1Enhanced carbon acquisition and use efficiency alleviate microbial carbon relative to nitrogen limitation under soil acidification显示文摘Background:Soil microbial communities cope with an imbalanced supply of resources by adjusting their element acquisition and utilization strategies.Although soil pH has long been considered an essential driver of microbial growth and community composition,little is known about how soil acidification affects microbial acquisition and utilization of carbon(C)and nitrogen(N).To close the knowledge gap,we simulated soil acidification and created a pH gradient by adding eight levels of elemental sulfur(S)to the soil in a meadow steppe.Results:We found that S-induced soil acidification strongly enhanced the ratio of fungi to bacteria(F:B)and microbial biomass C to N(MBC:MBN)and subsequently decreased the C:N imbalance between microbial biomass and their resources.The linear decrease in the C:N imbalance with decreasing soil pH implied a conversion from N limitation to C limitation.To cope with enhanced C versus N limitation,soil microbial communities regulated the relative production of enzymes by increasing the ratio ofβ-glucosidase(BG,C-acquiring enzyme)to leucine aminopeptidase(LAP,N-acquiring enzyme),even though both enzymatic activities decreased with S addition.Structural equation modeling(SEM)suggested that higher C limitation and C:N-acquiring enzyme stimulated microbial carbon-use efficiency(CUE),which counteracted the negative effect of metal stress(i.e.,aluminum and manganese)under soil acidification.Conclusions:Overall,these results highlight the importance of stoichiometric controls in microbial adaption to soil acidification,which may help predict soil microbial responses to future acid deposition.Tianpeng Li Ruzhen Wang Jiangping Cai Yani Meng Zhirui Wang Xue Feng Heyong Liu Ronald F.Turco Yong Jiang 2021Ecological Processes2021,10,1:2
2Nitrogen addition and mowing alter drought resistance and recovery of grassland communities显示文摘Nitrogen enrichment and land use are known to influence various ecosystems,but how these anthropogenic changes influence community and ecosystem responses to disturbance remains poorly understood.Here we investigated the effects of increased nitrogen input and mowing on the resistance and recovery of temperate semiarid grassland experiencing a three-year drought.Nitrogen addition increased grassland biomass recovery but decreased structural recovery after drought,whereas annual mowing increased grassland biomass recovery and structural recovery but reduced structural resistance to drought.The treatment effects on community biomass/structural resistance and recovery were largely modulated by the stability of the dominant species and asynchronous dynamics among species,and the community biomass resistance and recovery were also greatly driven by the stability of grasses.Community biomass resistance/recovery in response to drought was positively associated with its corresponding structural stability.Our study provides important experimental evidence that both nitrogen addition and mowing could substantially change grassland stability in both functional and structural aspects.Our findings emphasize the need to study changes across levels of ecological organization for a more complete understanding of ecosystem responses to disturbances under widespread environmental changes.Zhuwen Xu Heyong Liu Yani Meng Jinfei Yin Haiyan Ren Mai-He Li Shan Yang Shiming Tang Yong Jiang Lin Jiang 2023Science China(Life Sciences)2023,66,7:2
3Discrimination Between Foreshocks and Earthquake Swarm from the Change of First Motion Sign显示文摘Based on the data of first motion of 11 earthquake sequences with ML ≥ 3.0 recorded by the Telemetric Seismic Network of Shanxi since the 1980s, the first motion characteristics of each earthquake sequence were studied. It is known that earthquake sequences of different types have different consistency characteristics of focal mechanism. 