1Inhibition of SIRT6 in prostate cancer reduces cell viability and increases sensitivity to chemotherapeutics显示文摘SI RT6 is an important histone modifying protein that regulates DNA repair,telomere maintenance,energy me-tabolism,and target gene expression.Recently SIRT6 has been identifi ed as a tumor suppressor and is down-regulated in certain cancer types,but not in other can-cers.From deposited gene profi ling studies we found that SIRT6 was overexpressed in prostate tumors,compared with normal or paratumor prostate tissues.Tissue micro-array studies confi rmed the higher levels of SIRT6 in both prostate tumor tissues and prostate cancer cells than in their normal counterparts.Knockdown of SIRT6 in human prostate cancer cells led to sub-G1 phase arrest of cell cy-cle,increased apoptosis,elevated DNA damage level and decrease in BCL2 gene expression.Moreover,SIRT6-de-fi ciency reduced cell viability and enhanced chemothera-peutics sensitivity.Taken together,this study provides the fi rst evidence of SIRT6 overexpression in human prostate cancer,and SIRT6 regulation could be exploited for pros-tate cancer therapy.Yewei Liu Qian Reuben Xie Boshi Wang Jiaxiang Shao Tingting Zhang Tengyuan Liu Gang Huang Weiliang Xia 2013Protein & Cell2013,4,9:17
2Energy harvesting via nonlinear energy sink for whole-spacecraft显示文摘This paper presents nonlinear energy sink with giant magnetostrictive-piezoelectric material for energy harvesting of the wholespacecraft vibration reduction system. The whole-spacecraft vibration attenuation system can effectively reduce vibration and achieve self-tuning enhanced energy harvesting range. The open-circuit voltage generated at low frequency is affected by the magnetic field force, alternating magnetic field and relative displacement. In order to acquire a steady periodic solution of the energy harvesting system, a combination of the harmonic balance method and pseudo arc length continuation technique is used.The numerical outcomes are consistent with the analytical outcomes in a certain range, which also proves the accuracy and reliability of the results. The amplitude and voltage of the energy harvesting system are analyzed by parameters such as cubic stiffness, viscous damping, and external excitation acceleration. 3High quality reference genome of drumstick tree(Moringa oleifera Lam.), a potential perennial crop显示文摘The drumstick tree(Moringa oleifera Lam.) is a perennial crop that has gained popularity in certain developing countries for its high-nutrition content and adaptability to arid and semi-arid environments. Here we report a high-quality draft genome sequence of M. oleifera. This assembly represents 91.78% of the estimated genome size and contains 19,465 protein-coding genes. Comparative genomic analysis between M. oleifera and related woody plant genomes helps clarify the general evolution of this species, while the identification of several species-specific gene families and positively selected genes in M. oleifera may help identify genes related to M. oleifera's high protein content, fast-growth, heat and stress tolerance. 4Vibration reduction evaluation of a linear system with a nonlinear energy sink under a harmonic and random excitation显示文摘The nonlinear behaviors and vibration reduction of a linear system with a nonlinear energy sink(NES)are investigated.The linear system is excited by a harmonic and random base excitation,consisting of a mass block,a linear spring,and a linear viscous damper.The NES is composed of a mass block,a linear viscous damper,and a spring with ideal cubic nonlinear stiffness.Based on the generalized harmonic function method,the steady-state Fokker-Planck-Kolmogorov equation is presented to reveal the response of the system.The path integral method based on the Gauss-Legendre polynomial is used to achieve the numerical solutions.The performance of vibration reduction is evaluated by the displacement and velocity transition probability densities,the transmissibility transition probability density,and the percentage of the energy absorption transition probability density of the linear oscillator.The sensitivity of the parameters is analyzed for varying the nonlinear stiffness coefficient and the damper ratio.The investigation illustrates that a linear system with NES can also realize great vibration reduction under harmonic and random base excitations and random bifurcation may appear under different parameters,which will affect the stability of the system.Jiren XUE Yewei ZHANG Hu DING Liqun CHEN 2020Applied Mathematics and Mechanics(English Edition)2020,41,1:8
