAZ in osteoclasts has not yet been established.In this study,we demonstrated that global knockout and osteoclast-specific knockout of TAZ led to a low-bone mass phenotype due to elevated osteoclast formation,which was further evidenced by in vitro osteoclast formation assays.Moreover,the overexpression of TAZ inhibited RANKL-induced osteoclast formation,whereas silencing of TAZ reduced it.Mechanistically,TAZ bound to TGF-activated kinase 1(TAK1)and reciprocally inhibited NF-κB signaling,suppressing osteoclast differentiation.Collectively,our findings highlight an essential role of TAZ in the regulation of osteoclastogenesis in osteoporosis and its underlying mechanism.Wanlei Yang Xuanyuan Lu Tan Zhang Weiqi Han Jianlei Li Wei He Yewei Jia Kangxian Zhao An Qin Yu Qian 2021Bone Research2021,9,3:6
3Total body irradiation of donors can alter the course of tolerance and induce acute rejection in a spontaneous tolerance rat liver transplantation model显示文摘Liver transplantation is an established therapy for end-stage liver diseases. Graft rejection occurs unless the recipient receives immunosuppression after transplantation. This study aimed to explore the mechanism of acute rejection of liver allografts in rats pre-treated with total body irradiation to eliminate passenger lymphocytes and to define the role of CD4 + CD25 + regulatory T cells in the induction of immunotolerance in the recipient. Male Lewis rats were used as donors and male DA rats were recipients. Rats were randomly assigned to the following four groups: control group, homogeneity liver transplantation group, idio-immunotolerance group and acute rejection group. After transplantation, the survival time of each group, serum alanine aminotransferase, total bilirubin levels, number of Foxp3 + CD4 + CD25 + regulatory T cells, expression of glucocorticoid-induced tumor necrosis factor receptor on T cell subgroups, histopathology of the hepatic graft and spleen cytotoxic T lymphocyte lytic activity were measured. In the acute rejection group, where donors were preconditioned with total body irradiation before liver transplantation, all recipients died between day 17 and day 21. On day 14, serum alanine aminotransferase increased signifi- cantly to (459.2±76.9) U L -1 , total bilirubin increased to (124.1±33.7) μmol L -1 (P<0.05) and the ratio of Foxp3 + CD4 + CD25 + regulatory T cells decreased significantly to 1.50%±0.50% (P<0.05) compared with the other groups. Analysis of the T cell subpopulations in the acute rejection group varied from the other groups. Histological analysis showed typical changes of acute rejection in the acute rejection group only. 4Cloning of aminopeptidase N promoter and its activity in hematopoietic cell and different tumor cell lines显示文摘Aminopeptidase N (APN) promoter region was cloned and sequenced from peripheral blood mononuclear cells. The recombinant reporter construct containing the promoter and luciferase gene, designated pXPl-APNLuc, was introduced into myeloblastic cell line, T lymphocyte cell line and various tumor cell lines. Luciferase assay showed that APN upstream promoter is myeloid-specific for high expression in myeloblastic cell line and much lower expression in T lymphocyte cell line. The promoter activity was relatively high in lung adenoma cell line compared with other tumor cell lines including hepatoma cell line, tong cancer cell line and esophageal cancer cell line in which the promoter activity significantly diminished or was almost undetectable. 5Optimum design and experiment of composite leaf spring landing gear for electric aircraft显示文摘The composite leaf spring landing gear of an electric aircraft is optimized.With the strength and workability as constraints and the minimum structural weight as an objective,the two-stage optimization of the leaf spring landing gear with glass fiber unidirectional prepreg is carried out using a genetic algorithm,namely,the optimization of continuous thickness of layup,and the optimization of the layup sequence and discrete thickness.In the optimization process,the ground loads are calculated according to the structural stiffness of each chromosome,thus the stiffness constraints are relaxed,and the optimization results are compared with those using stiffness constraints.The static experiment verification reveals that the numerical simulation and experimental results are consistent,that is,the optimized leaf spring meets the strength requirements.The results show that the leaf spring landing gear based on two-stage optimization method achieves the objective of weight reduction.Donghui LI Zhenwei FAN Yewei ZHANG Jian ZANG Fengtian YANG 2020Chinese Journal of Aeronautics2020,33,10:2