4The binding epitope of sintilimab on PD-1 revealed by AbMap显示文摘PD-1 plays an important role as an immune checkpoint.Sintilimab is a newly approved PD-1 antibody for cancer immunotherapy with an unknown binding epitope on PD-1.In this study,to elucidate the molecular mechanism by which sintilimab blocks PD-1 activation,we applied Antibody binding epitope Mapping(AbMap)to identify the binding epitope of sintilimab.An epitope was successfully identified,i.e.SLAPKA,aa 127-132.By constructing a series of point mutations,the dominant residues S127,L128,A129,P130,and A132 of PD-1 were further validated by western blot analysis,biolayer interferometry,and flow cytometry.Structural analysis showed that the epitope is partially within the binding interface of PD-1 and PD-L1,and this epitope also partially overlaps with that of nivolumab and pembrolizumab.These results demonstrate that sintilimab can attenuate PD-1 activation by directly competing with the interaction between PD-1 and PD-L1 through binding with the key residues of the FG loop on PD-1.This study also demonstrates the high efficiency and accuracy of AbMap for determining the binding epitope of therapeutic antibodies.Mingliang Ma Huan Qi Chuansheng Hu Zhaowei Xu Fanlin Wu Nan Wang Danyun Lai Yang Li Hainan Zhang Hewei Jiang Qingfeng Meng Shujuan Guo Yani Kang Xiaodong Zhao Hua Li Sheng-ce Tao 2021Acta Biochimica et Biophysica Sinica2021,53,5:1
5Health benefits from the rapid reduction in ambient exposure to air pollutants after China’s clean air actions:progress in efficacy and geographic equality显示文摘Clean air actions(CAAs)in China have been linked to considerable benefits in public health.However,whether the beneficial effects of CAAs are equally distributed geographically is unknown.Using high-resolution maps of the distributions of major air pollutants(fine particulate matter[PM_(2.5)]and ozone[O_(3)])and population,we aimed to track spatiotemporal changes in health impacts from,and geographic inequality embedded in,the reduced exposures to PM_(2.5)and O_(3)from 2013 to 2020.We used a method established by the Global Burden of Diseases Study.By analyzing the changes in loss of life expectancy(LLE)attributable to PM_(2.5)and O_(3),we calculated the gain of life expectancy(GLE)to quantify the health benefits of the air-quality improvement.Finally,we assessed the geographic inequality embedded in the GLE using the Gini index(GI).Based on risk assessments of PM_(2.5)and O_(3),during the first stage of CAAs(2013 to 2017),the mean GLE was 1.87 months.Half of the sum of the GLE was disproportionally distributed in about one quarter of the population exposed(GI 0.44).During the second stage of CAAs(2017 to 2020),the mean GLE increased to 3.94 months and geographic inequality decreased(GI 0.18).According to our assessments,CAAs were enhanced,from the first to second stages,in terms of not only preventing premature mortality but also ameliorating health inequalities.The enhancements were related to increased sensitivity to the health effects of air pollution and synergic control of PM_(2.5)and O_(3)levels.Our findings will contribute to optimizing future CAAs.Tao Xue Ruohan Wang Meng Wang Yanying Wang Dan Tong Xia Meng Conghong Huang Siqi Ai Fangzhou Li Jingyuan Cao Mingkun Tong Xueqiu Ni Hengyi Liu Jianyu Deng Hong Lu Wei Wan Jicheng Gong Shiqiu Zhang Tong Zhu 2024National Science Review2024,11,2:0
6La_(0.7)Ca_(0.3)MnO_(3-δ):Ag nanocomposite thin films with large temperature coefficient of resistance(TCR)显示文摘In this work, oxygen vacancy-regulated La_(0.7)Ca_(0.3)MnO_(3-δ):Ag (LCMO:A) nanocomposite thin films onLaAlO_(3) (001) substrates were investigated to obtain films with large temperature coefficient of resistance(TCR) values. LCMO:A nanocomposite thin films were synthesized using pulsed laser deposition, andoxygen pressures during film deposition and annealing steps were optimized. As oxygen pressuresincreased, lattice parameter increased from 70 Pa to 100 Pa, Tp increased monotonically from 168 K to282 K, and average Mn^(4+) concentration in the film increased as indicated by X-ray photoemissionspectroscopy data. 7Simultaneous utilization of electro-generated O_(2)and H_(2)for H_(2)O_(2)production:An upgrade of the Pd-catalytic electro-Fenton process for pollutants degradation显示文摘The Electro-Fenton(EF)process is one of the promising advanced oxidation processes(AOPs)for environmental remediation.The H_(2)O_(2) yield of EF process largely determines its performance on organic pollutants degradation.Conventional Pd-catalytic EF process generates H_(2)O_(2) via the combination reaction of anodic O_(2) and cathodic H;.However,the relatively expensive catalyst limits its application.Herein,a hybrid Pd/activated carbon(Pd/AC)-stainless steel mesh(SS)cathode(PACSS)was proposed,which enables more efficie nt H_(2)O_(2)generation.It utilizes AC,the support of Pd catalyst,as part of cathode for H_(2)O_(2) generation via 