5Rat model of cholelithiasis with human gallstones implanted in cholestasis-induced virtual gallbladder显示文摘AIM:To facilitate translational research on cholelithiasis,we have developed a rat model of human gallstones by exploiting the unique biliopancreatic features of this species.METHODS:Under anesthesia,16 adult rats of equal genders underwent two times of abdominal surgery.First,their common bile duct(CBD)was ligated to cause cholestasis by total biliary obstruction(TBO).On day 0,1,3,7,14,21 and 28 after TBO,magnetic resonance imaging(MRI)was conducted to monitor the dilatation of the CBD,and blood was sampled to analyze total serum bilirubin(TSB).Secondly,on day 30,the abdomen was re-opened and gallstone(s)collected from human patients were implanted in the dilated CBD asa virtual gallbladder(VGB),which was closed by suture ligation.This rat cholelithiasis model was examined by MRI,clinical observation,microcholangiography and histology.RESULTS:All rats survived two laparotomies.After ligation,the CBD was dilated to a stable size of 4 to30 mm in diameter on day 21-28,which became a VGB.The rats initially showed signs of jaundice that diminished over time,which paralleled with the evolving TSB levels from 0.6±0.3 mg/d L before ligation,through a peak of 10.9±1.9 mg/dL on day 14,until a nearly normalized value after day 28.The dilated CBD with thickened wall allowed an incision for implantation of human gallstones of 1-10 mm in diameter.The rat cholelithiasis was proven by in vivo MRI and postmortem microcholangiography and histomorphology.CONCLUSION:A rat model cholelithiasis with human gallstones has been established,which proves feasible,safe,reliable,nontoxic and cost-effective.Given the gallstones of human origin,applications of this model may be of help in translational research such as optical detection and lysis of gallstones by systemic drug administration.Marlein Miranda Cona Yewei Liu Ting Yin Yuanbo Feng Feng Chen Stefaan Mulier Yue Li Jian Zhang Raymond Oyen Yicheng Ni 2016World Journal of Methodology2016,6,2:7
6TAZ inhibits osteoclastogenesis by attenuating TAK1/NF-κB signaling显示文摘Osteoporosis is an osteolytic disorder commonly associated with excessive osteoclast formation.Transcriptional coactivator with PDZ-binding motif(TAZ)is a key downstream effector of the Hippo signaling pathway;it was suggested to be involved in the regulation of bone homeostasis.However,the exact role of TAZ in osteoclasts has not yet been established.In this study,we demonstrated that global knockout and osteoclast-specific knockout of TAZ led to a low-bone mass phenotype due to elevated osteoclast formation,which was further evidenced by in vitro osteoclast formation assays.Moreover,the overexpression of TAZ inhibited RANKL-induced osteoclast formation,whereas silencing of TAZ reduced it.Mechanistically,TAZ bound to TGF-activated kinase 1(TAK1)and reciprocally inhibited NF-κB signaling,suppressing osteoclast differentiation.Collectively,our findings highlight an essential role of TAZ in the regulation of osteoclastogenesis in osteoporosis and its underlying mechanism.Wanlei Yang Xuanyuan Lu Tan Zhang Weiqi Han Jianlei Li Wei He Yewei Jia Kangxian Zhao An Qin Yu Qian 2021Bone Research2021,9,3:6
7Integration of a nonlinear energy sink and a piezoelectric energy harvester显示文摘A mechanical-piezoelectric system is explored to reduce vibration and to harvest energy. The system consists of a piezoelectric device and a nonlinear energy sink(NES), which is a nonlinear oscillator without linear stiffness. The NES-piezoelectric system is attached to a 2-degree-of-freedom primary system subjected to a shock load. This mechanical-piezoelectric system is investigated based on the concepts of the percentages of energy transition and energy transition measure. The strong target energy transfer occurs for some certain transient excitation amplitude and NES nonlinear stiffness. 9Dynamic design of a nonlinear energy sink with NiTiNOL-steel wire ropes based on nonlinear output frequency response functions显示文摘A novel vibration isolation device called the nonlinear energy sink(NES)with NiTiNOL-steel wire ropes(NiTi-ST)is applied to a whole-spacecraft system.The NiTi-ST is used to describe the damping of the NES,which is coupled with the modified Bouc-Wen model of hysteresis.The NES with NiTi-ST vibration reduction principle uses the irreversibility of targeted energy transfer(TET)to concentrate the energy locally on the nonlinear oscillator,and then dissipates it through damping in the NES with NiTi-ST.The generalized vibration transmissibility,obtained by the root mean square treatment of the harmonic response of the nonlinear output frequency response functions(NOFRFs),is first used as the evaluation index to analyze the whole-spacecraft system in the future.An optimization analysis of the impact of system responses is performed using different parameters of NES with NiTi-ST based on the transmissibility of NOFRFs.Finally,the effects of vibration suppression by varying the parameters of NiTi-ST are analyzed from the perspective of energy absorption.The results indicate that NES with NiTi-ST can reduce excessive vibration of the whole-spacecraft system,without changing its natural frequency.Moreover,the NES with NiTi-ST can be directly used in practical engineering applications.Yewei ZHANG Kefan XU Jian ZANG Zhiyu NI Yunpeng ZHU Liqun CHEN 2019Applied Mathematics and Mechanics(English Edition)2019,40,12:4