6Specific myeloprotection via multidrug resistance 1 gene controlled by aminopeptidase N myeloid promoter显示文摘In the treatment of tumor patients introduction of multidrug resistance genes into hematopoietic cells has been reported as an approach for reducing myelotoxicity created by antitumor drugs. However, the nonspecific expression of the genes can also increase the chemoresistance of the tumor cells invaded into bone marrow, which influences seriously the effectiveness of chemotherapy. In this study, a new strategy is described for specific myeloprotection. The recombinant retroviral vector containing multidrug resistance 1 (MDR1) gene regulated by aminopeptidase N (APN) myeloid promoter was constructed and then introduced into myeloblastic cells KGla and tumor cell line BEL7402. The specific transcript of MDR1 was detected in KGla cells transduced with MDR1 gene and rhodamine 123 was effectively extruded by Pgp, the protein of MDR1 gene. 7Design and transition characteristics of a standard model for hypersonic boundary layer transition research显示文摘To understand fundamental problems in hypersonic laminar-turbulent boundary layer transition for three-dimensional complex vehicles,a new standard model with typical lifting-body features has been proposed,named as hypersonic transition research vehicle(HyTRV).The configuration of HyTRV is fully analytical,and details of the design process are discussed in this study.The transition characteristics for HyTRV are investigated using three combined methods,i.e.,theoretical analyses,numerical simulations,and wind tunnel experiments.Results show that the fully analytic parameterization design of HyTRV can satisfy the model simplification requirements from both numerical simulations and wind tunnel experiments.Meanwhile,the flow field of HyTRV reveals typical transition mechanisms in six relatively separated regions,including the streamwise vortex instability,crossflow instability,secondary instability,and attachment-line instability.Therefore,the proposed HyTRV model is valuable for fundamental researches in hypersonic boundary layer transition.Shenshen Liu Xianxu Yuan Zhiyong Liu Qiang Yang Guohua Tu Xi Chen Yewei Gui Jianqiang Chen 2021Acta Mechanica Sinica2021,37,11:0
8Measurement of Factors behind the Decline of the Agricultural Labor Share in Total Labor Force of China(1990-2030)显示文摘On the basis of a simple model identifying the relationship between the agricultural labor share in total labor force and the determining factors,such as number of new participators to,and those leaving from agriculture and transferring to non-farming,this paper estimates the contribution of the determining factors for the changes of agricultural labor share during the period 1990~2030.Given the assumption about the average annual decline of the agricultural labor share in future,the number of new participates to agricultural labor is estimated to decline from 20.23 million during 2005~2010 to 11.42 million during 2010-2030,and the numbers of leaving and tranferring are estimated to change from 42.20 million and 38.43 million to 27.04 million and 29.49 million respectively during the same period. The factors of entrance,exit and transfer,which contribute to the decline of the agricultural labor share,are described to change from- 15.1%,34.7%and 38.1%respectively in 2005~2010 to-22.6%,54.3% and 49.8%in 2025~2030.The nonfarming job creation may decline from 69.69 million in 2005~2010 to 35.08 million in 2025~2030.The policy implications of the finding are also discussed.Lu Feng Yang Yewei 2012China Population Today2012,29,6:0
9Tumor microenvironment-activated theranostic nanoreactor for NIR-II Photoacoustic imaging-guided tumor-specific photothermal therapy显示文摘Theranostic agents that can be sensitively and specifically activated by the tumor microenvironment(TME)have recently attracted considerable attention.In this study,TME-activatable 3,3′,5,5′-tetramethylbenzidine(TMB)-copper peroxide(CuO_(2))@poly(lactic-co-glycolic acid)(PLGA)@red blood cell membrane(RBCM)(TCPR)nanoparticles(NPs)for second near-infrared photoacoustic imaging-guided tumor-specific photothermal therapy were developed by co-loading CuO_(2)NPs and TMB into PLGA camouflaged by RBCMs.As an efficient H_(2)O_(2)supplier,once exposed to a proton-rich TME,CuO_(2)NPs can generate H_(2)O_(2)and Cu^(2+),which are further reduced to Cu^(+) by endogenous glutathione.Subsequently,the Cu^(+)-mediated Fenton-like reaction produces cytotoxic·OH to kill the cancer cells and induce TMB-mediated photoacoustic and photothermal effects.Combined with the RBCM modification-prolonged blood circulation,TCPR NPs display excellent specificity and efficiency in suppressing tumor growth,paving the way for more accurate,safe,and efficient cancer theranostics.Nan Yang Hui Li Changyu Cao Lei Zhao Xuejiao Song Wenjun Wang Wenjing Xu Yewei Zhang Peng Chen Xiaochen Dong 2024Fundamental Research2024,4,1:0