2-electron anodic O_(2) reduction,and SS serve as a current distributor.Moreover,H_(2)O_(2) could be catalytically decomposed upon AC to generate highly reactive·OH,which avoids the use of Fe;.Compared with conventional Pd catalyst,H_(2)O_(2) concentration obtained by PACSS cathode is248.2%higher,the O_(2)utilization efficiency was also increased from 3.2%to 10.8%.Within 50 min,26.3%,72.5%,and 94.0%H_(2)O_(2) was decomposed by Pd,AC,and Pd/AC.Fluorescence detection results implied that Pd/AC is effective upon H_(2)O_(2) activation for·OH generation.Finally,iron-free EF process enabled by PACSS cathode was examined to be effective for reactive blue 19(RB19)degradation.After continuous running for 10 cycles(500 min),the PACSS cathode was still stable for H_(2)O_(2)generation,H_(2)O_(2)activation,and RB19 degradation,showing its potential application for organic pollutants degradation without increase in the running cost.Wei Zhou Xiaoxiao Meng Liang Xie Junfeng Li Yani Ding Yanlin Su Jihui Gao Guangbo Zhao 2022Chinese Journal of Chemical Engineering2022,35,4:0
8Octahedral silver oxide nanoparticles enabling remarkable SERS activity for detecting circulating tumor cells显示文摘The detection of circulating tumor cells(CTCs)is a crucial tool for early cancer diagnosis,prognosis,and postoperative evaluation.However,detection sensitivity remains a major challenge because CTCs are extremely rare in peripheral blood.To effectively detect CTCs,octahedral Ag_(2)O nanoparticles(NPs)with high dispersibility,good biocompatibility,remarkable surface-enhanced Raman scattering(SERS)enhancement,and obvious enhancement selectivity are designed as an SERS platform.Ag_(2)O NPs with many oxygen vacancy defects are successfully synthesized,which exhibit an ultra-high SERS enhancement factor(1.98×10^(6))for 4-mercaptopyridine molecules.The remarkable SERS activity of octahedral Ag_(2)O NPs is derived from the synergistic effect of the surface defect-promoted photo-induced charge transfer(PICT)process and strong vibration coupling resonance in the Ag_(2)O-molecule SERS complex,greatly amplifying the molecular Raman scattering crosssection.The promoted PICT process is confirmed using ultraviolet-visible(UV-Vis)absorption spectroscopy,demonstrating that obvious PICT resonance occurs in Ag_(2)O SERS system under visible light.An additional growth step of SERS bioprobe is proposed by modifying the Raman signal molecules and functional biological molecules on Ag_(2)O NPs for CTC detection.The Ag_(2)O-based SERS bioprobe exhibits excellent detection specificity for different cancer cells in rabbit blood.Importantly,the high-sensitivity Ag_(2)O-based SERS bioprobe satisfies the requirement for rare CTC detection in the peripheral blood of cancer patients,and the detection limit can reach 1 cell per m L.To our knowledge,this study is the first time that a semiconductor SERS substrate has been successfully utilized in CTC detection.This work provides new insights into CTC detection and the development of novel semiconductor-based SERS platforms for cancer diagnosis.Meng He Jie Lin Ozioma Udochukwu Akakuru Xiawei Xu Yanying Li Yi Cao Yanping Xu Aiguo Wu 2022Science China(Life Sciences)2022,65,3:0
9PCR Detection of Pilus-associated Genes and Serotype Identification of Pathogenic Escherichia coli Isolated from Chickens in the Jidong Area显示文摘[Objective] This study aimed to explore the presence of typeⅠpili(fim C gene) and P pili(pap C gene) and identify the serotype of pathogenic E.coli isolated from chickens in the Jidong Area.[Method]TypeⅠpili(fim C gene) and P pili(pap C gene) were detected by PCR.The serotype was identified by conventional agglutination test.[Result]The results showed that 100% of chicken-derived E.coli strains expressed typeⅠpili(fim C gene);39.1%(9/23) of chicken-derived E.coli strains expressed P pili(pap C gene).In addition,23 isolates of chicken-derived E.coli were assigned to 14 O serotypes,including O78,O93,O 45,O101,O38,O88,O24,O1,O163,O53,O15,O87,O34 and O29,among which O78 was the dominant serotype that accounted for 42.8%(6/14) of the total strain number.[Conclusion]Chicken-derived E.coli strains in the Jidong Area belonged to 14 serotypes,and O78 was the dominant serotype;83.3% of O78 serotype E.coli strains expressed both typeⅠpili and P pili.Qiumei SHI Yanying ZHANG Xiumin WANG Baoxin YANG Zhiqiang ZHANG Xiaojiao Hou Xinhua SHAO Xia MENG Yang YANG Guoqiang ZHU 2016Agricultural Biotechnology2016,5,2:0