10Association of the HTR2C-759CT polymorphism and antipsychotic-induced weight gain:a meta-analysis显示文摘Background Antipsychotic-induced weight gain(AIWG)is a crucial factor for the medication cessation of patients with schizophrenia.Multiple studies have shown that the functional polymorphism-759 C/T(rs3813929)in the HTR2C promoter region could possibly be correlated with AIWG.Aim To evaluate the genetic association of the HTR2C-759C/T polymorphism and AIWG in patients with schizophrenia with antipsychotic drugs(APDs)administration.Methods Eligible studies were identified by searching the following databases:PubMed,Embase,Web of Science,China Nation Knowledge Infrastructure(CNKI),VIP,Wanfang Data,Chinese Biomedical Literature Database(CBM)and the Airiti Library.The quality of studies was evaluated based on the Newcastle-Ottawa Scale.The pooled OR and 95%Cl were calculated for the dominant(CT/TT/T vs CC/C)mode,and subgroup analyses were performed based on ethnicity,antipsychotic medication and gender;ail statistical analyses were performed using the statistical software STATA V.12.0.Result A total of 17 studies with 3170 patients with schizophrenia were included in our meta-analysis.The result of the meta-analysis has shown that the association between the-759 C/T polymorphism and AIWG is statistically significant(OR 0.34,95%Cl:0.20 to 0.57,z=4.11,p<0.001).The subgroup analyses revealed significant correlations between the-759 C/T polymorphism and AIWG in the Caucasian population(OR 0.33,95%CI:0.14 to 0.77,z=2.55,p=0.011),the Asian population(OR 0.31,95%Cl:0.18 to 0.52,z=4.46,p<0.001),the patients with APDs administration(CT/JTfJ vs CC/C:OR 0.63,95%Cl:0.40 to 1.00,z=1.97,p=0.049)and the patients with atypical antipsychotic drug administration(CT/TT/T vs CC/C:OR 0.21,95%Cl:0.09 to 0.47,z=3.83,p<0.001).The sensitivity analysis showed that the results were stable.Begg's test(after correction z=1.07,p=0.287)and Egger's test(t=-2.41,p=0.029)show that the included articles have no significant publication bias.Conclusion There is a significant genetic association between HTR2C-759C/T and AIWG,and patients with T allele are less likely to have AIWG.Yan Chen Yewei Wang Xinyu Fang Yi Zhang Lisheng Song Chen Zhang 2020General Psychiatry2020,33,3:4
11Total body irradiation of donors can alter the course of tolerance and induce acute rejection in a spontaneous tolerance rat liver transplantation model显示文摘Liver transplantation is an established therapy for end-stage liver diseases. Graft rejection occurs unless the recipient receives immunosuppression after transplantation. This study aimed to explore the mechanism of acute rejection of liver allografts in rats pre-treated with total body irradiation to eliminate passenger lymphocytes and to define the role of CD4 + CD25 + regulatory T cells in the induction of immunotolerance in the recipient. Male Lewis rats were used as donors and male DA rats were recipients. Rats were randomly assigned to the following four groups: control group, homogeneity liver transplantation group, idio-immunotolerance group and acute rejection group. After transplantation, the survival time of each group, serum alanine aminotransferase, total bilirubin levels, number of Foxp3 + CD4 + CD25 + regulatory T cells, expression of glucocorticoid-induced tumor necrosis factor receptor on T cell subgroups, histopathology of the hepatic graft and spleen cytotoxic T lymphocyte lytic activity were measured. In the acute rejection group, where donors were preconditioned with total body irradiation before liver transplantation, all recipients died between day 17 and day 21. On day 14, serum alanine aminotransferase increased signifi- cantly to (459.2±76.9) U L -1 , total bilirubin increased to (124.1±33.7) μmol L -1 (P<0.05) and the ratio of Foxp3 + CD4 + CD25 + regulatory T cells decreased significantly to 1.50%±0.50% (P<0.05) compared with the other groups. Analysis of the T cell subpopulations in the acute rejection group varied from the other groups. Histological analysis showed typical changes of acute rejection in the acute rejection group only. 13Applications of multi-dimensional schemes on unstructured grids for high-accuracy heat flux prediction显示文摘The simulation of hypersonic flows with fully unstructured(tetrahedral)grids has severe problems with respect to the prediction of stagnation region heating,due to the random face orientation without alignment to the bow shock.To improve the accuracy of aero-heating predictions,three multi-dimensional approaches on unstructured grids are coupled in our Reynolds-averaged Navier-Stokes(RANS)solver,including multi-dimensional upwind flux reconstruction(MUP),multi-dimensional limiter(MLP-u2)and multi-dimensional gradient reconstruction(MLR).The coupled multi-dimensional RANS solver is validated by several typical verification and validation(V&V)cases,including hypersonic flows over a cylinder,a blunt biconic,and a double-ellipsoid,with commonly used prism/tetrahedral hybrid grids.Finally,the coupled multi-dimensional solver is applied to simulating the heat flux distribution over a 3D engineering configuration,i.e.a Hermes-like space shuttle model.The obtained numerical results are compared with experimental data.The predicted results demonstrate that the coupled multi-dimensional approach has a good prediction capability on aerodynamic heating over a wide range of complex engineering configurations.Yunbo Wan Nianhua Wang Laiping Zhang Yewei Gui 2020Acta Mechanica Sinica2020,36,1:2
14Effects of Epstein-Barr virus infection on the development of multiple myeloma after liver transplantation显示文摘Reduced cellular immune function in patients after liver transplantation easily results in many types of viral infections,such as Epstein-Barr virus.Epstein-Barr virus is a γ-herpesvirus and is related to many malignant diseases,especially epithelial and lymph tumors.The abnormal interaction of cluster of differentiation 40 with cluster of differentiation 40 ligand and expression of cluster of differentiation 40 ligand are considered closely related to the development of myeloma cells.This study explored the influence and mechanism of Epstein-Barr virus infection on the phenotype and biological behavior of myeloma cells after liver transplantation.Flow cytometry was used to detect coexpression of cluster of differentiation 40 and cluster of differentiation 40 ligand in 10 samples of freshly isolated multiple myeloma cells.Cluster of differentiation 40 and cluster of differentiation 40 ligand were coexpressed in sample Nos.5,8,9,and 10,particularly in sample No.5.Western blot analysis was used to detect the expression of the Epstein-Barr virus antigens latent membrane protein 1 and Epstein-Barr virus nuclear antigen 2 in the multiple myeloma cell line RPMI 8226 infected with Epstein-Barr virus.The antigen expression indicated that Epstein-Barr virus can infect multiple myeloma virus cells in vitro.Reverse transcription-polymerase chain reaction revealed upregulated expression of cluster of differentiation 40 ligand on the infected RPMI 8226 cells,which may be involved in the anti-apoptosis activity of the infected cells.Confocal microscopy showed that pairs of molecules of cluster of differentiation 40,cluster of differentiation 40 ligand,and latent membrane protein 1 were colocalized on the surface of the infected cells.CXC chemokine receptor 4 was upregulated on the RPMI 8226 cells after Epstein-Barr virus infection.The migratory ability of the infected cells improved in the presence of the chemokine stromal cell-derived factor-1α.Anti-apoptosis and migration are known important biological characteristics of malignant cells.Our results indicate the involvement of Epstein-Barr virus in the origin and development of multiple myeloma.The risk of multiple myeloma increases when Epstein-Barr virus infects B cells in the germinal center,which may result in an anti-apoptosis effect of B cells and an improved ability to migrate from the germinal center to peripheral blood.ZHANG YeWei ZHAO HeWei HE Xia ZHENG SuWen WANG TaiHong YAN DongLiang SUN JingFeng LU Xiang WEN JianFei LAU Wan Yee 2012Science China(Life Sciences)2012,55,8:2
15Optimum design and experiment of composite leaf spring landing gear for electric aircraft显示文摘The composite leaf spring landing gear of an electric aircraft is optimized.With the strength and workability as constraints and the minimum structural weight as an objective,the two-stage optimization of the leaf spring landing gear with glass fiber unidirectional prepreg is carried out using a genetic algorithm,namely,the optimization of continuous thickness of layup,and the optimization of the layup sequence and discrete thickness.In the optimization process,the ground loads are calculated according to the structural stiffness of each chromosome,thus the stiffness constraints are relaxed,and the optimization results are compared with those using stiffness constraints.The static experiment verification reveals that the numerical simulation and experimental results are consistent,that is,the optimized leaf spring meets the strength requirements.The results show that the leaf spring landing gear based on two-stage optimization method achieves the objective of weight reduction.Donghui LI Zhenwei FAN Yewei ZHANG Jian ZANG Fengtian YANG 2020Chinese Journal of Aeronautics2020,33,10:2
16Dynamics Analysis of Active Variable Stiffness Vibration Isolator for Whole-Spacecraft Systems Based on Nonlinear Output Frequency Response Functions显示文摘In order to improve the harsh dynamic environment experienced by heavy rockets during different external excitations,this study presents a novel active variable stiffness vibration isolator(AVS-VI)used as the vibration isolation device to reduce excessive vibration of the whole-spacecraft isolation system.The AVS-VI is composed of horizontal stiffness spring,positive stiffness spring,parallelogram linkage mechanism,piezoelectric actuator,acceleration sensor,viscoelastic damping,and PID active controller.Based on the AVS-VI,the generalized vibration transmissibility determined by the nonlinear output frequency response functions and the energy absorption rate is applied to analyze the isolation performance of the whole-spacecraft system with AVS-VI.The AVS-VI can conduct adaptive vibration suppression with variable stiffness to the whole-spacecraft system,and the analysis results indicate that the AVS-VI is efTective in reducing the extravagant vibration of the whole-spacecraft system,where the vibration isolation is decreased up to above 65%under different acceleration excitations.Finally,different parameters of AVS-VI are considered to optimize the whole-spacecraft system based on the generalized vibration transmissibility and the energy absorption rate.Kefan Xu Yewei Zhang Yunpeng Zhu Jian Zang Liqun Chen 2020Acta Mechanica Solida Sinica2020,33,6:1

18Research on linear/nonlinear viscous damping and hysteretic damping in nonlinear vibration isolation systems显示文摘A nonlinear vibration isolation system is promising to provide a high-efficient broadband isolation performance.In this paper,a generalized vibration isolation system is established with nonlinear stiffness,nonlinear viscous damping,and Bouc-Wen(BW)hysteretic damping.An approximate analytical analysis is performed based on a harmonic balance method(HBM)and an alternating frequency/time(AFT)domain technique.To evaluate the damping effect,a generalized equivalent damping ratio is defined with the stiffness-varying characteristics.A comprehensive comparison of different kinds of damping is made through numerical simulations.It is found that the damping ratio of the linear damping is related to the stiffness-varying characteristics while the damping ratios of two kinds of nonlinear damping are related to the responding amplitudes.The linear damping,hysteretic damping,and nonlinear viscous damping are suitable for the small-amplitude,medium-amplitude,and large-amplitude conditions,respectively.The hysteretic damping has an extra advantage of broadband isolation.Zhong ZHANG Muqing NIU Kai YUAN Yewei ZHANG 2020Applied Mathematics and Mechanics(English Edition)2020,41,7:1
19Microstructural formation and mechanical performance of friction stir double-riveting welded Al-Cu joints显示文摘A novel friction stir double-riveting welding(FSDRW) technology was proposed in order to realize the high-quality joining of upper aluminum(Al) and lower copper(Cu) plates,and this technology employed a Cu column as a rivet and a specially designed welding tool with a large concave-angle shoulder. The formations, interfacial characteristics, mechanical properties and fracture features of Al/Cu FSDRW joints under different rotational velocities and dwell times were investigated. The results showed that the well-formed FSDRW joint was successfully obtained.The cylindrical Cu column was transformed into a double riveting heads structure with a Cu anchor at the top and an Al anchor at the bottom, thereby providing an excellent mechanical interlocking.The defect-free Cu/Cu interface was formed at the lap interface due to the sufficient metallurgical bonding between the Cu column and the Cu plate, thereby effectively inhibiting the propagation of crack from the intermetallic compound layer at the lap interface between the Al and Cu plates. The tensile shear load of joint was increased first and then decreased when the rotational velocity and dwell time of welding tool increased, and the maximum value was 5.52 k N. 20Improved monochromatic pyrometry for synchronous measurement of full-field temperature and deformation显示文摘Measuring the temperature and deformation synchronously at elevated temperatures is technically challenging and has become a major concern in the evaluation of mechanical properties. In this study, a simple, easy-to-implement, yet effective monochromatic pyrometry is established for non-contact and full-field temperature measurements, which can significantly reduce the error caused by the camera’s channel crosstalk that commonly occurs in the existing improved two-color method. In addition, hightemperature digital image correlation, combined with band-pass filtering and monochromatic illumination, is applied for deformation measurement. Subsequently, an experimental system was set up to validate the accuracy of the proposed method,which consists of a CCD camera for image capturing, a blue bandpass filter for radiation suppression, blue light irradiation for light compensation, and an infrared pyrometer for temperature recording.
